      • Enhanced Electron-Transfer Reactivity of Nonheme Manganese(IV)–Oxo Complexes by Binding Scandium Ions

        <P>One and two scandium ions (Sc<SUP>3+</SUP>) are bound strongly to nonheme manganese(IV)–oxo complexes, [(N4Py)Mn<SUP>IV</SUP>(O)]<SUP>2+</SUP> (N4Py = <I>N</I>,<I>N</I>-bis(2-pyridylmethyl)-<I>N</I>-bis(2-pyridyl)methylamine) and [(Bn-TPEN)Mn<SUP>IV</SUP>(O)]<SUP>2+</SUP> (Bn-TPEN = <I>N</I>-benzyl-<I>N</I>,<I>N</I>′,<I>N</I>′-tris(2-pyridylmethyl)-1,2-diaminoethane), to form Mn<SUP>IV</SUP>(O)–(Sc<SUP>3+</SUP>)<SUB>1</SUB> and Mn<SUP>IV</SUP>(O)–(Sc<SUP>3+</SUP>)<SUB>2</SUB> complexes, respectively. The binding of Sc<SUP>3+</SUP> ions to the Mn<SUP>IV</SUP>(O) complexes was examined by spectroscopic methods as well as by DFT calculations. The one-electron reduction potentials of the Mn<SUP>IV</SUP>(O) complexes were markedly shifted to a positive direction by binding of Sc<SUP>3+</SUP> ions. Accordingly, rates of the electron transfer reactions of the Mn<SUP>IV</SUP>(O) complexes were enhanced as much as 10<SUP>7</SUP>–fold by binding of two Sc<SUP>3+</SUP> ions. The driving force dependence of electron transfer from various electron donors to the Mn<SUP>IV</SUP>(O) and Mn<SUP>IV</SUP>(O)–(Sc<SUP>3+</SUP>)<SUB>2</SUB> complexes was examined and analyzed in light of the Marcus theory of electron transfer to determine the reorganization energies of electron transfer. The smaller reorganization energies and much more positive reduction potentials of the Mn<SUP>IV</SUP>(O)–(Sc<SUP>3+</SUP>)<SUB>2</SUB> complexes resulted in remarkable enhancement of the electron-transfer reactivity of the Mn<SUP>IV</SUP>(O) complexes. Such a dramatic enhancement of the electron-transfer reactivity of the Mn<SUP>IV</SUP>(O) complexes by binding of Sc<SUP>3+</SUP> ions resulted in the change of mechanism in the sulfoxidation of thioanisoles by Mn<SUP>IV</SUP>(O) complexes from a direct oxygen atom transfer pathway without metal ion binding to an electron-transfer pathway with binding of Sc<SUP>3+</SUP> ions.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jacsat/2013/jacsat.2013.135.issue-24/ja403965h/production/images/medium/ja-2013-03965h_0014.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ja403965h'>ACS Electronic Supporting Info</A></P>

      • Gastro-protective effects of Brassica oleracea var. capitata extracts in HCl-Ethanol Induced gastritis rat model

        The purpose of this study was to evaluate the gastro-protective effect of cabbage extract (sulforaphane content was 5.19 mg/L and S-methylmethionine was 469.28 μg/L). In vitro study, the lipopolysaccharide (LPS)-treated group had an increased nitic oxide (NO) and interleukin (IL)-6 activity compared to normal group, and NO and IL-6 level was decreased when the cabbage extract was treated in the manner of dose. In the gastritis-induced mice group, severe hemorrhagic mucosal damage was observed in the entire area of the stomach due to bleeding marks. The degree of gastric mucosal damage was decreased in dose dependent manner. Mucosal erosion in the gastritis model was also ameliorated in the cabbage extract administration group. The blood level of histamine was significantly increased in the gastritis-induced animal model, and the secretion of histamine was reduced in the 23 mg/L treated group of cabbage extract. In conclusion, cabbage extract can help inhibit gastrointestinal disorders by improving the protective barrier.         Glial GABA, synthesized by monoamine oxidase B, mediates tonic inhibition

