      • A Study of D. H. Lawrence Sons and Lovers : A Truth Seeker's Character in Quest of Paul's Identity 자아 추구의 구도자적 성격

        This thesis aims to analyze and clarity Paul Morel's (main hero) life journey reflected in D. H. Lawrence's Sons and Lovers based on the mythological archetype approach. In Sons and Lovers, the writer of this thesis can find a literary trend of a buildungsroman, that is, literary, a "development novel" a novel which traces the growth of a single individual, usually a exceptionally talented and sensitive person from adolescence to maturity. Sons and Lovers obviously does this and as such, it can be traced as one of the most outstanding and most successful buildungsroman of our time. In this view the writer has determined to center his attention to a truth seeker, Paul Morel, as "Hero-Archetype" a protagonist embodied in the my theological-archetype approach. As hero-Archetype, Paul Morel must pass through following three steps : the quest, initiation, and the sacrificial scapegoat. in quest of his life journey, Hero Paul must perform impossible tasks, meet with various ordeals, solve unanswerable riddles, and at times overcome insurmountable obstacles in order to attain or secure his own goal or happiness. With these in mind, the writer has paid his attention to compose the body if the paper as follows : in chapter two, Paul's life aspects of background as a truth seeker, has been explored mainly on his home atmosphere and social environment including his intense attachment to his mother. In chapter three, Paul's initiation into society has been treated in connection with Paul-Miriam relation as well as Paul-Clara's relation, both of which has not been successful in their married life on account of different objectives in life goal among them. In initiation process, the hero undergoes a series of painful ordeal's in passing form ignorance and immaturity to social and spiritual adulthood, The initiation most commonly consists of three district phases : 1) separation 2) transformation 3) return, This is a variation of the death-and rebirth archetype. Based on this pattern, Paul determines to separate form his mother for getting married, firstly, to Miriam, a beautiful but spiritually prejudiced daughter of a neighboring farm, and, secondly, to Clara Dawes, Baxter's wife has not been successful, with Miriam, in a crippled being who loves Paul but cannot give himself to him on account of her own prejudiced character, and with Clara Baxter, in pointless promiscuity, An additional failure of Paul's married life will be his mother's strongest holds over him, and thus, as soon as Paul comes into contact with women, the battle goes on between the mother and the girl, with the son as object. The mother gradually proves stronger, because of the blood-tie. After the painful ordeals resulting form his unsuccessful married life, obviously talented, sensitive and artistic Paul pass through the transformation phase which serves to enlighten him form ignorance to the matured adulthood. finally Paul determined to return to his mother, symbolized as Great Mother who has string holds over Paul by the blood-tie. It is, however, surprising for him to find his mother's suffering from serious illness, to whom Paul take all the sleeping pills in the house, crushing them together in his mother's bed-milk and led her to die. Mother's death frees Paul form his mother's imprisoning love. But now that he is frees, the young man hardly knows what to do with himself, for the unique characteristic of his Oedipal prison has been its attractiveness. And then he has one time been sunken into a deep valley of despair, but at this moment Paul's reawakening to life from the blank and total despair of his grief for his mother. As a truth seeker, Paul has come to realize that a man is nothing but a minuscule living-things. At last, he recognizes, everything must fall before this final, triumphant reality of life, With this affirmation he may be redeemed form his deepest despair, not falling into an abyss of victim as a sacrificial scapegoat that is last destination in the hero-archetype quest journey.

      • A novel mode of TRPML3 regulation by extracytosolic pH absent in the varitint-waddler phenotype

