Alterations in DNA methylation patterns in regenerated Chinese cabbage (Brassica rapa ssp. pekinensis) plants derived from tissue culture

Plant tissue culture is an essential tool in biotechnology. However, tissue-cultured plants often exhibit variations that are either genetic or epigenetic in origin, termed somaclonal variations. Among these variations, DNA methylation is an important heritable epigenetic modification that plays a role in a wide variety of biological processes, including gene expression. In this study, we performed bisulfite sequencing of regenerated Chinese cabbage (Brassica rapa ssp. pekinensis) lines to identify DNA alterations induced by tissue culture. Sequencing data from each regenerated line were compared with reference genome sequences, and common differentially methylated regions (DMRs) were detected in the regenerants. To determine changes in expression levels of DMR-containing genes, we performed quantitative reverse transcription-polymerase chain reaction (qRT-PCR) of the target genes and PCR amplification with bisulfite-converted DNA. We identified DMRs between a non-regenerant line and regenerant lines and selected 10 DMR-associated genes that presented annotation information in Arabidopsis or Brassica rapa. Their expression levels were verified by qRT-PCR to determine the relation between methylation state and gene expression. We observed that genes positioned in DMRs significantly correlated with differential gene expression. We also observed similar methylation patterns in the selected DMRs by PCR-based methylation analysis. The results of this study are a valuable resource for the epigenetic analysis of regenerated lines, especially for Chinese cabbage.

A Study on Joke Misinterpretation in L2 Settings: Viewed from the Benign-Violation Theory

This paper examines cases in which Korean college students might misinterpret superiority, incongruity, and sarcastic English jokes. In parallel the writer tries to bring to light possible background reasons for their misinterpretation from the perspective of the Benign-Violation Theory’ concepts of norm violation, commitment, and psychological distance. Arguably Korean college students are likely to misinterpret English humor codes influenced by their ingrained belief system of strict moral standard as well as insufficient exposure to particular genres of English jokes.

명청교체기 인평대군 시문학 연구

Royal families were supposed to live idle lives, but the life of Prince Inpyeong was an exceptional case. Prince Inpyeong"s role has been evaluated as the absolute peace in the King Hyojong era. To solve sensitive diplomatic issues between Choson-Qing smoothly, Inpyeong took the critical role of an envoy. He took on almost all the difficult tasks in diplomacy. All his diplomatic journeys and achievements had the source in the close loving relationship with the king. The prince played a crucial role to cause the fall of Ming Dynasty and watched Qing Dynasty during the journeys. He provided information of the internal affairs of Qing Dynasty to King Hyojong. It was a significant source to make Hyojmg’s Plan to Conquer the North possible. His poetry was mainly to communicate with King Hyojong. The poems seek after a hero, enjoy the feeling of freedom elegantly, show sentiment of friendship and loyalty for his king and brother, Hyojong.

만성정신분열증의 정량화 뇌파검사 소견

본 연구는 만성 정신분열증 환자 30명과 정상대조군 21명의 뇌파 중 델타파와 쎄타파를 분석해 봄으로써 만성 정신분열증 환자의 국소뇌파 소견과 유병기간, 약물투여량 및 투여기간, 양성증상과 음성증상에 따른 뇌파소견을 보고자하였다. 연구결과는 다음과 같다. 첫째, 만성 정신분열증 환자와 정상대조군간에 델타파와 쎄타파의 국소뇌파량을 t 검증한 결과 델타파에는 유의한 차이가 없으나, 쎄타파는 전 부위에서 유의한 차이를 보였다. 둘째, 만성정신분열증 환자의 델타파 및 쎄타파는 유병기간에 따른 유의한 차이가 없고, 투약기간이 길어질수록 F7, T3, C3, C4의 델타가 감소하는 경향을 보이고, 항정신병약물 투여량이 많을수록 P3, P4, C4, T5, T6, O1, O2의 쎄타가 증가하는 경향을 보였다. 셋째, 양성증상의 유무에 따라 Fp2의 델타에 유의한 차이를 보이고, 음성증상의 유무에 따라서는 Fp2, T3, C3, P3, O1의 델타에 유의한 차이를 보였다. 넷째, 판별분석을 시행한 결과 qEEG의 델타파와 쎄타파의 국소 뇌파량만으로 만성정신분열증 환자 30명중 29명을 (96.7%) 만성 정신분열증으로 판별할 수 있었다. 이상에서 만성정신분열병 환자의 서파가 정상대조군과는 유의한 차이를 보이며 유병기간에 따른 차이는 없으나 투약기간과 투여량에 따른 차이를 보이고 음성 증상을 가진 환자에서 좌측 대뇌반구의 기능저하를 추측케한다. Electroencephalographic recordings in 30 medicated patients fulfilling the DSM-Ⅲ-R criteria for chronic schizophrenia and 21 normal healthy controls was studied, using computerized electroencephalography. The delta activity and theta activity were compared between the patients and the normal healthy controls, then the differences in delta activity and theta activity of the patients were analyzed according to duration of illness, duration of medication, dosage of medication, and the presence of positive and negative symptoms. In the comparison study of chronic schizophrenic patients and the normal healthy controls, the theta activity showed a significant difference in all regions, and a significant difference in abnormal electroencephalographic activity appearance rate. In the analysis of the delta and theta activities of the chronic schizophrenic patients, duration of illness had no significant influence, the delta activity in F7, T4, C3, and C4 regions showed a tendency to decrease as duration of medication increased, the theta activity in P3, P4, C4, T5, T6, O1, O2 regions showed a tendency to increased with the increment of dosage of neuroleptics. The delta activity in Fp2, T3, C3, P3, O1 regions showed a significant difference between those patients with and without negative symptoms. In summary, the theta activity of the chronic schizophrenic patients showed a significant difference in comparison with that of the normal controls. In chronic schizophrenic patients, no difference in electroencephalograph was apparent regarding the duration of illness, but significant differences were shown when analyzed regarding the duration and dosage of medication. According to the results of this study, dysfunction of left cerebral hemisphere could be presumed to be present in patients with negative symptoms.

