        인진호가 Hep G2 세포에서 에탄올 매개성 Cytokine 분비에 미치는 영향

        A human hepatoma cell line, Hep G2 cells, is reliable for the study of alcohol-induced hepatotoxicity. The aim of this study is to determine the relationship between TNF-${\alpha}$, IL-$1{\alpha}$ production and EtOH-induced cytotoxicity on Hep G2 cells. The cells were incubated with EtOH in the presence of Artemisia Capillaris Herba(AC) for 24 hours and in the absence of AC for 48 hours. Cytoviability and cytokines release were analyzed by MTT assay and enzyme linked immunosorbent assay (ELISA), respectively. After 24 hours of EtOH exposure, the cytoviability had markedly decreased, and the release of cytokines had increased. The increased amount of cytokines contributed to EtOH-induced cytotoxicity. Anti-TNF-${\alpha}$ and IL-$1{\alpha}$ antibodies almost abolished it. Interestingly, EtOH-induced cytotoxicity and cytokines production were inhibited by AC. Moreover, when AC was used in combination with antibodies, there was a marked inhibition of EtOH-induced cytotoxicity. These results suggest that EtOH-induced cytotoxicity may regulate, by various factors, and AC may prevent the cytotoxicity through partial inhibition of the $TNF-{\alpha}$ and IL-$1{\alpha}$ secretion.

        Acute and Sub-chronic Oral Toxicity Study of Ammonium Persulfate in Spraque-Dawley Rats

        The toxicity test of ammonium persulfate was conducted to ensure of its potential toxic effects according to the single-dose acute oral toxicity study (OECD Guideline 423) and 90-day repeated dose sub-chronic oral toxicity study guideline (OECD Guideline 408) for establishing national chemical management system, and matching in the Globally Harmonized Classification System (GHS) category. In acute oral toxicity study, pasty stool, perineal contamination and temporary body weight decrease were observed after dosing 1st and 2nd challenge (300 ㎎/㎏ body weight). All test animals were dead within 6 hours after dosing at 3rd challenge (2000 ㎎/㎏ body weight). Therefore, the GHS class of test substance is considered class 4. In sub-chronic toxicity study, body weight changes, food consumptions, hematological, biochemical and pathological examination did not show any noticeable and significant differences between the administered (5, 20, 80 ㎎/㎏ body weight) and control (vehicle only) group animals. Based on these results, the no observed adverse effect level (NOAEL) is considered above 80 mg/kg body weight.