        <P>TRPML3 belongs to the TRPML subfamily of the transient receptor potential (TRP) channels. The A419P mutation in TRPML3 causes the varitint-waddler phenotype as a result of gain-of-function mutation (GOF). Regulation of the channels and the mechanism by which the A419P mutation leads to GOF are not known. We report here that TRPML3 is a Ca(2+)-permeable channel with a unique form of regulation by extracytosolic (luminal) H(+) (H(+)(e-cyto)). Regulation by H(+)(e-cyto) is mediated by a string of three histidines (H252, H273, H283) in the large extracytosolic loop between transmembrane domains (TMD) 1 and 2. Each of the histidines has a unique role, whereby H252 and H273 retard access of H(+)(e-cyto) to the inhibitory H283. Notably, the H283A mutation has the same phenotype as A419P and locks the channel in an open state, whereas the H283R mutation inactivates the channel. Accordingly, A419P eliminates regulation of TRPML3 by H(+)(e-cyto), and confers full activation to TRPML3(H283R). Activation of TRPML3 and regulation by H(+)(e-cyto) are altered by both the alpha-helix-destabilizing A419G and the alpha-helix-favouring A419M and A419K. These findings suggest that regulation of TRPML3 by H(+)(e-cyto) is due to an effect of the large extracytosolic loop on the orientation of fifth TMD and thus pore opening and show that the GOF of TRPML3(A419P) is due to disruption of this communication.</P>

