T cells are an important line of defense against infections. Following antigen recognition they become activated and start to express the insulin receptor (InsR), which helps to cover the cells`s increased energy demand. Here we report that full activation of T cells could not be achieved when the InsR was ablated in transgenic knock-down rats. This was refi ected by a delayed up-regulation of CD25, a reduced secretion of pro-infiammatory cytokines and a diminished proliferation of CD4+ T cells. Interestingly, the observed effects were unrelated to differences in gene expression but rather seemed to be caused by alterations at the post-transcriptional level. CD8+ T cells, which are central to the clearance of viral infections, were also affected by the absence of the InsR. In particular their cytotoxic activity against foreign antigen was signifi cantly reduced. Preliminary experiments further suggested that adaptive immune responses in vivo were dampened when the InsR was absent in T cells. Collectively, our fi ndings indicate that insulin signaling fosters T cell activation. We would therefore expect that various T cell functions are compromised in patients with type 2 diabetes mellitus, which might explain why they are at greater risk of infections.
A redshifted codon-optimized firefly luciferase is a sensitive reporter for bioluminescence imaging

Bioluminescence imaging has evolved as a powerful tool for monitoring biological processes in vivo. As transmission efficiency of light through tissue increases greatly for wavelengths above 600 nm we examined whether a redshifted codon-optimized firefly luciferase (${\lambda}_{max}$ = 615 nm) could be successfully employed as a sensitive reporter in mammalian cells. To this end, unmodified codon-optimized luciferase (${\lambda}_{max}$ = 557 nm) as well as the red-emitting S284T mutant luciferase were expressed simultaneously in human glioma cells in vitro as well as in quadriceps muscles of mice in vivo. We show here that activity of the redshifted enzyme in human glioma cell culture approached approximately one-fourth of that seen with the unmodified enzyme. In contrast, light emission by the red-emitting luciferase in vivo was generally more efficient than that produced by its unmodified counterpart, most likely due to reduced absorption of red light by tissue. The mean ratio of light emission produced by the redshifted luciferase to that of the unmodified enzyme in vivo was~3. Application of this new redshifted luciferase together with other optical reporters may be of considerable importance to biological research as it allows for imaging of deeper tissues as well as simultaneous monitoring of two molecular events in vitro and in vivo if appropriate filter sets are employed.

Background: Working despite feeling ill – presenteeism – is a widespread behavioral phenomenon. Previous research has shown that presenteeism is influenced by various work-related and personal factors. It's an illness behavior leading to a range of negative but also positive consequences. Due to coronavirus disease 2019 (COVID-19) pandemic, remote work has become the “new normal” for many employees. But so far, little is known about presenteeism in remote work. This study aims to investigate presenteeism in remote work by looking at the extent of remote presenteeism, differences to presenteeism in on-site work, and associated factors. Methods: A nationwide cross-sectional online survey was conducted in Germany with N = 233 participants. Data were analyzed using descriptive statistics, t-tests, and correlation analysis. Results: The results reveal that presenteeism is prevalent in remote work x̅ = 4.13 days (Md = 3; D = 2; s = 4.95). A low ability to detach from work (r = -.17; p = .005) and low supervisor support (r = -.14; p = .02) is associated with more remote presenteeism days. Remote working conditions seem to facilitate presenteeism. Conclusion: This study provides empirical insights into a subject area of great societal relevance. The results show that awareness should be raised for presenteeism in remote work. It should be regarded as a behavior that can be functional or dysfunctional, depending on the individual situation. Supervisor support and detachment should be fostered to help reduce dysfunctional presenteeism. Promotion of health literacy might help remote workers to decide on a health-oriented illness behavior. Further research is vital to analyze to what extent and under which circumstances presenteeism in remote work is (dys)functional and to derive clear recommendations.
건강하고 활력 있는 고령화를 위한 독일의 지역사회 정책

인구 고령화의 부작용을 억제하고 시민의 수요를 충족할 수 있는 여건을 조성하려면 지방자치단체가 인구 고령화로 인한 변화를 해결하고 시민의 관심사에 맞춰 지역 구조를 조정해야 한다. 이와 관련해 지자체가 고려할 수 있는 선택지를 결정하는 것은 연방정부 및 주정부 간 협력과 재정 자원이다. 그러나 이를 계획하는 과정에서는 지역과 비공식 단체의 역할과 노력이 더 강조된다. 계획의 이행 과정에는 지역복지협회 대표, 시민, 행정부 관계자 등 다양한 이해관계자가 참여한다. 그 결과, 협력적 통합사회공간계획에 대한 논의가 부상하고 있다. 협력적 통합사회계획은 책임감 있는 주체들의 네트워크와 참여를 기반으로 하여 고령화 사회의 불평등을 줄이고 사회의 중심에서 시민이 자기 결정권이 있는 노후를 긍정적으로 맞이할 수 있도록 지원한다.
