        In Vitro Screening for Compounds Derived from Traditional Chinese Medicines with Antiviral Activities Against Porcine Reproductive and Respiratory Syndrome Virus

        Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol- 2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration (CC50) and 50% effective concentration (EC50) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The EC50 values were 270.8 ± 14.6 μg/ml and 28.21 ± 26.0 μg/ml and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.

        Studies on Residue Stress and Deformation Behavior of GCr15 Subjected to Whirlwind Milling

        Whirlwind milling is gaining extensive application for processing a large screw. Due to its specific forming principle, the profile of residual stresses and the deformation behavior would been given significant emphasis. Under conditions of large cutting depth (0.06–0.12 mm), the effect of cutting parameters on three characteristic parameters of the residual stress, such as surface residual stress, maximum residual stress and residual stress extreme value, was always different from that under small cutting depth (6–10 μm). Comparing to the right arc of the screw, the plastic deformation in the left arc is more obvious and far-reaching. The most extensive deformation region caused by WM cutting force was found for the topmost surface. As cutting speed increased, the nanohardness variation reflected the decreasing plastic deformation. Therefore, the research on physical change by WM is instructive for extending the fatigue life of the screw.

      • miRNA-1297 Induces Cell Proliferation by Targeting Phosphatase and Tensin Homolog in Testicular Germ Cell Tumor Cells

        To investigate the role of miR-1297 and the tumor suppressor gene PTEN in cell proliferation of testicular germ cell tumors (TGCT). MTT assays were used to test the effect of miR-1297 on proliferation of the NCCIT testicular germ cell tumor cell line. In NCCIT cells, the expression of PTEN was assessed by Western blotting further. In order to confirm target association between miR-1297 and 3'-UTR of PTEN, a luciferase reporter activity assay was employed. Moreover, roles of PTEN in proliferation of NCCIT cells were evaluated by transfection of PTEN siRNA. Proliferation of NCCIT cells was promoted by miR-1297 in a concentration-dependent manner. In addition, miR-1297 could bind to the 3'-UTR of PTEN based on luciferase reporter activity assay, and reduced expression of PTEN at protein level was found. Proliferation of NCCIT cells was significantly enhanced after knockdown of PTEN by siRNA. miR-1297 as a potential oncogene could induce cell proliferation by targeting PTEN in NCCIT cells.

      • Overexpression of NDRG2 Can Inhibit Neuroblastoma Cell Proliferation through Negative Regulation by CYR61

        Several recent studies have showed that the n-myc downstream regulated gene 2 (NDRG2) is a new tumor suppressor gene, and that it plays an important role in tumor suppression in several cancers or cancer cell lines. However, few studies focused on its function in neuroblastoma cells. In the present investigation, we demonstrated that NDRG2 overexpression inhibited their proliferation. Using a cDNA microarray, we found that overexpression of NDRG2 inhibited the expression of cysteine-rich protein 61 (CYR61), a proliferation related gene. From our research, CYR61 may partially hinder NDRG2-mediated inhibition of cell proliferation. Overexpression of NDRG2 resulted in accumulation of cells in the G1 phase, which was accompanied by upregulation of p21 and p27 and downregulation of CDK4 and cyclin D1. Taken together, these data indicate that NDRG2 inhibits the proliferation of neuroblastoma cells partially through suppression of CYR61. Our findings offer novel insights into the physiological roles of NDRG2 in neuroblastoma cell proliferation, and NDRG2 may prove to be effective candidate for the treatment of children with neuroblastoma.

      • Prognostic Evaluation of Categorical Platelet-based Indices Using Clustering Methods Based on the Monte Carlo Comparison for Hepatocellular Carcinoma

        Objectives: To evaluate the performance of clustering methods used in the prognostic assessment of categorical clinical data for hepatocellular carcinoma (HCC) patients in China, and establish a predictable prognostic nomogram for clinical decisions. Materials and Methods: A total of 332 newly diagnosed HCC patients treated with hepatic resection during 2006-2009 were enrolled. Patients were regularly followed up at outpatient clinics. Clustering methods including the Average linkage, k-modes, fuzzy k-modes, PAM, CLARA, protocluster, and ROCK were compared by Monte Carlo simulation, and the optimal method was applied to investigate the clustering pattern of the indices including platelet count, platelet/lymphocyte ratio (PLR) and serum aspartate aminotransferase activity/platelet count ratio index (APRI). Then the clustering variable, age group, tumor size, number of tumor and vascular invasion were studied in a multivariable Cox regression model. A prognostic nomogram was constructed for clinical decisions. Results: The ROCK was best in both the overlapping and non-overlapping cases performed to assess the prognostic value of platelet-based indices. Patients with categorical platelet-based indices significantly split across two clusters, and those with high values, had a high risk of HCC recurrence (hazard ratio [HR] 1.42, 95% CI 1.09-1.86; p<0.01). Tumor size, number of tumor and blood vessel invasion were also associated with high risk of HCC recurrence (all p< 0.01). The nomogram well predicted HCC patient survival at 3 and 5 years. Conclusions: A cluster of platelet-based indices combined with other clinical covariates could be used for prognosis evaluation in HCC.

