Two directional ring-closing metathesis (RCM) was applied successfully to the synthesis of 4'-vinylated carbocyclic nucleoside analogues from the trivinyl intermediate 12, which was readily made using a sequential Claisen rearrangement and ring-closing metathesis (RCM) starting from Weinreb amide 5. An antiviral evaluation of the synthesized compounds against various viruses such as HIV, HSV-1, HSV-2 and HCMV revealed that the guanine analogue 20 have moderate anti-HIV activity in the MT-4 cell line (EC50 = 10.2 m M).

Novel mercaptophenyl carbocyclic C-nucleoside analogue was synthesized via a cyclopentenol intermediate 10, which was prepared using a sequential [3,3]-sigmatropic rearrangement and ring-closing metathesis (RCM). Friedel-Crafts alkylation was then used to couple the thiophenol.
Stereoselective Synthesis of a Novel Cyclohexene Version of Carbovir

This paper describes a racemic and stereoselective synthetic route for a novel cyclohexenyl carbocyclic adenine analogue. The required stereochemistry of the target compound was controlled using a stereoselective glycolate Claisen rearrangement followed by a-chelated carbonyl addition. The introduction of 6-chloropurine was achieved using Mitsunobu conditions, and further modifications of the corresponding heterocycle gave the target cyclohexenyl nucleoside.

As a part of an ongoing effort to discover inhibitors of the Hepatitis C Virus RNA replication, we describe here the first synthetic route of 1'(α),2'(β)-C-dimethyl carbocyclic adenine analogue. The key intermediate cyclopentenyl alcohol 8(α) was prepared from aldehyde 4 using ring-closing metathesis (RCM) as a key reaction. Coupling of 8(α) with nucleosidic base via the regioselective Mitsunobu reaction followed by stereoselective dihydoxylation and deprotection afforded the target carbocyclic adenine analogue 12.

The influence of phenological stages onchemical composition of Scutellariae Radix (SR), theroot of Scutellaria baicalensis Georgi, was investigated. In order to deal with a large quantity of samples, arapid ultra-performance liquid chromatographic (UPLC)was first developed and validated for the simultaneousquantification of five flavonoids, namely baicalin(baicalein-7-O-b-D-glucuronide, BG), wogonoside(wogonin-7-O-b-D-glucuronide, WG), baicalein (BA),wogonin (WO), oroxylin A (OA) in the samples. Goodlinearity was obtained in the range of 0.742–389 ng(r2[0.9999) and satisfactory recoveries were obtained(101.72–104.56 %) with the RSD value below 5.0 %,for all analytes. Also, extraction conditions were optimizedto obtain maximum extractive contents of thefive flavonoids. Content variations of the five activeingredients in 225 samples from three different originswere investigated in five major phenological periods. Itwas found that the effect of phenology on the contentsof the tested five flavonoids was similar in the threeorigins. The contents of flavone O-glycosides, i.e., BGand WG accumulated to the highest level in leafexpansion period, while flavonoid aglycones, i.e., BA, WO and OA appeared a maximum concentration inflowering period. The UPLC method established in thisstudy was rapid and of good accuracy, repeatability andresolution, and hence can assist in the quality control ofSR.
ERTC Involved in HCC Growth and Metastasis through p53 and WNK1 Signaling Pathway

Aims: ERTC has been shown to be an important player in the regulation of centrosome/microtubule dynamics during mitosis and found to be deregulated in a variety of human malignancies. But, the functional role and signaling pathway of ERTC in hepatocellular carcinoma (HCC) remains elucidative. Methods: ERTC mRNA and miR-1324 expression was confirmed by real-time PCR analysis in human liver sample. ERTC protein expression was investigated by immunoblotting analysis in HCC cell lines and HCC tissues. The ERTC expression was infected with adenovirus, or knockdown by a delivery with short hairpin RNA (shRNA) or treatment with the potential ERTC inhibitor KHS101 hydrochloride, in Huh7, HepG2, SH-J1 and Alexander cell lines; cells were analyzed for proliferation, migration, and invasion. Tumor metastasis by ERTC and WNK1 was tested in vivo mouse model. Moreover, signaling pathways involved in invasion and metastasis were analyzed by Western blot. Results: ERTC was abundantly expressed in HCC cell lines and HCC tissues compared with non-tumor HCC tissues. ERTC mRNA was positively correlated with the tumor size, worsening differentiation status, lack of fibrous capsule formation, microvessel invasion, intrahepatic metastasis, AFP level and advanced stage of HCC. Knockdown of ERTC in SH-J1 and Alexander cells suppressed migratory and invasive behavior as well as the expression of EMT related markers. Silencing ERTC enhanced p53 expression and decrease phosphorylation of WNK1 in SH-J1 and Alexander cells. The cells with knockdown of ERTC using target shRNA reduced tumor metastasis in a lung metastasis mouse model. Moreover, knockdown of WNK1 also inhibit tumorigenicity and metastatic ability were examined in an orthotopic animal model. miR-1324 interacted with the 3’ untranslated region (3’ UTR) of ERTC. Levels of miR-1324 were correlated inversely with ERTC mRNA and in human HCC samples. Conclusions: Therefore, ERTC may be involved in HCC growth and metastasis through p53 and WNK1 signaling pathway, which may be useful therapeutic targets.