      • A형 간염의 자연항체와 예방접종을 통한 항체 생성률의 역가 비교분석

        2008년부터 A형 간염 환자들이 급속히 증가하고 본원에 내원하여 건진을 받는 대부분의 수검자들이 A형 간염(IgG) 항체 생성 유무에 관심이 많아지며 검사 건수가 증가하였다. 그에 따라 항체 검사결과가 cut-off값에 걸리는 검체가 많아져 원인을 분석하였더니 대부분 A형 간염 예방접종을 한 수검자들이었다. 이에 저자들은 건강증진센터에서 설문조사를 통하여 자연면역을 획득한 수검자들 그룹과 본원에서 A형 간염 예방접종(1차, 2차)을 실시한 직원들 그룹으로 나누어 검사를 시행하였고 cut-off값을 기준으로 항체 생성률과 그에 대한 역가를 비교하고 진단검사의학과와 핵의학과에서 사용하는 진단 시약간에 항체 생성률과 그에 대한 역가를 비교해 보고자 했고, 2012년 8월 한 달 동안 건진 수검자 185명을 설문조사하여 자연면역을 획득한 119명과 본원에서 예방 접종을 실시한 직원들을 대상으로 1차 접종자 53명, 2차 접종자 59명으로 대상을 분류했다. 항체 생성률은 cut-off값 1을 기준했을 때 0.90-1.10 (${\pm}$), 0.60-0.89 (1+), 0.30-0.59 (2+), 0.01-0.29 (3+)로 나누어 역가를 비교하고, 같은 기준으로 제조사별 백신 접종 후 항체 생성률에 대한 역가를 비교평가 해 보았다. 그 결과, 건진 수검자 중 자연 면역을 획득한 수검자는 cut-off값 1을 기준했을 때, 0.90-1.10 (${\pm}$)가 0%, 0.60-0.89 (1+)가 0%, 0.30-0.59 (2+)가 4.2%, 0.01-0.29 (3+)가 96%로 역가가 <0.60 ($${\geq_-}2+$$)가 100%였다. 그리고 예방접종을 실시한 직원들의 항체 생성률은 1차 접종자 중 ${\pm}$가 59.1%, 1+가 18.1%, 2+가 18.1%, 3+가 4.6%로 총 45.3%였고, 역가는 $${\geq_-}$$ 0.60 ($${\leq_-}1+$$)가 77.3%였다. 2차 접종자의 항체 생성률은 ${\pm}$가 1.9%, 1+가 15.4%, 2+가 36.54%, 3+가 46.2%로 총 88.1%였고 역가는 <0.60 ($${\geq_-}2+$$) 82.7%가 였다. 또한 제조사별로 비교 하였을 때 1차 접종자의 항체 생성률은 BNIBT 20.8% (${\pm}24.5%$), GB 15.7% (${\pm}7.8%$), RIAKEY 94.3% (${\pm}3.8%$), ROCHE 83% (${\pm}0%$), Abbot 73.1% (${\pm}5.8%$)였고, 2차 접종자의 항체 생성률은 BNIBT 86.4% (${\pm}1.7%$), GB 88.5% (${\pm}1.9%$), RIAKEY 100% (${\pm}0%$), ROCHE 98.3% (${\pm}0%$), Abbot 98.2% (${\pm}0%$)였다. 즉 자연면역 항체가 예방접종에 의한 항체보다 역가가 높다는 것을 알 수 있었고, 1차 접종 후 보다는 2차 접종 후 검사를 시행했을 때 항체 생성률과 역가가 대부분 높아짐을 알 수 있었다. 따라서 결과 보고시 negative, index (${\pm}$), weak positive (1+), positive (2+), strong positive (3+)로 역가를 나누어 보고를 하거나 결과값에 index값을 같이 적어서 결과를 상세히 보고한다면 과거결과와 비교도 가능할 것이다. 또 제조사별 비교 시 1차 예방접종 후의 항체 생성률과 역가에서 시약간에 많은 차이를 보이고 있었고, 매년 예방 접종률이 높아지고 있는 시점에서 이러한 차이를 줄이기 위해서 각 제조사들은 민감도나 재현성에 더 주의를 기울여야 하겠고, 자연면역항체와 예방접종을 통한 항체간에 생길 수 있는 미지의 차이를 감안하여 검사자들이 사용하는 시약을 신뢰할 수 있도록 더 연구하고 개발해야 할 것이다. Purpose: Since 2008, hepatitis A patients was rapidly increasing. So, Most of the health checkup examinees were interested in whether hepatitis A antibody was a lot. thereby The number of tests was increasing. In recent years, Antibody test results in the range of cut-off values were increased. According to the cause analysis, most examinees had a hepatitis A vaccine. This study was conducted to classify hepatitis A antibody as natural antibody and antibody after vaccination and compared the titer for seroconversion rate based on cut-off values. Materials and Methods: For a month in August 2012, First, We surveyed 185 health examinees and classified 119 health examinees who had acquired natural antibody. Second, for employees who were inoculated against hepatitis at our hospital, We classified into 53 primary inoculators and 59 secondary inculators. when the standard of cut-off value was 1, The seroconversion rate was compared the titer divided by 0.90-1.10 (${\pm}$), 0.60-0,89 (1+), 0.30-0.59 (2+), 0.01-0.29 (3+) and we compared the titer for seroconversion rate by each manufacturer after vaccination. Results: When the standard of cut-off value was 1, the titer of 119 health examinees who had acquired natural antibody was 0.90-1.10 (${\pm}$): 0%, 0.60-0.89 (1+): 0%, 0.30-0.59 (2+): 4.2%, 0.01-0.29 (3+): 96% and the titer of <0.60 ($${\geq_-}2+$$) was 100%. The titer of 53 primary inoculators was 0.90-1.10 (${\pm}:59.1%$), 0.60-0.89 (1+): 18.1%, 0.30-0.59 (2+): 18.1%, 0.01-0.29 (3+): 4.6% and the seroconversion rate was 45.3%. The titer of $${\geq_-}0.60$$ ($${\leq_-}1+$$) was 77.3%. The titer of 59 secondary inoculators was 0.90-1.10 (${\pm}:1.9%$), 0.60-0.89 (1+): 15.4%, 0.30-0.59 (2+): 36.54%, 0.01-0.29 (3+): 46.2% and the seroconversion rate was 88.1%. The titer of <0.60 ($${\geq_-}2+$$) was 82.7%. When we compared the titer for seroconversion rate by each manufacturer after vaccination, the seroconversion rate of 53 primary inoculators was BNIBT: 20.8% (${\pm}:24.5%$), GB: 15.7% (${\pm}:7.8%$), RIAKEY: 94.3% (${\pm}:3.8%$), ROCHE: 83% (${\pm}:0%$), ABBOTT: 73.1% (${\pm}:5.8%$) and the seroconversion rate of 59 secondary inoculators was BNIBT : 86.4% (${\pm}:1.7%$), GB: 88.5% (${\pm}:1.9%$), RIAKEY: 100% (${\pm}:0%$), ROCHE: 98.3% (${\pm}:0%$), ABBOTT: 98.2% (${\pm}:0%$). Conclusion: The study show that the titer of natural immune antibodies is higher than the titer of vaccination and the titer of secondary inoculation is mainly higher than the titer of primary inoculation. Consequently, if we know the titer of hepatitis A antibodies, it will help to give resullt reports. And then, when we compared the titer and the seroconversion rate by each manufacturer, There was a very distinct difference. As the test subjects inoculate against hepatitis A (HAV), it is considered BNIBT, GB will occur false negative rate and RIAKEY, ROCHE, ABOTT will occur false positive rate.

