      • Interleukin-1B(1L-1B) polymorphisms and gastric mucosal levels of IL-Iβ cytokine in Korean patients with gastric cancer

        Interleukin-1B and IL-1 receptor antagonist gene polymorphisms are associated with an increased risk of gastric cancer (GC) in Caucasian populations. However, recent studies could not find any association between IL-1B-511T polymorphism and the risk of GC in Asians. We tested for an association between IL-1 loci polymorphisms with increased gastric mucosal levels of IL-1β and an increased risk of developing GC in a Korean population. Polymorphisms of IL-1A-889, IL-1B-31, IL-1B-511 and IL-1RN were genotyped in 434 controls and 234 patients with GC. Mucosal IL-1β cytokine was measured using an ELISA. The frequencies of IL-1A, IL-1B-511, IL-1B-31 and IL-1RN were not statistically different between controls and all patients with GC. After subclassification of GC, only patients with intestinal-type GC showed a higher frequency of IL-1B-31T homozygotes (OR = 2.2; 95% CI = 1.1-4.3) compared with controls. Risk was also significantly increased in these patients for IL-1B-31T homozygotes compared with patients with diffuse-type GC (OR = 3.4; 95% CI = 1.5-7.7). As in Caucasian populations, linkage disequilibrium between IL-1B-31 and IL-1B-511 was nearly complete, but the pattern of haplotype related to the risk of GC (IL-1B-31T/IL-1B-511C) was opposite (IL-1B-511T/IL-1B-31C). Mucosal IL-1β levels in H. pylori-infected GC patients were higher in patients homozygous for IL-1B-31T compared with IL-1B-31C/T and IL-1B-31C/C. Thus, the combined effects of H. pylori infection and IL-1B-31T/IL-1B-511C polymorphisms with enhanced mucosal IL-1β production contributed to the development of intestinal-type GC in this Korean population.

        N/A Withdrawal of corticosteroids in patients with chronic type B hepatitis is frequently associated with enhanced cellular immune response to hepatitis B virus. This immunolgic rebound generally results in a transient reduction in levels of HBV-DNA and DNA polymerase, and seldom results in clearance of HBeAg. There was a pilot study that the combination of a short course of prednisone followed by ARA-AMP was potentially synergistic. The object of this study was to determine the effect of short term prednisolone followed by ARA-AMP on the immune system in patients with chronic active ltepatitis-B. Patients were started 40 mg of prednisolone daily for 4 weeks. After prednisolone therapy was discontinued, patients received no medication for 2 weeks. ARA-AMP was then given for 4 weeks at a dose of 500 mg daily. Immune parameters such as T cell subsets, IL, production, NK cell activeity, and LAK cell activity were tested before and during treatement period. The resullts were as follows. 1) Basal IL2 production in CAH-B (71.3 unit/ml) was lower than that of normal control (p<0.025). Basal NK activity, T4/T8 ratio and LAK activity of CAH-B were similar to those of normal control. 2) After prednisolone treatment, IL2 production, NK cell activity and LAK cell activity were significantly lower than those of basal value. 3) After induction of immune rebound, IL, production increased to 84.8 unit/ml (basal 71.3 unit/ml,

        특발성 염증성 근육병증 환자에서 IL-17 발현의 증가

        Objective: Idiopathic inflammatory myopathies (IIMs) are systemic autoimmune diseases characterized by infiltration of T lymphocytes, monocytes, and macrophages in muscle tissues. Interleukin-17 (IL-17), a Th17 cytokine, has potent pro-inflammatory actions and plays a role in autoimmune diseases. We investigated the expression of IL-17 in muscle tissues of patients with IIMs. Methods: We measured the IL-17 mRNA level of muscle tissues from 14 patients with IIMs (9 patients with dermatomyositis and 5 patients with polymyositis) by real-time RT-PCR and compared with controls. We also performed an immunohistochemical stain to detect IL-17 expression. Results: The expressions of IL-17 were significantly enhanced in IIMs than controls. In immunohistochemistry, IL-17 was expressed in perimysial, endomysial and perivascular infiltrating inflammatory cells. Conclusion: These results suggest that IL-17 plays a role in the immunopathogenesis of IIMs.

Four weeks ARA-AMP at a dose of 500mg daily showed potent immunosuppressive effect. It may be necessary to modulate dose, time and route of administration of ARA-AMP that show antiviral effect with no suppression of immune system.

        Analysis of histone deacetylase inhibitor-induced responses in human periodontal ligament fibroblasts.

