A mechanical model of vehicle-slab track coupled system with differential subgrade settlement

Post-construction subgrade settlement especially differential settlement, has become a key issue in construction and operation of non-ballasted track on high-speed railway soil subgrade, which may also affect the dynamic performance of passing trains. To estimate the effect of differential subgrade settlement on the mechanical behaviors of the vehicle-slab track system, a detailed model considering nonlinear subgrade support and initial track state due to track self-weight is developed. Accordingly, analysis aiming at a typical high-speed vehicle coupled with a deteriorated slab track owing to differential subgrade settlement is carried out, in terms of two aspects: (i) determination of an initial mapping relationship between subgrade settlement and track deflections as well as contact state between track and subgrade based on a semi-analytical method; (ii) simulation of dynamic performance of the coupled system by employing a time integration approach. The investigation indicates that subgrade settlement results in additional track irregularity, and locally, the contact between the concrete track and the soil subgrade is prone to failure. Moreover, wheel-rail interaction is significantly exacerbated by the track degradation and abnormal responses occur as a result of the unsupported areas. Distributions of interlaminar contact forces in track system vary dramatically due to the combined effect of track deterioration and dynamic load. These may not only intensify the dynamic responses of the coupled system, but also have impacts on the long-term behavior of the track components.

Although nucleolar protein nucleostemin (NS) is essential for cell proliferation and early embryogenesis and expression has been observed in some types of human cancer and stem cells, the molecular mechanisms involved in mediation of cell proliferation and cell cycling remains largely elusive. The aim of the present study was to evaluate NS as a potential target for gene therapy of human breast carcinoma by investigating NS gene expression and its effects on SKBR-3 cell proliferation and apoptosis. NS mRNA and protein were both found to be highly expressed in all detected cancer cell lines. The apoptotic rate of the pcDNA3.1-NS-Silencer group ($12.1-15.4{\pm}3.8%$) was significantly higher than those of pcDNA3.1-NS ($7.2-12.0{\pm}1.7%$) and non-transfection groups ($4.1-6.5{\pm}1.8%$, P<0.01). MTT assays showed the knockdown of NS expression reduced the proliferation rate of SKBR-3 cells significantly. Matrigel invasion and wound healing assays indicated that the number of invading cells was significantly decreased in the pcDNA3.1-NS-siRNA group (P<0.01), but there were no significant difference between non-transfected and over-expression groups (P>0.05). Moreover, RNAi-mediated NS down-regulation induced SKBR-3 cell G1 phase arrest, inhibited cell proliferation, and promoted p53 pathway-mediated cell apoptosis in SKBR-3 cells. NS might thus be an important regulator in the G2/M check point of cell cycle, blocking SKBR-3 cell progression through the G1/S phase. On the whole, these results suggest NS might be a tumor suppressor and important therapeutic target in human cancers.

By integrating useful insights from social cognitive theory and social capital theory, we aim to develop a model for better understanding people's behaviors related to the use of social networking sites (SNSs) and formalize the role of social capital in individuals' continuous SNS use. Propositions that emphasize the triadic interactive relationships among environmental, personal, and behavioral factors were highlighted in this study. After reviewing previous studies, in this paper we proposed the following: (1) the causation between SNS use and individuals' perceived social capital might be mutual; social capital may not only be the result of media selectivity, but could also be an essential stimulus initiating the start of using SNSs; (2) the influences of SNSs use on the generation of individuals' online social capital might be conditional upon particular patterns of use; (3) both the level of dependence on SNSs and the differentiated patterns of SNSs use vary according to individuals' perceived offline social capital and their personal characteristics, for instance, personality or self-construal, and social anxiety.

