      • HLA-DPB1 유전자 형별법 개발

        연구배경: HLA-DP 유전자는 DR 및 DQ와 같이 항원제공 세포(APCs), B 세포 및 활성화된 T 세포등의 표면에 발현되는 class Ⅱ 유전자로서 외인성 항원제공시 MHC 제한(restriction)과정을 통해 면역 반응에 관여한다. DP 항원 분자는 PLT(primed lymphocyte test)와 같은 세포학적 방법에 의해 처음으로 규명되었고, 이어서 혈청학적 형별이 시도되었으나, DR, DQ 분자에 비해 발현 정도가 낮아 형별에 어려움이 있었다. 최근 DP 유전자의 염기서열이 밝혀짐에 따라 DNA 수준에서 DP 형별이 가능하게 되었으며, 지금까지 DPA1 10개, DPBI 77개의 대립유전자가 밝혀졌다. 본 연구는 이식의 결과 및 질병 발생과 연관이 있다고 추정되는 HLA-DPB1 대립유전자를 DNA 수준에서 형별하고 정상 한국인의 DP 대립유전자 분포를 확인하기 위해 시도되었다. 방법: HLA-DPB1 유전자 중 초가변부위(hypervari-able region)가 있는 exon 2 부위를 일차적으로 증폭시킨후, 17종의 probe를 사용하여 PCR-SSOP를 실시하였고, PCR-SSP에는 5종의 primer를 사용하였다. 이들 방법은 DP형이 알려진 미국 UCLA 대학의 international cell exchange sample 88개를 표준 DNA 시료로 이용하여 확립하였다. 결과: 36종류의 HLA-DPBI 대립유전자를 형별할수 있음을 확인하였다. 그러나 DPBI*0402와 0601 대립유전자를 동시에 갖는 heterozygote인 경우에는 DPBI*0201와 2001을 동시에 갖는 heterozygous와 구분이 어려웠다. 100명의 정상 한국인에서 DPBI 대립 유전자형의 분포를 조사한 결과 한국인에서는 11종류의 HLA-DP 대립유전자가 존재함을 확인하였고, DPBI*0501(36.5%), 0201(27%), 0402(10.5%) 대립유전자의 순으로 분포되어 있었으며 검출율은 100%였다. 결론: 이상의결과로 골수 이식 등 이식시 공여자와 수혜자간의 조직적합성은 HLA-DP, DQ뿐만 아니라 DP도 확인 하게되었으며, 앞으로 질병과의 연관성 연구에서 이용될수 있을것으로 사료된다. Background: The HLA-DP genes are highly polymorphic, which encode heterodimeric cell surface glycoproteins that play a role in the immune response as restriction elements in antigen presentation. HLA-DP antigens were initially defined through the primed lymphocyte test (PLT), and the serological typing has been performed which is practically difficult because the expression of DP molecules is very low, comparing with that of DR and DQ. Recently, DNA sequencing and PCR have allowed the various and extensive study on the HLA-DP genes. We developed the molecular typing method for HLA-DPBI alleles, and studied the distribution in normal Korean population. Methods: After PCR amplification of hypervariable exon 2 regions in HLA-DPBI gene, both PCR-SSOP (sequence specific oligonucleotide probes) with 17 probes and PCR-SSP (sequence specific primers) by 5 Primers were used. And this method was tested using a international pannel DNA as standard. Results: Total 36 alleles of HLA-DPBI were defined in the standard DNA pannel. However, DPBI*0.402, and 0601 heterozygote could not be distinguished from DPBI*0201 and 2001 heterozygote. And 11 alleles were defined in 100 normal Koreans and the common alleles were DPBI*0501 (36.5%). 0201 (27%) and 0402 (10.5%). The detection rate was 100% in this study. Conclusion: As the results, the molecular typing of HLA-DPBI alleles is possible for the accurate matching between donor and recipient in born marrow transplantation and for the study of disease association.

        Detection of Puumala and Hantaan Virus among Bats in Korea by Nested RT - PCR

        Hantavirus is a genus of the Bunyaviridae family consisting following serotype groups: Hantaan, Seoul, Puumala, Prospect Hill, Thailand, Belgrade, Thotta palayam, Sin Nombre. Most of Hantavirus group have been associated with many clinically similar disease known collectively as hemorrhagic fever with renal syndrome (HFRS). Hantaan virus is the prototype of the genus hantavirus, originally isolated from Apodemus agrarius. Bat was found as a natural host for Hantaan virus in Lee's lab for the first time. Then, Hantaan-like virus was isolated Hantaan-like virus from bat. To identify hantaviruses that are present in Korea among bats, bats were collected from Jeong-Sun, Won-Joo, Chung-Ju and Hwa-Cheon area, RNA was isolated from lung and serum. RT-PCR was performed with a universal primer from M segment. Nested RT- PCR was carried out to differentiate Hantaan, Seoul and Puumala virus using serotype specific primers. As we expected, Hantaan viruses were detected in bats and Seoul virus was not detected. Interestingly, Puumala viruses were also detected in bats from Won-Ju, but not in other areas. Puumala virus is originally isolated from Clethrinomys glareolus, and cause light HFRS. Recently, Paradoxomis webbiana, a wild bird turn out to be a reservoir for Puumala virus in Korea. These data indicate that bat is a new natural reservoir of Puumala virus.

