C형 간염 바이러스(HCV)의 구조 유존자중 core 유전자를 fusion protein vector인 pGEX-2T vector에 cloning하여 core단백질을 발현시켜서 항원성을 증가시키고자 하였다. HCV의 core gene을 PCR (polymerase chain reaction) 기법을 이용하여 증폭한 후, 약 0.57kb 정도의 증폭된 core gene을 TA cloning vector에 cloning 한 후, restriction exzyme BamH I으로 잘라, gel elution method로 elution하여 pGEX-2T vector의 BamH I site에 cloning 시켰다. Cloning된 plasmid를 E coli cell인 JM105에 transformation시킨 후, IPTG에의해 protein induction 하였다. pGEX-2T에서 발현된 HCV core 단백질은 SDSPAGE및 Western-blotting 분석 결과, 양성 C형 간염 환자의 serum및 monoclonal antibody와 특이적으로 반응하였다.
Pocine Adenovirus-3의 E1B Region의 鹽基序列 分析

돼지 아데노바이러스(PAV-3). 6618주의 EIB region이 包含되어 있는 map unit 4.0에서 9.7까지의 유전자에 대한 1,984 bp의 염기서열을 決定하였으며, 이 結果를 알려진 여러 아데노바이러스 유전자와 비교하여 다음과 같은 결과를 얻었다. 1. PAV-3의 EIB유전자는 10개의 ORF로 구성되어 있으며, 그 중 아데노바이러스의 단백질과 유사성이 있는 것은 ORF1, ORF2 및 ORF3이었다. ORF1은 Ad41의 19kd 와 BAV-2에서의 EIB ORF2에서의 아미노산의 一致率은 각각 32%와 31%이었다. 2. ORF2는 Ad2 55kd protein과 tupaia adenovirus 44kd protein가 각각 34%로 아미노산 一致率이 가장 높았으며, Ad41의 52kd protein. BAV-3의 EIB ORF3에서도 33%의 一致率을 보였다. 3. ORF1은 61-666 uncleotide (606 bp), ORF 2에서는 429-1,850 uncleotide (1,422 bp)의 부위로 각각 202, 474 a.a로 構成되었으며, 예상되는 분자량은 20 kd와 52 kd이었다. 4. ORF3는 hexon-associated pIX유전자로 추정되며 내부에 1개의 polyadenylation signal(ATAAA)이 1938-1942 uncleotide에 위치하였으며, 이 부위는 TATA box (1937-1942 uncleotide)와 중복되어 존재하였다. Porcine adenovirus type 3 (PAV-3) does not cause severe infection in pigs. Adenovirus has been suggestive of live vaccine vector carrying foreign gene. One of insertion regions is delayed early (EIB) region. However, EIB region of PAV-3 has not been molecularly characterized to date. Nucleotide sequence of EIB of PAV-3 was determined. The EIB region was composed of 1,984 bp and located between 4.0 and 9.7 map units. Three potential open reading frames(ORFs) with low level of homology to other adenoviruses and a polyadenylation signal were identified in the rightward direction of genome. The nucleotide and the predicted amino acid sequences of EIB were compared to those of human and animal adenoviruses. One of the three potential ORFs. ORF1 encoded a polypeptide homologous to bovine adenovirus type 2(BAV-2) ORF2 and human adenovirus type 41(Ad41) 19 kd protein. ORF2 encoded a polypeptide homologous to human adenovirus type 2(Ad2) 55 kd protein, bovine adenovirus type 3(BAV-3) ORF3 and porcine adenovirus type 4(PAV-4) ORF2. The predicted protein of ORF1 had homology to those of Ad41 and BAV-2 with 32 and 31% respectively, whereas the deduced protein of ORF2 had homology to those of Ad2. BAV-3 and PAV-4 with 34, 33 and 29%, respectively.
소아에 있어서 뇌전산화단층 촬영상 결절성 두개강내 병변에 관한 임상적 연구

