        활락단(活絡丹)의 Adjuvant 유발 관절염 진행 억제 효과

        Objectives: This study was to investigate the antiarthritic effect of Hwallak-dan(Huoluo-dan) on the complete freund`s adjuvant(CFA)-induced arthritis in Lewis rats. Methods: Arthritis was induced by intradermal injection of CFA into base of tail. Experimental groups divided into normal(n=10), control(n=10) and treated(n=10) group. Animals of control group received normal saline for twenty days, treated group received extracts Hwallak-dan(Huoluo-dan) for same duration, light mineral oil only injected non-arthritic rats were served as normal group. The incidence of arthritis and arthritic index were observed after treatment. Body weight, paw edema volume and thickness of ankle joint were measured at 0, 11, 14, 17, 20 days after treatment. White blood cell(WBC) counts in blood were analysed at 20 days after treatment and tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), IL-6 contents in paw exudate were analysed by ELISA at 20 days after treatment. Histopathology on the ankle joint were performed at 20 days after treatment. Results: 1. Incidence of arthritis of treated group was 60% and control group was 100% at 20 days after treatment. 2. Arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. 3. Paw edema volume and thickness of ankle joint of treated group was significantly decreased compared with control group at 20 days after treatment. 4. Total WBC of treated group was significantly decreased compared with control group at 20 days after treatment. 5. Neutrophils of treated group was significantly decreased compared with control group at 20 days after treatment. 6. TNF-α content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 7. IL-1β content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 8. IL-6 content content in paw exudate of treated group was significantly decreased compared with control group at 20 days after treatment. 9. Histopathological arthritic index of treated group was significantly decreased compared with control group at 20 days after treatment. Conclusions: These results indicate that Hwallak-dan(Huoluo-dan) has inhibitory effect on the development and progression of CFA-induced arthritis in rats.

        방사선조사와 저칼슘식이가 치아형성시 Interleukin-1의 분포에 미치는 영향에 관한 연구

        Purpose : To elucidate the effects of the irradiation and calcium-deficient diet on expression of interleukin (IL)-1 during tooth formation of rat molar Materials and Methods : The pregnant three-week-old Spague-Dawley rats were used for the study. The control group was non-irradiation/normal diet group, and the experimental groups were irradiation/normal diet group and irradiation/calcium-deficient diet group . The abdomen of the rats on the 9th day of pregnancy were irradiated with single dose of 350 cGy. The rat pups were sacrificed on the 14th day after delivery and the maxillae tooth germs were taken. The specimen were prepared to make sections for light microscopy, and some of tissue sections were stained immunohistochemically with anti-IL-1 antibody. Results : In the irradiation/normal diet group, dental follicle showed fewer blood vessels, mononuclear cells, and fusions of mononuclear cells than in non-irradiation/normal diet group. Alveolar bone showed a few osteoblasts and osteoclasts. Periodontal ligament showed collagen fibers and fibroblasts with irregularity. Weak immunoreactivity for IL-1 was shown in dental follicle, alveolar bone, and periodontal ligament, In the irradiation/calcium-deficient diet group, dental follicle showed sparse cellularity. Alveolar bone showed diminished number of osteoblasts. Periodontal ligament showed irregular collagen fibers and atrophy of cementoblasts and fibroblasts. No immunore-activity for IL-1 was shown in dental follicle, alveolar bone, and periodontal ligament. Conclusion : Irradiation and calcium-deficient diet seems to cause disturbance of the expression of interleukin-1 during tooth formation of rat molar.         Different shapes of Al<sub>2</sub>O<sub>3</sub> particles induce differential cytotoxicity via a mechanism involving lysosomal destabilization and reactive oxygen species generation

