      • 악성흑색종과 양성멜라닌세포성모반에서 bcl-2 단백발현에 관한 면역조직화학적 연구

        Background: The bcl-2 protein, contrast to p53, has been shown to be suppress the apoptosis, and overexpression of the bcl-2 protein has been reported in several malignant tumors. Although how the bcl-2 protein takes part in tumorigenesis has not been proved, the bcl-2 positive tumors appeared to have good prognosis in some malignant tumors. The expression pattern of the bcl-2 in melanocytic nevi and malignant melanoma have not been well known. The authors analyzed immunohistochemically expression of the bcl-2 protein to investigate the role of the bcl-2 protein in developing malignant melanoma. Method: 10 malignant melanoma and 10 benign melanocytic nevi were immunostained with anti-bcl-2 monoclonal antibody. Results: In malignant melanoma, 1 case was 2+ positive, 9 cases were 3+ positive. In benign melanocytic nevi, 2 cases were 2+ positive, 8 cases were 3+ positive. Conclusion: Bcl-2 protein is expressed by benign melanocytic nevi and malignant melanoma. There is no difference of bcl-2 protein expression in melanocytic nevi and malignant melanoma.

      • Streptococcus mutans 로 부터 분리된 mutan의 화학적 특성

        Streptococcus mutans produces water-soluble and water-insoluble α-glucan from sucrose by multiple glucosyltransferase (GTFs). Mutan, primarily water-insoluble α-glucan plays an important role in the development of dental plaque. The study was conducted to investigate the chemical properties of mutan purified from crude extracellular polysaccharide of S. mutans. In batch culture of S. mutans, the percent yield(%) of crude extracellular polysaccharide was calculated as 0.026%. Total carbohydrate and protein contents of crude polysaccharide showed 67.3 and 24.3%, respectively. The contents of hexosamine from extracellular polysaccharide extract were found to be 2.4%. Sugar composition analysis showed that mutan obtained from purification of crude polysaccharide is mainly composed of glucose with a slight amount of galactose. In an infrared absorption spectrum of the mutan, the absorption peak in the range of 700 ∼ 900 cm^(-1) indicated that the mutan contained α-1,3-glucan. The ^(13)C NMR spectrum analysis shows that mutan has a high similarity with the α-1,3 linked glucan.

      • Na/Si(111)계의 초기 산화과정에 대한 XPS 연구

        X-선광 전자분광법을 이용하여 Na의 흡착에 의해 형성되는 Si(111)7×7-Na과 Si(111)3×1-Na 표면의 초기 산화과정에 대하여 연구하였다. Si(111)7×7-Na 표면은 깨끗한 Si(111)7×7 표면에 비하여 산화가 더욱 잘 일어나지만, Si(111)3×1-Na의 경우 산소와 거의 반응하지 않음을 알 수 있다. 또한 Si(111)7×7 표면과는 달리, Si(111)7×7-Na 표면에 1 ML의 Na를 흡착시키고 산소를 100L 노출한 경우 0 ls 내각 스펙트럼에 두 종류의 산소성분이 존재를 함을 알 수 있다. 산소의 노출량이 증가함에 따라 낮은 운동에너지 성분의 세기는 계속 증가를 하지만, 높은 운동에너지 쪽의 성분은 초기에 포화되었다. 이러한 0 ls 내각 스펙트럼의 변화와 Na KLL Auger peak intensity 증가로부터 Si(111) 7×7-Na 표면에 흡착되는 산소는 Si-0-Si, 그리고 Si-0-Na 두가지의 결합형태로 존재함을 예상 할 수 있다. 또한, Si(111)7×7-Na 표면에 주입되는 산소 노출량이 증가함에 따른 Na KLL Auger peak intensity가 증가하는 현상에 대하여 논의하였다. The initial oxidation of Si(111)7×7-Na and Si(111)3×1-Na surfaces have been studied by means of X-ray photoelectron spectroscopy(XPS). The Si oxidation rate of Si(111)7×7-Na surface was much enhanced more than that of the clean Si(111)7×7, but the Si(111)3×1-Na surface hardly reacted with oxygen. It shows that 0 1s peak of the Si(111)7×7-Na surfaces for 100L O_2 exposure consists of two components, coming from a different oxygen bonding configuration. From the change in intensity of two component of 0 ls and the increase of Na KLL peak intensity with increasing O_2 exposure, it is found that one component originates from Si-0-Si bonding at the lower kinectic energy and the other component comes from the Si-0-Na bonding at higher kinectic energy. Also, we have discussed a possible interpretation for the abnormally increase of Na KLL peak intensity.

