        폐포 대식세포 및 단핵구가 Interleukin-2 Enhanced Natural Killer 및 LAK Activity에 미치는 영향

        저자들은 폐포 대식세포 및 말초혈액내의 단핵구가 NK 활성도 및 LAK 활성도에 미치는 영향을 보기위하여, 임파구에 여러 가지 농도(0, 100 : 1, 10 : 1, 1 : 1)의 폐포 대식세포와 단핵구를 넣어 IL-2 enhanced NK 활성도 및 LAK 활성도를 비교하여 다음과 같은 결과를 얻었다. 1) 여러 가지 농도의 단해구는 IL-2 enchanced NK 활성도 및 LAK 활성도에 영향을 미치지 않았다. 2) 동량의 페포대식세포(임파구 : 폐포 대식세포= 1 : 1)는 IL-2 enhanced NK 활성도를 의의있게 억제하였으나(p<0.05), 소량의 폐포대식세포(임파구 : 폐포 대식세포-10 : 1과 100 : 1)는 IL-2 enhanced NK 활성도를 억제하지 못하였다. 3) 임팍와 폐포 대식세포의 비율이 1 : 1과 10 : 1에서는 LAK 활성도를 의의있게 억제하였으나, 소량의 폐포대식세포(임파구 : 폐포 대식세포=100 : 1)는 LAK 활성도를 억제하지 못하였다(p<0.05). 이상의 결과로 IL-2 enhanced NK 활성도 및 LAK 활성도는 폐포 대식세포의 양에 비례하여 억제되었으나, 말초혈액내의 단핵구에 의해서는 영향받지 않는 것을 알 수 있었다. Alveolar macrophages (AM) are thought to function as primary effector cells against tumors growing in the lung. Systemic administration of lymphokine activated killer (LAK) cells and IL-2 resulted in partial antitumor response in patients with advanced cancer. LAK activity is influenced by various factors. We studied the effects of AM and blood monocytes from healthy donors on IL-2 enhanced NK activity against K-562 cells and LAK activity against Raji cells utilizing a 4h ^(51)Cr release assay. The following results were obtained: 1) The addition of different doses of human blood monocytes showed no suppression or enhancement of IL-2 enhanced NK and LAK activity. 2) The addition of high dose of AM (Lymphocyte: AM=1:1) significantly suppressed IL-2 enhanced NK activity. Smaller doses of AM (Lymphocyte: AM= 10:1and 100:1) did not suppress IL-2 enhanced NK activity. 3) The addition of high dose of AM (Lymphocyte: AM = 1:1 and 10:1) significantly suppressed LAK activity. The smallest dose of AM (Lymphocyte: AM= 100:1) did not suppress LAK activity. In conclusion, IL-2 enhanced NK and LAK activity were dose-dependently suppressed by human alveolar macrophages. However IL-2 enhanced NK and LAK activity were not suppressed by blood monocytes.

        Triptolide suppresses interleukin-1beta-induced human beta-defensin-2 mRNA expression through inhibition of transcriptional activation of NF-kappaB in A549 cells.

        <P>The immunosuppressive effect of triptolide has been associated with suppression of T-cell activation. However, the immunosuppressive effects of triptolide on innate immunity in the epithelial barrier remain to be elucidated. Human beta-defensin (HBD)-2 is an inducible antimicrobial peptide and plays an important role in the innate immunity. We have previously demonstrated that IL-1beta induced HBD-2 mRNA expression in A549 cells through activation of nuclear factor-kappaB (NF-kappaB) transcriptional factor as well as p38 mitogen-activated protein kinase (MAPK), c-Jun N-terminal kinase (JNK), or phosphatidylinositol-3-kinase (PI3K). In this study, we investigated effects of triptolide on IL-1beta-induced HBD-2 mRNA expression in A549 cells. Triptolide inhibited IL-1beta-induced HBD-2 mRNA expression in a dose-dependent manner. Addition of triptolide did not suppress activation of p38 MAPK, JNK, or PI3K in response to IL-1beta. Triptolide inhibited IL-1beta-induced MAPK phosphatase-1 expression at the transcriptional level and resulted in sustained phosphorylation of JNK or p38 MAPK, explaining the little effect of triptolide on IL-1beta-induced phosphorylation of these kinases. Although triptolide partially suppressed IL-1beta-mediated degradation of IkappaB-alpha and nuclear translocation of p65 NF-kappaB, triptolide potently inhibited NF-kappaB promoter-driven luciferase activity in A549 cells. These results collectively suggest that the inhibitory effect of triptolide on IL-1beta-induced HBD-2 mRNA expression in A549 cells seems to be at least in part mediated through nuclear inhibition of NF-kappaB transcriptional activity, but not inhibition of p38 MAPK, JNK, or PI3K. This inhibition may explain the ability of triptolide to diminish innate immune response.</P>

