      • Induction of heme oxygenase-1 protects against podocyte apoptosis under diabetic conditions

        Heme oxygenase-1 (HO-1) is an anti-oxidant enzyme normally upregulated in response to oxidant injury. Here we determined the role of HO-1 in podocyte apoptosis in glomeruli of streptozotocin-treated rats and in immortalized mouse podocytes cultured in media containing normal or high glucose. HO-1 expression, its activity, the ratio of Bax/Bcl-2 protein, and active caspase-3 fragments were all significantly higher in isolated glomeruli of diabetic rats and in high glucose–treated podocytes. These increases were inhibited by zinc protoporphyrin treatment of the rats or by HO-1 siRNA treatment of the podocytes in culture. The number of apoptotic cells was also significantly increased in the glomeruli of diabetic rats and in high glucose–treated podocytes. Inhibition of HO-1 accentuated the increase in apoptotic cells both in vivo and in vitro. Our findings suggest that HO-1 expression protects against podocyte apoptosis under diabetic conditions.

        Upregulation of heme oxygenase-1 by ginsenoside Ro attenuates lipopolysaccharide-induced inflammation in macrophage cells

        Background: The beneficial effects of ginsenoside species have been well demonstrated in a number of studies. However, the function of ginsenoside Ro (GRo), an oleanane-type saponin, has not been suffi- ciently investigated. Thus, the aim of the present study was to investigate the anti-inflammatory effects of GRo in vitro using the Raw 264.7 mouse macrophage cell line treated with lipopolysaccharide (LPS), and to clarify the possible mechanism of GRo involving heme oxygenase-1 (HO-1), which itself plays a critical role in self-defense in the presence of inflammatory stress. Methods: Raw 264.7 cells were pretreated with GRo (up to 200mM) for 1 h before treatment with 1 mg/ mL LPS, and both cell viability and inflammatory markers involving HO-1 were evaluated. Results: GRo significantly increased cell viability in a dose dependent manner following treatment with LPS, and decreased levels of reactive oxygen species and nitric oxide. GRo decreased inflammatory cytokines such as nitric oxide synthase and cyclooxygenase-2 induced by LPS. Moreover, GRo increased the expression of HO-1 in a dose dependent manner. Cotreatment of GRo with tin protoporphyrin IX, a selective inhibitor of HO-1, not only inhibited upregulation of HO-1 induced by GRo, but also reversed the anti-inflammatory effect of GRo in LPS treated Raw 264.7 cells. Conclusion: GRo induces anti-inflammatory effects following treatment with LPS via upregulation of HO-1.

      • Protective Effects of Hyperoside against Carbon Tetrachloride-Induced Liver Damage in Mice

        <P>In this study, the hepatoprotective effects of hyperoside (<B>1</B>), a flavonoid glycoside isolated from <I>Artemisia capillaris</I>, have been examined against carbon tetrachloride (CCl<SUB>4</SUB>)-induced liver injury. Mice were treated intraperitoneally with vehicle or <B>1</B> (50, 100, and 200 mg·kg<SUP>−1</SUP>) 30 min before and 2 h after CCl<SUB>4</SUB> (20 μL·kg<SUP>−1</SUP>) injection. Levels of serum aminotransferases were increased 24 h after CCl<SUB>4</SUB> injection, and these increases were attenuated by <B>1</B>. Histological analysis showed that <B>1</B> prevented portal inflammation, centrizonal necrosis, and Kupffer cell hyperplasia. Lipid peroxidation was increased and hepatic glutathione content was decreased significantly after CCl<SUB>4</SUB> treatment, and these changes were reduced by administration of <B>1</B>. Protein and mRNA expression of tumor necrosis factor-α (TNF-α), inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and heme oxygenase-1 (HO-1) and nuclear protein expression of nuclear factor erythroid 2-related factor 2 (Nrf2) significantly increased after CCl<SUB>4</SUB> injection. Compound <B>1</B> suppressed TNF-α, iNOS, and COX-2 protein and mRNA expression and augmented HO-1 protein and mRNA expression and Nrf2 nuclear protein expression. These results suggest that <B>1</B> has protective effects against CCl<SUB>4</SUB>-induced acute liver injury, and this protection is likely due to enhancement of the antioxidative defense system and suppression of the inflammatory response.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jnprdf/2011/jnprdf.2011.74.issue-5/np200001x/production/images/medium/np-2011-00001x_0006.gif'></P>

      • Azithromycin으로 치료한 임신 중 쓰쓰가무시병 1예

        We report a case of tsutsugamushi disease in a 26 year-old pregnant woman who was treated with azithromycin. Her gestation period was 27 weeks and she admitted with fever, rash, and eschar on the right shoulder. Currently recommended medications for the treatment of scrub typhus are doxycycline or chloramphenicol. But, these drugs are class D drugs according to the U.S. Food and Drug Administration (FDA) Fetal Risk Summary, so they couldn't be used to treat pregnant women. Recently, a few case reports suggested that azithromycin, a relatively new macrolide antibiotic, was effective and safe for the treatment of scrub typhus in pregnant women. And, there is no evidence that azithromycin causes harmful effects to the developing fetus or to children. On the basis of current in vivo test that confirms the effectiveness of azithromycin, it may be the drug of choice for the treatment of scrub typhus in pregnant women and children.         The laser welding process is one of the most advanced manufacuring technologies owing to its high speed and deep penetration. Welding characteristics of Inconel 600 Alloy using a continuous wave Nd:YAG laser are experimentally investigated. The major process parameters studied in the present laser welding experiment were position of focus, laser power, travel speed. The gap and offset maintained as small as possible. Optical microscope were used to investigate the microstructures of the welded zone. The follow conclusions can be drawn the laser power and travel speed have a pronounced effect the fusion zone size and shape.