        <P><B></B></P><P>GABA is the major inhibitory transmitter in the brain and is released not only from a subset of neurons but also from glia. Although neuronal GABA is well known to be synthesized by glutamic acid decarboxylase (GAD), the source of glial GABA is unknown. After estimating the concentration of GABA in Bergmann glia to be around 5–10 m<SMALL>m</SMALL> by immunogold electron microscopy, we demonstrate that GABA production in glia requires MAOB, a key enzyme in the putrescine degradation pathway. In cultured cerebellar glia, both Ca<SUP>2+</SUP>‐induced and tonic GABA release are significantly reduced by both gene silencing of MAOB and the MAOB inhibitor selegiline. In the cerebellum and striatum of adult mice, general gene silencing, knock out of MAOB or selegiline treatment resulted in elimination of tonic GABA currents recorded from granule neurons and medium spiny neurons. Glial‐specific rescue of MAOB resulted in complete rescue of tonic GABA currents. Our results identify MAOB as a key synthesizing enzyme of glial GABA, which is released via bestrophin 1 (Best1) channel to mediate tonic inhibition in the brain.</P>

        Intravenous albumin for the prevention of contrast-induced nephropathy in patients with Liver cirrhosis and chronic kidney disease undergoing contrast-enhanced CT

        Background: The purpose of this study was to evaluate the incidence of contrastinduced nephropathy (CIN), and the effect of intravenous albumin for prophylaxis of CIN in patients with liver cirrhosis (LC) and chronic kidney disease (CKD). Methods: We conducted a retrospective study of 81 subjects with LC and CKD (estimated glomerular filtration rate (eGFR)o60 mL/min/1.73 m2) who underwent contrast-enhanced computed tomography (CT). Patients received either isotonic sodium bicarbonate solution (3 mL/kg for 1 h before CT and 1 mL/kg/h for 6 h after CT) or albumin (20% albumin, 25 mL for 1 h before CT and 75 mL for 6 h after CT). CIN was defined as an increase of Z25% or Z0.5 mg/dL in serum creatinine level. Results: Overall, CIN developed in three patients (3.7%). Of the 81 subjects, 43 received sodium bicarbonate solution and 38 received albumin. Both groups were comparable with regard to age, sex, diabetes mellitus, and baseline eGFR. The albumin group showed a significantly poorer liver function profile. CIN incidence did not differ significantly between the groups: it occurred in one (2.3%) of the 43 subjects receiving sodium bicarbonate and two (5.3%) of the 38 subjects receiving albumin (P¼0.6). However, the albumin group showed a significantly smaller increase in body weight (P¼0.03). Conclusion: The incidence of CIN in patients with LC and CKD undergoing contrast-enhanced CT after preventive measures was relatively low. The incidence of CIN was not significantly different between sodium bicarbonate and albumin groups.

      • Enhancement of antitumor metastasis activity of red ginseng extracts by synergistic immuno-stimulating activity of γ-aminobutyric acid in vivo

        The effects of red ginseng extracts (RG) and γ-aminobutyric acid (GABA) on immunostimulatory activity and tumor metastasis inhibition in mice were investigated. The proliferative activity of splenocytes was significantly higher in the RG + GABA (RG-GA) group than in RG or GABA alone. The RG-GA-administered group showed increased production of interleukin (IL)-6 and GM-CSF compared to that in the RG or GABA groups. BM cell proliferation was significantly induced when the supernatant of Peyer’s patch cells obtained from GA-RG group was added. In the experimental tumor metastasis model, RG-GA treatment produced a significant anti-tumor metastatic effect. In addition, oral administration of RG-GA not only increased the cytotoxic activity of NK cells against YAC-1 cells, but also significantly increased IFN-γ and granzyme B concentrations in the culture supernatant. The blood level of interferon-γ, IL-17, and IL-21 were significantly higher in RG-GA group compared to monotreated group. Therefore, RG has a synergistic effect with GABA that enhances the host defense system against foreign materials. This research was funded by Ministry of SMEs and Startups.

