Objective: The aim of this research was to evaluate the effect of different concentrate levels in diets based on cactus Opuntia Stricta (Haw.) Haw cladodes on the performance of lactating Girolando cows. Methods: The experiment involved 10 Girolando multiparous dairy cows at 512.6 kg of body weight (BW) and producing 13.2 kg milk/d, allocated into two 5×5 Latin squares. The experimental treatments consisted of control diet composed by cactus Nopalea cochenillifera. Salm-Dyck. cladodes (Nopalea), forage sorghum silage and concentrate at 20% on dry matter (DM) basis, and four concentrate levels diets (20%, 24%, 28%, and 32%) plus cactus Opuntia stricta (Haw.) Haw. cladodes (Opuntia) and forage sorghum silage. Results: Regarding cows fed control diet, the nutrients intake were greater than for cows fed with cactus Opuntia and concentrate. Regarding concentrate levels, intakes of DM, organic matter (OM), crude protein (CP), non-fiber carbohydrates (NFC), and total digestible nutrients of cows increased linearly. Organic matter, CP, and NDF digestibilities were similar in between to control diet and cactus Opuntia-based diets. The digestibility of NFC increased linearly when the concentrate was inserted. The N balance was the same for control diet and cactus Opuntia-based diets, irrespective the concentrate levels. Conclusion: For cows producing 14 kg/d with 3.5% of fat, it is recommended 32% of concentrate to be included in cactus Opuntia-based diets, and the increase in concentrate level promotes a linear increase in milk yield.

This paper presents a new methodology to apply a kinetic hydrate inhibitor (KHI) to prevent hydrateplugging considering the system availability of the topside system in an offshore gas platform. Theproposed methodology comprises three steps: the system availability analysis by Monte Carlosimulation, subsea multiphaseflow simulation to evaluate the risk of hydrate plugging, and application ofKHI to prevent hydrate plugging. The topside system availability and its longest downtime wereestimated to be 99.3% and 47 h, respectively. As a result of the multiphaseflow simulation, thetemperature in theflowline decreases from 40 C to 5 C during the system downtime, creating thecondition for hydrate formation. Thus, polyvinylcaprolactam (PVCap) was used to delay hydrate pluggingfor a period longer than the cooldown time. As a result, the injection rate of 0.75 wt% of PVCap wasrequired 0.6 times per year to prevent hydrate plugging, while 30 wt% of mono ethylene glycol (MEG) wasrequired under the same hydrate formation condition. This approach provides the guideline for KHIinjection according to the system downtime andflowing performance in subsea gasflowline as a riskmanagement strategy to mitigate operation risks in the offshore gas platform.

Background: The Wnt/${\beta}$-catenin signaling pathway is an important regulator of cellular functions such as proliferation, survival and cell adhesion. Wnt/${\beta}$-catenin signaling is associated with tumor initiation and progression; ${\beta}$-catenin mutations explain only 30% of aberrant signaling found in breast cancer, indicating that other components and/or regulation of the Wnt/${\beta}$-catenin pathway may be involved. Objective: We evaluated AXIN2 rs2240308 and rs151279728 polymorphisms, and expression profiles of ${\beta}$-catenin destruction complex genes in breast cancer patients. Materials and Methods: We collected peripheral blood samples from 102 breast cancer and 102 healthy subjects. The identification of the genetic variation was performed using PCR-RFLPs and DNA sequencing. RT-qPCR was used to determine expression profiles. Results: We found significant association of AXIN2 rs151279728 and rs2240308 polymorphisms with breast cancer risk. Significant increase was observed in AXIN2 level expression in breast cancer patients. Further analyses showed APC, ${\beta}$-catenin, CK1${\alpha}$, GSK3${\beta}$ and PP2A gene expression to be associated to clinic-pathological characteristics. Conclusions: The present study demonstrated, for the first time, that AXIN2 genetic defects and disturbance of ${\beta}$-catenin destruction complex expression may be found in breast cancer patients, providing additional support for roles of Wnt/${\beta}$-catenin pathway dysfunction in breast cancer tumorigenesis. However, the functional consequences of the genetic alterations remain to be determined.
Can the body composition of crossbred dairy cattle be predicted by equations for beef cattle?

Objective: The aim of the study was to evaluate the efficiency of the Hankins and Howe (HH46), Valadares Filho (V06), and Marcondes (M12) equations for predicting the physical and chemical composition of dairy crossbred bulls carcasses, as well as the chemical composition of their empty bodies. Methods: This study was conducted using 30 dairy crossbred bulls. One group of five animals was slaughtered at the beginning of the experiment, and the remaining were slaughtered 112 days later. Animals were distributed in a completely randomized design into treatments consisting different levels of concentrate (0%, 17%, 34%, 51%, and 68%). The physical and chemical compositions of the cattle were obtained from the right half of the carcass and using samples taken between the 9th and 11th ribs of the left half of the carcass. The estimated and experimentally determined values were compared using the correlation and concordance coefficient, as well as the mean square error of prediction (MSEP) and its components. Results: The HH46 equations were better at estimating the amount of muscle plus fat in the carcass. The amount of bone in the carcasses could not be well estimated by the HH46 and M12 models. The M12, HH46, and V06 equations were worst at estimating the amounts of protein, ether extract, and water in the carcass, respectively. In the empty body, the amounts of protein and water were well estimated by the HH46 equations. Protein, ether extract, and water were accurately estimated by the V06 equations, and ether extract by the M12 equations. Conclusion: The physical and chemical composition of dairy crossbred bull carcasses, as well as the chemical composition of their empty bodies, can be predicted using the equations tested here. The amount of bone in these carcasses could not be accurately predicted.
