      • 석이에서 분리한 GE974의 α-Glucosidase 저해효과

        This study was conducted to search for the α-glucosidase inhibitor from the natural products. In the previous study, the water extract of Gyrophora esculenta exhibited a potent inhibitory effect on α-glucosidase activities. Then, by bioassay-guided fractionation followed by chromatographic seperation of the water extract of Gyrophora esculenta, α-glucosidase inhibitor was isolated as GE974. GE974 showed significant inhibitory activities on some kinds of α-glucosidases in vitro. Its inhibitory mechanism seemed to be competitive for disaccharides. Also, it markedly inhibited α-glucosidases of intestine separated from both nondiabetics and diabetics.

      • 포러스 콘크리트를 利用한 보도용 콘크리트판 적용에 關한 實驗的 硏究

        Although porous concrete is recently used as a pavement at a parking lot, a public square and a bicycle road, we use without enough examining. Most the maintenance of quality for porous concrete is only dealt with strength and color tone. Also, there is not yet enough fundamental data about dynamical properties for strength, rate of void and water permeability, even when it is applied to at the scene, it is been a matter. Accordingly in this study analyzed influences through the dynamical properties, combination, and time to vibration. After the analyzation, they investigated peculiar dynamics which are strength of porous concrete, rate of void, and water permeability follow the rate of increase on an admixture making pozzolan reaction. This is purpose to improve capacity of the sidewalk permeable concrete and development of the economical technique for pavement.

        체인형 합성고무탄성재의 탄성력 감쇄

        본 연구는 체인형 합성고무탄성재의 시간에 따른 탄성력 감쇄의 양상을 평가하여, 이들 재료의 임상적용시 적절한 교정력의 적용을 위해 참고가 될 만한 기준을 구해보려는 목적으로 시행되었다. 공간폐쇄시 사용되는 체인형 합성고무탄성재의 탄성감쇄정도를 알아보기 위하여, 임상에 널리 사용되는 두가지의 체인형 합성고무탄성재를 구강내 상태와 유사한 조건하에서 4주간 신장시켜 관찰한 결과 다음과 같이 요약할 수 있었다. 1. 시간에 따른 탄성력 감쇄양상은 전형적인 log함수의 형태를 보이며, 4주후 잔존탄성력은 원래의 41.2~64.6 %이며 제품에 따라 차이가 있었다. 2. 신장후 초기 10분동안 탄성력의 급격한 감소를 보여서, 초기힘의 20~25 %를 상실하였으며, 이후 감소량이 줄어들어 1주이후부터 4주까지의 평균감소량은 1.5 %정도로 거의 일정한 힘의 크기를 보여주었다. 3. 동일한 재료를 사용하더라도, 탄성재의 신장량이 증가할수록 잔존탄성력이 감소하여 탄성감쇄율이 증가하였다. The purpose of this study was to evaluate the force degradation rate of synthetic elastomeric chains during space closing phase of orthodontic treatment. Two kinds of synthetic elastomeric chains(RMO, 3M) were selected which were commonly used in clinics. All of the samples were extended and tested for 4 weeks under the simulated intraoral condition. The results can be summarized as follows : 1. Time related residual force showed typical logarithm function. Residual force after 4 weeks was 41.2~64.6 % of original force, and difference between two kinds of elastomeric chain existed. 2. Elastic force decreased greatly during first 10 minutes, so 20-25 % of original force disappeared After that, this decreasing tendency was diminished significantly, average rate of elastic force after 1 week to 4 weeks were 1.5 % demonstrating rather constant force. 3. Even though the same brand of elastomeric chiain was used, as extension rate of elastomeric chain increased, force degradation rate increased by decreasing of residual force.

