        Foxp3 is a key downstream regulator of p53-mediated cellular senescence

        <P>The downstream events and target genes of p53 in the process of senescence are not fully understood. Here, we report a novel function of the forkhead transcription factor Foxp3, which is a key player in mediating T-cell inhibitory functions, in p53-mediated cellular senescence. The overexpression of Foxp3 in mouse embryonic fibroblasts (MEFs) accelerates senescence, whereas Foxp3 knockdown leads to escape from p53-mediated senescence in p53-expressing MEFs. Consistent with these results, Foxp3 expression resulted in the induction of senescence in epithelial cancer cells, including MCF7 and HCT116 cells. Foxp3 overexpression also increased the intracellular levels of reactive oxygen species (ROS). The ROS inhibitor N-acetyl-L-cysteine rescued cells from Foxp3-expression-induced senescence. Furthermore, the elevated ROS levels that accompanied Foxp3 overexpression were paralleled by an increase in p21 expression. Knockdown of p21 in Foxp3-expressing MEFs abrogated the Foxp3-dependent increase in ROS levels, indicating that Foxp3 acts through the induction of p21 and the subsequent ROS elevation to trigger senescence. Collectively, these results suggest that Foxp3 is a downstream target of p53 that is sufficient to induce p21 expression, ROS production and p53-mediated senescence.</P>

        Beijing urban particulate matter-induced injury and inflammation in human lung epithelial cells and the protective effects of fucosterol from <i>Sargassum binderi</i> (Sonder ex J. Agardh)

        <P><B>Abstract</B></P> <P>Particulate matter (PM) air pollution has gradually become a widespread problem in East Asia. PM may cause unfamiliar inflammatory responses, oxidative stress, and pulmonary tissue damage, and a comprehensive understanding of the underlying mechanisms is required in order to develop effective anti-inflammatory agents. In this study, fine dust collected from Beijing, China (CPM) (size < PM13 with majority < PM2.5) was evaluated for its oxidative stress- and inflammation-inducing effects, which cause cell damage, in A459 human lung epithelial cells. Oxidative stress was marked by an increase in intracellular ROS levels and the production of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), and heme oxygenase-1 (HO-1). Upon induction of oxidative stress, a marked increase was observed in the expression of key inflammatory mediators such as COX-2 and PGE<SUB>2</SUB> and the pro-inflammatory cytokines TNF-α and IL-6 via NF-kB and MAPK pathways. Cellular damage was marked by a reduction in viability, increased lactate dehydrogenase (LDH) release, formation of apoptotic and necrotic bodies, accumulation of sub-G1 phase cells, and DNA damage. Apoptosis was found to be mediated via the activation of caspases through the mitochondria-mediated pathway. Fucosterol, purified from the brown alga <I>Sargassum binderi</I> (Sonder ex J. Agardh) by bio-assay-guided fractionation and purification, exhibited potential therapeutic effects against CPM-induced detrimental effects. Further studies could focus on developing fucosterol, in forms such as steroidal inhalers, against PM-induced pulmonary tissue inflammation.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Fine dust air pollution is a major reason of pulmonary complications in East Asia. </LI> <LI> Dust particles induce oxidative stress and inflammation damaging the lung epithelial cells. </LI> <LI> Fucosterol suppressed the dust induced cell damage by inhibiting oxidative stress and inflammation. </LI> <LI> Fucosterol may have beneficial effects in alleviating adverse respiratory effects of air pollution. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        p34 is a novel regulator of the oncogenic behavior of NEDD4-1 and PTEN

        PTEN is one of the most frequently mutated or deleted tumor suppressors in human cancers. NEDD4-1 was recently identified as the E3 ubiquitin ligase for PTEN; however, a number of important questions remain regarding the role of ubiquitination in regulating PTEN function and the mechanisms by which PTEN ubiquitination is regulated. In the present study, we demonstrated that p34, which was identified as a binding partner of NEDD4-1, controls PTEN ubiquitination by regulating NEDD4-1 protein stability. p34 interacts with the WW1 domain of NEDD4-1, an interaction that enhances NEDD4-1 stability. Expression of p34 promotes PTEN poly-ubiquitination, leading to PTEN protein degradation, whereas p34 knockdown results in PTEN mono-ubiquitination. Notably, an inverse correlation between PTEN and p34/NEDD4-1 levels was confirmed in tumor samples from colon cancer patients. Thus, p34 acts as a key regulator of the oncogenic behavior of NEDD4-1 and PTEN.

