        The Effect of Amino Density on the Attachment, Migration, and Differentiation of Rat Neural Stem Cells In Vitro

        Artificial extracellular matrices play important roles in the regulation of stem cell behavior. To generate materials for tissue engineering, active functional groups, such as amino, carboxyl, and hydroxyl, are often introduced to change the properties of the biomaterial surface. In this study, we chemically modified coverslips to create sur-faces with different amino densities and investigated the adhesion, migration, and differentiation of neural stem cells (NSCs) under serum-free culture conditions. We observed that a higher amino density significantly promoted NSCs attach-ment, enhanced neuronal differentiation and promoted excitatory synapse formation in vitro. These results indicate that the amino density significantly affected the biological behavior of NSCs. Thus, the density and impact of functional groups in extracellular matrices should be considered in the research and development of materials for tissue engineering.

        ESA in High-order Descriptor Linear Systems via Output Feedback

        This paper considers eigenstructure assignment (ESA) in high-order descriptor linear systems via output feedback. Parametric expressions for the left and right closed-loop eigenvectors associated with the finite closed-loop eigenvalues and two simple and complete parametric solutions to the feedback gain matrix are obtained on the basis of the parametric solution to the generalized high-order Sylvester matrix equation. This approach does not impose any restriction on the closed-loop eigenvalues. The numerical example illustrates the effectiveness of the proposed approach.

        Associations of Plasma Glucagon Levels with Estimated Glomerular Filtration Rate, Albuminuria and Diabetic Kidney Disease in Patients with Type 2 Diabetes Mellitus

        Background: Type 2 diabetes mellitus (T2DM) is characterized by elevated fasting glucagon and impaired suppression of postprandial glucagon secretion, which may participate in diabetic complications. Therefore, we investigated the associations of plasma glucagon with estimated glomerular filtration rate (eGFR), albuminuria and diabetic kidney disease (DKD) in T2DM patients.Methods: Fasting glucagon and postchallenge glucagon (assessed by area under the glucagon curve [AUCgla]) levels were determined during oral glucose tolerance tests. Patients with an eGFR <60 mL/min/1.73 m2 and/or a urinary albumin-to-creatinine ratio (UACR) ≥30 mg/g who presented with diabetic retinopathy were identified as having DKD.Results: Of the 2,436 recruited patients, fasting glucagon was correlated with eGFR and UACR (r=–0.112 and r=0.157, respectively; P<0.001), and AUCgla was also correlated with eGFR and UACR (r=–0.267 and r=0.234, respectively; P<0.001). Moreover, 31.7% (n=771) presented with DKD; the prevalence of DKD was 27.3%, 27.6%, 32.5%, and 39.2% in the first (Q1), second (Q2), third (Q3), and fourth quartile (Q4) of fasting glucagon, respectively; and the corresponding prevalence for AUCgla was 25.9%, 22.7%, 33.7%, and 44.4%, respectively. Furthermore, after adjusting for other clinical covariates, the adjusted odds ratios (ORs; 95% confidence intervals) for DKD in Q2, Q3, and Q4 versus Q1 of fasting glucagon were 0.946 (0.697 to 1.284), 1.209 (0.895 to 1.634), and 1.521 (1.129 to 2.049), respectively; the corresponding ORs of AUCgla were 0.825 (0.611 to 1.114), 1.323 (0.989 to 1.769), and 2.066 (1.546 to 2.760), respectively. Additionally, when we restricted our analysis in patients with glycosylated hemoglobin <7.0% (n=471), we found fasting glucagon and AUCgla were still independently associated with DKD.Conclusion: Both increased fasting and postchallenge glucagon levels were independently associated with DKD in T2DM patients.

        The effi cacy of daily chlorhexidine bathing for preventing healthcare-associated infections in adult intensive care units

        Background/Aims: Healthcare-associated infections (HAIs) in critically ill patients with prolonged length of hospital stay and increased medical costs. The aim of this study is to assess whether daily chlorhexidine gluconate (CHG) bathing will significantly reduce the rates of HAIs in adult intensive care units (ICUs). Methods: PubMed, EMBASE, and the Cochrane Central Register of Controlled Trials were systematically searched until December 31, 2014 to identify relevant studies. Two authors independently reviewed and extracted data from included studies. All data was analyzed by Review Manager version 5.3. Results: Fifteen studies including three randomized controlled trials and 12 quasi- experimental studies were available in this study. The outcomes showed that daily CHG bathing were associated with significant reduction in the rates of primary outcomes: catheter-related bloodstream infection (risk ratio [RR], 0.44; 95% confidence interval [CI], 0.32 to 0.63; p < 0.00001), catheter-associated urinary tract infection (RR, 0.68; 95% CI, 0.52 to 0.88; p = 0.004), ventilator-associated pneumonia (RR, 0.73; 95% CI, 0.57 to 0.93; p = 0.01), acquisition of methicillin-resistant Staphylococcus aureus (RR, 0.78; 95% CI, 0.68 to 0.91; p = 0.001) and vancomycin-resistant Enterococcus (RR, 0.56; 95% CI, 0.31 to 0.99; p = 0.05). Conclusions: Our study suggests that the use of daily CHG bathing can signifi- cantly prevent HAIs in ICUs. However, more well-designed studies are needed to confirm these findings.