      • 웹 브라우져를 통한 서버 클라이언트 환경의 영상처리 실습 시스템 설계

        웹 기반의 환경은 인터넷과 컴퓨터의 접목으로 실생활에서 연구활동에 이르기까지 다양하게 그 효용성의 범위를 굳혀가고 있다. 교육기관에서 활용하는 가상강좌는 이러한 웹의 장점을 활용한 학습방법의 다양화의 결과이다. 본 논문에서 설계한 웹 브라우져를 통한 서버 클라이언트 환경의 영상처리 실습 시스템은 가상강좌나 웹 환경이 구축된 장소에서 교육자와 피교육자사이의 교육효과를 증진시키도록 상호 의사전달의 효율성을 가지게 하는 시스템이다. 각 클라이언트가 다운로드 받은 컴포넌트를 실행하여 구동하게 되면 클라이언트들이 자신의 실습을 위한 프로그램을 로컬에서 가동할 수 있도록 하였기 때문에, 서버측의 부하가 얼어서,실습 결과영상을 로컬 클라이언트에서 부하시간 없이 얻을 수 있게 하였다. 또한 서버의 수정 혹은 갱신으로 바뀌어진 컴포넌트는 클라이언트 측에서 접속시 업데이트가 가능하므로 각 클라이언트에 대한 업 데이트를 따로 고려하지 않아도 되는 장점이 있다. 또한 컴포넌트가 가지는 여러 가지 메쏘드들은 다양한 영상처리의 결과를 볼 수 있게 해 줌으로써 고가의 소프트웨어를 따로 구입하지 않아도 되는 장점을 가지게 된다. The environment of web is applied variously from our life to research activities thanks to the internet and the computer. The virtual lecture utilized by an educational institution is the result of application of a learning system with the advantage of web. In this paper, a design of the image processing training system at the environment of server-client through web browser is the system of high degree of efficiency at the point of communication between lecturer and student. A client can get the result image from another client without load balancing of the server, because the processer for image processing is operated at the container of a client after downloaded as the form of a component. Clients can get the new version of component when they contact, so clients do not need to consider the status of updated component locating in the server. Furthermore, the client can handle an image with the many operations included in component with no purchase of the software additionally, that is another merits of this system.

      • 노인근로자를 위한 고용친화적 제도

        현대 사회가 고령화 현상을 겪음에 따라 크게 관심을 가져야 하는 것이 노인의 노동시장 참여이다. 하지만 현재의 노동 현실은 노인 근로자에게 불리하게 나가고 있다. 그러나 현재의 나이 차별에 대한 뿌리는 현실적인 경제적 고려에 있기보다는 경영진이 노인 근로자에 대해 밑바탕에서 인식하는 부정적 편견에 있는 경우도 많다. 이런 맥락에서 본 글에서는 노인에 대한 고용 여건과 노인 근로자의 고용에 대한 인식을 살펴보고, 노인 근로자가 퇴직 전과 퇴직 후에 경제활동에 참여하는데 도움이 되는 산업복지 관련 프로그램을 살펴본다. With the coming of ageing society in advanced countries and Korea, broad attention has been increasingly paid to participation of older workers in the labor market. Yet thus far the prospect for employability has not been good for older people. To a large extent, it seems that the root of current age discrimination lies not in the practical economic consideration but in the employers' negative streotypes on older workers. Therefore this study discusses the employment status of older workers and the employers' perception on them. Then some programs and schemes which can be useful in industrial welfare for the promotion of employment of older workers before and after retirement are suggested.

