        Evaluation of reference genes for miRNA expression analysis in Galeruca daurica (Coleoptera: Chrysomelidae) using qRT‐PCR

        The selection of suitable reference genes (RGs) for data normalization is a crucial step for obtaining reliable results from gene expression analysis using the quantitative real-time polymerase chain reaction (qRT-PCR). Expression stability of eleven putative RGs (miR-100-5p, miR-92a-3p, miR-998-3p, miR-279d-3p, miR-305-5p, miR-276a-3p, miR-275-3p, miR-9a-5p, miR-2a-3p, U6 snRNA and 5S rRNA) was assessed by using a web-based tool: RefFinder, which integrates four algorithms (Delta Ct, geNorm, NormFinder, and BestKeeper), and the best combination number of RGs was calculated by geNorm. Validation of selected RGs was performed by an expression analysis of let-7-5p under different experimental conditions. The best combinations of RGs were shown as follows: miR-100-5p + miR-275-3p for temperature treatment, miR-100-5p + miR-305-5p for adult tissues, miR- 276a-3p + miR-9a-5p for genders, miR-2a-3p + miR-276a-3p for days after eclosion, U6 snRNA + miR-9a-5p + miR-2a-3p for developmental stages, and U6 snRNA + miR-279d-3p for all above conditions, and there was no one RG suitable for all experimental conditions. Incorrect RG selection affected the evaluation of let-7-5p abundance. Our findings demonstrate that an integrated approach of multiple algorithms is necessary to identify reliable, stable RGs for miRNA evaluation, and provide a basis for further studies on expression and function analysis of miRNAs and their target genes in Galeruca daurica, a new pest with great outbreaks in Inner Mongolia since 2009.

      • 중국 베이징 지역의 준 제로에너지 주택의 에너지 성능 최적화 방안

        중국은 급속한 도시화 시대에 경제와 사회의 급속한 발전과 사람들의 생활수준의 지속적인 개선으로 인해 에너지와 환경 사이에 갈등이 갈수록 심화되고 있다. 건물 총 에너지 소비량과 에너지 소비 강도에 대한 압력이 계속 증가하고 있다. 현재 속도로 계속 성장한다면 건물 관련 탄소 배출량은 2050년까지 두 배가 될 것이므로 건물 에너지 효율 표준의 지속적인 개선을 통해 건물에 대한 특별한 에너지 절약 배출 감소 작업을 수행해야한다. 한랭 지역의 지역 특성에 적합한 설계 전략을 모색하고 주택의 에너지 수요를 줄일 수 있는 녹색 외곽을 개발한다는 목표를 달성할 수 있으며 주택의 초기 계획단계에서 활용을 목적으로 한다. 본 연구에서는 DesignBuilder 해석 시뮬레이션 프로그램을 통해 준제로 에너지 주택의 시뮬레이션 분석 모델을 구축하였으며, 중국 한랭 지역 대표 도시의 기후 데이터를 EnergyPlus 프로그램을 통한 시뮬레이션 분석에 사용 하였다. 입력 및 출력 데이터를 얻다. <준 제로에너지 건축 기술 표준>의 지표에 따라 EnergyPlus 프로그램을 사용하여 최적화 시뮬레이션 분석을 수행하였다. 연구를 통해 외곽의 준제로 에너지 주택에 적용된 성능에 따른 재료두께 및 에너지 요구량 분석을 시행하였으며 에너지 및 성능이 동시에 최적 평가기준을 만족하는 것으로 분석되었다. At the 21st United Nations Climate Change Conference, the whole life cycle of buildings account for 30% of the total global carbon emissions. If this rate continues, carbon emissions of buildings will double by 2050. With the rapid urbanization, economic and social development as well as continuous improvement of people’s living standards, contradictions between energy and environment in China are increasingly prominent, and the upward pressure on total energy consumption and intensity of buildings is increasing. As a consequence, it is necessary to improve the energy-saving standards of buildings, and conduct special energy conservation and emission reduction of buildings. The study with DesignBuilder program to establishes a simulation analysis model of Nearly Zero Energy House; based on the climate data of representative cities in the cold regions of China, it makes a simulation analysis with DesignBuilder program, gaining the input and output data; and according to the index of "Technical Standard for Nearly Zero Energy Building", it conducts an Optimization simulation analysis with EnergyPlus program. larger amount is required in order to be optimal, obtaining the Optimization Building Envelope performance with minimal use of energy.