        Biochanin A의 미백 효과에 관한 연구

        본 연구는 미백 소재 개발을 위하여 강진향(Dalbergia odorifera T. Chen) 추출물에서 분리한 활성물질인 biochanin A의 멜라닌 생성에 연관된 효과를 알아보았다. Biochanin A는 B16F1 melanoma 세포에서 농도의존적으로 멜라닌 양을 억제하였으며, 멜라닌 생합성 저해 효과는 10 μg/mL 농도에서 멜라닌 생성 억제율이 48%로 나타났다. Western blot을 이용하여 microphthalmia associated transcription factor (MITF), Tyrosinase, Trp-1와 Trp-2의 발현을 억제함을 확인하였다. 따라서 biochanin A는 미백 효능을 갖는 화장품 소재로서의 개발 가능성이 클 것으로 기대된다. To develop a new whitening agent for cosmetics, we investigated the inhibitory effects of biochanin A on melanogenesis. The biochanoin A is an active component in the extracts of Dalbergia odorifera T. Chen. It reduced melanin contents of B16F1 melanoma cells in a dose-dependent manner and decreased to about 48% at a concentration 10 μg/mL. It also inhibited the expression in microphthalmia associated transcription factor (MITF), Tyrosinase, Tyrosinase related protein 1 (Trp-1), and Tyrosinase related protein 2 (Trp-2) in melanocytes. The results suggested that biochanin A has considerable potential as a cosmetics ingredient with a whitening effect.

