        In present study, we tested antifungal effects of 11 different kinds of compounds isolated from Phellodendri Cortex (PC), Lonicerae Flos (LF), Lonicerae Folium et Caulis (LFC) and Araliae Continental is Radix (ACR). These herbal medicines are generally applied for the treatment of inflammations. Prior to the test, antifungal effect of 70%-ethanol extracts of the medicines was examined. Results showed that the three extracts of PC, LF and LFC, but not ACR extract, inhibited Candida albicans growth, in-vitro. Among the 11 compounds, berberine and palmatine isolated from PC showed the most antifungal activity (P < 0.05). Lonicerin and loganin from LF and chlorogenic acid from LFC also retarded growth of C. albicans at high dose of 250 ㎛/ml, but their activity was much less than berberine or palmatine. However, ent-Pimara-8(14),15-diene-19-oic acid and 16α,17- Dihydroxy-ent-kauran-19-oic acid from ACR had no antifungal activity as expected. All together, these data imply that most of oriental herbal medicines having anti-inflammatory activity could have anticandidal effect, suggesting that PC, LF and LFC be effective for the treatment of septic arthritis by killing of C. albicans.

      • 3大營養素가 흰쥐 十二指腸의 Goblet Cell에 미치는 影響

        In our experiments we made use of male and female Swiss albino rats weighing about 60gm each for the sake of investigating the duodenal goblet cells which mostly bring about secretory work. The mice fed on three unbalanced diets consisting mostly of carbohydrate, protein and fat were the chief objects of study. on the 20th day after feeding, and after one or three-day fasting, Specimens were taken from the upper, middle and lower portions of the duodenum. tne specimens were fixed in 10% formalin and made into paraffin sections of 5μ and stainings were done with Hematoxylin-eosin. all of the goblet cells in each 20 crypts and villi, baths of which were cut in the axial directions were counted and averaged numbers were considered as total relative number of one animal. The results were gained as follows: 1. The relative number of the goblet cells in the duodenum of the mice fed 'on normal food averaged 22.5. these cells were found more in the villi than in the crypts. 2. The relative number of the goblet cells in the mice fed on fat was larger than the normal value, and was tne largest in the three groups fed on unbalanced diets. 3. The protein diet group was the smallest value among three groups. 4. The group's goblet cells fasting 1-day increased a little in number relative to the normal. 5. The group's goblet cells fasting 3-days increased the most in number of all groups.

      • 치은섬유모세포 및 치주인대세포에 의한 교원질 수축에 관한 연구

        The purpose of this in vitro study was to evaluate contraction of the human gingival and periodontal ligament fibroblast in the collagen matrix. The cell cultures were prepared using human gingiva and tooth. The collagen gels were observed daily with an inverted microscope and their area was determined by measuring the diameters of the collagen gel. The configurations of human gingival and periodontal ligament fibroblasts were done by SEM observation. The results as follows; 1. The contraction was found to be proportional to seeding cell without seeding cells, the contraction was not occured. 2. Periodontal ligament fibroblast was shown more fast contraction than gingival fibroblast. 3. The collagen matrix was translucent initially, but with fully contraction, the structure become opaque. 4. The collagen matrix with fibroblast were aggregated to form bundles and reorganized collagen fiber.

