      • L1210 세포증식에 대한 Glycyrrhizin의 억제작용 기전

        The purpose of this research was to investigate the mechanism of inhibitory action of Glycyrrhizin(GZ) on the proliferation of mouse leukemia cell-line, L1210 cells. The cytotoxic activity was tested using a colorimetric tertrazolium assay(MTT assay), the apoptosis was tested using flow cytometry. Nitric oxide(NO) production form mouse peritoneal macrophage was tested using a Griess method and the phagocytosis of human polymorphonuclear cells was tested using a lucigenin chemiluminescence. GZ ingibited the proliferation of L1210, BALB/c 3T3 cells and mouse thymocytes at 50 ug/ml/ GZ did not affect nitric oxide production from mouse peritoneal macrophages in vitro, but ingibited nitric oxide production from lipopolysaccharide and y-interferon treated macrophages. Macrophages of GZ-administered mice accelerated NO production. The proliferation of L1210 cells apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. The apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. GZ increased the phagocytosis of human polymorphonuclear cells. These results suggest that GZ inhibit the proliferation of L1210 cells not only as a direct cytotoxic agent o tumor cells, but also by the enhancement of NO production and phagocytic activity.

      • 傷寒論중 汗, 下法을 중심으로 한 少陰人 病證論 考察

        이제마는 사상의학의 생리, 병리를 설명함에 있어서 상한론을 많이 인용하였는데 치법에 있어서는 상한론의 것과 일치하지 않으며 특히 소음임편에서는 상한론의 한법이나 하법에 대해서 많은 비판을 가하고 있다. 이에 동의수세보원 소음인편에 나타난 상한론 인용문과 동무의 의론을 한법과 하법을 위주로 비교하여 본 결과 사상의학과 상한론은 공통적인 사기의 개념과 병증구성상의 표리구조를 가지고 있었으며, 한과 하의 개념에 있어서도 별다른 차이가 없었다. 그러나 치료법에 있어서 사상의학에서는 소음인에 대하여 승양이 안되는 체질상의 부족을 보충하는 승양익기법을 중요시하여 상한론과 차이가 남을 알 수 있었다. The main purpose of this study is to lay the groundwork of mutual assistance between Shnngknlun and Sa-sang Constitutional Medicine by looking into their differences and similarities in the concept and the medical treatment in the diaphoretic therapy and the purgative therapp. The study compared Shanghan Jomun quoted in Soeumin, Dongy-Soose-Boum m:ith Je-ma, Lee s Theory of Medicine. And its conclusion is the following: 1. Both Shanghlun and Sa-sang Constitutional Medicine connote the concept of pathogenic factors in the Nae-Kyung hfedicine as the medicinal substances. And both of them have the fundamental structure of exterior-to-interior movements in the symptoms of disease to appear when pathogenic factors penetrate into the bodies. 2. Sa-sang Constitutional Medicine has treated the causes and the mechanism of disease in the symptoms of disease differently from other therapies: focusing on the body's constitutional symptom of disease, namely Soeumin's lack of the Yang-Qi and the poor Ascending Yang, not an attack on pathogenic factors as a interior-exterior symptom. Therefore Ascending-yang Suppiling Qi. which fills up the lacking parts of the body and keeps the balance of the body fitted in one's constitution, has been used rather than the diaphoretic and purgative therapy. 3. As Shanghlun and Sa-sang Constitutional Medicine have the similarities in the interior-to-exterior structure, pathogenic factors, and the notion of diaphoresis and purgation, they do so in the principle of the medicinal substance and the therapy. However, there are actually differences of carrying out the diaphoretic and purgative therapy and other treatments between Shangtnnlun and Sa-sang Constitutional Medicine by the reason of the dissimilarities in the mechanism of disease and focal points.