        중대산업사고 예방을 위한 종합위험관리체제(IRMS) 구축에 관한 연구

        The Process Safety Management (PSM) by the Law of Industry, Safety and Health has been performed for preventing major accidents of chemical plants since 1996. In terms of preventing chemical accidents more precisely, it is essential to develop a tool for quantitative risk assessment. For this, KOSHA (Korea Occupational Safety and Health Agency) developed an Integrated Risk Management System (IRMS) . The system is designed to assimilate data on chemical plant hazards from external database, to integrate these data with location information (topographic and demographic), and to make them user-friendly accessible. The system consists of several main functions: display of five major Korean petrochemical complex layout, display of equipment layout with its information utilizing the external database, zonation of the hazard effected area with consequence analyses, the most probable accident scenario generation, accident/incident database and calculation of frequency of accident using equipment reliability database, etc. The highlight of IRMS is to provide the risk contours using GIS(Geographical Information System) technology. IRMS is intended to manage hazardous installation more systematically and effectively, to reduce the number of accident remarkably, further minimizing production loss in the plant. The system is now under application to about 500 PSM sites as well as and emergency authorities in Korea by KOSHA (Korea Occupational Safety and Health Agency)

        In previous study, Gyrophora esculenta showed significant inhibitory effect on α-glucosidases in vitro and blood glucose elevation in vivo In the isolating process of active substance, orcinol was separated from Gyrophora esculenta. Orcinol is known to be toxic, therefore, in this study, it was analyzed by the TLC densitometry method for quantitative determination from Gyrophora esculenta. The average amount of orcinol of Gyrophora esculenta was 0.2%. For the purpose of removing orcinol, the water extract of Gyrophora esculenta was sequentially fractionated by organic solvents, and the acute toxicity of each fraction was assessed in mice. Among them, the LD50 of butanol fraction was 1.19 g/kg(p.o.) and the weight increase of mice in that group was somewhat retarded.

      • Cheese whey를 利用한 醱酵乳製造에 關한 硏究

        The fluid yoghurts were prepared by using cheese whey (6% T.S.) as a main material and added 0, 3, 6, 9% of skimmilk powder, respectively. After that they were incubated with L. bulgaricus inoculation. The test added amount of skimmilk powder, the test inoculated amount of starter, the best incubating time, and the quality of products were investigated. The results obtained were as follows: (1) As the added ammounts of solids-not-fat in whey were increased, the number of lactic acid bacteria and the acidity of whey were increased. (2) When the 5% starter was inoculated, it produced a good quality product. (3) As the incubating time was extented, the production of acid an? the number of lactic acid bacteria were increased in whey yoghurt, but the best quality of products was produced at incubation far 48 hours. (4) The product added 6% skimmilk to whey was similar to thee product added more than 6% skimmilk in properties.

      • 다제 내성 폐렴 구균에 대한 CFC-222 의 시험관내 항균력 및 살균 속도

        목적 : 새로이 개발된 quinolone 제제인 CFC-222와 함께 기존의 항균제들의 MIC를 비교하여 CFC-222가 항균제 내성 폐렴 구균의 치료에 사용될 수 있는 약제인지를 검증하고 아울러 in vitro time kill assay를 시행하여 CFC-222의 살균 속도를 검증하고자 하였다. 방법 : 국내 및 아시아 지역 국가에서 분리 확인된 폐렴 구균 균주 200주를 대상으로 하여 cefotaxime, CFC-222, ofloxacin, levofloxacin, sparfloxacin, ciprofloxacin, 그리고 meropenem에 대한 한천희석법을 이용하여 MIC를 결정하였다. In vitro time kill assay는 총 8균주(2 penicillin-susceptible, 2 intermediate, 4 resistant)를 대상으로 시행하였고 대상이 되는 항균제는 CFC-222, sparfloxacin, penicillin G로 하였다. 결과 : 폐렴 구균 균주 200주에 대한 한천 희석법을 이용한 MIC 결과는 페니실린 MIC가 증가함에 따라 cefotaxime과 meropenem의 MIC가 증가하는 경향을 보였다. Quinolone제제 중에서는, CFC-222와 sparfloxacin의 MIC_(90)이 페니실린 내성과 관계없이 0.5㎍/mL 이하로 가장 낮았다. time kill assay 결과 CFC-222는 MIC의 4배 농도에서 페니실린 고도내성 4균주 전체에 대해 8시간째 살균 효과를 보였다. 반면 sparfloxacin과 penicillin은 MIC의 4배 농도에서 12시간째 살균효과를 보였다. 결론 : CFC-222와 sparfloxacin이 가장 우수한 시험관내 항균 효과를 보였다. In vitro time kill assay 결과 CFC-222가 sparfloxacin에 비해 페니실린 고도 내성 균주에 대해 더 빠른 살균속도를 보였다. Background : CFC-222 is a new fluoroquinolone which possesses a broad antibacterial activity, being especially potent against gram-positive bacteria as well as gram negative bacteria. To evaluate the in vitro activity and the bactericidal killing rate of CFC-222 against Streptococcus pneumoniae, agar dilution test and lime-kill study was performed. Method : 200 clinical isolates were tested by agar dilution method to determine MICs of penicillin, cefotaxime, meropenern, ciprofloxacin, ofloxacin, levofloxacm, sparfloxacin, and CFC-222. Time-kill studies were performed with 8 pneumococcal isolates (2 peni-cillin-susceptible, 2 intermediate, and 4 resistant) to compare the bactericidal activity of penicillin, sparfloxacin, and CFC-222. Results : MICs of cefotaxime and meropenem showed increasing tendency as penicillin MIC is increased. Of the quinolones tested, CFC-222 and sparfloxacin had the lowest MIC (MIC90 = 0.5 ㎎/mL), regardless of the penicillin-susceptibility of the strains. CFC-222 yielded 99.9 % killing of all penicillin-resistant strains after 8 h at 4 MIC, while penicillin and sparfloxacin were bactericidal after 12 h at 4 MIC. Conclusion : By MIC and time-kill combined, CFC-222 and sparfloxacin were the most potent agents according to the MICs, whereas CFC-222 was more rapidly bactericidal than sparfloxacin against penicillin-resistant isolates.

