Indocyanine green-incorporated exosomes for improved in vivo imaging of sentinel lymph node

For improved physical stability and fast accumulation in sentinel lymph nodes in vivo, indocyanine green (ICG) was incorporated into exosomes (EXOs) to form a novel ICG-based near-infrared fluorescence probe. Compared with free ICG, prepared ICG-TBAI-encapsulated EXOs (ICG/EXOs) smaller than 50 nm had greater physicochemical stability in aqueous solution when exposed to light at 37 C. In addition, sentinel lymph nodes were visualized within minutes with ICG/EXOs at a low ICG dose (0.05 mg/kg) owing to its well-controlled size distribution. Taken together, the results show that ICG/ EXOs are effective biocompatible and sensitive near-infrared probes for real-time sentinel lymph node imaging in vivo.

Comparative evaluation of cell- and serum-derived exosomes to deliver immune stimulators to lymph nodes

<P><B>Abstract</B></P> <P>To determine whether exosomes are efficient carriers for immune stimulating molecules into lymph nodes, comparative studies of exosomes (EXOs) derived from different origins (cells and serums) in terms of physicochemical properties and delivery efficiency were performed. Serum-derived EXOs were of a preferable size and generated higher yields than RAW264.7 cell-derived exosomes (RAW-EXO). In particular, fetal bovine serum-derived exosomes (bo-EXO), with a size below 50 nm, were delivered not only to surface zones (subcapsular sinus (SCS) macrophage zone) but also to inner paracortex zones (T cell zone) of lymph nodes, which allowed an efficient delivery of immune stimulating molecules to antigen presenting cells and T cells. The encapsulation of immune stimulating biomolecules (monophosphoryl lipid A (MPLA) and CpG oligodeoxynucleotides (CpG ODN)) within EXOs greatly increased intracellular delivery to macrophages via phagocytic pathways, which induced higher TNF-α and IL-6 secretion than free MPLA and free CpG ODN. MPLA-incorporated exosomes activated and differentiated T cells after subcutaneous injection, which elevated cytokine IFN-γ and TNF-α induction for CD3<SUP>+</SUP> T cells. Taken together, bo-EXOs might serve as efficient carrier systems of immune stimulators to lymph nodes for desired immune responses.</P>

Purpose: The purpose of this study to verify the symptom clusters of colorectal cancer patients and also identify the influence of the symptom clusters upon colorectal patient symptom clusters identify acording exper. Methods: 101 colorectal cancer patients were the subjects of this study. As for a research tool, the global health status/QOL was used including the sub-scales of QLQ-C30. In the functional scales and the symptom scales, the sub-scales of QLQ-C30 and QLQ-CR29 were used. Results: The result of this study shows us there are 7 kinds of symptom clusters. Results: The result of this study shows us there are 7 kinds of symptom clusters, bowel movement problems, abdominal-pelvic problems, feces problems, urination problems, dry mouth-taste problems, side effect problems, and elimination problems. Conclusion: Based on the result, the symptom clusters in colorectal cancer patients were confirmed to be influencing the subjects’ quality of life and functions. The efficient management of symptom clusters that subjects experience leads to promoting colorectal cancer patients" physical and emotional status, thereby contributing to an improvement in their quality of life.

CpG oligonucleotide and a-D-mannose conjugate for efficient delivery into macrophages

For improved intracellular uptake of CpG ODN into macrophages, we developed CpG ODN and mannose conjugate (CpG–Man conjugate) via simple conjugation of a-D-mannose to CpG ODN. CpG–Man conjugate showed greatly enhanced intracellular uptake by 2.1-fold into macrophages, compared to CpG ODN in a TLR-9 receptorspecific manner. In addition, internalized CpG–Man conjugate successfully triggered TNF-a release in macrophages. Taken together, the CpG–Man conjugate can serve as the promising delivery systems of CpG ODN into immune cells as a candidate adjuvant.

『海と毒藥』論 : <語り>と登場人物像を中心に <이야기>와 등장인물 상을 중심으로

『海と毒藥』は昭和32年、『文學界』に、それぞれ「海と毒藥」「裁かれる人??」「夜のあけるまで」として3回億にわたって連載され、翌33年4月、文藝春秋新社から單行本として刊行された。 この小說の題材は、周知のように第2次大戰中に九州帝國大學文學部で起った米軍捕虜生體解剖事件を取り扱っている。しかし、作者は實際のできごとを背景としなから、「私はその事件そのものを書く氣持ちは毛頭なかった。私の內部にあるもので事件を變容させ、別の次元に移しかえてみるつもりだった」(『出世作のころ』)と語っているように、作品自體を現實の事件とは違った次元のもとにつくりあげたフィクションである。だが、事件關係者からの抗議などが起こり、その背景となった事件の眞相を巡って事實と小說との相違点を分析する試みなども行われている。 遠藤周作の問題意識は、常に上記したような場に始終している。このような作者自らの問題は、さらに一神論と汎神論の比較、對立、相克、という關係論へと展開され、彼の文學の基調音となっていくといえよう。しかし、下山孃子が遠藤がキリスト作家として「西歐の基盤をふまえた一神論の基督敎を日本文學に消化することが如何に至難ぞあるか」(「基督敎と日本文學」)と身をもって體驗し、苦鬪した作家であるとしても、「キリスト敎や聖書についての何をかの知識や情報を持ったない限り遠藤文學の讀解は困難である」という事態を招くのではないか、それはまた「遠藤自身も嫌う護敎文學となる危險性をもつことになるのではないか」と指摘しているように、彼の文學をキリスト敎的次元から讀み解こうと腐することにおいてのみ、遠藤文學を理解しようとすることは、護敎文學としての讀みの强制となる危險性を示すものであることも見ておく必要がある。ここではそうした問題点を踏まえて、構造主義を經た現在の文學硏究の一つの生き方としてナラロジ-(語り·語り手)の着目しつつ、登場人物の人物像とともに、彼らが示している神の正義について考察してみた。