      • 子官癌細胞(HeLa)에 대한 血府逐瘀湯과 數種 抗癌劑의 倂用投與 效果

        The studies were conducted to investigate the combined effects of Hyulbu Chuko Tang (HCT) and several anti-cancer drugs. The effects of HCT and several anti-cancer drugs on the proliferation of HeLa Colls, human cervical cell line, was estimated by MTT colorimetric assays. The 50% inhibitory concentrations of mitomycin C(MMC), cisplatin(CPT), mercaptopurine (MCP) and 5-fluorouracil(5-FU) on HeLa cell were 0.313, 0.565, 22.745 and 28.325/㎖, repectively. HCT extract inhibited the proliferation of HeLa cell at 10^-6-10^-3g/㎖. The inhibitory action of the MCP treated group on the proliferation of HeLa cell was increased by HCT and MCP. When the mice were treated by MMC, the number of leukocyte was decreased significantly at the 1st and 3rd day, but recovered at the 7th day. In the groups of MMC treated with HCT, the number of leukocytes was increased significantly than the group of MMC treated only at the 1st and 3rd day. The HCT extract increased the number of plaque forming cells(PFC), but the MMC treated group decreased the number of PFC. The combined treatment of MMC and HCT increased the number of PFC significantly than the MMC treated group. The HCT extract increased the proliferation of T cells, but the MMC treated group decreased the proliferation of T cells. The combined treatment of MMC and HCT increased the T cell proliferation significantly than MMC treated group.

      • 韓國産 납자루 亞科 魚類의 染色體와 ARM NUMBER

        Karyotypes of 12 species of the Acheilognathine fishes were studied using chromosomes of gill slit and kidney cells prepared by the flame drying technique. The results were as follows : 1. Numbers of chromosomes were classified into three patterns as 2n=48, 44, and 46. 2. In the 2n=48, No. of two-arm chromosome (TAC) and one-arm chromosome (OAC)were 12 or 14 pairs and 10 or 12 pairs, respectively. In the 2n=44, TAC and OAC were 14 or 12 and 8 or 10, severally, In the 2n=46, TAC was 2 pairs and OAC was 21 pairs. 3. Above results indicate that patterns of arm number(AN) were classified into four groups as AN=76, 72, 68, and 50. In the view of these results, we suggest that primitive chromosomal type of Acheilognathine fishes(cyprinidae) was 2n=46 and AN=50(R. notatus, R. suigensis).

      • Euonymus alatus추출물의 실험적 당뇨발생 억제효과

        Insulin-dependent diabetes mellitus (IDDM) is majorly caused by an immune system-mediated destruction of the beta cells in the islets of Langerhans. Patients with IDDM face major changes in lifestyle and the possibility of debilitating and life-threatening complications. In Korean traditional medicine, Euonymus alatus (EA) extract has been widely used for preventing and curing ulcer and inflammation. A previous study has shown that EA itself has an antidotic activities against inflammation, suggesting possibility that EA can exert this beneficial effects to IDDM by an initial protection against diabetes caused by pancreatic inflammation. The present study was undertaken to evaluate the prophylactic effect of EA extract on the diabetogenesis of streptozotocin (STZ) and alloxan (ALX), using animal model, and to explore the possible mechanisms of its antidiabetic activity. Multiple low doses of STZ (45 mg/Kg for 5 days) in ICR mice and single dose of ALX (60 mg/Kg) in SD rats induced high level of hyperglycemia (defined as plasma glucose level > 300 mg/dl) and remarkably diminished body weight. But, animals that received diabetogenic agents in combination withsEA were maintained plasma glucose level as a normal state ( < 150 mg/dl), and appeared in good health and had similar weights to those of normal control. These results strongly indicated that EA prevented and blocked diabetogenesis of chemicals. EA significantly inhibited the nitric oxide- and tumor necrosis factor-production of mouse peritoneal macrophages. However, EA enhanced interleukin 1 production of macrophages at lower concentration but not at higher. EA itself increased phagocytic activity of unstimulated monocytes, but it did completely block that activity of zymosan- or platelet activating factor-stimulated cells. Taken together, this study led to conclusion that EA itself has a significant prophylactic effects against diabetes, and also suggested that its antidiabetic effects might be manifested by the downregulation of inflammatory process in diabetogenesis.