      • 치아재식후 치유과정에서 fibronectin의 분포에 관한 면?조직화학적 연구

        Periodontalhealing requires the formation of new periodontal tissues comprised of various extracellular components. The present study investigated the formation and distribution of the fibronectin FN), a major adhesive glycoprotein. Rats weighing 130`150 gm were fed 0.4% R -aminopropionitrile for 5 days to achieve gentle tooth extraction. The maxillary first molars were extracted under anesthesia with pentobarbital, washed in sterile distilled water, treated with bacterial collagenase to digest collagen fibers on the root surfaces. After washing in water overnight, the mesial root surfaces were demineralized by application of citric acid, washed, dried and stored at 4 t. Immediately after tooth extraction and bleeding control, the treated molars extracted previously from other rats were reimplanted. Ten animals each were sacrificed 45 min, 1, 3, 6, 10 days after reimplantation of teeth by intracardiac perfusion with 4% paraformaldehyde. The replanted molars and surrounding tissues were cut, demineralized, dehydrated and embedded in paraffin. Imrnunostaining of the FN was achieved by the avidin-biotin complex method. The results as follows; 1. The control group showed gradual healing from 1 day and complete regeneration of the periodontium at 10 days. 2. At early healing stage, intense staining for FN was observed in the newly formed connective tissues, granulation tissue, especially around blood vessels as well as inflammatory cells. 3. At later healing stage, FN was expressed later in the periodontal ligament, cementum, osteoid, and newly formed connective tissue and its attachment site to the root surface. 4. Strong immunolabeling for FN was found in cementoblasts, osteoblasts, and new bone and cementum, which weaken staining was observed on the dentin and alveolar bone. 5. These results suggest that the healing after replantation is achieved through a series of cell differentiation and formation of FN.

      • 발치와의 치유과정에서 교원질 및 비교원단백질의 분포에 관한 면역조직화학적 연구

        During tooth socket healing, the coagulum in the socket is replaced by fibrous connective tissue which undergoes mineralization and eventually becomes bone. Using this model, the healing process of the tooth socket and the role of type I collagen(CI), fibronectin(FN), bone sialoprotein(BSP), and osteopontin(OP) in the process were studied. Thirty Sprague-Dawley rats weighing 130-150gm were fed fi -aminopropionitcille for 5days before extraction of the first maxillary molars, and sacrified by perfusion with 4% paraformaldehyde at 1, 3, 5, 7, and 10days after tooth extraction. The socket and surrounding tissues were cut, demineralized, dehydrated and embedded in paraffin. For morphological observation, the sections were stained by Azan. Immunostaining of the ECM components was achieved by the avidin-biotin complex method. The results as follows; 1. Morphology of the tooth socket 1day after tooth extraction. The socket was filled with blood coagulum which was composed of densly aggregated red blood cells, platelets and fibrin network. 2. Morphology of the tooth socket 3days after tooth exyraction. A large portion of the coagulum was replaced by fibrous connective tissue contains a large number of fibroblasts, come from periphery of the socket. 3. At 5days, the newly formed bone demonstrated the highest proliferation activity. At 7days, the soclet was occupied with new bone. 4. Type I collagen was observed in the newly formed connective tissue and around the new bone. fiber. 5. Intensive staining for fibronectin was observed in granulation tissue, especially around blood vessels as well as inflammatory cells. 6. Strong immunostaining for bone sialoprotein and osteopontin was found in osteoblasts and new bone, while weaker staining was observed on dense connective tissue. 7. These data suggest that collagen and noncollagen proteins (FN, BSP, OP) an important role daring socket healing.

      • Chlorogenic acid Rutin의 Macrophage 및 T-임파구 증식에 대한 효과

        In the present study, we examined immunoregulatory activitres of cholrogenic acid (CA) and rutin that are isolated from Lonicera Flos (A family of Caprifoliaceae). For the examination, NO production of LPS-treated RAW264.7 monocyte/macrophage cell line and effect on the T-cells proliferation were determined. Results showed that CA inhibited NO production at low concentrations below 10 ㎛/ml, but at higher concentration ranged from 10 ㎛/ml to 40 ㎛/ml the inhibitory activity was lost. However, rutin blocked the NO production in a dose-dependancy. In case of T-cell proliferation, both of the compounds suppressed the proliferation (P < 0.05). Combined together, it is assumed that the compounds might be effective to arthritic inflammation, possibly, by removal of CD4+ T-cells, but rutin might be more effective against the inflammation because of the blockage of NO production from macro phages which is responsible for the pain during the process of arthritis.