        Streptococcus dysgalactiae로부터 분리된 히알루론산과 황화된 유도체의 구조와 항염증 활성

        히알루론산(HA, Hyaluronic acid)은 β-1, 3-N-acetyl glucosamine과 β-1, 4-glucuronic acid가 반복된 선형 폴리머 고분자로서 생물학적 활성 및 생체친화성 특성 때문에 의약 및 약학분야에서 중요한 분자로 여겨지고 있다. 본 연구는 HA을 S. dysgalactiae으로 얻고, 화학적 방법을 통해 황화된 히알루론산(S-HA, Sulfated hyaluronic acid)유도체를 합성하여 그 구조와 항염증 활성을 비교하였다. HA의 생산은 S. dysgalactiae를 5 l 생물반응기를 이용하여 대량 배양하여 수용성 히알루론산(HA-WS, water soluble hyaluronic acid)과 비수용성 히알루론산(HA-WI, water insoluble hyaluronic acid)을 분리 정제하였다. 특히 HA-WI를 황화시켜 황화된 히알루론산(S-HA) 유도체를 합성하였으며, 그 수율은 90%로 나타났다. 합성된 S-HA의 구조를 FT-IR 및 ¹H/<SUP>13</SUP>C-NMR를 통해 S. dysgalactiae 로부터 생산된 표준 HA, HA-WS 및 HA-WI와 비교 분석한 결과, 황으로 치환된 양상을 확인하였다. 또한, S-HA의 항염증 활성을 RAW 264.7 대식세포를 통해 확인한 결과, S-HA는 천연 형태의 HA (HA, HA-WS)보다 nitric oxide (NO)와 COX-2 및 PGE₂ 유전자 발현이 유의하게 낮게 발현되었다. 염증 매개 cytokine인 TNF-α (<80 pg/ml) 및 IL-6 (<100 pg/ml)의 생성도 S-HA가 천연 HA보다 낮은 수준으로 정량되었다. 이 같은 결과에서 황화된 S-HA은 천연 히알루론산보다 용해성이 우수하고 염증관련 사이토카인의 생성 억제를 통해 항염증 효과를 나타내므로 염증치료제, 성형 및 생체 적용 약물전달 소재로 그 활용이 기대된다. Hyaluronic acid (HA) is an important macromolecule in medical and pharmaceutical fields. HA is a natural and linear polymer composed of repeating disaccharide units of β-1, 3-N-acetyl glucosamine and β-1, 4-glucuronic acid. This work aimed to confirm the structural characteristics and anti-inflammatory activities of HA and its chemically sulfated-HA. HA was produced from a fed-batch fermentation process using Streptococcus dysgalactiae in a 5 l bioreactor. HA was isolated water-soluble form (HA-WS) and water-insoluble form (HA-WI) from culture medium, and was obtained chemically sulfated-derivative (S-HA) that resulted in a 90% yield from HA-WI. The structural features of the sulfated- HA (S-HA) were investigated by FT-IR and ¹H-NMR spectroscopy. The FT-IR and NMR patterns revealed the similarity in both the FTIR spectrum as well as NMR spectrum of both reference standard and purified HA from S. dysgalactiae. The anti-inflammatory activities of HA and S-HA were examined on LPS-induced RAW 264.7 cells. S-HA was significantly inhibited production of pro-inflammatory mediators such as nitric oxide (NO) and PGE₂ and the gene levels of iNOS and COX-2, which are responsible for the production of NO and PGE₂, respectively. Furthermore, S-HA also suppressed the overproduction of pro-inflammatory cytokine TNF-α (<80 pg/ml) and IL-6 (<100 pg/ml) compared to that of HA-WI. The present study clearly demonstrates that HA-S exhibits anti-inflammatory activities in RAW 264.7 macrophage cells.

        A 12-b Asynchronous SAR Type ADC for Bio Signal Detection

        This paper describes a low power asynchronous successive approximation register (SAR) type 12b analog-to-digital converter (ADC) for biomedical applications in a 0.35 ${\mu}m$ CMOS technology. The digital-to-analog converter (DAC) uses a capacitive split-arrays consisting of 6-b main array, an attenuation capacitor C and a 5-b sub array for low power consumption and small die area. Moreover, splitting the MSB capacitor into sub-capacitors and an asynchronous SAR reduce power consumption. The measurement results show that the proposed ADC achieved the SNDR of 68.32 dB, the SFDR of 79 dB, and the ENOB (effective number of bits) of 11.05 bits. The measured INL and DNL were 1.9LSB and 1.5LSB, respectively. The power consumption including all the digital circuits is 6.7 ${\mu}W$ at the sampling frequency of 100 KHz under 3.3 V supply voltage and the FoM (figure of merit) is 49 fJ/conversion-step.

        This paper discusses about the implementation of a simulator for the Navigation Control System(NCS) of bimodal tram. To operate a NCS of bimodal tram, values of all sorts of sensors installed in bimodal tram should be transmitted to the NCS, and the NCS calculates the measured sensor values to determine traveling direction, traveling speed, current position etc. The implementation of the simulator consists of a device applying driver’s input transaction function & virtual sensor program and a sub-rack device that controls communication with the NCS to evaluate navigation control function. The virtual sensor program can create routes (map), traveling profiles & seat information et cetera in order to transmit to the NCS, analyzes driver's input values and NCS output values to create virtual sensor values. The sub-rack device takes charge of communication with the NCS using CAN-OPEN, CAN-J1939, MVP protocols. This paper discusses about the implementation of the simulator and afterwards analyzes and evaluates the NCS simulation results.