        Garments and garment behavior is influenced by ethical, aesthetical standards and also values and skills, therefore different types of garments exist according to different statuses, positions, and values. The type of garment one chooses to wear represents his/her status, position and values. It plays an important role as a non-verbal medium for delivering information about the wearer that enables visual communication. Therefore choosing what to wear and expressing ourselves through what we wear all contribute in an activity of selecting preferences and understanding what it means. Symbols are ``media that have meanings``. In order to be able to understand this fully, the dictionary definition of media is required and it is an object that delivers an action from one place to another. But a symbol is primarily for replacement, therefore it is very difficult to explain just as it is. For a better understanding of a symbol, further research needs to be done and an area of study on this matter is called semiology. The purpose of semiology is on creating the meaning and interpretation. The definition of semiology is, a study on the act and process of defining the relationship between the symbols and the way people use to rule the symbols, and a research on extracting and interpreting the meanings through symbols. Therefore this research is an attempt to examine the phenomenon of garments structurally using a semiotic approach on the film costumes and through an understanding of the structure of the meanings so that we can look into how the status, position, and values of different characters are expressed by different type of garments.

        The initial framework of developing semantic knowledge base of Mongolian idioms

        As a significant component of Mongolian language resources, idioms have long history and carry rich cultural heritage. They provide a valuable resource for the research fields of Mongolian civilization. But, with the development of internet and information society, vocabulary of Mongolian idioms has shrunk, resulting in declining on the frequency of usage, gradual disappearance of Mongolian culture that Mongolian language has carried, and thus facing a serious challenge. Conducting a digital research on Mongolian idioms and developing the Semantic Knowledge Base of the Mongolian Idioms is not only the best way to protect, develop and utilize Mongolian idioms, but also the basic project of constructing a full-scale platform of Mongolian language. For the unsatisfactory status quo of the research in the semantics of Mongolian idioms at present, the paper will dissertates the initial framework of the digital research on the semantic knowledge of Mongolian idioms from the perspectives of importance of research, methods, contents, basic works and anticipative results.

        국내에서 분리된 다제 내성 결핵균의 katG 와 inhA 변이 다양성 및 그 빈도

        연구배경: INH 내성은 katG 와 inhA(ORF와 promoter)의 변이에 의한 것으로 알려져 있다. 유전자 변이는 지역적으로 종류와 빈도가 다르게 나타날 수 있는데 기존 국내의 연구보고들은 흔하다고 알려진 katG의 463 코돈만을 추적한 것들이었다. 따라서 본 연구는 국내에서 분리된 INH 내성균들의 두 유전자에서 나타날 수 있는 변이의 종류와 빈도를 확인하고자 하였다. 연구방법: 대한결핵협회 결핵연구원에서 MDR-TB로 판명된 INH 내성 결핵균 29주로부터 bead beater-phenol법으로 DNA를 추출하여 katG(2,223 bp), inhA ORF(-77∼897, 975 bp) 및 inhA promoter(-168∼80, 248 bp) 염기서열 결정 및 분석은 ABI PRISM 3730 XL Analyzer 및 MegAlign package를 사용하였다. 결과: 모든 균주들은 분석 표적으로 사용한 세 유전자 부위 중에서 적어도 한 개 이상의 유전자 부위에 변이가 있었다. INH 내성균은 거의 대부분(>93%) katG의 변이를 갖고 inhA 유전자 변이만 있는 경우는 드물어 INH 내성을 결정하는 중요한 요인은 katG의 변이 인 것을 확인할 수 있었다. katG 부위에서 Arg463Leu 변이와 Ser315Thr 변이가 높은 빈도(62.1% 및 55.2%)로 발견되었고, katG 완전결실과 inhA promoter-15(C→T) 변이도 일정한 빈도로 나타남을 볼 수 있었다. 그 외 inhA ORF 변이도 1주에서 1종류의 변이가 발견되었다. 결론: 기존 연구결과에서는 보고되지 않고 본 연구에서 처음으로 확인된 변이들도 14 종류나 있어서, INH 내성은 주로 katG 혹은 일부 inhA 특정 부위의 변이가 주도하지만 이들 외에도 다양한 변이가 존재한다는 것을 알 수 있었다. 이들 새로이 확인된 변이들은 염기서열 분석에 의한 INH 내성 여부 판단 시, 기존 알려진 변이 외에 보조 자료로 사용할 수 있을 것으로 생각한다. Backgrounds: Mutations of katG and inhA (ORF and promoter) are known to be related to isoniazid (INH) resistance of Mycobacterium tuberculosis. Because reports on these mutations in Korean isolates are limited (i.e. only the frequency of katG codon 463 was evaluated.), we tried to know the kinds of mutations of two genes and their frequencies in INH resistant Korean M. tuberculosis strains. Methods: PCR was performed to amplify katG (2,223 bp), inhA ORF (-77~897, 975 bp), and inhA promoter (-168~80, 248 bp) from 29 multidrug resistant M. tuberculosis (MDR-TB) DNAs prepared by bead beater-phenol method. Their sequences were determined and analyzed by ABI PRISM 3730 XL Analyzer and MegAlign package program, respectively. Results: All of the isolates had more than one mutation in katG or inhA gene. Twenty seven (93%) of 29 tested strains had katG mutations, which suggests that katG is a critical gene determining INH resistance of M. tuberculosis. Amino acid substitutions, such as Arg463Leu and Ser315Thr, due to point mutations of the katG were the most frequent (62.1% and 55.2%) mutations. In addition, deletion of the katG gene was frequently observed (17.2%). Analyzed Korean MDR-TB isolates also had variable inhA mutations. Point mutation of inhA promoter region, such as -15 (C→T) was frequently found. Substitution of amino acid (Lsy8Asn) due to point mutation (AAA→AAC) of inhA ORF was found in 1 isolate. Interestingly, 14 point mutated types that were not previously reported were newly found. While four types resulted in amino acid change, the others were silent mutations. Conclusions: Although it is not clear that the relationship of these newly found mutations with INH resistance, they show marked diversity in Korean MDR-TB strains. It also suggests their feasibility as a molecular target to supplement determining the INH resistance of clinical isolates because of the possible existence of low-level INH resistant strains.       • HDAC6 siRNA Inhibits Proliferation and Induces Apoptosis of HeLa Cells and its Related Molecular Mechanism

