        LiMn<sub>2</sub>O<sub>4</sub>의 Mn용출 현상 연구를 위한 전기화학적 Mn(II) 정량 분석법

        [ $LiMn_2O_4$ ] $Mn^{2+}$이온 용출현상 연구를 위한 간단하고 신속한 전기화학적 $Mn^{2+}$이온 분석법을 정립하였다. 이 분석법은 완충용액에서 $Mn^{2+}$이온이 $Mn^{4+}(MnO_2)$로 산화되는 원리를 이용한다. 최적조건 (pH 8.9 0.04 M $NH_3-NH_4Cl$ 완충용액 및 glassy carbon 작업전극)에서, Linear sweep voltammetry(LSV) 측정에 대해 $5{\mu}M-100{\mu}M$ (0.275-5.5 ppm) $Mn^{2+}$이온 범위에서, differential pulse voltammetry (DPV) 측정에 대해 $0.2{\mu}M-10{\mu}M$ (0.011-0.55 ppm) 범위에서 선형적 응답 특성을 확인하였다. 또한, 측정용액에 리튬 이차전지용 전해액 ($LiPF_6$, EC, EMC)이 첨가할 경우, 첨가량에 비례하여 $Mn^{2+}$ 이온의 산화 전류 감소하였는데, 이러한 감소의 주요 원인은 EMC보다는 $LiPF_6$와 EC성분임을 확인하였다. A simple and rapid electrochemical method for the quantitative analysis of $Mn^{2+}$ ion is demonstrated with a view to examine the $Mn^{2+}$ dissolution behavior of $LiMn_2O_4$. The method described herein is based on the oxidation reaction of $Mn^{2+}$ to $Mn^{4+}(MnO_2)$ in aqueous buffer solution. Under the optimum condition (pH 8.9 0.04 M $NH_3-NH_4Cl$ buffer solution and glassy carbon working electrode), the linear range of $5{\mu}M-100{\mu}M$ (0.275-5.5 ppm) [$Mn^{2+}$] is obtained for the Linear sweep voltammetry(LSV) and $0.2{\mu}M-10{\mu}M$ (0.011-0.55 ppm) [$Mn^{2+}$] for the differential pulse voltammetry (DPV), respectively. It is also noted that the oxidation reaction of $Mn^{2+}$ ion is reduced with increasing amount of the electrolyte ($LiPF_6$, EC, EMC) added to the measuring solution, which is found to be mainly due to $LiPF_6$ and EC rather than EMC.

        SPF Ktc : ICR 마우스의 자연발생 종양에 대한 병리학적 연구

        The study was carried out to obtain the basic data for types, incidence and histopathological features of the spontaneous tumors of the specific pathogen free (SPF) Ktc : ICR mice bred in a barrier system in Technology Research Center, Korea Research Institute of Chemical Technology. One hundred of the mice consisted of 50 males and 50 females were examined for 18 months. 1. The overall incidence rate of spontaneous tumors was 51(51%) of 100 heads tested. The male mice showed slightly higher incidence(28%) than the female(23%), and the incidence rate and the number of affected organs were increased with the increasing age of mice. 2. The incidence rate of primary tumor was 59(59%) of 100 heads tested, consisted of 30 cases (50.8%) of benign tumors and 29 cases(49.2%) of malignant tumors. Among the malignant tumors twenty cases were metastasized to various organs. 3. In tumor incidence rates by systems and organs, the male mice showed the high incidence rate in the liver (18%), hematopoietic system (16% ) and lung (14%), while the female mice, in the hematopoietic system(18%), lung(12%), liver(8%) and uterus(8%). 4. The tumors showing the particularly low incidence rates (<1.0%) were rhabdomyosarcoma in the skeletal muscle, malignant schwannoma in the peripheral nerve, cortical adenoma in the adrenal gland, transitional cell carcinoma in the urinary bladder, tubular cell adenoma in the kidney and adenoma in the pituitary gland and harderian gland.

        Caffeine이 N-bis(2-hydroxypropyl)nitrosamine과 sulfadimethoxine에 의해 유발된 갑상선 피막의 섬유성 증식에 미치는 영향

        Caffeine (1,3,7-trimethylxanthine), a central nervous system stimulant, is contained in various foods, beverages and over-the-counter medications. Sulfadimethoxine (SDM) is one of the anti-thyroid agents and induces proliferation of thyroid capsule in two stage thyroid carcinogenesis model using N-bis(2-hydroxypropyl)nitrosamine (DHPN). In this study, we examined the effect of caffeine on fibrous proliferation of thyroid capsule in DHPN and SDM-treated rats. Five-week-old male F344 rats were given a single subcutaneous injection of DHPN (2,800 mg/kg, body weight). Starting one week thereafter, SDM (1,000 ppm in drinking water) with or without caffeine (1,500 ppm in diet) was administered for 12 weeks. All animals were autopsied and histopathological examination of the thyroid glands was performed. Thyroid follicular proliferative changes were induced in all rats treated with DHPN+SDM. In addition, the proliferation of perithyroidal fibrous tissue and pleomorphic thyroid follicular cells within the capsule were observed in DHPN+SDM treated group. Caffeine would not be related to these lesions in this experimental condition. although pentoxifylline, a methyl xanthine derivative, has an anti fibrotic effects.

