        Type-specific Amplification of 5S rRNA from Panax ginseng Cultivars Using Touchdown (TD) PCR and Direct Sequencing

        Generally, the direct sequencing through PCR is faster, easier, cheaper, and more practical than clone sequencing. Frequently, standard PCR amplification is usually interpreted by mispriming internal or external regions of the target template. Normally, DNA fragments were eluted from the gel using Gel extraction kit and subjected to direct sequencing or cloning sequencing. Cloning sequencing has often troublesome and needs more time to analyze for many samples. Since touchdown (TD) PCR can generate sufficient and highly specific amplification, it reduces unwanted amplicon generation. Accordingly, TD PCR is a good method for direct sequencing due to amplifying wanted fragment. In plants the 5S-rRNA gene is separated by simple spacers. The 5S-rRNA gene sequence is very well-conserved between plant species while the spacer is species-specific. Therefore, the sequence has been used for phylogenetic studies and species identification. But frequent occurrences of spurious bands caused by complex genomes are encountered in the product spectrum of standard PCR amplification. In conclusion, the TD PCR method can be applied easily to amplify main 5S-rRNA and direct sequencing of panax ginseng cultivars.

        Evaluating the Immunological Adjuvant Activities of Carrageenan and Degraded Carrageenan

        Carrageena은 전세계적으로 안전한 식품첨가물로 승인되어 오랜 기간 다양한 식품, 기타 가공품에 사용되어지고 있다. 다른 한편으로, 이 Carrageenan은 동물 실험에서 염증 유도 물질로 확인되어 염증 유발 실험에 현재까지도 매우 빈번히 사용 되고 있다. 또한 이 Carrageenan을 고온과 강산에서 처리하여 부분적으로 분해한 degraded Carrageenan은 염증 유도 능이 Carrageenan 보다 더 강한 것으로 알려져 있다. 면역 보강제의 중요한 특성 가운데 하나는 선천면역(대표적으로 염증반응)의 활성화 인 것이 잘 알려져 있다. 그러나 현재까지 Carrageenan이나 degraded Carrageenan의 면역 보강제로서의 효과에 관하여 상세한 비교 연구는 수행되어 지지 않았다. 본 연구의 목적은 Carrageenan과 degraded Carrageenan의 면역 보강제로서의 효과를 비교 분석하는데 있다. 실험 동물은 마우스를 사용하였으며, 난 알부민을 항원으로, 피하면역을 수행하여 각각의 면역 보강제 효과를 항체 형성 정도로 조사하였다. Carrageenan이나 degraded Carrageenan 모두 항원 단독으로 면역한 것과 비교할 때 유의적으로 높은 IgG 생성 능을 보였다. 추가적으로 항원 특이적 IgG1과 IgG2a를 조사한 결과, 이들 Carrageenan, degraded Carrageenan은 본 실험에서 양성 대조 군으로 사용한 보강제, Complete Freund`s adjuvant와 비교 할 때 IgG2a 보다는 IgG1 생성 능이 높게 유도되는 것이 확인되었다. 이들 결과를 종합하면 염증유발 능이 보다 강한 degraded carrageenan의 면역 보강제 효과는 carrageenan과 유사한 정도로 확인되었으며, 이들 모두 IgG2 보다는 IgG1 생성 효과가 강한 것으로 나타났다. Carrageenan (CGN) has been used as a safe food additive for several decades. CGN has also been widely used to induce inflammation in various animal models. Likewise, degraded CGN (dCGN), which is produced by subjecting CGN to acid hydrolysis, also induces inflammation and does so more effectively than CGN. One of the most important characteristics of an immunological adjuvant is its ability to activate innate immunity. The immune-adjuvant effects of CGN and dCGN have not yet been studied in detail. The purpose of this study was to evaluate the immunological adjuvant activities of both CGN and dCGN, which was done by comparing the levels of an ovalbumin (OVA)-specific antibody after treatment with OVA in the absence or presence of CGN or dCGN in plasma from immunized mice. CGN and dCGN showed similar levels of adjuvant activity, as evidenced by increased antibody titer. Specifically, both CGN and dCGN significantly increased the levels of OVA-specific IgG, IgG1, and IgG2a antibodies in the plasma as compared with OVA alone (the control). However, compared to the positive control (Freund’s adjuvant), both CGN and dCGN caused greater increases in IgG1 than in IgG2a. These results suggest that CGN and dCGN have similar adjuvant activities and produce more IgG1 antibodies than IgG2a.

        본 논문에서는 선형 2-norm 공간에 대한 strict convexity와 strict 2-convexity의 특성들을 아래에 정의한 duality mapping types I(x,y)와 ??에 의해서 주어진다. 첫째, 선형 2-norm 공간이 strictly 2-convex 공간이 되기 위한 필요충분조건은 I(x,y)∩I(y,z)∩I(z,x)≠Φ 이면 어떤 양의 실수 α,β에 대해서 z=αχ+βy이고, 둘째, 선형 2-norm 공간이 strictly 2-convex 공간이기 위한 필요충분조건은 ??이면 어떤 양의 실수 α,β에 대해서 z=αχ+βy이다. 이러한 결과들은 몇개의 보조정리을 얻는다.