      • Search for light tetraquark states in ϒ(1S) and ϒ(2S) decays

        <P>We search for the J(PC) = 0(--) and 1(+-) light tetraquark states with masses up to 2.46 GeV/c(2) in gamma(1S) and gamma(2S) decays with data samples of (102 +/- 2) million and (158 +/- 4) million events, respectively, collected with the Belle detector. No significant signals are observed in any of the studied production modes, and 90% credibility level (C. L.) upper limits on their branching fractions in Upsilon(1S) and Upsilon(2S) decays are obtained. The inclusive branching fractions of the Upsilon(1S) and Upsilon(2S) decays into final states with f(1)(1285) are measured to be B(Upsilon(1S) -> f(1)(1285) + anything) = (46 +/- 28(stat) +/- 13(syst)) x 10(-4) and B(Upsilon(2S) -> f(1)(1285) + anything) = (22 +/- 15(stat) +/- 6.3(syst)) x 10(-4). The measured chi(b2) -> J/Psi + anything branching fraction is measured to be (1.50 +/- 0.34(stat) +/- 0.22(syst)) x 10(-3), and 90% C. L. upper limits for the chi(b0;b1) -> J/Psi + anything branching fractions are found to be 2.3 x 10(-3) and 1.1 x 10(-3), respectively. For B(chi(b1) -> omega + anything), the branching fraction is measured to be (4.9 +/- 1.3(stat) +/- 0.6(syst) x 10(-2). All results reported here are the first measurements for these modes.</P>

      • Search for Λc+→ϕpπ0 and branching fraction measurement of Λc+→K−π+pπ0

        <P>We have searched for the Cabibbo-suppressed decay Lambda(+)(c) -> pi p(0) in e(+) e(-) collisions using a data sample corresponding to an integrated luminosity of 915 fb(-1). The data were collected by the Belle experiment at the KEKB e(+) e(-) asymmetric-energy collider running at or near the (4S) and (5S) resonances. No significant signal is observed, and we set an upper limit on the branching fraction of B(Lambda(+)(c) -> phi p(0)) < 15.3 x 10(-5) at 90% confidence level. The contribution of nonresonant Lambda(+)(c) -> K+ K- p pi(0) decays is found to be consistent with zero, and the corresponding upper limit on its branching fraction is set to be B(Lambda(+)(c) ->. K+ K- p pi(0))(NR) < 6.3 x 10(-5) at 90% confidence level. We also search for an intermediate hidden-strangeness pentaquark decay P-s(+) -> phi p. We see no evidence for this intermediate decay and set an upper limit on the product branching fraction of B(Lambda(+)(c) -> P-s(+) pi(0)) x B(P-s(+) -> phi p) < 8.3 x 10(-5) at 90% confidence level. Finally, we measure the branching fraction for the Cabibbo-favored decay Lambda(+)(c) -> K- pi(+) p pi(0); the result is B(Lambda(+)(c) -> K- pi(+) p pi(0)) = (4.42 +/- 0.05(stat)+/- 0.12(syst)+/- 0.16(norm))%, which is the most precise measurement to date.</P>

        Detection and genotyping of Korean porcine rotaviruses

        Porcine group A rotavirus (GARV) is considered to be an important animal pathogen due to their economic impact in the swine industry and its potential to cause heterologous infections in humans. This study examined 475 fecal samples from 143 farms located in 6 provinces across South Korea. RT-PCR and nested PCR utilizing primer pairs specific for the GARV VP6 gene detected GARV-positive reactions in 182 (38.3%) diarrheic fecal samples. A total of 98 porcine GARV strains isolated from the GARV-positive feces were analyzed for G and P genotyping. Based on the sequence and phylogenetic analyses, the most predominant combination of G and P genotypes was G5P[7], found in 63 GARV strains (64.3%). The other combinations of G and P genotypes were G8P[7] (16 strains [16.3%]), G9P[7] (7 strains [7.1%]), G9P[23] (2 strains [2.0%]), and G8P[1] (1 strain [1.0%]). The counterparts of G or P genotypes were not determined in three G5, five P[7], and one P[1] strains. Interestingly, phylogenetic analysis indicated that all Korean G9 strains were more closely related to lineage VI porcine and human viruses than to other lineages (I-V) of GARVs and to Korean human G9 strains (lineage III). These results show that porcine GARV infections are common in diarrheic piglets in South Korea. The infecting strains are genetically diverse, and include homologous (G5P[7]), heterologous (G8P[1]), and reassortant (G8P[7]), as well as emerging G9 GARV strains.