        The biological effects of nano- and micro-sized $Al_2O_3$ particles are hypothesized to differ according to the shapes as well as the sizes of the particles. Thus, the mechanisms of interleukin (IL)-$1{\beta}$ production and the association between the shape of the $Al_2O_3$ particle and its cytotoxicity in macrophage-like THP-1 cells were investigated using particles with three different shapes [$N-Al_2O_3$ (<30 nm), $S-Al_2O_3$ ($2-4nm{\times}100-800nm$), $L-Al_2O_3$ ($2-4nm{\times}2800nm$)]. Levels of IL-$1{\beta}$ production and cytotoxicity were concentration-dependent and were in the following order: $S-Al_2O_3$ > $N-Al_2O_3$ > $L-Al_2O_3$. Stimulus of THP-1 cells by $Al_2O_3$ particles led to lysosomal destabilization, generation of intracellular reactive oxygen species (ROS), and subsequent release of cathepsin B. The magnitude of the stimulus was dependent on the shapes and aspect ratios of the particles. Additional results suggested that caspase-1 (NALP3 inflammasome) activation and IL-$1{\beta}$ production followed cathepsin B release. In addition, the cell death induced by $Al_2O_3$ particles was closely related with cathepsin B leakage. The mode of cell death was necrotic as much as apoptotic. We conclude that $Al_2O_3$ particles induce different levels of IL-$1{\beta}$ production and cytotoxicity depending on their particle shapes or aspect ratios. The current finding may support the development of safe forms of $Al_2O_3$ particles.

        Tobacco Smoking Could Accentuate Epithelial-Mesenchymal Transition and Th2-Type Response in Patients With Chronic Rhinosinusitis With Nasal Polyps

        Tobacco smoking (TS) has been known as one of the most potent risk factors for airway inflammatory diseases. However, there has been a paucity of information regarding the immunologic alteration mediated by TS in patients with chronic rhinosinusitis with nasal polyps (CRSwNP). To identify the effect of TS, we harvested human tissue samples (never smoker: n=41, current smoker: n=22, quitter: n=23) and analyzed the expression of epithelial-derived cytokines (EDCs) such as IL-25, IL-33, and thymic stromal lymphopoietin. The expressions of Th2 cytokines and total serum IgE showed a type-2 inflammatory alteration by TS. In addition, the epithelial marker E-cadherin and epithelial-mesenchymal transition (EMT)-associated markers (N-cadherin, α-SMA, and vimentin) were evaluated. Histological analysis showed that EDC expressions were upregulated in the current smoker group and downregulated in the quitter group. These expression patterns were consistent with mRNA and protein expression levels. We also found that the local Th2 cytokine expression and IgE class switching, as well as serum IgE levels, were elevated in the current smoker group and showed normal levels in the quitter group. Furthermore, the expressions of E-cadherin decreased while those of N-cadherin, α-SMA, and vimentin increased in the current smoker group compared those in the never smoker group. Taken together, these results indicate that TS contributes to the deterioration of pathogenesis by releasing local EDCs and Th2 cytokines, resulting in EMT in patients with CRSwNP. We verified that alterations of immunological response by TS in sinonasal epithelium can play a vital role in leading to CRSwNP.

      • 비용종 상피세포와 섬유아세포 동시배양 모델에서 시토카인의 발현

        Background and Objectives: Nasal polyposis is a chronic inflammatory disease characterized histologically by edematous fluid with sparse fibrous cells, few mucous glands and proliferation of stromal and epithelial cells. There have been many studies done separately about epithelial cells, fibroblasts, and other inflammatory cells in nasal polyps. However, there is no usable study model for the cell-to-cell interaction. Therefore, we tried to make a new model for the study of nasal polyp by coculture of epithelial cells and fibroblasts. Subjects and Method: Nasal polyp was collected during endoscopic sinus surgery. Almost half of them were used for epithelial cell culture and the remaining portion was used for fibroblast culture in the same polyp. Using a transwell insert fibroblasts were placed in the bottom and epithelial cells were placed on the insert respectively. RNA were isolated both from cpithelial cells and fibroblasts of 12,24, and 48 hours' coculture. The expressions of IL-8,IL-6, and eotaxin from coculturcd cells were compared with cells that were cultured alone by semiquantitative RT-PCR. Results: IL-8 was highly expressed in fibroblasts than in epithelial cells, especially in cocultured cells. The expression of IL-6 in fibroblasts was much higher than in epithelial cells, especially in cocultured fibroblasts. Eotaxin is weakly expressed in fibroblasts but not in epithelial cells. Conclusion : These findings indicate that cocultured cells show different expression of the mRNA of various cytokines compared with cultured cells alone. And this coculture system may be more ideal than the mono-cell culture system for the study of cell-to-cell interactions and to reveal the pathogenesis of oasal polyposis.

