        Using Periostin as a Biomarker in the Treatment of Asthma

        Periostin acts both as an extracellular matrix protein belonging to the fasciclin family and as a matricellular protein functioning in cell activation by binding to its receptors on the cell surface. It has been established that periostin is a downstream molecule of interleukin (IL)-13, a signature type 2 cytokine, and that periostin plays an important role in the pathogenesis of allergic diseases, including asthma. Based on these findings, much attention has been paid to periostin as a biomarker useful in the treatment of asthma. Periostin is a surrogate biomarker for type 2 immunity; it has been shown that serum periostin can predict the efficacy of anti-IL-13 antibodies (lebrikizumab) and anti-IgE antibodies (omalizumab), and that this usefulness can be potentially expanded to other type 2 antagonists. Moreover, it has been shown that periostin is not a simple surrogate biomarker for type 2 immunity; periostin-high asthma patients have several unique characteristics, including eosinophilia, high fraction of nitric oxide, aspirin intolerance, nasal disorders, and late onset. These characteristics are likely to be correlated with the involvement of periostin in the tissue remodeling of asthma. Periostin is also associated with hyporesponsiveness to inhaled corticosteroids, probably reflecting tissue remodeling. Thus, periostin has 2 characteristics as a biomarker for early diagnosis of asthma: surrogate biomarkers for type 2 immunity and tissue remodeling. Based on these characteristics, we will be able to apply serum periostin to treatment of asthma.

        In August of 1998, Toyota city put in the operation of the “ITS model district experiment, In Toyota”, appointed by the “Traffic and Motor Vehicle Intelligence Enforcing Committee” in japan. The content of the experiment is composed of 4 themes and 3 social experiments utilizing ITS technologies and researched facts on operation probabilities for the enforcement of 3 social experiments. The 3 types of experiments are ①Experiment on Car-Navigation of ITS as well as the VICS(Vehicle Information and Communication System) and the Traffic Information and Communication System for higher technologies. ②P&P experiment of hourly events. ③The EV collaborated utilization experiments. These experiments have been enforced in 1999. Furthermore, Toyota city investigated the expected outcomes by analyzing the test cases as it's main source on ITS experiments in 1999.

        Periostin and Interleukin-13 Are Independently Related to Chronic Spontaneous Urticaria

        <P>Chronic spontaneous urticaria (CSU) is a complex idiopathic disease of the skin with various cellular infiltrations. Although mast cells are key effector cells in the pathogenesis of CSU, CD4+ T helper 2 cells also have particular roles in the development and maintenance of CSU. Periostin is known as a downstream molecule of interleukin (IL)-4 and IL-13, key cytokines of type 2 immune responses. In this study, we examined periostin and IL-13 levels in the sera of patients with CSU (n=84) and healthy normal controls (NCs, n=43). Periostin levels were significantly lower in the CSU group than in NCs (71.4±21.8 vs 85.1±22.4 ng/mL, <I>P</I>=0.04). Periostin levels were also lower in the severe CSU group than those in mild CSU (59.7±18.0 vs 73.4±22.0 ng/mL, <I>P</I>=0.04). However, IL-13 levels were significantly higher in patients with CSU than in NCs (508.5±51.2 vs 200.7±13.3 pg/mL, <I>P</I>=0.001). In conclusion, periostin and IL-13 may be independently related to the pathogenesis of CSU.</P>

        AKARI OBSERVATIONS OF DUSTY TORI OF ACTIVE GALACTIC NUCLEI

        The dusty torus of Active Galactic Nuclei (AGNs) is one of theimportant components for theunification theory of AGNs. The geometry and properties ofthe dusty torus are key factors in understanding the nature of AGNs aswell as the formation and evolution of AGNs. However, they are stillunder discussion. Infrared observation is useful for understanding the dustytorus as thermal emission from hot dust with the dust sublimationtemperature ($\sim$ 1500 K) has been observed in the infrared. We have analyzed infrared spectroscopic data of low-redshift andhigh-redshift quasars, which are luminous AGNs. For the low-redshift quasars, we constructed the spectral energydistributions (SEDs) with AKARI near-infrared and Spitzer mid-infraredspectra and decomposed the SEDs into a power-lawcomponent from the nuclei, silicate features, and blackbody components withdifferent temperatures from the dusty torus. From the decomposition,the temperature of the innermost dusty torus shows the range between900-2000 K. For the high-redshiftquasars, AKARI traced rest-frameoptical and near-infrared spectra of AGNs. Combining with WISE data,we havefound that the temperature of the innermost dusty torus in high redshift quasarsis lower than that in typical quasars. The hydrogen H$\alpha$ emission line from the braod emission line region in thequasars also shows narrow full widthat half maximumof 3000$-$4000 km s$^{-1}$. These results indicatethat the dusty torus and the broad emission line region are more extendedthan those of typical quasars.

        Streptomyces lividasn 의 Blasticidin S 에 대한 내성 Mechanism

        A mechanism of resistance to blasticidin S in Streptomyces lividans 66 was investigated. Although the microorganism was highly resistant to the drug in vivo, the in vitro protein-synthesizing system was significantly sensitive. The possible resistance mechanism was discussed and suggested.

        Serum Periostin Is Negatively Correlated With Exposure to Formaldehyde and Volatile Organic Compounds in Children

        Epidemiological studies have shown that exposure to tobacco smoke causing irritation and inflammation in the airways tends to reduce serum periostin concentrations in adults. We now investigate prospective cross-sectional study on 135 Korean students aged 7 years in the first grade who were participating in the Seongnam Atopy Project for Children's Happiness 2016 (SAP2016) cohort. To the best of our knowledge, this is the first study to show significant inverse correlations between serum periostin concentration and exposure to xylene and formaldehyde in children. Our findings suggested the need for caution in using the serum periostin level as a marker for allergic diseases, since exposure to volatile organic compounds and formaldehyde may confound the interpretation of these results.

        Serum Periostin Levels: A Potential Serologic Marker for Toluene Diisocyanate-Induced Occupational Asthma

        Purpose: Toluene diisocyanate (TDI) is a leading cause of occupational asthma (OA). Periostin is a matricellular protein implicatedin type 2 immunity-driven asthma. Its pathogenic role in TDI-OA has not been completely elucidated. The present study was performed to investigate the role of periostin in TDI-OA. Materials and Methods: Serum periostin levels were measured in subjects with TDI-OA, asymptomatic TDI-exposure controls (AECs), non-occupational asthmatics (NAs), and unexposed normal controls (NCs). To understand the mechanism by which TDI induces periostin production, primary small airway epithelial cells (SAECs) were cultured under stimulation of TDI and neutrophilsfrom asthmatic patients. Results: Fifty-three subjects with TDI-OA, 71 AECs, 67 NAs, and 83 NCs were enrolled. Serum periostin levels were significantly higher in TDI-OA subjects than in AECs (p=0.001), NAs (p<0.001), and NCs (p<0.001). In TDI-exposed subjects (TDI-OA and AEC), the PC20 methacholine levels were significantly lower in subjects with a higher periostin level than in those with a lower periostin level. TDI exposure did not increase periostin production directly by SAECs; however, periostin production increased significantly after co-culture with TDI and neutrophils, which was suppressed by an antioxidant. In addition, increased release of TGF-β1 was noted from SAECs when exposed to TDI and neutrophils, which was also suppressed by an antioxidant. Conclusion: These results suggest that an increased periostin level may contribute to the progression of airway inflammation to remodeling in TDI-exposed workers. A high serum periostin level is a potential serologic marker of the phenotype of TDI-OA.