        The phagocytic activity of murine peritoneal macrophage was determined by lucigenin chemiluminescence and engulfment of fluorescein-conjugated E. coli particles. The activity-guided fractionation upon the methylenechloride fraction of Aurantii immaturi pericarpium led to the isolation of a flavonoid, isosinensetin, as a suppressive component of phagocytosis, lsosinensetin suppressed the lucigenin chemiluminescence and the engulfment of fluorescein-conjugated E. coli particles and enhanced the production of nitric oxide in murine peritoneal macrophage.

      • 자궁암세포(HeLa)에 대한 사군자탕과 수종 항암제의 병용투여 효과

        The studies were conducted to investigate the combined effects of Sa Kunja Tang(SKT) and several anti-cancer drugs. The effects of SKT and several anti-cancer drugs on the proliferation of HeLa cells, human cervical cell line, was estimated by MTT colorimetric assays. The SKT extract inhibited the proliferation of HeLa cell at 10^-4g/ml. The inhibitory action of the mitomycin C(MMC), mercaptopurine(MCP) and 5-fluorouracil(5-Fu) treated group, respectively, on the proliferation of HeLa cell was increased by the SKT. When the mice were treated by the MMC, the number of leukocyte was decreased significantly at the 1st and 3rd day, but recovered at the 7th day. In the groups of the MMC treated with the SKT, the number of leukocytes was increased significantly than the group of the MMC treated only at the 3rd day. The SKT extract decreased the thymus weight of mice. The SKT extract increased the number of plaque forming cells(PFC), but the MMC treated group decreased the number of PFC. The combined treatment of MMC and the SKT increased the number of PFC significantly than the MMC treated group. The SKT extract increased the proliferation of T cells, but the MMC treated group decreased the proliferation of T cells. The combined treatment of MMC and the SKT increased T cell proliferation significantly than the MMC treated group. In conclusion, the results presented in this paper suggest that the SKT extract can decrease the dose of anti-cancer drugs on HeLa cell, and recover the side dffects of the MMC, such as leukopenia and immunosuppresion, without any intercalating the anti-proliferative action of the MMC in vivo.

      • 사군자탕이 항암제를 투여한 마우스의 면역세포에 미치는 영향

        The purpose of this research was to investigate effects of Sa-Kunja-Tang(SKT) on immune cells of antitumor drugs administered mice. The apoptosis and T lymphocytes subpopulation were tested using a flow cytometry, and the proliferation was tested using a MTT assay. The administration of etoposide. vincristine or doxorubicin increased the apoptosis of T-lymphocytes, but the action of doxorubicin was decreased by the administration of SKT. The administration of etoposide or vincristine decreased helper T and cytotoxic T cells population of T lymphocytes, but the action of vincristine was recovered by the administration of SCT. The administration of etoposide, vincristine or doxorubicin decreased the proliferation of T-lymphocytes, but the action of doxorubicin was increased by the administration of SKT. These results suggest that SKT has a regulative function of T-lymphocytes in anti-tumor drugs administered mice.

      • 사군자탕이 L1210 세포를 이식한 마우스의 면역세포에 미치는 영향

        The purpose of this research was to investigate effects of Sa-Kunja-Tang(SKT) on immune cells of L1210 cell-transplanted mice. The apoptosis and T lymphocytes subopoulation were tested using a flow cytometry, and the proliferation was tested using a MTT assay. Nitric oxide production from mouse peritoneal macrophage was tested using a Griess reagents, and the phagocytic activity of mouse peritoneal macrophage was tested using a lucigenin chemiluminescence. SKT suppressed apoptosis of T-lymphocytes induced by L1210 transplantation. SKT decreased nitric oxide production from mice peritoneal macrophages increased by L1210 transplantation, and the phagocytic activity decreased by L1210 transplantation. These results suggest that SKT suppresses T lymphocyte apoptosis and macrophage activity in L1210 transplanted mice.