To make guideline to the diagnosis and treatment and or the intracranial nodular lesions, the authors made a clinical analysis on 81 pediatric patients who had showed nodular lesions on brain CT scan at Severance Hospital from Mar. 1979 to Feb. 1989 and the results were summarized as follows. 1) The peak incidence of intracranial nodular lesions were found in the 2nd decade(61.7%). There was no sexual difference. 2) The most common clinical symptom was seizure(92.6%) and the predilection site was parietal area. The nodular enhancing type on brain CT scan showed not only the most common type but also the most favorable clinical course. 3) As the etiology of the intracranial nodular lesions, tuberculosis, paragonimiasis, cysticercosis, tumor(meningioma), and aspergillosis were confirmed or suspected, but tuberculosis was the most common. CSF routine examination, chest PA, PNS X-rays, skin test for paragonimiasis and tuberculosis, CSF ELISA test for cysticercosis and paragonimiasis should be routine as the diagnostic studies. 4) We made treatment principle for intracranial nodular lesions. In cases that initial brain CT can show low density or calcified density without contrast enhancement, we treat those patients with anticonvulsants and steroid with regular check-up. If the lesion increase in size or new enhancement develops on follow up CT scan, medications for the suspected etiology should be added. In cases that initial brain CT scan show enhancing lesions, treatment is started with anticonvulsants and medications for the etiology regular check up. If the lesion increase in size on follow up CT scan or seizure is uncontrolled, surgery should be considered.
해수에서 분리한 Vibrio sp. M-96 균주의 열감수성 alkaline phosphatase 성질

A thermolabile alkaline phosphatase has been purified through steps of osmotic shock, ammonium sulfate salting-out, and DEAE-cellulose chromatography from the cultured broth of the marine Vibrio sp. M-96 strain. The optimal temperature for the enzyme activity was 35℃. The optimal pH was pH 11.0, and the range of pH stability was pH 10.4 to 12.0. Thermal inactivation occured within 6 minutes at 60℃. The enzyme was considerably inactivated by 0.1mM concentrations of Hg^2+, Ni^2+ and Zn^2+, whereas activated up to 234% by 1mM of Mn^2+. The activation energy and deactivation energy by the Arrhenius equation were 4.02 Kcal/mol and 9.09 Kcal/mol, respectively. The Km and Vmax values of the enzyme for p-nitrophenylphosphate were found to be 0.0465mM and 0.001335mM/min. respectively. Active from of the enzyme had a molecular weight of 57,000 dalton determined by the Sephadex G-200 gel filtration method.

Emphysematous pyelonephritis is a rare disease complicated from diabetes or obstructive uropathy. The characteristic of this disease is the presence of gas within the renal parenchyma, which may enter the perinephric space by extension. It was first reported in 1898 by Kelly and MacCallum. Up to 1992 only about 90 cases have been reported in English literature. In Korea, Shin et al. reported the first case of emphysematous pyelonephritis in 1982, and only about 28 cases reported in the next decade. In most cases, surgical therapies were recommended for emphysematous pyelonephritis. We report on a case of emphysematous pyelonephritis in diabetic patient who had been treated with medical therapy (antibiotics and blood glucose control by open loop insulin pump). Without surgical intervention, the patient (34-year-old female) recovered from emphysematous pyelonephritis.

The formation of biofilms on the enamel surface of teeth by Streptococcus mutans is an important step in dental plaque formation, demineralization, and early caries because the biofilm is where other bacteria involved in dental caries attach, grow, and proliferate. The objectives of this study were to determine the effect of phosvitin (PSV) on the biofilm formation, exopolysaccharides (EPS) production, adherence activity of S. mutans, and the expression of genes related to the compounds essential for biofilm formation (quorum-sensing inducers and components of biofilm matrix) by S. mutans. PSV significantly reduced the biofilm-forming activity of S. mutans and increased the degradation of preformed biofilms by S. mutans. PSV inhibited the adherence activity of S. mutans by 31.9%-33.6%, and the production of EPS by 62%-65% depending upon the strains and the amount of PSV added. The expressions of genes regulating the production of EPS and the quorum-sensing-inducers (gtfA, gtfD, ftf, relA, vicR, brpA, and comDE) in all S. mutans strains were down-regulated by PSV, but gtfB was down-regulated only in S. mutans KCTC 5316. Therefore, the anti-biofilm-forming activity of PSV was accomplished through the inhibition of biofilm formation, adherence activity, and the production of quorum-sensing inducers and EPS by S. mutans.