        Stenosis of central vein is a common complication arising after percutaneous subclavian vein catheter insertion performed for temporary vascular access in chronic renal failure patients undergoing hemodialysis. There are several treatment methods for the condition like percutaneous angioplasty(PTA), stent insertion, and surgery, but recent trend is toward PTA and stents. Among the patients diagnosed with chronic renal failure from March 1993 to May 2002 and undergoing hemodialysis through AV fistula, the 14 Patients in whom central vein stenosis arose were selected for the study. A total of 28 percutaneous interventions(5 PTA and 23 stent placement) were performed, and restenosis rate and the time taken till the restenosis in de novo lesions and instant lesions were compared. All 28 cases were operated successfully. The 14 cases that received both anigioplasty and stent placement initially. (de novo lesion : 14 cases), Among the 10 cases with de novo lesion that followed up more than 1 year, 3 cases are currently undergoing hemodialysis without restenosis, and the remaining 7 cases have recurred stenosis with the mean time to restenosis of 10.9 months. In the 7 cases in whom stenosis recurred, 11 interventions were done(instent lesion: 11 cases). 4 of these were using only ballon angioplasty with 100% restenosis rate and the mean time of 3 months until restenosis. The remaining 7 cases were using both balloon angioplasty and stent placement, also with 100% restenosis rate but with the mean time of 12 months until restenosis, which was later than the group receiving only balloon angioplasty. In treating the patients with central vein stenosis, stent placement seems to be more advantageous over PTA in terms of restenosis rate and the mean duration of patency. In the case of instent lesion, inserting the stent for the second time after stenosis recurred lengthened the duration of patency compared to performing balloon angioplasty alone.

Such an urban design has both side an objective and precess. The objective is a qulitative phase to concerned with methodology to accomplish the urban design goal. This urban design as a Process is actually to realize urban design plan in urban space. Methodology to materialize the urban design is expressed as urban design control technique , and the adequate urban design control should be utilize to accomplish the urban plan efficiently. In order to realize this urban design, according to the action from the legislation, it is desirable to establish the urban planning rule or urban design rule having equal level with architecture rule, according to the action from the administration, it is hopeful to set up the administration department in charge the urban design and bring up the specialist of urban design, and it is necessary to raise the inhabitants participation condition and able to select for themselves the desirable welcome of urban. when these systematic improvement actions are accomplished , the urban design can get the actual effect and contribute to create the desirable urban environment more and more.

        역사적 도심지구내 터줏대감의 정주의식에 관한 연구 : 청주시 성안동·중앙동의 사례 In Case of Seongan-dong and Jungang-dong in Cheongju City

        The purpose of this paper is to clarify the longest-tern householder's ideas of settlement, that is, a house guardian deity, in traditional urban area focused on Seongan-dong and Jungang-dong in Cheongju City of Korea. As a result, the summaries of findings are as follows : 1. The longest-term householders have been existing over 35 years(in case of resident registration) in the historical urban area. Most of them are over 60 years old and have been succeeded by family members. More than half of householders occupy their dwelling houses combined with shop and business uses. 2. We found those who are proud of a house site and asked them about ideas of settlement by questionnaire, and it made sure that they hope to live in there for a inheritance, site conditions, and so on, while we could understand that the site not be succeeded to next generations in near future. 3. We found those who are not proud of their dwelling houses and asked them about renovations by questionnaire, and it made sure that they hope to remodel and build their dwelling for deterioration, small-sized rooms, and so on. These new-built houses occupy family members and houses combined with shop.

        역사적 도심지구의 집주모델시안에 나타난 전통성과 지역성의 표현 : 건축설계경기의 제안작품을 통하여

        The purpose of this paper is to analyze and evaluate the tendency of collective habitat model in traditional urban area proposed by the architectural competition. The analysis and evaluation of these works is made by reference-frame such as community, locality, and economy. As results, the summaries of findings are as follows: 1. In the tentative plan of the collective habitat models, these have a tendency to encourage community life as the characteristic of traditionality and locality. That is, these concepts are represented by the consideration of environs, the positive use of site conditions, and so on. 2. In evaluation of these works, the critics are composed of these groups-specialist, developer, residents. The specialist groups make an emphasis on the community life and environs, the developer groups on the plan and structure system, and the residents on the present life-cycle and conditions. 3. The collective habitat models find their traditional and local expression in the alley, court, arrangement, exterior space, and traditional space.

        淸州市 舊都心內 터줏대감의 住宅改造 및 新築 樣相

        The purpose of this paper is to clarify an aspect on the housing reconstruction of the longest-tenn householders In traditional urban area focused on Seongan-dong and Jungang-dong In Cheongju City of Korea As a result, the summaries of findings are as follows 1. The longest-term householder's dwellings have been divided by two typical types One of them IS a pure 'House' type, which has been located In the residential districts On the other hand, the other type is a 'House with Shop', Which has been located min the districts, where had been inhabited by the storekeeper class And these are also classified by the existing or new-built houses 2. In the reconstruction of pure houses, the longest-term householders would maintain or make alterations the existing house types With the proceeding of the evolution of the housing plan, the two stoned houses and the houses With multi families have been newly built But these have succeeded the placement of the traditional house, especially the location of gate and entrance 3. In the reconstruction of shop houses, these present, for the most part, are newly built except a few old shop houses Perhaps the most notable aspect of shop house be 'multi-stoned process' With the evolution of multi-story, the dwelling space has been shifted from the first floor to the second/third floor But some of the longest-term householders would maintain the old shop house type-small and medium Size, separation between dwelling and shop space, and types of old industry-around the street network of the existing market