        CD19 chimeric antigen receptor T-cell (CAR-T) therapy, a genetically engineered cell therapy, showed unprecedented efficacy in the treatment of relapsed or refractory diffuse large B-cell lymphoma. Two agents, axicabtagene ciloleucel and tisagenlecleucel, were approved by the Food and Drug Administration in 2017. However, CAR-T therapy is a treatment with complex logistics and high costs, as well as inherent adverse events, including cytokine-release syndrome and neurotoxicity. In addition, predictive biomarkers for efficacy and toxicity are lacking. Industry-academy cooperation is urgently required to develop CAR-T therapy that is effective, safe, and affordable for patients in Korea. (Korean J Med 2019;94:152-158)

        The Effect of Mental Rotation on Surgical Pathological Diagnosis

        <P><B>Purpose</B></P><P>Pathological diagnosis involves very delicate and complex consequent processing that is conducted by a pathologist. The recognition of false patterns might be an important cause of misdiagnosis in the field of surgical pathology. In this study, we evaluated the influence of visual and cognitive bias in surgical pathologic diagnosis, focusing on the influence of “mental rotation.”</P><P><B>Materials and Methods</B></P><P>We designed three sets of the same images of uterine cervix biopsied specimens (original, left to right mirror images, and 180-degree rotated images), and recruited 32 pathologists to diagnose the 3 set items individually.</P><P><B>Results</B></P><P>First, the items found to be adequate for analysis by classical test theory, Generalizability theory, and item response theory. The results showed statistically no differences in difficulty, discrimination indices, and response duration time between the image sets.</P><P><B>Conclusion</B></P><P>Mental rotation did not influence the pathologists' diagnosis in practice. Interestingly, outliers were more frequent in rotated image sets, suggesting that the mental rotation process may influence the pathological diagnoses of a few individual pathologists.</P>

      • Hypometabolism and altered metabolic connectivity in patients with internet gaming disorder and alcohol use disorder

        <P><B>Abstract</B></P> <P>Internet gaming disorder (IGD) has become the subject of growing concern as an addictive behavior and has been compared with substance/non-substance-related addiction. Although IGD show clinical impairments and social dysfunction, neurobiological alterations in IGD have not been clearly elucidated. We used <SUP>18</SUP>F-fluorodeoxyglucose PET to investigate differences in glucose metabolism and metabolic connectivity in young men [thirty-six patients with IGD, twenty-six patients with alcohol use disorder (AUD) and thirty-nine healthy controls (HC)].</P> <P>Compared with the HC, IGD showed hypometabolism in the anterior cingulate cortex (ACC), temporal, frontal, parietal and striatum and AUD exhibited hypometabolism in the occipital, temporal and parietal lobule. Furthermore, IGD showed negative correlations between the ACC and game duration and between the orbitofrontal cortex and impulsivity. Also, IGD had lower metabolic connectivity between temporal and limbic regions and between the motor area and occipital region. And AUD showed greater metabolic connectivity between the orbitofrontal and parietal regions, and between the somatosensory or parietal and temporal regions, but lower metabolic connectivity in the fronto-striatal or fronto-limbic regions. Our results provide evidences that hypometabolism and altered metabolic connectivity in IGD might be related to the abnormal sensory function by longtime gaming and dysfunction of impulsive/motivational states.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We assessed with PET in patients with internet gaming disorders (IGD) or alcohol use disorder (AUD). </LI> <LI> Addiction duration was associated with hypometabolism in IGD. </LI> <LI> Abnormal metabolism and different metabolic connectivity were observed in IGD and AUD. </LI> </UL> </P>