Objectives: To investigate the effects of betaine on HeLa cell growth and apoptosis and molecular mechanisms. Materials and Methods: Concentrations of 0.1, 1.0, 5.0, 20.0, 100.0 mg/ml of betaine were used to evaluate the anticancer efficacy for HeLa cells respectively, and MCF-10A was also detected as a normal diploid cell control. Results: We found that proliferation of HeLa cells was inhibited significantly upon exposure to increasing betaine levels with the MTT test (p<0.05). The percentage of S phase cells in the low dose groups (<5mg/ml) were distinctly higher than in high dose groups, and the rates of Sub-G1 phase were the opposite (p<0.01); A high concentration of betaine (>5.0mg/ml) significantly promoted the apoptosis of HeLa cells (p<0.01). SOD activities of the low dose groups were slightly higher than the control group (p<0.05) and there were obvious synchronicity and correlation among the expression of promoting apoptosis genes Bax, P53, Caspase 3 and apoptosis suppression gene Bcl-2. In response to an apoptosis-inducing stimulus, p53 and cyclin D1 could be activated with blockage of the cell cycle at G1/S or S/G2 checkpoints. Conclusions: Our data showed that betaine could promote HeLa cells proliferation in vitro at low concentrations. In contrast, high concentrations could significantly inhibit cell growth and migration, and induce apoptosis of HeLa cells through caspase 3 signaling and further promoted necrosis. This might imply that betaine exhibits tumoricidal effects and acts as a biological response modifier in cancer treatment by inducing apoptosis and cell cycle arrest in a dose and time-dependent manner.

Developed from Digital Multimedia Broadcasting (DMB), DMB+ changes the application focus fundamentally. The plus symbol “+” indicates that DMB+ is no longer for digital radio/videos, instead it works as a general data transmission technique. The merits of wireless transmission, broad coverage, high quality, information security and low building/maintenance cost make DMB+ suitable for a wide range of applications with numerous terminals and users, such as public information release, alarm and administration etc. The concept of DMB+ base station enables DMB+ to be used in building independent networks and for remote control and management, as well as for Industry 4.0. DMB+ will access into a new field that till now has been dominated by communication techniques, will open a huge commercial opportunity.

An Internet of Things (IOT) sensor network is an effective solution for monitoring environmental conditions. However, IOT sensor networks generate massive data such that the abilities of massive data storage, processing,and query become technical challenges. To solve the problem, a Hadoop cloud platform is proposed. Using the time and workload genetic algorithm (TWLGA), the data processing platform enables the work of one node to be shared with other nodes, which not only raises efficiency of one single node but also provides the compatibility support to reduce the possible risk of software and hardware. In this experiment, a Hadoop cluster platform with TWLGA scheduling algorithm is developed, and the performance of the platform is tested. The results show that the Hadoop cloud platform is suitable for big data processing requirements of IOT sensor networks.

We investigated the influence of interphase interaction on unsteady sheet/cloud cavitating flows around a 2D hydrofoil numerically. Inthe computations, the homogeneous (without the consideration of interphase interaction) and inhomogeneous (with the consideration ofinterphase interaction) multiphase flow models are used, respectively. The inhomogeneous model assumes that both phases co-exist atevery point in the flow field and each phase is governed by its own set of conservation laws. The exchanges of mass, interphase dragforce and momentum transfer induced by interphase mass transfer are treated explicitly as transfer terms. The results show that the distributionsof interphase drag force and momentum transfer induced by mass transfer are highly related with the velocity difference of twophases and interphase mass transfer rate, respectively. Without the consideration of interphase interaction, the over-prediction of turbulenteddy viscosity in the rear of attached cavity causes the reentrant jet to lose its momentum and becomes too weak to cut across thecavity sheet. However, when considering the interphase interaction, the predicted turbulent kinetic energy in the rear of attached cavitycan be reduced considerably; thus, turbulent eddy viscosity drops significantly in this region, and the reentrant jet becomes much stronger,so typical cloud cavitation shedding occurs. Compared with those predicted without the consideration of interphase interaction, a betteragreement with experimental results concerning the time evolution of cavity shape, time-averaged lift/drag coefficient, frequency andmean u-velocity profiles is obtained with the consideration of interphase interaction.