『홋케겐키』는 쵸큐년간(1040~1044년) 히에잔 요카와 슈료공인(比??山橫川首??嚴院)려 친겐(鎭源)이 편찬한 법화경 영험담이다. 『홋케겐키』는 『곤쟈쿠모노가타리』와 『고쿠라쿠키』 이후에 편찬된 헤이안 시대의 왕생전의 중요한 전거가 되고 있다. 그러나 『홋케겐키』에 관한 기존의 연구를 살펴보면 헤이안 시대의 법화신앙사를 연구하기 위한 연구사료나 또는 『곤쟈쿠모노가타리』와 『고쿠라쿠키』 이후에 편찬된 여러 왕생전의 전거문헌으로만 간단하게 취급하고 있을 뿐 『홋케겐키』가 가지고 있는 독자적인 편찬의식에 대한 연구는 전무한 상황이다. 따라서 이 논문에서는『홋케겐키』의 편자인 친겐(鎭源)의 편찬의식을 고찰하기 위한 연구의 하나로써 『홋케겐키』에 나타난 관음설화의 의의에 주목했다. 『홋케겐키』의 내용을 살펴보면 법화신앙 뿐만 아니라 내세이익을 나타내는 왕생연험설화와 현세이익을 나타내는 관음신앙설화, 밀교신앙 등이 기술되어 있다. 이들 신앙이 『홋케겐키』가 편찬될 당시인 헤이안시대에 일본사회에서 독자적인 종교로서 번창했다는 것은 이미 알려진 사실이다. 따라서 편자의 편찬의도를 고찰하기 위해서는 『홋케겐키』에 묘사된 신앙에 대한 설화들을 종합적으로 연구할 필요가 있다. 그러나 지면의 제약상 내세이익을 나타내는 왕생영험설화와 밀교에 대해서는 다음 기회에 논하기로 하고, 본 논문에서는 『홋세겐키』의 영험담 중 현세이익을 나타내는 관음(觀音)설화에 주목함으로써, 관음설화와 『법화경』과의 관계 특히 『법화경』의 경전명(經典名)이 어떻게 기술되어 있는가에 대해 고찰했다. 또 『홋케겐키』에 기술된 관음신앙을 『콘쟈쿠모노가타리』·『니혼료이키』와 비교·검토하였다. 그 결과 『곤쟈쿠모노가타리』와 『니혼료이키』에 서술된 관음신앙은 반드시 『법화경』제8권「관세음보살보문품」에 종속된 것이 아니라 『법화경』과 대등한 신앙이었다는 사실을 알 수 있었다. 즉 『홋케겐키』는 이 작품이 편찬될 당시 융성했던 현세이익적인 관음신앙을 『법화경』의 일부로 새롭게 위치를 부여함으로써 『법화경』의 현세이익적인 효과를 더욱 강하게 표명할 수가 있었던 것이다. 정리해 보면, 『홋케겐키』는 기본적으로 『법화경』 영험담으로써 현세이익을 표방하고 있다. 그러나 다른 한편으로는 관음신앙과 같은 헤이안 시대(平安時代)의 대표적인 현세이익 신앙을 설화소재로 채택함으로써 그들을 전부 『법화경』에 종속된 일부분으로 규정하고 있다. 그로 인해 『홋케겐키』는 사람들에게 현세이익을 가져다 주는 원천으로써 법화신앙의 위대한 힘을 강조하기 위해 편찬된 작품이라고 볼 수 있다.

接續表現「で」の意味機能

本稿は現代日本語の口語體にみられる獨特な表現のうち、實際の會話で多くの部分を占めている接續表現「で」に焦点をしぼって考察を行ったものである。これまで「で」は「それで」の宿約形の一種と見なすのが一般的であったが、本稿では接續表現「で」の現れる用例を中心に、それがどのような接續表現に置き換えられるかを檢討した後、意味·機能的な側面から話題接續と話題轉換に分けて考察した。分類の結果は次のようである。 Ⅰ. 話題接續機能 (ⅰ) 話題歸結(總括): それで, だから, そういうわけで (ⅱ) 話題誘導: そこで, それじゃ (ⅲ) 累加: そして, それから, それに, しかも, さらに, また (ⅳ) 解說: なぜなら, たとえば, つまり, ただし (ⅴ) 逆接: ところが, しかし, でも, だけど, けれども Ⅱ. 話題轉換機能 · 典型的な用法 (ⅵ) 話題轉換: ところで · 周邊的な用法 (ⅶ) 話題展開: ええと, まあ (ⅷ) 話題喚起: まず これによって「で」が本來の意味領域を擴大し、新たに逆接や話題轉換と同じ機能を獲得し、口語體では欠かせない接續表現として定着していく過程がうかがわれる。さらに、いままで「それで」の宿約形ク)宿約形、または異形として分類されてきた「で」はむしろ複合的な意味を持つ、一つの獨立した接續表現として見なすべきであることを明らかにした。