        중국 웨딩 산업의 맞춤형 정보 제공을 위한 중국 웨딩 · 뷰티 이미지 분석연구

        본 연구는 향후 중국 웨딩 산업의 주체로 부상하고 있는 80후와 90후의 여성을 대상으로 웨딩 · 뷰티 스타일 트렌드를 조사 분석하여 그들의 니즈에 맞는 패션 · 뷰티 정보를 중국시장에 진출하려고 하는 한국웨딩업체에 제공하는데 그 목적이 있다. 연구방법으로는 문헌연구와 실증적 연구를 병행하였는데, 문헌연구에서는 선행연구 및 관련 산업 현황을 조사하였고, 실증적 연구에서는 웨딩 · 뷰티 트렌드의 특성을 파악하기 위하여 중국의 대표 웨딩 전문잡지인 시상신양(;Cosmo Bride)의 2011년 1월-2013년 12월까지 총 36개월의 자료와 중국의 결혼과 관련해 가장 보편화된 웨딩 관련 사이트와 웨딩 박람회 자료를 선택한 후 패션 · 뷰티 이미지 분석을 전문가 집단을 통하여 실시하였다. 연구의 분석결과는 다음과 같다. 중국 웨딩패션뷰티 이미지 분석 결과 엘레강스 이미지가 가장 높게 나왔으며, 메이크업은 유사 색상 배색을 통해 단아하고 차분한 이미지를 연출하는 것을 선호하는 것으로 나타났다. 헤어에 있어서는 업스타일 형태가 주를 이루었다. 형태적인 면에 있어서는 실루엣은 A라인 실루엣, 네크라인은 베어드탑, 웨이스트 라인은 노멀 웨이스트 라인, 소매는 슬리브리스가 높은 빈도를 보였다. 웨딩드레스의 소재는 시스루 소재, 드레스 색채는 무채색이 높은 비율을 보였고, 장신구는 착용하지 않는 형태가 가장 높은 빈도를 보였다. 이를 이미지별로 살펴보면, 내추럴 이미지의 경우 유사색상배색의 메이크업과 내추럴 헤어, 웨딩드레스는 A라인 실루엣, V넥, 하이웨이스트, 슬리브리스 형태, 시스루 소재의 무채색 웨딩드레스에 베일이나 장신구가 없는 형태로 코디네이션되면서 높은 빈도를 보였다. 모던이미지의 경우 유사색상배색의 메이크업, 업스타일 헤어, 웨딩드레스는 A라인 실루엣, 베어드탑, 노멀 웨이스트, 슬리브리스, 시스루 소재의 무채색 웨딩드레스에 장신구가 없는 형태로 코디네이션되어 연출되었다. 로맨틱 이미지의 경우 유사색상배색의 메이크업, 업스타일 헤어, 웨딩드레스는 A라인과 X라인 실루엣, 베어드탑, 노멀 웨이스트, 슬리브리스, 시스루 소재의 무채색 웨딩드레스에 헤드드레스가 착용되어 코디네이션되고 있었다. 마지막으로 엘레강스 이미지의 경우 유사색상배색의 메이크업, 업스타일 헤어, 웨딩드레스는 A라인과 X라인 실루엣, 베어드탑, 노멀 웨이스트, 슬리브리스, 시스루 소재의 무채색 웨딩드레스에 장신구가 없는 형태로 코디네이션되고 있었다. 결론적으로 중국의 80후와 90후 세대를 타켓으로 하는 웨딩 트렌드는 지역적, 문화적 차이가 있는 한국의 웨딩트렌드와는 미묘한 차이가 있기에 우리 기업이 중국에서 웨딩시장에 성공적으로 진입하기 위해서는 반드시 필요한 연구임을 알 수 있었다. 무엇보다도 중국 웨딩 소비시장의 규모가 상승세를 타고 있는 시점에서 막연한 한류보다는 그들의 구체적인 선호 트렌드를 파악함으로써 또 다른 웨딩한류를 기획하는 마케팅 전략을 개발하는데 유용한 정보를 제공할 수 있을 것으로 생각된다. The purpose of this study is to provide information on fashion and beauty which satisfy the needs of 80hou and 90hou women who are becoming the main agent of the Chinese wedding industry in the future for the Korean wedding companies willing to enter Chinese market by investigating and analyzing their wedding and beauty style trends. As for the research method, literature reviews and empirical studies were arranged together. In the literature review, the precedent studies and present conditions of the relevant industry were investigated. And in the empirical studies, the materials of Cosmo Bride from Jan. 2011 to Dec. 2013 (total 36 months), the most common wedding web sites related to Chinese wedding, and wedding fairs were selected and an analysis of fashion and beauty images was carried out by an expert panel to understand the characteristics of the wedding and beauty trends. The analytical results of the study are as follow: The result which analyzed Chinese wedding fashion and beauty images has found that elegant image is highest. Makeup has found that graceful and calm image by arrangement of similar colors are preferred and hair was mainly up hairdos. As for form aspects, high frequency was shown in A line silhouettes for silhouettes, bared top for necklines, normal waistlines for waistlines, and sleeveless for sleeves. And high frequency in wedding dress" materials was shown in see-through materials and in colors were achromatic colors. As for accessories, high frequency was shown in the form of not wearing accessories. To look at these by images, natural images showed high frequency of coordination with makeup with arrangement of similar colors, natural hair, and wedding dress of A line silhouettes, V neck, high waistline, sleeveless, see-through materials and achromatic colors without veil or accessories. As for modern images, makeup with arrangement of similar colors, up hairdos, and wedding dress of A line silhouettes, bared top, normal waistline, sleeveless, see-through materials and achromatic colors without accessories were coordinated. As for romantic images, makeup with arrangement of similar colors, up hairdos, and wedding dress of A line and X line silhouettes, bared top, normal waistline, sleeveless, see-through materials and achromatic colors with head dress were coordinated. Lastly, elegance images were coordinated with makeup with arrangement of similar colors, up hairdos, and wedding dress of X line silhouettes, bared top, normal waistline, sleeveless, see-through materials and achromatic colors without accessories. In conclusion, this is a necessary research for Korean companies to successfully enter Chinese wedding market because the Chinese wedding trend targeting 80hou and 90hou has subtle differences with the Korean wedding trends which have regional and cultural differences. Above all, as the scale of Chinese wedding consumption market are on the rise, it gives useful information to develop marketing strategies of planning Korean wedding wave by understanding their concrete preferred trend not just depending on Korean wave.