        규산질 비료의 참외 흰가루병 발생 억제 효과

        토양에 시용한 규산의 참외 흰가루병에 대한 직접적인 억제 효과와 살균제의 방제 효과를 증진시킬 수 있는 보조효과를 조사하였다. 규산질 비료의 처리는 철가루병 균에 의해 감염된 참외 잎에서 균사의 생장과 분생포자의 형성을 억제함으로써 흰가루병 발생을 일부 억제할 수 있는 것으로 조사되었다. 물론 규산질 비료 자체만의 흰가루병 발생 억제 효과는 매우 미약한 수준이었으나 살균제의 흰가루병 방제 효과를 크게 증진시켜줄 수 있는 것으로 나타났다. 따라서 현재 농가 현장에서 밝혀지고 있는 규산질 비료의 참외 병해 발생 억제 효과는 주로 이러한 살균제의 효과를 증진시킬 수 있는 규산의 보조적인 작용에 기인하는 것으로 판단된다. 따라서 토양에 대한 적절한 수준의 규산질 비료의 시용은 살균제의 방제 효과 증진을 통하여 과다한 약제의 사용과 그에 따른 부작용을 경감시킬 수 있을 것으로 보인다. 규산질 비료의 흰가루병 발생 억제 효과를 증진시키기 위해서는 앞으로 병의 발생을 최대로 억제할 수 있는 참외 잎 중의 적정 규소 함량에 대한 검토 등의 추가적인 연구가 필요할 것이다. Silicon is known to accumulate in plants and results in greater resistance to diseases and insect pests. In this study, we investigated the effect of silicate fertUizer applied in soil on the development of powdery mildew of oriental melon Oriental melon seedlings of four-leaf stage were transplanted and grown in a plastic film house. silicate fertilizer was applied to maintain soil available SiO₂ level of 200 ㎎/㎏ one week before transplanting. Fungicide trinumizol was sprayed three times; one, two, and three weeks after transplanting. Sphaerotheca fuliginea was inoculated 2 weeks after transplanting. The number of infected leaf and the number of fungal colony in leaves were measured one, two, and three weeks after the inoculation. Three weeks after the fungal inoculation, in the treatment of fungicide triflwnhl, infected leaf numbem and number of colony per leaf were reduced by 10 and 58% respectively. In the silicate fertilizer treatment, infected leaf numbers and numbers of colony per infected leaf were suppressed only by 6 and 16%, respectively, and the efficacy was lower than that of the fungicide triflumizol. The combined treatment of silicate fertilizer and the fungicide suppressed powdery mildew more effectively, and infected leaf numbem and numbem of colony per leaf were reduced by 31 and 80%, respectively. These results indicate that although silicate fertilizer itself is not much effective in the suppression of powdery mildew, it can significantly enhance the efficacy of the fungicide.

      • 선형파괴역학에 의한 Aluminum Foil과 종이의 파괴거동 해석

        크랙이 내재된 시편을 크랙으로부터 멀리 떨어진 단면에 대해 균일한 크기의 응력과 변형량이 발생하도록 길이방향으로 당겨주는 장치를 이용하여, 선형 파괴 역학적 관점에서 알루미늄 호일과 종이의 파괴거동을 평가하였다. 상사형의 균열을 가진 파괴시편의 파괴하중을 차원해석으로 예측하였고, 실험을 통하여 예측된 하중과 실제하중을 비교, 검토하였다. The fracture behaviors of aluminum foils and sheet papers were analyzed on the basis of linear elastic fracture mechanics(LEFM). The fracture loads of the Al foil and paper were calculated by dimensional analysis. The actual fracture loads were measured using the simple tension equipment. The predicted fracture loads were compared with the experimental results, respectively.

      • 신축 공동주택의 VOCs와 알데히드류 성분에 대한 실태조사

        In comparison with other studies, in particular, the emission of indoor HCHO in new apartments were very serious and Vacs showed below the Japan's guidelines (260 ㎍/㎥) excepting toluene (324.31 ㎍/㎥). Concentration of volatile organic compounds (VOCs) including aldehydes in indoor air was analyzed at 3 new apartments (at the lower, middle and upper floors of each apartment) during the Spring in 2004. The sample gases of Vacs and aldehydes were collected by Silicon canister (6 L) and 2,4-DNPH cartridges, and the analysis was made by the GC/MSD and PLC. As a result of the analysis, formaldehyde (HCHO) amongst aldehydes was found the highest (874.63 ppb) at the upper floor of C Apt., and the aldehydes' concentrations were S > C > K Apt. each. Also it did not show an apparent variation of characteristics according to floor. The concentration of VOCs was the highest (819.22 ppb) at the upper floor of C Apt. which exhibited in proportion with height.

      • 영지 (Ganoderma lucidum) 자실체 다당류의 분자량에 따른 생리활성

        The study was conducted to investigate the biological activities of an protein-bound polysaccharide (GLP) isolated from the fruit body of Ganoderma lucidum. GLP was extracted by hot water and then it was purified high (GLP-HM) and low (GLP-LM) molecular weight fraction by ethanol precipitation. Both of GLP-HM and GLP-LM at doses of 20 mg/kg in vivo had the ability to inhibit the tumor growth of Sarcoma 180 tumor-bearing mice. Inhibition rates of tumor (%) were inhibited 60.6% and 38.7% by GLP-HM and GLP-LM, respectively. In assay of anti-complementary activity in vitro, the GLP-HM were higher than that of the GLP-LM polysaccharide. And also, the GLP-HM enhanced nitric oxide production of mouse bone marrow cells. When the GLP-HM was injected intraperitoneally in tumor-bearing mouse at doses of 20 mg/kg, the concentration of TNF-α in vivo was 140.9 pg/ml in the serum, 269.6 pg/ml in sarcoma tumor mass and 879.4 pg/ml in liver. This study shows that the high molecular polysaccharide from mushroom can enhance more the bioactivities than that of the low molecular polysaccharide.