      • 디지털 무선 모뎀용 적응 결정귀환 등화기 설계

        This work describes an ASIC implementation of adaptive decision-feedback equalizer (ADFE) which is applicable to wide band digital wireless modems. A new approach based on redundant binary (RB) arithmetic is devised for an efficient implementation of complex-number filter which is a key functional block of ADFE. The proposed RB complex-number filter structure reduces the critical path delay of ADFE. as well as leads to a more compact realization than conventional methods. Also, the carry-propagation free (CPF) operations of the RB arithmetic enhance its speed. To demonstrate the proposed approach, a prototype chip set which is composed of feed-forward equalizer module (FFEM) and decision-feedback equalizer module (DFEM) has been designed. Each module contains two complex-number filter taps along with their coefficient update circuits, and can be cascaded to implement longer filter taps for high bit-rate applications. The chip set was modeled and verified using VHDL. and synthesized using 0.8-㎛ sea-of-gate (SOG) array library.

      • 도시거주자의 혈중 카드뮴 농도와 간 및 신장 기능과의 상관성 평가

        The possibility that liver and kidney function is adversely affected by current levels of environmental exposure to cadmium (Cd) as investigated in adult men and women in the general population in Seoul. From February to August in 2001, blood and morning spot urine samples were collected from 136 not occupationally exposed group (age range 20~75 years) at 4 survey sites throughout Seoul. Liver and kidney function parameters in serum and urine were examined by conventional methods. The questionnaire included factors, i.e. sex, age, smoking, alcohol, diet habit etc. The geometric mean values for Cd in blood (Cd-B) were 1.43 μg/l. It seemed prudent to conclude that liver and kidney function as not disturbed by the current environmental exposure to Cd in study.

        The purpose of this study is to provide some basic data which can be helpful for improving the methods of mathematics teaching-learning emphasizing the affective factors by investigating the effect on affective characteristics and mathematical problem solving ability of the students on the elementary school sixth grade. In order to achieve these purpose, the following research problems were set up for the present study: The results of this study are as follows: First, it was appeared that there was not any difference on attitude on mathematics, belief, self-confidence and the degree of mathematical problem solving ability by sex. Second, it was appeared that there was some statistical meaningful difference on attitude on mathematics, belief, self-confidence and the degree of mathematical problem solving ability according to high middle low ability on mathematics which the students think that they have. Based on the results of this study, I'd like to give some suggestions as followings to provide some help to teaching and learning mathematics. First, the succeeding studies are needed by selecting different population from the population of this study, classifying low-ranking factors consisting of affective characteristics differently from this study, compounding each low-ranking factors in various manners like using new items, etc. Second, the succeeding studies are needed in this part, because what mathematical belief teachers have mathematics can have an effect on students' mathematical belief and action as well as on teachers' thinking process and action in the class. Third, the succeeding studies are needed to develop concrete field study and program which can be helpful for improving actually students' affective behavior characteristics in school field.

      • 複合組織鋼의 母相粒徑과 軸斷面形狀이 破壞擧動에 미치는 影響

        The weight reduction and reliability, stability of various steel materials used in industrial machinery have become a great importance as industry developes. For this reason a study on dual phase steel has been performed by the reporter with great interest. It was pointed out that fatigue fracture behaviours of MEF dual phase steel were dependent upon a unit of ferrite grain size when the plastic constraint effect and the internal stress were constant. However, when the plastic constraint effect and the internal stress were constant, fracture behaviours of MEF dual phase steel might be expected to depend upon the ferrite grain size and the shape of section. Specimens were prepared the combined microstructures having different size of ferrite grain which has approximatly the constant plastic constraint effect and the constant internal stress through careful heat treatments. Each of tensile specimens and fatigue specimens was conducted by universal testing machine and rotary bending fatigue test machine. They were prepared to be able to apply the stress intensity factor equations(K₁=σ? πℓ. F(?): solid shaft, K₁=k₁.k₂σ? πℓ:hollow shaft). The relationship between fatigue crack length, fatigue crack propagation velocity, stress intensity factor and scanning electron microscope investigation of tensile fracture surface and fatigue fracture surface was investigated during the test. The results of the experiment are as follows. 1. Fracture behaviours of MEF dual phase steel with the constant plastic constraint effect and the constant internal stress depend on great and small size of ferrite grain. It has been found that Hall-petch equation is applied dual phase-poly crystallization. 2. The crack propagation length and number of cycles of hollow specimen and solid specimen approaches nearly and makes a gentle slope in case of small ferrite grain size under low stress level. 3. The difference of fatigue crack propagation velocity is large about the influence of shape of specimen at the same △K value under high stress level and that is approached under low stress level. 4. It can be had for light weighted purpose as use of hollow shaft with MEF dual phase and MEF dual phase steel demands minute ferrite grain size under low stress level. 5. It has been found that the shape of striation changes according to shape difference of specimen. The space of striation is narrow as the ferrite grain size and the stress level are small.