        <P>Periodontal ligament (PDL) fibroblasts play critical roles in the regeneration of periodontal tissues damaged by periodontitis. Histone deacetylase inhibitors (HDIs) have been suggested to be potential tools in tissue engineering. The feasibility of using the HDI, sodium butyrate (NaB) for periodontal regeneration was examined by evaluating its effect on the osteogenic differentiation of human PDL fibroblasts and its modulation of the inflammatory responses to lipopolysaccharide (LPS). NaB did not cause significant cell death at 100?μM but promoted the expression of the osteoblast phenotype (Runx2, osterix, osteocalcin, and bone sialoprotein). NaB significantly inhibited the LPS-induced production of reactive oxygen species and the expression of pro-inflammatory cytokines (IL-1β and TNF-α). These results suggest that HDIs can offer a potential therapeutic agent for periodontal regeneration.</P>

        검정콩 껍질 유래 안토시아닌의 RBL-2H3 세포에서 탈과립화와 사이토카인 생성 저해 효과

        안토시아닌(anthocyanins)은 플라보노이드에 속하고 항산화 활성들을 포함한 그들의 다양한 건강 유익성에 대해 알려져 있다. 그들 중 검정콩 껍질에서 분리한 주요 안토시아닌은 glycopyranose를 함유하고 있는 배당체이다. 천식은 호염기성 세포(basophils)와 비만세포(mast cells)를 포함한 다양한 면역세포와 관련된 알레르기 관련 질병이다. 호산구 (eosinophils), 호염기성 세포, 비만세포는 탈과립화에 의한 천식-특이적 보조 2(T-helper 2) 사이토카인 분비 그리고 계속해서 일어나는 증폭과 같은 염증을 일으키는 매개체의 증가를 통해 알레르기 천식(allergic asthma)에서 중요한 역할을 한다. 흰쥐 호염기성 백혈병(rat basophilic leukemia) RBL-2H3 세포는 알레르기 반응을 측정하기 위해 가장 일반적으로 사용되는 in vitro 모델이다. 본 연구에서는 검정콩 껍질 안토시아닌이 항원으로 자극한 RBL-2H3 세포에서 탈 과립 그리고 Th2 사이토카인 생산에 미치는 효과를 조사하였다. 세포 탈과립은 β-hexosaminidase의 방출을 검출함으로써 평가하였다. IgE-항원 복합체로 자극한 RBL-2H3 세포내 β-hexosaminidase 방출과 Th2 사이토카인 생산이 무 처리군의 그것과 비교하여 더 높았다. 안토시아닌은 RBL-2H3 세포의 IgE-항원 복합체 유도 탈과립을 현저하게 억제 시켰고 RBL-2H3 세포내 IgE-항원 복합체-매개체 interleukin( IL)-4, IL-13 그리고 TNF-α 생산을 저해하였다. 이러한 결과는 검정콩 껍질 유래 안토시아닌이 알레르기 반응(allergic reaction)을 현저히 저해하는 효과가 있음을 보였고, 이들 안토시아닌이 향후 알레르기 천식을 억제하거나 개선하는데 유용한 물질로 사용될 가능성이 있을 것으로 판단되었다. Anthocyanins belong to a group of flavonoid compounds and are well known for their various health beneficial effects, which include antioxidative activities. Among them, the major anthocyanins isolated from seed coat of black soybean (Glycine max L.) were previously characterized as glycosides containing glucopyranose. Asthma is an allergic disease that is strongly associated with various immune cells, including basophils and mast cells. Eosinophils, basophils, and mast cells play important roles in allergic asthma through the release of inflammatory mediators such as asthma-specific T-helper 2 (Th2) cytokines and subsequent amplification of asthma symptoms via degranulation. Rat basophilic leukemia RBL-2H3 cells are the most common in vitro models for evaluating allergic reactions. In this study, we examined the effects of anthocyanin from seed coat of black soybean on antigen-stimulated degranulation and Th2 cytokine production in RBL-2H3 cells. Cell degranulation was evaluated by measuring the release of β-hexosaminidase. β-Hexosaminidase release and Th2 cytokine production in RBL-2H3 cells was much higher upon stimulation with IgE-antigen complex than those in untreated control cells. Anthocyanins significantly suppressed IgE-antigen complex-induced degranulation of RBL-2H3 cells and inhibited IgE-antigen complex-mediated interleukin (IL)-4, IL-13, and tumor necrosis factor α (TNF-α) production in RBL-2H3 cells. These findings suggest that anthocyanins from seed coat of black soybean effectively inhibit allergic reactions and may have beneficial effects against allergic asthma.

