        Photosensitizing properties of chlorins in solution and in membrane-mimicking systems

        The photosensitizing properties of three chlorins, meso-tetra(3-hydroxyphenyl)chlorin (m-THPC), chlorin e6 (Ce6) and meso-tetraphenylchlorin substituted by two adjacent sulfonated groups ($TPCS_{2a}$) are compared in solution and when incorporated in dioleoyl-sn-phosphatidylcholine (DOPC) liposomes. In solution, the three chlorins possess a similar efficacy to generate singlet oxygen (quantum yield ~0.65). The formation of conjugated dienes was used to determine their ability to induce the peroxidation of methyl linoleate as a target of singlet oxygen. In ethanol solution, the apparent quantum yield for this process is the same for the three chlorins and its value agrees with that expected from the known rates for the decay of singlet oxygen and its reaction with methyl linoleate. When incorporated in liposomes, the order of efficacy is m-THPC > $TPCS_{2a}$ > Ce6. This order is tentatively assigned to the relative embedment of the photosensitizer within the lipidic bilayer, $TPCS_{2a}$ and Ce6 being anchored by their negative chains nearer to the water-lipid interface. The photoinduced permeation of the lipidic bilayer by these chlorins was investigated by measuring the release of carboxyfluorescein entrapped into DOPC liposomes. The charged chlorins, in particular $TPCS_{2a}$, are the most efficient, a result discussed in relation with the technology of photochemical internalization, PCI.

      • Poster Session : PS 0615 ; Others ; Are We Losing Empathy in the Medical Profession?

        Background: There are many valuable skills in the medical profession, and empathy is the one most sought by patients from their physicians. Despite strong support from medical education, empathy in medical residents` declines with years of training. The primary objective for our study is to evaluate the empathy level among our internal medicine residents. Methods: Our program is a community-based, university-affi liated residency program with 12 residents in each post-graduate year (PGY) 1-3. We developed and implemented behavioral science and geriatric medicine curricula with the main goal to teach and provide empathetic patient care. From May to October 2013 we administered to all of our residents a voluntary, self-reported, validated and standardized empathy survey. The empathy scores and statistical analysis were obtained from the Jefferson Scale of Physician Empathy report. Results: Twenty-two participants (45%) responded to the survey. There were men responding (67%) than women (33%), ages ranged from 21-30 years (71%) and 31- 40 years (28%). The range of possible physicians` empathy scores are between 50-140 with a mean of 120 and standard deviation (SD) of +/- 12. Our sample scores ranged between 96-136, with a mean of 117.4 and SD of 10.1. The mean for PGY1-3 and new coming residents were 117.2, 114.3, 124.0 and 109.7 respectively. There was no statistical difference between males and females and between PGY-1 and PGY-2 residents. However, a statistically signifi cant difference in mean empathy scores was found between PGY-3 and new, incoming residents. Conclusions: Although results based on such a small sample size may not be reliable, our survey showed that empathy scores in our program are higher with a higher level of training. We conclude that empathy is teachable, can be evaluated and we strongly support teaching empathy in residency programs.

        Background: Grieving after the loss of a loved one is a normal process. People diagnosed with dementia also experience grief, but the manifestation may be different. Very little has been studied with regard to grief in an individual with dementia. Methods: We present the case study of a nursing home patient with advanced Alzheimer`s dementia diagnosed with depression after the sudden death of his wife. When asked about their relationship the patient usually answered “This is my wife, we are married 69 years”, and her typical response was “I love you honey”. Following her death, patient became withdrawn, had poor appetite with weight loss, and only responded to simple questions with ‘yes’ or ‘no’. We discuss this case example, along with a review of the literature, to develop several conclusions regarding dementia and grief. Results: Our search yielded little on this topic. Based on a review of the dementia literature, we offer several Conclusions: Many thoughts regarding grief and dementia are expressed in the literature, however, they are mainly related to caregivers` grief. The areas of the brain responsible for memory are distinct from those associated with emotion. Although those with dementia may be unable to logically express why they are experiencing various emotional states, it is quite clear that they do continue to experience these emotions, including grief. Conclusions: Although Alzheimer`s Dementia has signifi cant cognitive affects on those in which it is diagnosed, individuals do not lose all of their faculties immediately. Many components remain long after the degenerative process has begun. We should expect that those with Alzheimer`s are capable of experiencing emotional reactions to the loss of loved ones. Individuals should be allowed to experience and express their grief, as part of a healthy bereavement process.