        실험연구 : Methylmethacrylate Monomer의 흰쥐 기관 평활근 이완 효과

        Background: Methylmethacrylate monomer (MN) bone cement is commonly employed in orthopedic procedures, particularly total hip and knee replacement, to anchor prosthetic devices to bone. Numerous cardiopulmonary complications can occur just after injection of MN. And MN produces direct relaxation of vascular smooth muscle in vitro. The purpose of this study was to determine if MN could have relaxation effect in tracheal smooth muscle too. Methods: Each ring of rat trachea was suspended on wire supports in a bath with Tris Tyrode solution. Dose response curves of MN were recorded after contraction of tracheal ring with acethylcholine (Ach) 10-5 M or cabachol (Cch) 10-8 M. MN was administered in denuded tracheal rings and compared it`s effect with intact tracheal rings to see the effect of epithelium for contraction. And MN dose response curves were recorded after pretreatment of nitric oxide synthase inactivator (L-NAME), muscarinic receptor blocker (atropine), beta-adrenaline receptor blocker (propranolol), adenylyl cyclase inhibitor (SQ22536) respectively. The effects of MN on cellular Ca2+ and K+ migration in rat tracheal preparations were studied. Results: MN significantly inhibited acetylcholine or carbachol induced contractions of tracheal rings dose-dependently (P < 0.05). This relaxation effect of MN was not recovered in denuded tracheal rings. And pretreatment with L-NAME, propranolol, atropine, SQ22536 or tetraethylammonium respectively did not recover the relaxation effect of MN. MN inhibited both intracellular calcium release and extracelluar calcium influx. Conclusions: The relaxation effects of MN on rat tracheal rings are not related with epithelium, nitric oxide, muscarinic, or beta-adrenergic receptor. Methylmethacrylate monomer inhibits both intracellular calcium release and extracelluar calcium influx. (Korean J Anesthesiol 2007; 52: 194~201)

      • 경요도 절제술을 시행받는 환자에서 초음파기를 이용한 폐쇄신경 차단 - 서혜부 주름부위 접근법

        연구배경: 경요도절제술을 시행받을 환자 중에서 초음파기와 신경 자극기를 이용하여 폐쇄신경 차단 을 시행하는 것이 환자에게 안전하고 효과적인지 알 아보고자 한다. 방법: 척추 마취하 경요도절제술을 시행 받는 26 명의 환자들에게 초음파기와 신경 자극기를 사용하 여 폐쇄신경 차단을 시행하였다. 초음파로 해부학적 구조를 확인하고 근수축 확인 후 1% lidocaine 10 ml 를 폐쇄신경의 anterior와 posterior branch에 각각 투 여하였다. 초음파기를 이용하여 해부학적 위치감별 에 걸리는 시간, 전체 신경차단에 걸리는 시간, 바늘 깊이, 수술 중 근수축의 정도, 실패율, 합병증등을 평가하였다. 결과: 초음파기를 이용하여 fascial plane을 감별하 는 데 걸리는 시간은 21±14.9초, 폐쇄신경의 anterior branch와 posterior branch를 확인하는 데 걸리는 시간 은 각각 39.6±27.4와 24.3±11.7초였으며 전체 신경차 단에 걸리는 시간은 174±65.1초, 바늘 깊이는 anterior branch가 4.8±0.8 cm, posterior branch가 6.2±1.1 cm였 다. 실패율은 anterior branch가 2/26 (7.7%), posterior branch가 7/26 (26.9%)이었다. Grade 1의 근수축은 22/26 (84.7%)에서 나타났으며 폐쇄신경술의 성공률 은 23/26 (88.5%)이었다. 결론: 서혜부 위치에서 초음파로 신경의 위치를 확인하는 방법은 쉬우나 초음파와 신경 자극기를 사 용한 폐쇄신경 차단술의 성공률은 큰 차이가 없었다.