        인간 복막 중피 세포의 Transforming Growth Factor-β1(TGF-β1) 합성에 관한 연구

        Objective:to investigate the effect of high glucose and spent peritoneal dialysate on the TGF-β1 synthesis of cultured human peritoneal MC(HPMC); to examine the effect of costimulation with high glucose or dialysate and cytokines, interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α), on transforming growth factor(TGF-β1) synthesis of HPMC. Design:HPMCs were exposed to different concentrations of glucose(30, 60 & 90 mM/L) or spent peritoneal dialysate for 48 hours in the absence or presence of IL-1β(1ng/ml) and TNF-α(1ng/ml). TGF-β1 mRNA expression was assessed by Northern blot analysis and TGF-β1 protein synthesis and release by Western blot analysis with immunoprecipitation. Results:Exposure of MC to high glucose condition(30mM, 60mM & 90mM of D- glucose) induced 2.3-, 3.6- and 4.0-fold increases in TGF-β1 mRNA expression of MC with enhanced TGF-β1 protein synthesis and secretion into the media. Incubation with spent dialysate also significantly increased TGF-β1 mRNA expression & protein pared to control media(P$lt;0.05) Stimulation with IL-1β(1ng/ml) or TNF-α(1ng/ml) significantly increased TGF-β1 mRNA expression after 48 hours above the control level by 2.7-fold and 2.1-fold, respectively. However, TNF-α-induced increase in TGF-β1 mRNA expression was not translated into TGF-β1 protein secretion whereas IL-1β stimulation induced a significant increase in TGF-β1 protein secretion as well as TGF-β1 mRNA expression. Combined stimulation of high glucose or spent dialysate together with IL-1β or TNF-α showed a greater increase in TGF-β1 mRNA expression and protein secretion compared to stimulation with high glucose or spent dialysate alone. Conclusion:Our results clearly show that high glucose concentration of peritoneal dialysate and spent dialysate themselves might be sufficient to stimulate the production of TGF-β1 by peritoneal mesothelial cell. This state of chronic induction of TGF-β1 is further exaggerated in the presence of peritonitis because of stimulatory effect of proinflines, resulting in the augmented TGF-β1 synthesis, thus promoting peritoneal fibrosis.

      • Aberrant expression of interleukin-10 and activation-induced cytidine deaminase in B cells from patients with Behçet's disease

        <P>Despite extensive studies, the pathogenesis of Behçet's disease (BD) remains unclear. In particular, the roles of B cells in patients with BD have not been elucidated. Activation-induced cytidine deaminase (AID) is a critical enzyme for immunoglobulin (Ig) heavy chain class switching and somatic hypermutation in B cells and the abnormal expression of AID in various immune conditions has previously been studied. B10 cells, an interleukin (IL)-10-secreting subset of regulatory B cells, function to downregulate inflammation and autoimmunity. Thus, in the present study, the relevance of B cells in patients with BD was investigated. The plasma levels of IL-10 and IgA and the proportions of cluster of differentiation (CD)43<SUP>+</SUP> B cells, excluding naïve B cells, were measured in 16 patients with BD and 16 age- and sex-matched healthy controls (HCs). Additionally, the mRNA levels of IL-10 and AID were assessed in B cells from fresh peripheral blood samples of the BD patients and HCs. The plasma level of IL-10 in patients with BD did not differ significantly from that in HCs. Similarly, there was no significant difference in the plasma level of IgA, although a slight increase was observed in patients with BD compared with that in HCs. There were no differences in CD43<SUP>+</SUP>CD19<SUP>+</SUP> B cell numbers between patients with BD and HCs. However, IL-10 mRNA levels were significantly reduced (P<0.05), while AID mRNA levels were significantly increased (P<0.01) in the B cells of patients with BD compared with those in HCs. These results provide insight into the role of B cells in patients with BD.</P>