        Identification of Proteomic Components Associated with Resistance to Fusarium Head Blight in Rye

        Rye was used here to dissect molecular mechanisms of resistance to Fusarium head blight (FHB) and to go deeper with our understanding of that process in cereals. F. culmorum-damaged kernels of two lines different in their potential of resistance to FHB were analyzed using two-dimensional gel electrophoresis and mass spectrometry to identify resistance markers. The proteome profiling was accompanied by measurements of a- and b-amylase activities and mycotoxin content. The proteomic studies indicated a total of 18 spots with clear differences in protein abundance between the more resistant and more susceptible rye lines after infection. Eight proteins were involved in carbohydrate metabolism of which six proteins showed a significantly higher abundance in the resistant line. The other proteins recognized here were involved in stress response and redox homeostasis. Three remaining proteins were associated with protease inhibition/resistance and lignin biosynthesis, revealing higher accumulation levels in the susceptible rye line. After inoculation, the activities of a- and b-amylases, higher in the susceptible line, were probably responsible for a higher level of starch decomposition after infection and a higher susceptibility to FHB. The presented results could be a good reference for further research to improve crop resistance to FHB.

        Novel biocidal N-halamine plastic based on poly(vinyl chloride): Preparation and characteristics

        Preparation method of new biocidal plastics based on plasticized PVC has been elaborated. Thederivative of poly(4-vinylbenzyl chloride) containing N-halamine substituents has been mixed with PVCgranules and processed by dry blend extrusion and injection moulding. The properties of obtainedmaterial have been tested using SEM/EDX and thermogravimetry. The biocidal efficacy of modified PVCresin has been tested against two strains of bacteria: Escherichia coli (Gram-negative) and Staphylococcusaureus (Gram-positive). It was found that even small amount of biocidal component yields to efficient biocidal activity of thewhole plastic. Chemical composition and other properties of PVC remain almost unchanged

      • Prostate Cancer, High Cortisol Levels and Complex Hormonal Interaction

        Prostate cancer (PCa) is one of the most common diseases in men. It is important to assess prognostic factors and whether high cortisol levels and complex hormonal interactions could be responsible for PCa development. We evaluated the relationship between cortisol, leptin and estrogens in 141 men, 71 with PCa and the remaining 70 constituting a low risk group (LRG). They were recruited for this study from a total of 2906 middle-aged men (ages 45-70 years) who completed an evaluation for prostatic diseases at the Urology Division, Hospital de Clínicas "$Jos{\acute{e}}$ de San $Mart{\acute{i}}n$", University of Buenos Aires, in May 2009. In this cross sectional study, cortisol, PSA, total-testosterone, free-testosterone, bioavailable testosterone, LH and estradiol were measured in serum. We observed increased cortisol levels in PCa patients as compared to LRG cases (p=0.004,). Leptin and estradiol levels were also higher in PCa patients (p=0.048; p<0.0001, respectively). Logistic regression analysis indicated that serum cortisol (OR: 1.110 (95% CI 1.016-1.213), p=0.022), estradiol (OR: 1.044 (95% CI 1.008-1.081), p=0.016) and leptin (OR: 1.248 (95% CI 1.048-1.487), p=0.013) explained 27% of the variance of dependent variables, even after adjusting for age, smoking, BMI and waist circumference. We found increased cortisol levels in PCa patients as compared to LRG, as well as an altered circulating hormonal profile.

        Characterization of Lupinus Iuteus Chloroplgsl Gene Coding for Components of a Chloroplastic NADH Dehydrogenase

        The plastid genomes of several plants contain ndh genes homologues of genes encoding subunits of the mitochondrial complex I. We sequenced the part of lupin ndhB, ndhD and ndhF genes in order to compare the structure of these genes with those of Nicotiana tabaum, Arabidopsis thaliana, Zea mays and Oryza sativa with the idea to detect the presence of stretches with identical aminoacid composition. We were only able to find one or two stretches of this kind of about 16 aminoacid- long in the analyzed fragments of the ndh genes. The total number of such stretches was different in particular gene products: for ndhc 1, ndhB 9, ndhD 3 and ndhF 6. We have also examined the transcription pattern of ndhC, ndhK and ndhJ genes during lupin development. We show that the greatest amount of ndhC, ndhK and ndhJ transcripts are observed in 7- to 14 day- old lupin seedlings. We also studied the level of transcription of those genes in plants growing at low temperature. All the data confirmed that the abundance of transcription of ndhC, ndhK, and ndhJ genes increased under chill conditions. It has to be noted that the level of transcription of the ndhC gene was higher than the other genes probably due to higher stability of this transcript.