        1,3-Dichloropropanol 흡입에 의한 랫드간의 조직변화에 관한 연구

        1,3-Dichloro-2-propanol (1,3-DCP) is known as chloride chemicals and causes severe hepatotoxic agent. The Ito cells and Kupffer's cells of the liver in the 5 old F344 Rats were exposed to 1,3-DCP gas chamber for 6 hours/a day, 5 days/a week, and 13 weeks, in the 0, 5, 20, 80 ppm, respectively. After then the body weights, liver weights, and relative liver weight to body weight were measured, and the hepatic tissues were prepared by the routine and Immunostain method, and observed by the LM, and EM. In the results, there were severe body weight decrease (p<0.05) in the 80 ppm of the male and female rats. The relative liver weights to the body weight were increased relate with exposed 1,3-DCP concentration (P<0.001). Inflammatory cells, infiltration was observed at the perivascular area in the 20 ppm exposed group, and bilirubin pigment infiltration, bile duct hyperplasia, inflammation hepatocytic necrosis, fibrosis were observed in the 80 ppm exposure group. In the 80 ppm exposure group, disarrangement of the endothelial cells, erythrocytes and hepatic cell fragment in the Disse space and numerous migration macrophages were observed in the necrotic area by EM observation. In the immunostained hepatic tissues positive stained ED1 cells were extremely increased (P<0.05) in central vein area, but ED2 was weakly positive immunostained in the 80 ppm exposed group. Immunostained desmin was observed in the Ito cell. It was no difference in the low and medium exposed group but it was typical increase in the necrotic area. In conclusion, These results suggest that NOAEL of 1,3-DCP may be 5 ppm in rats and the Immunostained of desmin, ED1 and ED2 positive cells activated in the inflammatory liver were related to the exposure volume and density. The increase of the lto cells were related to the severe phagocytosis of the Kupffer's cells.

        Ktc:Sprague-Dawley 랫드의 안구에 자연발생한 amelanotic melanoma

        Two intraocular tumors were identified in low and medium dosed groups of a carcinogenicity study using 200 males and 200 females Sprague-Dawley rats in Screening & Toxicology Research Center, Korea Research Institute of Chemical Technology. The tumors were grossly observed as white or yellow, unilateral nodules. They were approximately $1-2{\times}3-5mm$ in size. The tumors located in the region of iris and/or ciliary body invaded peripheral cornea. The microscopic features were usually composed of spindle cells arranged in parallel, forming gently curving bundles or whorls. The spindle cells had poorly defined cell boundaries, scant to moderate cytoplasm, fusiform nuclei and indistinct nucleoli. Mitotic figures were rare and areas of necrosis were present. The spindle cells had positive immunoreactivity for S-100 protein and vimentin but negative for desmin, collagen and HMB-45 antibody. In special histochemical studies, the spindle cells react with Gomori's stain for argyrophih fibers, Prussian blue stain for iron but negative with Masson-Fontana's stain for melanin granules. Ultrastructurally, cytoplasmic premelanosomes were not observed in the tumor cells due to the poor preservation of tumor masses. Based on the results, the tumors were diagnosed as amelanotic melanoma.

        Si<sub>3</sub>N<sub>4</sub>-코팅 유/무기 복합 분리막을 통한 리튬이온전지용 분리막의 제조 및 평가

        리튬 이차전지의 대표적인 분리막인 polyethylene(PE) 분리막은 열에 의한 수축 및 기계적 파열의 단점을 가지고 있다. 본 연구에서는 이러한 기존 PE 분리막을 개선하기 위해 $Si_3N_4$ 코팅 분리막 (SCS, Silicon-nitride Coated Separator)을 제작하였다. $Si_3N_4$ 코팅이 분리막의 열적/기계적 수치안정성, 이온전도도, 및 전지의 출력 특성에 미치는 영향을 알아보았다. $Si_3N_4$ 분말을 polyvinylidene fluoride(PVdF) 결착재를 이용하여 PE 분리막의 한 쪽 면에 10 ${\mu}m$ 두께로 코팅하여 SCS를 제작하였다. SCS는 PE 분리막보다 $100{\sim}150^{\circ}C$에서 우수한 열적안정성을 나타냈으며, 특히 $150^{\circ}C$에서의 수축률은 10~20% 감소를 보였다. 또한, SCS의 인장강도는 PE 분리막에 비해 증가를 보였다. SCS는 PE 분리막에 비해 다소 낮은 이온 전도도를 보였지만, $LiCoO_2$/Li 코인전지의 C-rate(0.2~3C) 특성 평가에서는 유사한 결과를 보였다. Polyethylene (PE) separator is the most popular separator for lithium-ion batteries. However, it suffers from thermal contraction and mechanical rupture. In order to improve the thermal/mechanical dimensional stabilities, this study investigated the effects of $Si_3N_4$ coating. SCS (Silicon-nitride Coated Separator) has been fabricated by applying 10 ${\mu}m$-thick $Si_3N_4$/PVdF coating on one side of PE separator. SCS exhibits enhanced thermal stability over $100{\sim}150^{\circ}C$: its thermal shrinkage is reduced by 10~20% compared with pristine PE separator. In addition, SCS shows higher tensile strength than PE separator. Employing SCS hardly affects the C-rate performance of $LiCoO_2$/Li coin-cell, even though its ionic conductivity is somewhat lower than that of PE separator.