      • Effect of MLN8237, a Novel Aurora A Kinase Inhibitor, on the Spontaneous Fragmentation of Ovulated Mouse Oocytes

        Aurora A kinase is a mitotic serine/threonine kinase whose proposed functions include the maturation of centrosomes, G2/M transition, alignment of chromosomes at metaphase, and cytokinesis. In this study, we investigated the effect of MLN8237, an aurora A kinase inhibitor, on the postovulatory aging of oocytes based on the frequency of oocyte fragmentation, cdk1 kinase activity, and cyclin B degradation. The fragmentation of ovulated oocytes during prolonged culture was inhibited by treatment with MLN8237 in a concentration-dependent manner. The frequency of fragmented oocytes was significantly lower in oocytes treated with 2 ${\mu}M$ MLN8237 (13%) than in control oocytes (64%) after two days of culture. Most of the control (non-fragmented) oocytes (91%) were activated after two days of culture. In comparison, only 22% of the MLN8237-treated oocytes were activated; the rest of the oocytes (78%) were still in metaphase with an abnormal spindle and dispersed chromosomes. Next, cdk1 activity and the level of cyclin B were examined. The level of cyclin B and cdk1 activity in MLN8237-treated oocytes were nearly equal to those in control oocytes. Our results indicate that MLN8237 inhibited the fragmentation of ovulated oocytes during prolonged culture, although it blocked the spontaneous decrease in activity of cdk1 and degradation of cyclin B. This mechanism of inhibition is different from that in oocytes treated with nocodazole, which have high levels of cdk1 activity and cyclin B.

      • On Ordered Normal Bases and Ordered Compactifications

        T₁-공간의 Wallman-type Compactification을 일반화 시킨 순서 Compactification을 취급하고 있다. 순서위상공간(X, τ, ??)의 순서 normal base의 개념을 소개하여 X가 dense embedding되는 compact 순서위상공간(β0(X), τ*, ??)을 만들었고, 순서 normal base βo을 가지고 연속순서를 가진 compact순서위상공간은 (β0(X), τ*, ??)와 iseomorphic임을 밝혔다. The purpose of this paper is to construct an ordered compactification for a topological ordered space with semicontinuous order which generalizes the Wallman-type compatification of a T₁-space. In order to do this, we introduce the concept of an ordered normal base on a topological ordered space, and obtain that for a topological ordered space (X, τ, ??) with semicontinuous order having an ordered normal base β0, there exists a compact topological ordered space (β0(X), τ*, ??) with continuous order, into which X is densely embeded. It is shown that compact topological ordered space (X, τ, ??) with a continuous order having an ordered normal base β0 is iseomorphic with (β0(X), τ*, ??). Hong [1] has introduced the concept of 0-completely regular filters on a completely regular ordered space in order to construct the ordered compactification. Moreover, in [4], Y.S. Park has introduced the concept of bifilter on a topological ordered space and constructed an ordered compactification for a convex ordered topological space with semicontinuous order. In this paper, we introduce the concept of an ordered normal base for a topological ordered space (X, τ, ??) with semicontinuous order and obtain the compact topological ordered space with continuous order which is an ordered compactification for the space (X, τ, ??). For a topological ordered space we refer to [3, 5]. we assume all our topological ordered spaces to be topological ordered space with semicontinuous order. Let β₁ and β₂ be the families of sets. Then we write β₁∪β₂={A|A∈β₁ or A∈β₂} β₁∪β₂={A∩B|A∈β₁ and B∈β₂}.

        Lavandulyl Flavanones from <i>Sophora flavescens</i> Protect Mouse Hippocampal Cells against Glutamate-Induced Neurotoxicity <i>via</i> the Induction of Heme Oxygenase-1

        <P>Lavandulyl flavanones of <I>Sophora flavescens</I> roots are anti-malarial, anti-inflammatory, and cytotoxic. Here, we examined whether four lavandulyl flavanones, (2<I>S</I>)-2′-methoxykurarinone (1), sophoraflavanone G (2), leachianone A (3), and (−)-kurarinone (4), isolated from <I>S. flavescens</I> could protect HT22 immortalized hippocampal cells against glutamate-induced oxidative stress. Compounds 1 and 2 induced the expression of heme oxygenase (HO)-1 and increased HO activity dose- and time-dependently. These two compounds also suppressed glutamate-induced reactive oxygen species generation in HT22 cells, whereas compounds 3 and 4 were not protective. These two lavandulyl flavanones (compounds 1, 2) may protect against glutamate-induced neurotoxicity <I>via</I> HO-1 induction.</P>