        한국인에서 5,10-Methylenetetrahydrofolate Reductase 유전자 다형성 분석 및 임신의 결과에 미치는 영향

        목적: 본 연구는 한국인 임신 여성에서 MTHFR 유전자형 변이와 혈청내 호모시스테인 및 엽산의 농도와 이들이 임신에 미치는 영향을 알아보고자 하였다. 방법: 임신부 600명에게서 혈액을 채취하여 얻어진 DNA로 MTHFR C677T와 A1298C 유전자의 다형성을 PCR-RELP로 분석하였고 혈청내 호모시스테인은 HPLC 방법으로, 엽산은 RIA 방법으로 측정하였다. 결과: 혈청내 호모시스테인은 MTHFR C677T 유전자형이 C/C형이나 C/T인 군에 비해 T/T인 군에서 유의하게 높았으며 MTHFR A1298C 유전자형이 A/C형이나 C/C인 군에 비해 A/A인 군에서 역시 유의하게 높았다( p<0.05). 혈청 호모시스테인은 모든 MTHFR 유전자형에서 혈청 엽산농도와 역상관관계를 보였으나, 변이형 유전자형(T/T와 A/A유전자형)에서 더 큰 역 상관관계를 보였다. 15?mol/L 이상의 고호모시스테인혈증의 임신부에서 임신 주수와 출생체중이 36.1주, 3053.8 g으로 15 ?mol/L미만인 군의 38.3 주, 3215.3 g 보다 유의하게 낮았다( p<0.05). 결론: 혈청 호모시스테인의 농도는 MTHFR C677T와 A1298T 유전자의 다형성에 의해 영향을 받으며, MTHFR C677T와 A1298T 유전자의 다형성과 혈청 호모시스테인은 모두 임신주수와 신생아의 체중에 영향을 미친다. Objective: The purpose of this study was to analyze MTHFR polymorphism among the Korean population and to evaluate the relationship between serum levels of homocysteine and MTHFR polymorphism and also to investigate the effect on pregnancy outcomes. Methods: DNA was extracted from whole blood of 600 pregnant women. All samples were genotyped for the C677T and A1298C polymorphisms in MTHFR gene by PCR-RELP assay. Serum levels of homocysteine and folate were measured by high performance liquid chromatography for homocysteine and radioassay for folate. Pregnancy outcomes were estimated by gestational weeks and birth weights of newborns. Results: Serum homocysteine was higher in women with the T/T genotype than those with the C/T or C/C genotype of the MTHFR C677T polymorphism (p<0.05). And also serum homocysteine was higher in women with the A/A genotype than those with the A/C or C/C genotype of the MTHFR A1298C polymorphism (p<0.05). Serum homocysteine was negatively correlated with serum folate in all MTHFR genotypes, especially prominent in T/T genotype of MTHFR C677T polymorphism and A/A genotype of MTHFR A1298C polymorphism. Gestational age and the birth weight of infant from hyperhomocysteinemic mothers whose homocysteine levels higher than 15 μmol/L were 36.1 weeks, 3053.8 g, respectively, which were significant lower than those from normohomocysteinemic mothers (38.3 weeks, 3,215.3 g) (p<0.05). Conclusion: Serum homocysteine was influenced significantly by MTHFR C677T polymorphism and MTHFR A1298C polymorphism. MTHFR C677T and A1298C polymorphism and serum homocysteine levels affect pregnancy outcomes, although not mainly by serum folate level.

      • A microfluidic electrochemical aptasensor for enrichment and detection of bisphenol A.