      • 게이트 산화전 RCA세정 횟수에 따른 Si/SiO₂계의 절연특성에 관한 연구

        The purity of wafer surface is an essential requisite for the successful fabrication of VLSI silicon circuits. The solution used in the cleaning step of silicon wafer surfaces known as "RCA standard clean" has the volume ratios NH₄OH : H₂O₂: H₂O=1:1:5. This RCA cleaning process for bare wafer can affect the state of wafer surfaces. This can cause the degeneration of gate oxide layer. For the investigation of this problem, we measured the Dit(interface trap density) of Si/SiO₂system using the C-V method. The Dit is in the range of 0.89∼2.13×10¹¹[cm-²eV-¹]. This measure is increased proportionally to the number of cleaning times. Leakage current is in the range of 1×10-¹∼1×10(-8)[A] according to the cleaning times, especially, we notice that additional RCA cleaning increases the leakage current in the order of hundred times compared to one times cleaning under 10[V] gate voltage. The electrical breakdown field using the breakdown obtained by I-V method is in the range of 8~12[MV/cm]. This measure is decreased proportionally to the cleaning times. This increment of leakage current by additional RCA cleaning result in the 0.8% decrement of yield. Since cleaning process of before gate oxidation directly affect the device performance and reliability, we think that further investigation is necessary for the clear understanding and of cleaning process.

      • 말라리아 환자 32예의 임상적 고찰

        목적 : 삼일열 말라리아는 국내에서 1986년 이후에는 박멸된 것으로 알려졌으나 1993년 이후 증가하는 양상을 보이고 있다. 이에 말라리아의 임상적 고찰을 통하여 이 질환을 이해함에 있다. 방법 : 1991년 1월부터 1999년 10월까지 반복적 고열과 오한을 주소로 본 병원에 내원하여 말초혈액 도말검사에 의해 말라리아 감염으로 확진된 32예의 환자에 대해서 후향적으로 조사하였다. 결과 : 1991년부터 1999년 10월까지 총 32명의 환자의 연도별 발생빈도는 각각 1991년에 1명, 1996년에 1명, 1997년에 3명, 1998년에 13명, 1999년에 14명이었다. 월별발생빈도는 7월과 8월에 집중적으로 발생하였다.(46.9%) 환자의 거주지는 경기 연천과 파주에 주로 분포되었다(53.1%). 성별발생분포는 32예의 환자중에 여자가 5명, 남자가 27명으로 남자가 더 많았다. 발병연령은 22세에서 87세까지 분포되었고 20-30대에 많이 발생하였으며 20대가 가장 많았다. 군인 및 제대자가 17명(59.4%)에 해당하였으며, 제대자의 평균 제대후의 기간은 10개월 정도였다. 가장 흔한 임상증상은 발열 및 오한이었다.(100%) 비장종대 및 간장종대는 각각 26명(81.3%), 3명(9.4%)에서 관찰되었다. 검사실 소견은 평균적으로 WBC 5.317/㎕, 혈색소 12.5±7.1g/㎗, 혈소판 78,906.3±68391.4㎕, GOT/GPT 43.7±26.7/49.6±57.3 IU/ℓ, Total bilirubin 1.6±0.9㎎/㎗를 보였다. 말초혈액 도말검사상 모두 P. vivax였다. 치료는 hydroxychloroquine과 primaquine의 치료에 잘 반응하였다. 결론 : 말라리아는 7월과 8월에 한국에서 호발하는 중요한 급성 열성 질환으로 조기 진단 및 치료에 보다 관심을 기울여야 할 것이다. Background : In Korea malaria has been rare, but since reemergence of indigenous vivax malaria in 1993, the number of cases of malaria have recently increased. We analyzed the cases of malaria who were treated in Kyung-Hee University College of Medicine with regard to define the clinical characteristics of malaria. Methods : We retrospectively analyzed of 32 cases of malaria admitted to Kyung-Hee University Hospital between 1991 and 1999. Malaria was diagnosed by peripheral blood smear. Results : Among a total of 32 cases, the number of yearly incidence of 1991, 1996. 1997, 1998 and 1999 were 1,1,3,13,14 respectively. It demonstrated the high incidence especially between July and August. The most patients (53.1%) resided in Yeonchon and Pajoo. Among them, 5 cases were female and 27 cases were male. The age distribution ranged from 22 to 76 year old. The high incidence of age was between 20's and 30's. Military solidiers and ex-soldiers are 17 cases(59.4 %). The most frequent symptom was fever and chills (100%). Splenomegaly and hepatomegaly were also observed in 81.3 %(26 of 32) and 9.4 %(3 of 32) respectively. The laboratory findings were WBC 5.317/㎕. Hb 12.5±2.1 g/dl. platelet 78,906±68,391.4/㎕. GOT/GPT 43.7±26.7/49.6±57.3 IU/L. LD 678.3±50.1 IU/L. total bilirubin 1.6±0.9mg/dl- The all cases were treated suscessfully with hydroxychloroquine and primaquine. There were no central nervous system complications and pulmonary edema. Conclusion : Malaria is one of the important acute febrile illness with the high incidence between July and August.