        알칼리장석-일라이트가 육용오리의 생산성 및 육질에 미치는 영향

        본 연구는 3주령의 육용오린 사료에 알칼리장석-일라이트를 0, 0+ 항생제, 0.5, 1.0 및 $1.5\%$ 첨가한 5처리구에 3반복으로 각각 12수씩 배치하여 43일간 급여하여 생산성 및 육질에 미치는 영향을 알아보고자 실행하였다. 육성오리의 일당 증체량은 알칼리장석-일라이트 1.0와 $1.5\%$ 첨가구에서 약간 증가하였다(p>0.05). 사료섭취 량은 알칼리장석-일라이트 첨가구에서 증가하는 경향이었다(p>0.05). 혈중 글루코스 농도는 알칼리장석-일라이트 $0.5\%$ 처리구에서 약간 감소한 반면에(p>0.05) 혈중 요소태 질소 함량은 알칼리장석-일라이트 $0.5\%$ 첨가구에서 유의적으로 증가하였다(p<0.05). 콜레스테롤 함량은 알칼리장석-일라이트 $0.5\%$ 첨가구에서 유의적으로 감소하였다(p<0.05). 도체중과 도체율은 알칼리장석-일라이트 첨가수준에 따라 증가하는 경향이었다(p>0.05). 알칼리 장석-일라이트 급여에 의한 육용오리 가슴육의 조지방 함량은 알칼리장석-일라이트 $1.5\%$ 첨가구에서는 유의적으로 감소하였다(p<0.05). 육색의 명도와 황색도는 알칼리장석-일라이트에서 높게 나타났으며(p>0.05), 콜레스테롤 함량은 알칼리장석-일라이트 첨가구에서 감소하였다(p>0.05). 지방산 패도는 알칼리장석-일라이트 첨가구에서 약간 감소하였다(p>0.05). 알칼리장석-일라이트 첨가에 의한 포화지방산 비율이 약간감소하는 경향인 반면에 불포화지방산 비율이 약간 증가하는 경향을 나타내었으나 유의적인 차이는 없었다(p>0.05). 알칼리장석-일라이트 첨가에 의한 육성오리 간의 중금속 함량은 납 축적량이 비교적 높게 나타났다(p>0.05). 관능 평가(appearance)에서 알칼리장석-일라이트 1.0와 $1.5\%$ 첨가구에서 외관의 유의적인 개선 효과를 나타내었다 (p<0.05). 이상의 결과를 종합해 볼 때 육성오리에 대한 알칼리장석-일라이트 급여는 증체량의 개선효과와 더불어 가슴육의 조지방 함량의 감소 그리고 관능평가에서 외관의 개선 효과가 있음을 알 수 있었다. This experiment was conducted to investigate the effect of the supplemental alkali feldspar-ilite(feldspar) on growth performance and meat quality in broiler ducks for 43 days. One hundred eighty broiler ducks were divided into 5 groups of 12ducks. Dietary levels of feldspar 0, 0+antibiotics, 0.5, 1.0 and $1.5\%$ were added to experimental diets of each of the groups. Daily weight gain was slightly increased in 1.0 and $1.5\%$ feldspar treatments. Feed intake was slightly increased at all feldspar treatments. Glucose concentration of serum profile was decreased whereas BUN concentration was significantly increased (p<0.05) at $0.5\%$ feldspar. Cholesterol concentration was decreased at all feldspar treatments, this difference was especially observed in supplemental levels of $0.5\%$ feldspar(p<0.05). Carcass weight was increased at all feldspar treatments. Moisture and crude fat contents of proximate chemical composition in duck meat were decreased at all feldspar treatment, this difference especially was observed in supplemental levels of $1.5\%$ feldspar(p<0.05) on crude fat content. Lightness and yellowness was increased at all feldspar treatment. Cholesterol contents and TBA in meat were decreased, but this parameters were not difference by feldspar treatment. The composition of saturated fatty acids(SFA) was decreased, whereas unsaturated fatty acids(USFA) was slightly increased by feldspar treatment. The Pb content of heavy metal concentrations was increased with compared control, but not difference. The appearance of sensory evaluation was improved by supplemental feldspar, especially in supplemental feldspar, 1.0 and $1.5\%$(p<0.05). The results of this study indicate that the supplemental alkali feldspar may improve the production and meat quality of broiler ducks.