Endometriosis is the abnormal growth of endometrial tissue outside the uterine cavity. Endometriosis in the abdominal scar following cesarean section is very rare condition among the extrapelvic endometriosis. The frequency of abdominal wall endometriosis secondary to cesarean section is from 0.03% to 0.8% in some reports. It is difficult to diagnosed in spite of the typical symptoms. The typical symptom is aching swelling in the area of the surgical scar, which is influenced by the phases of menstruation. The definitive diagnosis is established by pathologic analysis. Surgical excision remains the treatment of choice. The majority of patients presented from 1 to 2 years after the precipitating operation. We experienced an unusual case of abdominal wall endometriosis presented over 10 years after cesarean section, who was admitted to our hospital complaining of recurrent left lower abdominal pain. So, we report this case with a brief review of the concerned literatures.

Genetic Algorithm을 이용한 철근콘크리트보의 최적설계

Simple Genetic Algorithm (SGA) is refined to take into account for the optimization of reinforced concrete beams under positive and negative moments. GA has beneficial aspects compared to conventional numerical or analytical optimization techniques in that ; it can handle discrete variables, does not need continuity or derivatives of a function, has more probabilities of convergence to global minimum; and is relatively easy to include the equations in structural design code. Considered in optimization are : discrete set of widths, depths and number of rebars both in positive moment and in negative moment region for beam members. In addition to the strength constraints, requirements for reinforcement arrangement including minimum cover thickness and necessary spacings between rebars in a beam section are considered. Within a reasonable and finite number of searching, the developed algorithm was able to yield optimum solutions for reinforced concrete beams.

공분산을 고려한 조립공차의 민감도 해석

The most commonly used variation simulation analysis methods for assemblies have been worst case analysis, root sum square method, and Monte Carlo simulation, which the individual parts of assemblies are considered as rigid bodies. Assembly tolerance, sometimes, requires the consideration of its compliance because of potentially significant deformation during the spot-weld assembly process in automobile industry. A relationship, called sensitivity, between the assembled part deviation vector for incoming components and tooling hence has been found for a unified linear variation model. In this method, it is important to take into account of the covariance between the sources of variation as they are closely located, which is the case in most auto body assembly. This research is done on the CAVA (Compliant Assembly Variation Analysis) model, which is a variation simulation model for single station level, In the CAVA model, only results for part representation are obtained and the contact effects will be solved in the future research.

Epstein-Barr virus(EBV)와 연관된 Nasal type T/Natural Killer(NK)-cell lymphoma 1예

Nasal type T/NK-cell lymphoma는 병리조직학적으로 응고성 괴사가 동반된 혈관중심성 침윤을 특징으로하며 T-cell과 NK-cell 항원을 모두 표현하는 면역조직화학염색 소견을 보이는 매우 드문 피부 림프종으로서 EBV와의 연관성이 흔히 보고되고있다. 저자들은 42세 여자에서 발생한, 전신적인 홍반성 구진, 판, 결절의 임상양상을 보였으며 피부 병변에서의 EBV-encoded RNA(EBER)에 대한 in situ hybridization상 양성소견을 보였고 전산화단층촬영, 골주사, 갈륨주사 등의 검사상 비강, 위장관 및 다른 내부장기 침범소견은 보이지 않아서 피부에만 국한된 경우로 사료되는 nasal type T/NK-cell lymphoma 1예를 보고하는 바이다.

복수과 진료 환자의 중복 처방에 대한 분석 및 평가

In an aging society, there is an increasing possibility of the duplication of the drugs given to patients because they take many kinds of drugs. Among the prescriptions given to the patients who was treated at multiple ambulatory clinics in St. Mary's hospital for one month of September, 2002, in which drugs that has identical or similar effects are prescribed, we analysed and evaluated them by patients' characteristics, kinds of medication, severity of side effects on a case by case basis. More, we assessed the potential additional costs. As a result of this study, the duplication rate turned out to be 6.69% and it was shown that the proportion of the elderly patients over 60 was high. The gastro-intestinal medications took up a large part and most of the cases showed that the danger caused by double taking of medicine was slight, but there were some examples in which the serious side effects were predicted. Expected additional costs were the average 11.349 won and it ranges from the minimum of 56 won to the maximum of 135,720 won. In conclusion, the management of drug histories of the patients who need the plural treatments is very important and the necessity of the individual and professional guidance of taking medicines for the elderly patients is emerged, too.
