        ABT-737 ameliorates docetaxel resistance in triple negative breast cancer cell line

        Purpose: This study aimed to validate the synergistic effect of ABT-737 on docetaxel using MDA-MB-231, a triple negative breast cancer (TNBC) cell line overexpressing B-cell lymphoma-2 (Bcl-2). Methods: Western blot analysis was performed to assess expression levels of Bcl-2 family proteins and caspase-related molecules. Cell viability was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Cell cycle distribution was determined by flow cytometry analysis. Benzyloxycarbonyl-Val-Ala-Asp(OMe)-fluoromethylketone (z-VAD-fmk) was used for pretreatment to assess the role of caspases. Results: Cell viability of MDA-MB-231 after combination treatment with ABT-737 and docetaxel was significantly lower than that after docetaxel or ABT-737 monotherapy based on MTT assay (both P < 0.001), with a combination index of 0.41. The proportion of sub-G1 population after combination treatment was significantly higher than that after docetaxel or ABT-737 monotherapy (P = 0.001, P = 0.003, respectively). Pretreatment with z-VAD-fmk completely restored cell viability of MDA-MB-231 from apoptotic cell death induced by combination therapy (P = 0.001). Although pro-caspase-8 or Bid did not show significant change in expression level, pro-casepase-9 showed significantly decreased expression after combination treatment. Cleaved caspase-3 showed increased expression while poly (ADP-ribose) polymerase cleavage was induced after combination treatment. However, hypoxia-inducible factor 1-alpha and aldehyde dehydrogenase 1 totally lost their expression after combination treatment. Conclusion: Combination of ABT-737 with docetaxel elicits synergistic therapeutic effect on MDA-MB-231, a TNBC cell line overexpressing Bcl-2, mainly by activating the intrinsic pathway of apoptosis. Therefore, adjunct of ABT-737 to docetaxel might be a new therapeutic option to overcome docetaxel resistance of TNBCs overexpressing Bcl-2.

        This study aims to analyze the priority of regional climate change policy utilizing AHP (analytic hierarchy process) at the area of Metropolitan City of Incheon. It derives four factors at first hierarchical level, at which level the analysis of pair wise comparison indicates that industrial sector, energy sector, climate change response, and green culture policy are considered important in that order. It also ends up with sixteen factors at second level. The result of comparison analysis between all factors reveals that investment promotion in green technology R&D is considered the most significant factor of all, followed by establishment of green enterprise support system, electricity-efficiency enhancement support project and build-up of green culture policy governance. The result implies that diverse promotional policies have to incorporate business, institutional, and cultural aspects for sustainable climate change policy of regions. The contribution of this study is that it highlights the need to include regional characteristics in deciding priority among policy options for them to be effective.

        The Effect of Selective Attributes of Customized Cosmetics on Purchase Intention: Focusing on the Mediating Effect of Perceived Natural Raw Materials

        Purpose: This study presents the direction for the development of the cosmetics industry and the revitalization of the customized cosmetics industry. By using natural raw materials that are harmless to the human body as raw materials for customized cosmetics, It was conducted with the aim of raising positive awareness among consumers and inducing purchases. Customized cosmetics can contribute to solving the problem of the unemployed as small-scale start-ups are possible, Since only those with professional qualifications can manufacture customized cosmetics, We want to research how we can fulfill consumers' desire for safety and their desire to own cosmetics that are right for their skin. Method: A total of 306 questionnaires were administered to men and women in their 20s and 60s who had experience in purchasing cosmetics living in Korea, and a frequency analysis, descriptive statistical analysis, exploratory factor analysis, and reliability analysis using Cronbach's alpha coefficient were conducted using SPSS 25.0 program as the analysis data, and an independent sample t test was conducted to test the hypotheses. We also conducted correlation analysis, multiple regression analysis, and mediation analysis using SPSS process macros to verify mediation effects. Results: First, it can be said that the higher the Awareness of selective attributes and Reliability of selective attributes of customized cosmetics, the higher the Purchase intention. Second, emphasizing the use of natural ingredients in the formulation of customized cosmetics will further enhance purchase intent. Third, since the awareness of customized cosmetics is high enough, supporting it in various ways will not only expand the market, but also allow customers to purchase customized cosmetics with confidence. Fourth, it means that the trust in the customized cosmetics formulation manager who received professional education the state had a positive effect on the choice of customized cosmetics. Conclusion: Using natural raw materials for customized cosmetics can contribute to solving international environmental problems, For the first time in the world, only those with professional qualifications can prepare customized cosmetics. It will be able to satisfy consumers' desire to own cosmetics that are safe and suitable for their skin. Therefore, in a situation where customized cosmetics are attracting worldwide attention, this study aims to increase consumers' positive awareness and help skin health by using harmless natural raw materials as raw materials for customized cosmetics. It will be able to suggest a direction as a new growth engine for K-beauty, Even as an international subject, it is considered to have original value.