        가미소염탕(加味消炎湯)이 Monosodium Iodoacetate로 유발된 흰쥐의 골관절염에 미치는 영향

        Objectives : This study was to investigate the effects of Gamisoyeoum-tang(Jiaweixiaoyan-tang) on the monosodium iodoacetate-induced osteoarthritis in rats. Methods : Monosodium iodoacetate induced arthritic rats were divided into control and treated group. Control group was taken distilled water for 20 days. Treated group was taken extracts of Gamisoyeoum-tang(Jiaweixiaoyan-tang) by orally for same duration. Normal group was injected with normal saline and was taken distilled water. Body weights were measured at 0, 5, 10, 15, 20 days after injection. At the end of experiment, gross and histopathological examination on the articular cartilages of the knee joints were performed. Contents of TNF-α, IL-1β, IL-6 in synovial fluids and proteoglycan contents of articular cartilages were analysed. COX-2 and iNOS immunohistochemical examination on the knee joints were performed. Results : 1. Body weights of the treated group were significantly increased compared with control group at 20 days after injection. 2. Grossly, the severity of osteoarthritis in the treated group were alleviated compared with control group. 3. PG contents in articular cartilages of the treated group were significantly increased compared with control group. 4. Histopathologically, degenerative and necrotic lesion of articular cartilages in the treated group were alleviated compared with those of the control group. 5. IL-1β contents in synovial fluids of the treated group were significantly decreased compared with control group. 6. Positive reactions of COX-2 in chondrocytes and synovial membranes of the treated group were decreased compared with the control group. Conclusions : On the basis of these results, we concluded that Gamisoyeoum-tang(Jiaweixiaoyan-tang) has anti-arthritic effects on the monosodium iodoacetate-induced osteoarthritis in rats. And it`s effects were related with reduced secretion of IL-1β and COX-2 from osteoarthritic chondrocytes and synovial membranes.

        Objectives : This study was to investigate the effects of GCP treatment on the monosodium iodoacetate-induced osteoarthritis in rats. Methods : Arthritis was induced by injection of Monosodium Iodoacetate(0.5mg) into knee joints of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. Control group was taken distilled water for 20 days. Treated group was taken extracts of GCP by oraly for same duration. Normal group(n=8) was injected with normal saline and was taken distilled water for 20 days. Body weight was measured at 0, 5, 10, 15, 20 days after injection. Macroscopic examination and histopathological study on articular cartilage of knee joint were operated at 20 days after injection. Proteoglycan(PG) content of articular cartilages of knee joint was represented by safranine O staining, was measured at 20 days after injection. Tumor necrosis factor-α, Interleukin-1β, Interleukin-6 in synovial fluid were measured with ELISA kit at 20 days after injection. Immunohistochemical staining of COX-2, iNOS in knee joints were observed at 20 days after injection. Results : 1. Body weight of the treated group increased compare with control group at 20 days after injection. 2. Macroscopically, degree of osteoarthritis in the treated group were evaluated compared with the control group. 3. PG content in articular cartilage of the treated group was significantly increased compared with the control group. 4. Histopathologically, osteoarthritic scores of the treated group was significantly decreased compared with the control group. 5. TNF-α content in synovial fluid of the treated group was decreased compared with the control group. 6. IL-1β content in synovial fluid of the treated group was significantly decreased compared with the control group. 7. Positive reaction of COX-2 in chondrocytes and synovial membrane of the treated group was faint compared with the control group. Conclusions : On the basis of these results, we concluded that GCP has inhibiting effects on the IL-1β and COX-2 secretion of chondrocytes and synovial membrane in Monosodium Iodoacetate-Induced osteoarthritis model of rats.