      • AISI 316 스테인리스강의 크리프-피로 상호작용시 수명예측에 관한 연구

        The effect of tensile hold time on the creep-fatigue interaction in AISI 316 stainless steel was investigated. To study the fatigue characteristics of the material, strain controlled low cycle fatigue(LCF) tests were carried out under the continuous triangular waveshape with three different total strain ranges of 1.0%, 1.5%, and 2.0%. To study the creep-fatigue interaction, 5 min., 10 min., and 30 min. of tensile hold times were applied to the continuous triangular waveshape with the same three total strain ranges. The creep-fatigue life was found to be the longest when the 5 min. tensile hold time was applied and was the shortest when the 30 min. tensile hold time was applied. The cause for the shortest creep-fatigue life under the 30 min. tensile hold time is believed to be the effect of the increased creep damage per cycle as the hold time increases. Under the total strain range of 1%, significant reduction in life was observed as the tensile hold time was increased compared to the result of 2% total strain range. The theoretical prediction obtained by the Modified Coffin-Manson method and the experimental results of creep-fatigue life were found to be in resonable agreement with each other.

      • 기-액병류상향 고정층반응기에서 액체의 흐름과 축방향분산

        For upward cocurrent gas-liquid flow in a fixed bed reactor, the flow and axial dispersion of liquid were investigated in an air-water-Zeolite particle system. Based on experimental residence time distributions, parameters of mixing models were evaluated with two proposed liquid mixing models: axial dispersion model with stagnant zone(DSM) and plug flow model with stagnant zone(PFM). Accordingly, the applicability and parameters of each mixing model were analyzed for various operating conditions. We found that the flow and axial mixing characteristics of liquid could be analyzed well by DSM and parameters were well estimated by curve fitting using trial and error method. Also, the fraction of stagnant zone was decreased with increasing liquid velocity, while the dimensionless mass transfer coefficient between dynamic and stagnant zones was increased, and these parameters estimated by DSM were lower than those by PFM. In dynamic zone, calculated Peclet number were increased with increasing liquid velocity and axial mixing characteristics of liquid tended to access to plug flow for the high liquid Reynolds number(about Re1>50). For the porous catalyst particle system the flow and axial mixing characteristics of liquid had nearly same tendency that for the nonporous particle(glass bead) system, while dimensionless mass transfer coefficient was lower value than that for it. In these conditions, correlations of these parameters were obtained for various operating conditions.

      • 유지시간 변화에 따른 크리프-피로 수명예측 방법에 관한 연구

        In this study, creep-fatigue tests were carried out under the various test conditions using AISI 316 stainless steel. Life prediction was made by three different methods which are the modified Coffin-Manson method, the modified Ostergen method and the artificial neural network method with the adaptive learning rate. The predicted lives were compared with the experimental results and following conclusions were obtained within the scope of this study. Creep-fatigue life prediction by the modified Coffin-Manson method and the modified Ostegren method had the average error of 47.3% and 50.8% respectively whereas the artificial neural network method had only 28.8% for compressive and tensile+compressive hold time. From the obtained results, the artificial neural network method with the adaptive learning rate was proved to be more accurate and effective than the other two methods. The validity of artificial neural network method for life prediction checked with 3 verification data points proved also to be very satisfactory.

      • 오이흰가루병의 환경보전형 방제법 개발

        Potassium silicate, 광합성 세균, 그리고 몇가지 전착제들을 수용액 상태로 만들어 오이 잎에 1주 간격으로 4회 분무접종하며 병 발생 조사를 한 결과, 처리 초기 부터 오이 잎당 흰가루병균의 균총수와 병반면적지수가 크게 억제되었다. 조사 종료 시점에서 무처리구는 오이 잎당 균총수가 70개 이상, 병반면적은 30% 이상으로 병이 심하게 발생한 반면, 균총 수와 병반면적 모두 80% 이사의 억제율을 보인 처리는 potassium silicate 8mM과 16mM, 광합성 세균, 그리고 전착제 카바와 Tween 20이었다. 모든 처리에서 병반면적지수 억제율과 균총 수 억제율은 거의 비례하였다. 처리구 중 광합성 세균을 제외하고는 모두 phytotoxicity를 일으키지 않았지만, 광합성 세균은 잎의 가장자리 근처가 약간 쭈그러들며 충분히 전개하지 못하는 가벼운 phytotoxicity를 유발하였다. Potassium silicate와 Tween 20은 오이 흰가루병의 환경보전형 방제제로서의 가능성이 매우 큰 것으로 나타났다.