        청서 시상하부의 growth hormone-releasing factor 및 somatostatin 면역반응신경세포의 분포

        This study was attempted to investigate the topographical distribution, shape and immunoreactivity of growth hormone-releasing factor(GRF)- and somatostatin(SOM)-immunoreactive neurons in the hypothalamus of the Korean squirrels(Sciurus vulgalis coreae). For the light microscopical examination of immunohistochemistry, the brains were fixed with 4% paraformaldehyde solution by means of intracardiac perfusion. And the frozen sections($40{\mu}m$ thick) were stained immunohistochemically by ABC method. Distribution of GRF immunoreactive neurons($12-17{\mu}m$) was highest in the paraventricular nucleus, moderate in the periventricular and supraoptic nuclei, and low in the arcuate nucleus and lateral hypothalamic area. Their immunoreactive fibers were found very high in the median eminence, moderately in the supraoptic, paraventricular and periventricular nuclei, and low in the arcuate nucleus and lateral hypothalamic area. SOM immunoreactive perikarya($14-18{\mu}m$) were found moderately in the periventricular nucleus near the subependymal layer of the third ventricle, and low in the arcuate and suprachiasmatic nuclei. SOM immunoreactive fibers were found high in the median eminence, and moderately or low in the arcuate and periventricular nuclei.

        발효 인삼꽃 추출물의 경구 투여가 마우스 사이토카인 IL-6, TNF-α의 생성에 미치는 영향

        생체 내 실험에서 발효 인삼꽃 추출물(FM), 발효하지 않은 인삼꽃 추출물(FD)과 대조군으로 생리식염수를 2주간 마우스 체중 kg 당 100, 200 mg/kg B.W.의 농도로 마우스에 경구 투여한 후 LPS에 의해 활성화된 복강 대식세포가 분비하는 염증성 사이토카인 IL-6, TNF-${\alpha}$의 생성량을 측정하였다. 그 결과, IL-6는 발효 LPS로 자극한 경우, 두 가지 농도에서 모두 처리한 군에 비해 높은 증식능을 나타내었고, LPS로 자극한 결과, 특히 발효 인삼꽃 추출물 200 mg/kg B.W. 농도에서 유의적으로 낮은 IL-6 분비량을 보였다. TNF-${\alpha}$의 경우, 100 mg/kg B.W.와 200 mg/kg B.W. 두 농도 모두에서 LPS로 자극하지 않은 경우, 낮은 증식 효과를 보여주었고, 자극한 경우, 인삼꽃 시료를 첨가한 군이 대조군보다 낮은 TNF-${\alpha}$ 분비량을 보였으며, FM에서 FD보다 더 TNF-${\alpha}$를 억제하는 효과가 큰 것을 볼 수 있었다. 이상의 결과에 따르면 FD의 사이토카인 IL-6, TNF-${\alpha}$ 생성 효과는 200 mg/kg B.W. 농도 투여 시 효과적으로 면역 세포와 면역 기관의 주요 기능을 증진시킬 가능성이 있을 것으로 사료된다. 이러한 연구결과를 토대로 앞으로 인삼꽃 발효를 이용한 기능성 사료 개발과 더불어 산업적 측면에서 보다 긍정적인 효과를 얻을 수 있을 것으로 판단된다. Panax ginseng C.A. Meyer has been used as a traditional medicinal ingredient and the ginseng flower-buds also proved to have good medicinal properties. In this study, in order to enhance immune activities of ginseng flower-bud, the ginseng flower-bud extract was being fermented by Bacillus subtilis KCTC 1022 (BS), Lactobacillus plantarum KCTC3 and Saccharomyces cerevisiae strain CHY1011 (SC). Mice were orally administered daily for two weeks at two different concentrations (100 and 200 mg/kg B.W.). Treatment samples were water extracts of ginseng flower-buds (FD), water extracts of fermented ginseng flower-buds (FM) and controls for saline solution. Cytokine production (IL-6, TNF-${\alpha}$) either stimulated with LPS or not stimulated with LPS was detected by the ELISA assay when using the cytokine kit. Cytokine was statistically increased at supplemented groups with LPS in both the 100 and the 200 mg/kg B.W. and treatment with FM significantly decreased the LPS-induced TNF-${\alpha}$ and IL-6 production more than the treatment with FD. The results of this study may suggest that supplementation with FM increases the immune function by regulating cytokine production capacity for activated macrophages.