        <P><B>Abstract</B></P> <P>Bisphenol A (BPA) is an organic monomer used to make common consumer goods such as plastic containers, sports equipment, and cosmetics which are heavily produced worldwide. A growing interest has been drawn to general public as BPA is one of the major endocrine disrupting chemicals threating human health. To date, numerous BPA sensors have been attempted to be developed but important challenges still remained such as limited linearity range, easy to use, and long term response time. To address the present issues, a microfluidic channel should be integrated into an electrochemical aptasensor and it is called Geometrically Activated Surface Interaction (GASI) chip. The vigorous generation of the micro-vortex in the GASI fluidic chamber provides the high collision chances between BPA and anti-BPA aptamer (BPAPT) and consequently more BPA molecules can be captured on the aptasensor surface, which finally results in high sensitivity of the aptasensor. To construct the integrated aptasensor, a miniaturized gold electrode is fabricated using shadow mask and e-beam evaporation process. Afterward, BPAPT is immobilized on a nanostructured gold electrode via thiol chemistry, and other terminus of the aptamer is labeled with a ferrocene (Fc) redox probe. Then, the microfluidic channel is mounted over the miniaturized gold electrode to introduce and enrich BPA to the aptasensor. Upon the specific interaction between BPA and its aptamer, configuration of aptamer is changed so that Fc tag approaches to the electrode surface and direct oxidation signal of Fc and BPA are followed as analytical signals. The unique microfluidic integrated electrochemical aptasensor delivers a wide linear dynamic range over 5 × 10<SUP>–12</SUP> to 1 × 10<SUP>−9</SUP> M, with a limit of detection 2 × 10<SUP>–13</SUP> M. This aptasensor provides a precise platform for simple, selective and more importantly rapid detection of BPA.       • S-510 A case of peritoneal dialysis-associated peritonitis caused by Agromyces mediolanus

        Introduction:?Agromyces mediolanus (A.mediolanus) is an unusual pathogen in human infection. No case of peritoneal dialysis (PD)-associated peritonitis by A.mediolanus has been reported. This is the first case report of PD-associated peritonitis by A.mediolanus in the world. We describe a case of PD-associated peritonitis by A.mediolanus that was cured after catheter removal.?Case: A 59-year-old man who had been treated with CAPD was admitted to our hospital because of turbid peritoneal effluent accompanied by constant diffuse abdominal pain. The patient had an exit-site infection with purulent secretion. The laboratory findings showed PD peritonitis: the white blood cell (WBC) count of the peritoneal effluents was 1157/mm3 with a neutrophil predominance (91.3%). Cefazolin and ceftazidime were given intraperitoneally per day. The peritoneal WBC decreased, but the patient’s clinical condition didn’t improve on the 5th day after starting intraperitoneal antibiotics. Culture of the peritoneal dialysate revealed Agromyces species, which was susceptible to meropenem, imipenem, vancomycin, but resistant to ceftriaxone. Thus, we changed antibiotics to intraperitoneal vancomycin and meropenem on the 5th day. Despite the intraperitoneal antibiotics for 4 days, the patient’s abdominal pain persisted and the peritoneal WBC count became elevated. Therefore, we removed the PD catheter and the patient was switched to hemodialysis on the 9th day. Vancomycin and imipenem was continued for another 2 weeks and the patient’s clinical condition improved. The patient has been maintained on hemodialysis.?Conclusions: There is no previous report of peritonitis associated with A.mediolanus in a CAPD patient. In this case, we used intraperitoneal cefazolin and ceftazidime and changed to vancomycin and imipenem according to the result of peritoneal dialysate. However the patient improved after removal of catheter. Therefore, early removal of the PD catheter in A. mediolanus related peritonitis without prompt reaction to antibiotic therapy should be considered.