        광고매체로써 공유자전거 디지털 사이니지의 수용가능성에 관한 탐색적 연구

        본 연구는 기술수용모델을 활용하여 중국 시장에서 새로운 교통수단으로 부상하고 있는 공유자전거의 디지털 사이니지 광고에 대한 수용가능성을 살펴보았다. 이와 함께 기술 수용에 대한 소비자의 지각된 위험이 아직 실행되기 전인 공유자전거 디지털 사이니지의 수용과정에 어떤 영향이 있는지에 대해서도 살펴보았다. 이 과정에서 소비자의 공유자전거 이용여부가 디지털 사이니지의 수용에 영향을 미치는지에 대해서도 살펴보았다. 먼저 공유자전거 디지털 사이니지에 대한 지각된 유용성과 지각된 사용 용이성이 디지털 사이니지 이용태도 및 사용의도에 영향을 미치는 것으로 나타났다. 한편 지각된 위험은 지각된 유용성이나 지각된 사용용이성과 마찬가지로 혁신수용에 대한 태도와 수용의도에 부정적인 영향을 미치는 것으로 나타났다. 이와 같은 결과를 바탕으로 새롭게 도입하고자 하는 공유자전거 디지털 사이니지 광고에 있어서 지각된 유용성, 지각된 사용용이성, 지각된 위험의 효과를 파악할 수 있었다. The purpose of this study is to investigate the acceptability of digital signage, a shared bicycle advertising signage, which is emerging as a new means of transportation in the Chinese market. For this research objective, this study used the technology acceptance model. In addition, this study examined the influence of a number of factors on the acceptance process of the shared bicycle digital signage. These factors are perceived risk, perceived usefulness, and perceived easiness. The results of this study are as follows: First, the assumption that the perceived usefulness and perceived easiness of using of the shared bicycle digital signage would affect the usage attitude and intention was statistically significant in this study. In other words, both perceived usefulness and perceived easiness have positive effects on usage attitude, and perceived usefulness has a positive effect on intention to use. Second the usage attitude showed a positive influence on intention to use, and the perceived easiness of use had a positive effect on perceived usefulness. Finally, this study showed that perceived risk had a negative effect on the usage attitude and intention to use. Based on these results, it is suggested that the perceived usefulness, perceived easiness of use, and perceived risk of new advertising medium are grasped in the future based on the technology acceptance model.

        Characterization of the Methylation Status of Pax7 and Myogenic Regulator Factors in Cell Myogenic Differentiation

        Epigenetic processes in the development of skeletal muscle have been appreciated for over a decade. DNA methylation is a major epigenetic modification important for regulating gene expression and suppressing spurious transcription. Up to now, the importance of epigenetic marks in the regulation of Pax7 and myogenic regulatory factors (MRFs) expression is far less explored. In the present study, semi-quantitative the real-time polymerase chain reaction (RT-PCR) analyses showed MyoD and Myf5 were expressed in activated and quiescent C2C12 cells. MyoG was expressed in a later stage of myogenesis. Pax7 was weakly expressed in differentiated C2C12 cells. To further understand the regulation of expression of these genes, the DNA methylation status of Pax7, MyoD, and Myf5 was determined by bisulfite sequencing PCR. During the C2C12 myoblasts fusion process, the changes of promoter and exon 1 methylation of Pax7, MyoD, and Myf5 genes were observed. In addition, an inverse relationship of low methylation and high expression was found. These results suggest that DNA methylation may be an important mechanism regulating Pax7 and MRFs transcription in cell myogenic differentiation.