        This thesis aims at studying one of the ways which grasps exactly environmental qualities. For this purpose, this paper examines the cognitive dimensions of environmental identity which is the identity that a person or group has with his environment, in particular when he is experiencing it as an insider. There are two important dimensions: the vertical dimension and the horizontal dimension. The vertical dimension is classified by the degree of relationship between the observer and the environment. The horizontal dimension is classified by the scale of the environment concerned with its users.

      • 흰쥐에서 Divalent Metal Transporter 1의 조직내 분포와 Iron에 의한 조절

        Iron (Fe) is an essential metal in biological processes, which maintains a homeostasis in the human body Divalent metal transporter 1 (DMT1) has been known as an iron transporting membrane protein, which is involved in the uptake Fe at the apical portion of intestinal epithelium, and may transport Fe across the membrane of acidified endosome in peripheral tissues In this study, we studied the tissue distribution of DMT1 in the Fe supplemented (FeS) diet fed rats, and the regulation of DMT1 expression by depleting body Fe Sprague-Dawley rats were divided into two groups, and fed FeS (120mg Fe/kg) diet or Fe deficient (FeD, 2~6mg Fe/kg) diet for 4 weeks The evaluation of body Fe status was monitored by measuring sFe, UIBC and tissue Fe concentration Additionally, DMT1 mRNA levels were analyzed in the peripheral tissues by using the quantitative real time RT-PCR method In the FeS diet fed rats, the tissue Fe was maintained at a relatively high level, and DMT1 was eventually expressed in all tissues studied DMT1 was highly expressed in the testis, kidney and spleen, while a moderate levels of DMT1 expression was detected in the brain, liver and heart In the digestive system, the highest level of DMT1 was found in the duodenum Feeding the FeD diet caused a reduced body weight gain and depletion of body Fe with finding of decreased sFe, increased UIBC and decreased tissue Fe concentration The depletion of body Fe upregulated DMT1 expression in the peripheral tissue The expression of DMT1 was very sensitive to the body Fe depletion in the small intestine, especially in the duodenum, showing dramatically higher levels in the FeD rats than those of the FeS group In the FeD diet fed animals, the expression of DMT1 was low significantly in other tissues compared with the duodenum The expression of DMT1, however, was 60~120% higher in the testis, kidney and spleen, and 30~50% higher in the lung, liver and heart, compared to the FeS diet fed rats In summary, DMT1 expression was ubiquitous in mammalian tissue, and the level of expression was the organ-dependent The expression of DMT1 in peripheral tissues was upregulated by depletion of body Fe Duodenum was the most sensitive tissue among organs studied during Fe depletion, and expressed the greatest level of DMT1, while other tissues were less higher than in duodenum This study supports that DMT1 plays a role in maintaining the body Fe level through intestinal uptake as well as homeostasis of Fe in the peripheral tissue.