        Status of the KSTAR superconducting magnet system development

        <P>The aim of the Korea superconducting tokamak advanced research (KSTAR) project is to develop a steady-state-capable advanced superconducting tokamak for establishing a scientific and technological basis for an attractive fusion reactor. Since the KSTAR mission includes the achievement of a steady-state-capable operation, the use of superconducting coils is an obvious choice for the magnet system. The KSTAR superconducting magnet system consists of 16 toroidal field (TF) and 14 poloidal field (PF) coils which include 8 central solenoid coils. Both the TF and PF coil systems use internally-cooled cable-in-conduit conductors (CICC). The TF coil system provides a magnetic field of 3.5 T at the plasma centre and the PF coil system provide a flux swing of 17 V s. The major achievement in the KSTAR magnet system development includes the development of CICC, a full size TF model coil, a background magnetic field generation coil system and the construction of a large scale superconducting magnet and the CICC test facility. TF and PF coils are at the stage of fabrication for the KSTAR completion in the year 2007.</P>

      • Finite gap behaviour in the superconductivity of the ‘infinite layer’ n-doped high-<i>T</i><sub>c</sub> superconductor Sr<sub>0.9</sub>La<sub>0.1</sub>CuO<sub>2</sub>

        <P>We report on the first small-angle neutron scattering measurements from the flux line lattice (FLL) in the high-<I>T</I><SUB>c</SUB> cuprate superconductor Sr<SUB>0.9</SUB>La<SUB>0.1</SUB>CuO<SUB>2</SUB>. Using a polycrystalline sample, the scattered intensity decreases monotonically with scattering angle away from the undiffracted beam, independently of the azimuthal angle around the beam. The absence of clear peaks in the intensity suggests the establishment of a highly disordered FLL within the grains. We find that the intensity distribution may be represented by the form factor for a single flux line in the London approximation, with some contribution from crystal anisotropy. Most interestingly however, we find that, over the observed field range, the temperature dependence of the diffracted intensity is best represented by s-wave pairing, with lower limits of the gap values being very similar to the Bardeen–Cooper–Schrieffer value of Δ(0) = 1.76<I> k</I><SUB>B</SUB><I>T</I><SUB>c</SUB>. However, a qualitative consideration of corrections to the observed intensity suggests that these gap values are likely to be higher, implying strong-coupling behaviour.</P>

        AzTEC millimetre survey of the COSMOS field – I. Data reduction and source catalogue

        <P>ABSTRACT</P><P>We present a 1.1 mm wavelength imaging survey covering 0.3 deg<SUP>2</SUP> in the COSMOS field. These data, obtained with the AzTEC continuum camera on the James Clerk Maxwell Telescope, were centred on a prominent large-scale structure overdensity which includes a rich X-ray cluster at <I>z</I>≈ 0.73. A total of 50 mm-galaxy candidates, with a significance ranging from 3.5 to 8.5σ, are extracted from the central 0.15 deg<SUP>2</SUP> area which has a uniform sensitivity of ∼1.3 mJy beam<SUP>−1</SUP>. 16 sources are detected with S/N ≥ 4.5, where the expected false-detection rate is zero, of which a surprisingly large number (9) have intrinsic (deboosted) fluxes ≥5 mJy at 1.1 mm. Assuming the emission is dominated by radiation from dust, heated by a massive population of young, optically obscured stars, then these bright AzTEC sources have far-infrared luminosities >6 × 10<SUP>12</SUP> L<SUB>⊙</SUB> and star formation rates >1100 M<SUB>⊙</SUB> yr<SUP>−1</SUP>. Two of these nine bright AzTEC sources are found towards the extreme peripheral region of the X-ray cluster, whilst the remainder are distributed across the larger scale overdensity. We describe the AzTEC data reduction pipeline, the source-extraction algorithm, and the characterization of the source catalogue, including the completeness, flux deboosting correction, false-detection rate and the source positional uncertainty, through an extensive set of Monte Carlo simulations. We conclude with a preliminary comparison, via a stacked analysis, of the overlapping MIPS 24-μm data and radio data with this AzTEC map of the COSMOS field.</P>