        Insight into adsorption mechanism of cationic dye onto agricultural residues-derived hydrochars: Negligible role of π-π interaction

        Hydrochars derived from golden shower pod (GSH), coconut shell (CCH), and orange peel (OPH) were synthesized and applied to remove methylene green (MG5). The results indicated that the hydrochars possessed low specific surface areas (6.65-14.7m2/g), but abundant oxygen functionalities (1.69-2.12mmol/g). The hydrochars exhibited cellular and spherical morphologies. Adsorption was strongly dependent on the solution pH (2-10) and ionic strength (0-0.5M NaCl). Equilibrium can be quickly established in the kinetic study (60-120 min). The maximum Langmuir adsorption capacities at 30 oC followed the order GSH (59.6mg/g)>CCH (32.7mg/g)>OPH (15.6mg/g)> commercial glucose-prepared hydrochar (12.6mg/g). The dye adsorption efficiency was determined by the concentrations of oxygen-containing functionalities on the hydrochar surface. The adsorption process occurred spontaneously (−ΔGo) and exothermically (−ΔHo). Desorption studies confirmed the reversible adsorption process. Oxygenation of the hydrochar surface through a hydrothermal process with acrylic acid contributed to increasing MG5 adsorption and identifying the negligible role of π-π interaction to the adsorption process. The analysis of Fourier transform infrared spectrometry demonstrated that the aromatic C=C peak did not significantly decrease in intensity or shift toward higher/lower wavenumbers after adsorption, which further confirms the insignificant contribution of π-π interaction. Electrostatic attraction played a major role in adsorption mechanisms, while minor contributions were accounted for hydrogen bonding and n-π interactions. The primary adsorption mechanisms of MG5 onto hydrochar were similar to biosorbent, but dissimilar to biochar and activated carbon (i.e., π-π interaction and pore filling).

        Water and carbohydrate levels at different developmental stages and dynamics in hibernating pupae of Pieris melete (Lepidoptera: Pieridae)

        For insight into the physiological indicators of diapause in Pieris melete, water and carbohydrate (glycogen and trehalose) levels were measured under both natural and laboratory conditions. The highest water content (3.71–3.79 mg/mg dry weight) was found in larvae and developing pupae, which was substantially higher than in diapausing pupae (2.59 mg/mg dry weight). Water content was almost stable during diapause, except for individuals approaching diapause termination (3.43–3.58 mg/mg dry weight). The total carbohydrate level was significantly higher in pre‐pupae (47.41 μg/mg) compared to larvae (22.80 μg/mg) and developing pupae (21.48 μg/mg). The highest level of trehalose was detected in winter diapausing pupae, and no trehalose was found in larvae or developing pupae. Levels of glycogen were highest in pre‐pupae and lowest in diapausing pupae. Levels of total carbohydrate decreased as diapause proceeded, and no significant changes were found in trehalose levels for diapausing pupae under natural conditions or treated for 60–90 days at 5°C. Pupae treated at 20°C for 60–90 days had significantly lower levels of trehalose than those treated for 30 days. Glycogen content was relatively stable at 5°C, but increased after treatment under natural conditions and 20°C for more than 60 days. These results suggest that the dynamics of water and carbohydrate levels are potential physiological diapause indicators, which show metabolic differences between trehalose and glycogen during diapause development.

        Study on Mechanical Properties of Polycaprolactone Modified Cement-Based Material

        With the continuous development of economy, engineering construction is developing towards deep high-stress area, and the problems of large-deformation engineering are becoming more and more serious. Anchoring grouting is a very effective support reinforcement method. The anchoring grouting materials used in the project are mainly cement paste and cement mortar. The characteristics of the grouting material have a decisive influence on the grouting effect. However, it is difficult for the existing grouting materials to satisfy the requirements of strength and deformation at the same time, and improvement is urgently needed. Polycaprolactone (PCL) is an organic polymer material of good engineering properties, including high ductility, good stability and strong impact resistance. In this paper, PCL was added as an auxiliary additive to cement-based materials innovatively, and the mechanical test and micro-test of PCL-cement-based materials were carried out. The results show that adding PCL can significantly increase the compressive strength of cement paste. By adjusting the content of PCL, the strength and ductility of cement mortar can be effectively improved. When the content of PCL is 15%, ideal high performance cement mortar material can be obtained. Anchor grouting is a very effective reinforcement and support method.
