Effectiveness of Green Tea Tannin on Rats with Chronic Renal Failure

The effects of green tea tannin on nephrectomized rats were examined. There were increases in blood urea nitrogen, serum creatinine, and urinary protein, and a decrease in creatinine clearance in the nephrectomized control rats, whereas better results for these parameters were obtained in rats given green tea tannin after nephrectomy, demonstrating a suppressed progression of the renal failure. When the renal parenchyma was partially resected, the remnant kidney showed a decrease in the activity of radical scavenger enzymes. Green tea tannin, however, was found to lighten the kidney under such oxidative stress. Mesangial proliferation and glomerular sclerotic lesions, which were conspicuous in the rats that were not given green tea tannin after nephrectomy, were also relieved.

The effect of magnesium lithospermate B purified from Salviae Miltiorrhizae Radix on the kinin-prostaglandin E_2 system was investigated using kidney slices and isolated kidney microsomes. Magnesium lithospermate B markedly increased the amount of thiobarbituric acid-reactive substances produced by incubation of arachidonic acid in renal slices. Enhancement of arachidonic acid-mediated oxygenation of 1,3-diphenylisobenzofuran was also observed in renal microsomes from rats treated with magnesium lithospermate B, indicating an increase of prostaglandin biosynthesis. Furthermore, magnesium lithospermate B significantly increased the synthesis of prostaglandin E_2 in renal slices, but the magnesium lithospermate B-mediated response was blunted with kinin antagonist. On the basis of the above results, it is apparent that magnesium lithospermate B exerts an influence on the prostaglandin system via kinin receptors.

인삼(人蔘)사포닌중 소미량성분인 $ginsenoside-Rf,\;-Rg_2$ 및 $-Rh_1$을 조제용(造製用), 준조제용(準造製用) 및 분석용(分析用) HPLC를 사용(使用)하여 분리(分離)하였다. 본(本) 방법(方法)은 신속하며 이들 소미량 ginsenoside의 분리(分離) 및 동정(同定)에 매우 유효(有效)하였으며 재(再)순환 방식을 사용하여 달성되었다. The minor components of $saponin-ginsenoside-Rf,\;-Rg_2\;and\;-Rh_1$ were isolated from Panax ginseng C.A. Meyer by preparative, semi-preparative and analtical high performance liquid chromatography. The rapid method developed in this work was proved to be very effective in separation and isolation of these minor ginsenosides. A further isolation was achieved by using the recycling technique.
Ursolic Acid Inhibits Aflatoxin B_1-Induced Mutagenicity in a Salmonella Aassay System

An attempt was made to isolate the active component of Eriobotrya japonica, which inhibits aflatoxin B_1-induced mutagenicity in the Salmonella assay system. The number of revertants per plate was significantly decreased when a MeOH extract of Eriobotrya japonica was added to the assay system using Salmonella typhimurium TA 100 or TA98. Furthermore, we examined the effect of each fraction purified from the MeOH extract, and an EtOAc fraction was found to be the most effective. Ursolic acid isolated from the EtOAc fraction makedly and significantly decreased the numbers of Salmonella typhimurium TA100 revertants per plate, thus showing antimutagenic activity.

인삼의 유효약리성분으로 밝혀진 saponin중의 각 ginsenosides를 효과적이고 능률적으로 분리하기 위하여 대량분취전용 고속액체 chromatograph인 preparative HPLC의 응용을 검토하였다. 조(粗) saponin획분을 preparative HPLC인 Prep LC/system-500를 사용하여 부분분획을 하고 각 획분에 함유되어 있는 ginsenosides의 조성을 Analytical HPLC로 동정한 후 Semi-preparative HPLC를 사용하여 인삼주성분 saponin을 단리했다. 그 결과 인삼 주성분 saponin인 $ginsenoside-Rb_1,\;-Rb_2,\;-Rc,\;-Rd,\;-Re$ 및 $-Rg_1$은 약 20 mg / 2.0 ml / injection으로 chromatography를 행하여 $300{\sim}400mg/day$로 대량분취가 가능하였다. 따라서 ginsenosides의 약리 및 임상효능 연구에 크게 기여하게 될 것이다. Relatively large quantity of the major components of saponin, $ginsenoside-Rb_1,\;-Rb_2,\;-Rc,\;-Rd,\;-Re\;and\;-Rg_1$ from Panax ginseng C.A. Meyer were isolated using preparative and semipreparative high performance liquid chromatography, and analyzed by analytical HPLC. The application of HPLC for isolation of ginsenosides was not only very effective for rapid analysis but also reduced the isolation time. The isolation capacity of pure ginsenosides was $30{\sim}50mg/hr$.
Antitumor Effects of Ursolic Acid Isloated from the Leaves of Eriobotrya japonica

Mice bearing experimentally transplanted sarcoma showed a higher survival rate after administration of ursolic acid isolated from Eriobotrya japonica than untreated control mice, and decrease in tumor weight was also found. These antitumor effects were also observed in in vitro experiments using sarcoma 180 cells.

During aging, its chance of tissue damage by active oxygen species increases, which causes the abnormal aging as well as diseases like cancer. In biological system, there are enzymes such as superoxide dismutase, catalase and glutathione peroxidase which scavenge active oxygen species as well as antioxidant agents such as ceruloplasmin, albumin and nonprotein-bound SH including glutathione related to defense mechanism. In this study, superoxide dismutase, catalase, serum albumin, protein-bound SH and nonprotein-bound SH levels were increased in SAM-R/1 mice treated with 2.5 or 5.0 ㎎/㎏ ginsenoside Rb₂. But malondialdehyde which is the product of lipid peroxidation was significantly decreased. These results suggest that increase of antioxidants by ginsenoside Rb₂ results in the protective effects against active oxygen species.