        The Effect of Extract from Sea Buckthorn on DNCB-induced Atopic Dermatitis in NC/Nga Mice

        Sea Buckthorn (Hippophae rhamnoides L.) has been used in traditional medicine for the treatment of cough, indigestion, circulatory problems and pain. The associated anti-inflammatory effect of this agent is achieved via the inhibition of Nf-${\kappa}B$ signaling, a property that has been demonstrated to effectively control the symptoms of various skin disorders, including atopic dermatitis. Accordingly, the purpose of this study was to assess the efficacy of Sea Buckthorn in reducing the production of lipopolysaccharide (LPS) activated nitric oxide (NO) by inhibiting the Nf-${\kappa}B$ pathway, as measured by the symptoms of atopic dermatitis (AD) occurring secondarily to inflammation and immune dysregulation. Our data demonstrate that Sea Buckthorn significantly decreased the LPS-induced production of NO (p<0.001). Atopic dermatitis was induced by repeated application of 2,4-dinitrochlorobenzene to the dorsal skin of mice. Topical application of 5% Sea Buckthorn extract improved the symptoms of AD, specifically reducing disease severity scores, scratching behaviors and epidermal thickness. When compared to the control group, animals treated with Sea Buckthorn exhibited increased serum IL-12 levels and decreased serum TNF-${\alpha}$, IL-4 and IL-5 levels. Such a modulation of biphasic T-helper (Th)1/Th2 cytokines may result in a reduction in serum IgE levels. Our findings suggest that mechanism of action of Sea Buckthorn in the treatment of AD is associated with a marked anti-inflammatory effect as well as an inhibition of Th2-mediated IgE overproduction via the modulation of biphasic Th1/Th2 cytokines. Such results suggest that topical Sea Buckthorn extract may prove to be a novel therapy for AD symptoms with few side effects.

        국제 원유가의 지속적인 상승에 따라 화석연료 고갈을 대비한 대체에너지 및 온실가스배출 감소를 위하여 바이오연료의 시용 및 상용보급은 전세계적인 추세이다. 우리나라의 경우 바이오디젤은 2002년부터 시범보급사업(Demonstration & disseminatio을 거쳐 2000년 7월부터 전국주유소를 통하여 경유 중에 바이오디젤 0.5%를 혼합한 BD0.5를 수송용 연료로 도입하여 아시아 최초로 상용보급화를 시행하고 있다. 또한 휘발유 중 바이오에탄올 혼합 연료유 도입을 위한 실증평가연구를 2006년 8월부터 2008년 7월까지 수행중이다. 자동차용 휘발유의 옥탄가 향상을 위해 함산소 기재로 사용되는 MTBE(Methyl Tertiary Butyl Ether)를 바이오에탄올로 대체한 바이오에탄올 혼합연료유는 수분 혼입에 의한 상 분리(Phase separation)와 금속에 대한 부식성 문제를 야기 시킬 수 있다. 바이오에탄올을 서브옥란가솔린(Sub-octane gasoline)에 혼합하여 상 분리 모사실험, 금속류 부식시험, 고무류 침지실험 등 다양한 품질특성평가를 수행하였으며, 이런 결과들을 바탕으로 국내실정에 알맞은 최적의 혼합량(E3, E5)을 도출하였다. 또한 전국에 4개 시범주유소를 운영하여 바이오에탄올 혼합 연료유의 유통 및 보급을 통해 최적의 유통인프라(Distribution infrastructure) 보완 및 구축 방안을 도출 하고자 한다.

      • Characterization of OsJAC1 which is responding to different types of ionizing radiation

        Ionizing radiation affects gene expression from plant genomes. To monitor the genome-wide transcriptional changes induced by three types of ionizing radiation, we used the rice RNA sequencing to identify genes that are up- or down-regulated by gamma rays (GAs), proton (PRs) and ion beams (IBs). The Oryza sativa jacalin-like lectin domain containing proteins (OsJAC1) gene was highly induced by GAs, PRs and IBs. OsJAC1 was selected based on the expression patterns of a genome-wide dataset of RNA sequencing. Many jacalin-related lectin genes have been shown to be associated with disease resistance, biotic and abiotic stress signaling. Therefore, we studied its expression pattern in response to different abiotic stress and phytohormone treatments. The expression patterns of OsJAC1 under two different abiotic stress conditions (salt and heat stress) and phytohormones (salicylic acid and methyl jasmonate) were examined. The transcripts of OsJAC1 were significantly induced in response to abiotic stress conditions, including salt and heat treatments. In addition, it was induced in response to the salicylic acid and methyl jasmonate treatments, respectively. To investigate the sub-cellular localization of OsJAC1, the gene was expressed as a fusion protein tagged with GFP, in tobacco leaf epidermis and examined under confocal microscope. The OsJAC1 was clearly localized at the nucleus. These results provide critical insights into the molecular functions of the rice jacalin-like lectin domain containing proteins as receptors of external signals.