      • The neural correlates of cross-modal interaction in speech perception during a semantic decision task on sentences: A PET study

        <P><B>Abstract</B></P><P>Speech perception in face-to-face conversation involves processing of speech sounds (auditory) and speech-associated mouth/lip movements (visual) from a speaker. Using PET where no scanner noise was present, brain regions involved in speech cue processing were investigated with the normal hearing subjects with no previous lip-reading training (<I>N</I>?=?17) carrying out a semantic plausibility decision on spoken sentences delivered in a movie file. Multimodality was ensured at the sensory level in all four conditions. Sensory-specific speech cue of one sensory modality, i.e., auditory speech (A condition) or mouth movement (V condition), was delivered with a control stimulus of the other modality whereas speech cues of both sensory modalities (AV condition) were delivered during bimodal condition. In comparison to the control condition, extensive activations in the superior temporal regions were observed bilaterally during the A condition but these activations were reduced in extent and left lateralized during the AV condition. Polymodal region such as left posterior superior temporal sulcus (pSTS) involved in cross-modal interaction/integration of audiovisual speech was found to be activated during the A and more so during the AV conditions but not during the V condition. Activations were observed in Broca's (BA 44), medial frontal (BA 8), and anterior ventrolateral prefrontal (BA 47) regions in the left during the V condition, where lip-reading performance was less successful. Results indicated that the speech-associated lip movements (visual speech cue) rendered suppression on the activity in the right auditory temporal regions. Overadditivity (AV?>?A?+?V) observed in the right postcentral region during the bimodal condition relative to the sum of unimodal speech conditions was also associated with reduced activity during the V condition. These findings suggested that visual speech cue could exert an inhibitory modulatory effect on the brain activities in the right hemisphere during the cross-modal interaction of audiovisual speech perception.</P>

      • Control of Optical Hysteresis in Block Copolymer Photonic Gels: A Step Towards Wet Photonic Memory Films

        <P><B>Abstract</B></P><P>Polystyrene‐<I>block</I>‐poly(2‐vinyl pyridine) (PS‐<I>b</I>‐P2VP) block copolymer photonic gels are fabricated that exhibit controllable optical hysteresis in response to a cyclic pH sweep. The optical hysteresis is tuned by controlling the ion‐pairing affinity between various anions and the protonated pyridinium ions on the P2VP block, which is highly dependent on the hydration energy of the ions, the dielectric constant of the solvent, and the ionic strength of the medium. The pH coercivity defining the magnitude of hysteresis of the photonic gels could be varied from 0.26 to 7.4. Photonic gel films with strong optical hysteresis can serve as wet photonic memory films where information can be cyclically recorded and erased at least 15 times and maintained for at least 96 h. The memory colors can be further tuned by selection of the copolymer molecular weight.</P>

      • New therapeutic agent against Propionibacterium acnes for the treatment of acne

        Background: Conventional therapeutic agents used for treatment of Acne are associated with various adverse effects necessitating development of safe and effective alternative therapeutic agents. In this study, a polyherbal formulation HPE/HVW was developed for treatment of Acne. Objectives: To evaluate antimicrobial activity of HPE/HVW. Methods: The disk diffusion susceptibility method was performed with P. acnes. After identifying the plant extracts as potential antibiotics through disk diffusion tests, broth dilution test was conducted to determine the minimal inhibitory concentration (MIC). Minimum bactericidal concentration (MBC) was also tested. Results: HPE/HVW formulation exhibited an antimicrobial activity against P. acnes. The best bacteriostatic effect of the HPE/HVW formulation was against P. acnes with MIC and MBC values ranging between 5.21 and 5.21 μg/mL, respectively. This activity was also comparable to that of Minocyclin with MIC and MBC values ranging between 4.17 and 16.67 μg/mL, respectively. Conclusion: Our data indicated the HPE/HVW mixture had a strong inhibitory effect on P. acnes. Further study justifies its role in the management of Acne vulgaris.

      • Development of natural cosmetic preservatives from plant

        Background: Parabens have been extensively used as cosmetic preservatives, because of their broad antimicrobial spectrum. As the concern of consumers on chemical materials grows highly and many research data showed side-effects of parabens, recently government announced that several paraben derivatives are going to be prohibited. New ingredients of preservatives need to be developed. Objectives: Studies to find the potential alternatives from plants extract as cosmetic preservatives are performed. Methods: The disk diffusion susceptibility method was performed with Staphylococcus aureus, Bacillus subtilus, Escherichia coli, Candida albicans, Pseudomonas aeruginosa. After identifying the plant extracts as potential preservatives through disk diffusion tests, broth dilution test was conducted to determine the minimal inhibitory concentration (MIC). Minimum bactericidal concentration (MBC) was also tested. Results: The plant extract named as HNE-0034 in our study showed weak antimicrobial effects against Staphylococcus aureus, Bacillus subtilus. NV-015 is also known as a component of a non-preservative hydrating ingredient. However, the combination of HNE-0034 and NV-015 displayed noticeable synergistic prevention against Staphylococcus aureus, Bacillus subtilus. Conclusion: Our data showed the possible application of combination of plant extracts and moisturizer as new preservatives. For the development of new preservatives in cosmetics, plants extract showed unlimited possibilities.