        Agile Networking in Smart Grids

        Recently, the smart grid (SG) has been introduced to solve the serious network issues caused by the increasing electrical demand and the complex nonlinear nature of the electric power distribution network. The SG, regarded as the next generation power grid, can transmit power in more efficient ways by building an automatic and distributed energy delivery network. In this paper, we first assess how various existing networking technologies, such as IEEE 802.11 (WiFi) and IEEE 802.15.4 (ZigBee), meet the requirements of the SG communication protocols. Specifically, we classify the existing network protocols into three categories: WSN-based networking, WiFi-based networking, and wireline-based networking. We then survey the security issues regarding the SG. Finally, we propose an agile SG networking architecture and show the effectiveness of different adopted networking technologies and, as a result, present a candidate solution to implement agile networking in SGs.

      • Achieving high mass loading of Na<sub>3</sub>V<sub>2</sub>(PO<sub>4</sub>)<sub>3</sub>@carbon on carbon cloth by constructing three-dimensional network between carbon fibers for ultralong cycle-life and ultrahigh rate sodium-ion batteries

        <P><B>Abstract</B></P> <P>The mass loading of the active materials in most flexible electrodes is relatively low, which greatly impedes their practical application. Here, we report a facile strategy to achieve high mass loading of Na<SUB>3</SUB>V<SUB>2</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>@carbon (NVP@C) supported on carbon cloth (NVP@C-CC) by a two-step coating followed by an annealing treatment and the resultant NVP@C-CC membrane can be used as a binder-free cathode for sodium ion batteries (SIBs). The NVP@C is not only uniformly anchored on the surface of carbon fibers of CC, but also filled between carbon fibers of CC in interconnected three-dimensional (3D) macroporous structure. It is because of the full use of the spaces between carbon fibers of CC that we achieve a high NVP@C mass loading. Thus-obtained NVP@C-CC exhibits excellent cyclability (82.0% capacity retention over 2000 cycles at 20C) and high rate capacity (96.8mAhg<SUP>−1</SUP> at 100C and 69.9mAhg<SUP>−1</SUP> at 200C) for sodium half cells and meanwhile the high mass loading of NVP@C on CC also endows the cell with fairly high energy and powder densities of 396Whkg<SUP>−1</SUP> and 97kWkg<SUP>−1</SUP>. Furthermore, it also presents superior cycling stability and rate performance when evaluated as full battery (NaTi<SUB>2</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>@C as the anode) cathode.         Iterative Learning Control for a Class of Mixed Hyperbolic-parabolic Distributed Parameter Systems

        This paper deals with the problem of iterative learning control algorithm for a class of mixed distributedparameter systems. Here, the considered distributed parameter systems are composed of mixed hyperbolicparabolicpartial differential equations. The domain of the system is divided into two parts in which the system ishyperbolic and parabolic, respectively, with transmission conditions at the interface. According to the characteristicsof the systems, iterative learning control laws are proposed for such mixed hyperbolic-parabolic distributedparameter systems based on P-type learning scheme. Using the contraction mapping method, it is shown that thescheme can guarantee the output tracking errors on L2 space converge along the iteration axis. A simulation exampleillustrates the effectiveness of the proposed method.

        Mechanical behavior of rock-coal-rock specimens with different coal thicknesses

        To explore the influence of coal thickness on the mechanical behavior and the failure characteristics of rock-coal-rock (RCR) mass, the experimental investigation of uniaxial compressive tests was conducted first and then a systematic numerical simulation by particle flow code (PFC2D) was performed to deeply analyze the failure mechanical behavior of RCR specimens with different coal thicknesses in conventional compression tests. The overall elastic modulus and peak stress of RCR specimens lie between the rock and the coal. Inter-particle properties were calibrated to match the physical sample strength and the stiffness response. Numerical simulation results show that the deformation and strength behaviors of RCR specimens depend not only on the coal thickness, but also on the confining pressure. Under low confining pressures, the overall failure mechanism of RCR specimen is the serious damage of coal section when the coal thickness is smaller than 30 mm, but it is shear failure of coal section when the coal thickness is larger than 30 mm. Whereas under high confining pressures, obvious shear bands exist in both the coal section and the rock section when the coal thickness is larger than 30 mm, but when the coal thickness is smaller than 30mm, the failure mechanism is serious damage of coal section and shear failure of rock section.
