Expression of Sara2 Human Gene in Erythroid Progenitors

A human homologue of Sar1, named Sara2, was shown to be preferentially expressed during erythropoiesis in a culture stimulated by EPO. Previous studies, in yeast, have shown that secretion-associated and Ras-related protein (Sar1p) plays an essential role in protein transport from the endoplasmic reticulum to the Golgi apparatus. Here, we report the molecular analysis of Sara2 in erythroid cell culture. A 1250 bp long cDNA, encoding a 198 amino-acid protein very similar to Sar1 proteins from other organisms, was obtained. Furthermore, we also report a functional study of Sara2 with Real-time quantitative PCR analysis, demonstrating that expression of Sara2 mRNA increases during the initial stages of erythroid differentiation with EPO and that a two-fold increase in expression occurs following the addition of hydroxyurea (HU). In K562 cells, Sara2 mRNA was observed to have a constant expression and the addition of HU also up-regulated the expression in these cells. Our results suggest that Sara2 is an important gene in processes involving proliferation and differentiation and could be valuable for understanding the vesicular transport system during erythropoiesis.

Purpose: This study aimed to describe the growth, body protein status, and micronutrient biomarkers of Brazilian infants with cow’s milk allergy (CMPA) at baseline and at 18 months of follow-up in comparison with their healthy peers. Methods: Thirty infants with CMPA younger than six months of age were included in this longitudinal study, and their nutritional status was compared with that of 24 non-allergic age-matched children. Anthropometric measurements were used to assess growth, and blood and urine samples were analyzed for protein and micronutrient status. Mixed linear models adjusted for birth weight, socioeconomic status, infant feeding at baseline, weightfor- age, C-reactive protein, serum albumin, micronutrient dietary supplementation, and salt consumption were employed to evaluate the evolution of nutritional parameters throughout the follow-up period. Results: Overall, the mean age of the children at enrolment was 2.9 (standard deviation 1.7) months, and 29 children (53.7%) were male. Infants with CMPA showed a higher prevalence of functional iron depletion (transferrin saturation <20) (p=0.027), lower serum ferritin (p=0.009), and lower urinary iodine (p=0.034) levels than non-allergic children at baseline. Patients with CMPA showed a higher increment in weight-for-age and length-for-age over time than those in the control group (p<0.01). Mixed linear analyses showed a significantly lower increase in serum vitamin B12 (s-B12) (p=0.001) and urinary iodine (p<0.001) concentrations over time compared to the control group. Conclusion: Infants with CMPA on a cow’s milk elimination diet had a higher weight and length at 18 months of follow-up but showed signs of inadequate iron, iodine, and B-12 vitamin status.

We present a case of osteoradionecrosis treated with leukocyte- and platelet-rich fibrin (LPRF) and surgery and followed up with clinical and tomographic investigations. A 65-year-old woman presented with pain in the posterior region of the right palate. Her medical history included cardiovascular disease and squamous cell carcinoma in the anterior region of the floor of the mouth that had been treated with intensity-modulated radiation therapy. Measurements of isodose curves showed a full dosage of 6,462.6 cGy in the anterior mandibular region, whereas that in the posterior region on the right side of the maxilla reached 5,708.1 cGy. Osteotomy was performed using rotary instruments, and debridement and placement of two LPRF membranes were also carried out. New gum tissue with no bone exposure was noted 14 days postoperatively. Tissue repair was complete, and the patient had no further complaints. During a 39-month follow-up period, the oral mucosa remained intact, and the patient was rehabilitated with a new upper denture. Since there is no consensus regarding the best protocol to treat osteoradionecrosis, LPRF might be an interesting adjuvant to a surgical approach. The use of LPRF is simple and reduces operational costs, time of handling, probability of technical failure, and associated morbidities for patients with osteoradionecrosis.

Leukemia is the most common neoplastic disease of the white blood cells which is important as a pediatric malignancy. Oral manifestations occur frequently in leukemic patients and may present as initial evidence of the disease or its relapse. The symptoms include gingival enlargement and bleeding, oral ulceration, petechia, mucosal pallor, noma, trismus and oral infections. Oral lesions arise in both acute and chronic forms of all types of leukemia. These oral manifestations either may be the result of direct infiltration of leukemic cells (primary) or secondary to underlying thrombocytopenia, neutropenia, or impaired granulocyte function. Despite the fact that leukemia has long been known to be associated with oral lesions, the available literature on this topic consists mostly of case reports, without data summarizing the main oral changes for each type of leukemia. Therefore, the present review aimed at describing oral manifestations of all leukemia types and their dental management. This might be useful in early diagnosis, improving patient outcomes.

Phenotyping is the major bottleneck in the efort to develop varieties of rice (Oryza sativa L.) suitable for growing under a water-saving irrigation management, such as alternate wetting and drying irrigation techniques (AWD). To analyze if the genotypic variability for carbon isotope discrimination (CID) in rice leaves could be used as a relatively high-throughput tracer to early select superior genotypes highlighting improved root architecture traits when submitted to AWD, a set of twenty varieties grown under semi-natural conditions were submitted to two water irrigation regimes, continuous fooding (CF) and AWD cycles. Coefcients of genetic variance (π2g) obtained for root architecture, micro-morphological and physiological traits were signifcant for all of them regardless of the adopted irrigation system, except to mean root diameter. The three signifcant principal components (PCs) with eigenvalue>1, explain most of the total variation across cycles and water regimes. For most of analyzed traits, the values of heritability coefcients were higher regardless of adopted irrigation management and trait category; for CID, the magnitudes of broad heritability at an individual level (greater than 0.80) were similar in the two irrigation techniques, evidencing that the success of the selection is independent of irrigation management. The higher CID values after three AWD cycles are associated with varieties with higher total root length and volume. To our knowledge, this is the frst study demonstrating the potential application of CID as a tracer to select root architecture traits in rice when water-saving irrigation management is of concern
Cytogenotoxic study of aqueous fruit extract of Morinda citrifolia in Wistar albino rats

The use of herbal products and/or their derivatives as an alternative or complementary medicine becomes globally popular. To date, various biological activities have been reported in Morinda citrifolia Linn, especially antioxidants, antimicrobial, anti-infammatory and antitumor efects. This study aimed to investigate the genotoxicity and DNA repair capacity of the aqueous fruit extract of M. citrifolia (AEMC). Additionally, we checked the mutagenic capacity of AEMC by the comet assay and micronucleus tests in rats. For this, rats (n=10; 5 males and 5 females) were divided into fve groups as: negative control (distilled water); positive control (25 mg/kg of cyclophosphamide, i.p.); and three groups for AEMC (2.5, 5 and 10 mg/kg, i.p.). The results suggest that, the AEMC induced genotoxicity in white blood cells, but a DNA repair capacity was also observed after 72 h of AEMC exposure. AEMC also showed cytotoxicity and mutagenicity in liver and kidney cells of rats at 5 and 10 mg/kg doses. In conclusion, AEMC exhibited a dose-dependent cytotoxicity, genotoxicity and mutagenicity along with a time-dependent DNA repair capacity in rats. Further studies are necessary regarding the safe use of this medicinal plant for human consumption.