        금화규(Abelmoschus manihot) 꽃 추출물의 In Vitro 피부 미백 및 주름 개선 효능 연구

        본 연구는 화장품 소재로서 금화규(Abelmoschus manihot) 꽃의 미백 활성율 및 항주름 활성을 평가하기 위해 수행하였다. 금화규 꽃은 물과 70% 에탄올을 이용하여 추출하였다. Tyrosinase 억제 활성은 물과 70% 에탄올 추출물 모두 농도 의존적으로 억제 활성이 증가 된다고 평가되었으며, B16F10 세포주에서 생존율에 영향을 미치지 않은 농도인 10, 25, 50, 75, 100 μg/mL에서 멜라닌 생합성을 저해하는 것으로 확인되었다. 금화규 꽃의 항주름 효능으로는 collagenase와 elastase의 활성 저해 능력을 측정한 결과 70% 에탄올 추출물이 물 추출물보다 높은 항주름 활성을 보였다. 본 연구 결과를 통하여 금화규 꽃 추출물은 미백 활성과 항주름 활성을 가지는 기능성 화장품 소재로 사용 가능할 것으로 사료된다. In this study, the skin whitening and anti-wrinkle activity of Abelmoschus manihot (A. manihot) flowers were evaluated as a cosmetic material. A. manihot flowers were extracted using water and 70% ethanol. Tyrosinase inhibitory activity was evaluated to increase concentration-dependent inhibitory activity in both water (AMW) and 70% ethanol extracts (AME), and was found to inhibit melanin biosynthesis at concentrations that did not affect survival in B16F10 cell lines at 10, 25, 50, 75, and 100 μg/mL. As a result of measuring the anti-wrinkle effectiveness of A. manihot flowers, 70% ethanol extract (AME) showed higher anti-wrinkle activity than water extract (AMW). Through the results of this study, it is considered that the A. manihot flower extract can be used as a functional cosmetic material with whitening and anti-wrinkle activity.

        다양한 레진시멘트로 합착한 섬유포스트의 결합강도 비교

        이 연구는 다양한 접착시스템을 사용하는 6가지 레진시멘트를 이용하여 근관에 합착한 섬유포스트를 push-out 검사에 의해 포스트와 치근 상아질 간의 결합강도를 상호 비교하기 위하여 시행되었다. 발거된 하악 소구치의 치관부를 제거하고 치근부를 근관충전한 후 9 mm의 포스트 룸을 형성하였다. 사용된 레진시멘트의 종류에 따라서 무작위로 6개씩 선택하여 Duo-Link 군 Variolink Ⅱ 군, Panavia F 군, Multilink Automix 군, RelyX Unicem 군, Maxcem 군으로 분류하였다. 제조사의 지시에 따라서 각각의 레진시멘트를 Uni-dose needle tip을 이용하여 근단부에서부터 주입하여 광투과성 섬유 포스트인 no. 2 DT Light Post를 근관에 합착하였다. 증류수에 24시간동안 보관한 후, 주수 하에 저속의 Diamond Wheel Saw를 이용하여 각 치근을 횡단면으로 1 mm두께로 절단하여 연속적인 절편 6개를 얻었다. 각 군의 시편을 universal testing machine에 고정하고 push-out 검사를 시행하여 push-out 강도를 통계 분석한 결과, 서로 다른 접착시스템을 사용한 레진시멘트 군 간의 push-out 강도는 유의한 차이가 나타나지 않았다 (p>0.05). The purpose of this study was to compare the push-out strength of a fiber post cemented with various resin cements. Newly extracted 36 human mandibular premolars which had single root canal were selected and their crown portions were removed. The root canal was instrumented using PROTAPER^(TM) system and obturated using continuous wave technique, In each root, a 9-mm deep post space was prepared. #2 translucent fiber post (DT Light post, Bisco Inc., Schaumburg, IL, U.S.A.) was cemented using injection technique with Uni-dose needle tip (Bisco) and six different resin cements. The tested resin cements were Duo-Link(Bisco Inc., Schaumburg, IL, U.S.A.), Variolink Ⅱ (Ivoclar-Vivadent AG, Schann, Liechtenstein), Panavia F (Kuraray Medical Inc., Okayama, Japan), Multilink Automix (Ivoclar-Vivadent AG, Schann, Liechtenstein), RelyX Unicem (3M ESPE Dental Products, St. Paul, MN, U.S.A.), and Maxcem (Kerr Co.,CA, U.S.A.). After storage in distilled water for 24 hours, each root was transversally sectioned into approximately 1-mm thick sections. This procedure resulted in 6 serial sections per root. Push-out test was performed using a universal testing machine (EZ Test, Shimadzu Co.) with a crosshead speed of 1 mm/min. The data were analyzed with one-way ANOVA and Tukey HSD (p=0.05). The push-out strength of the groups which cemented fiber post with Panavia F and Multilink Automix were lower than those of the other groups. But, there were no statistically significant difference among groups at a probability level of 0.05.