        Objective: To investigate the effects of histone deacetylase 6 (HDAC6) siRNA on cell proliferation and cell apoptosis of the HeLa cervical carcinoma cell line and the molecular mechanisms involved. Methods: Division was into three groups: A, the untreated group; B, the control siRNA group; and C, the HDAC6 siRNA group. Lipofectamine 2000 was used for siRNA transfection, and Western blot analysis was used to determine the protein levels. Cell proliferation and apoptosis were characterized using a CCK-8 assay and flow cytometry, respectively. Results: HDAC6 protein expression in the HDAC6 siRNA-transfection group was significantly lower (P < 0.05) than in the untreated and control siRNA groups. The CCK-8 kit results demonstrated that the proliferation of HeLa cells was clearly inhibited in the HDAC6 siRNA transfection group (P < 0.05). In addition, flow cytometry revealed that the early apoptotic rate ($26.0%{\pm}0.87%$) was significantly elevated (P < 0.05) as compared with the untreated group ($10.6%{\pm}1.19%$) and control siRNA group ($8.61%{\pm}0.98%$). Furthermore, Western blot analysis indicated that bcl-2 protein expression in the HDAC6 siRNA-transfection group was down-regulated, whereas the expression of p21 and bax was up-regulated. Conclusion: HDAC6 plays an essential role in the occurrence and development of cervical carcinoma, and the down-regulation of HDAC6 expression may be useful molecular therapeutic method.

        Associations of hypoxia inducible factor-1a gene polymorphisms with susceptibility to digestive tract cancers: a case–control study and meta-analysis

        We aim to investigate the correlations of hypoxia inducible factor-1a (HIF-1a) C1772T (rs11549465) and G1790A (rs11549467) gene polymorphisms with digestive tract cancers. A sum of 267 digestive tract cancers patients were hospitalized in Taizhou Central Hospital of Zhejiang Province as case group between December 2012 and December 2014. Additionally, 275 healthy people who had a physical examination in our hospital at the same time were selected as control group. Polymerase chain reaction-restriction fragment length polymorphism was utilized for detecting allele and genotype frequency of different locus in case and control group. Meta-analysis was performed using Comprehensive Metaanalysis 2.0 (Biostat Inc., Englewood, New Jersey, USA). Our result showed statistical significance only exists in family history of cancer between case and control group (P\0.05). Both C1772T (rs11549465) and G1790A (rs11549467) polymorphisms showed positive correlations with an increasing risk of digestive tract cancers. The frequencies of TT genotype of C1772T (rs11549465) and GA, AA genotypes of G1790A (rs11549467) polymorphisms in case group were evidently higher compared with the controls (all P\0.05). Besides, the comparison of allele and dominant models of HIF-1a C1772T (rs11549465) and G1790A (rs11549467) between two groups showed a significant difference (all P\0.05). Meta-analysis results further confirmed that the onset risk of digestive tract cancers may be improved under allele and dominant models of HIF-1a C1772T (rs11549465) and G1790A (rs11549467) (all P\0.05). Single nucleotide polymorphisms of HIF-1a C1772T (rs11549465) and G1790A (rs11549467) may play a role in development of digestive tract cancers.
