        실험적 급성 허혈성 신부전에서 Verapamil 이 혈중 Polyamine 동태에 미치는 영향

        N/A It has been proposd that calcium entry from an external medium increases intracellular free calcium to toxic levels during ischemic acute renal failure, and verapamil (ARF) has been suggested as the agentblocking calicium entry into renal cells and protecting renal function during ischemic injury. Polyamines, anorganic cations that play various roles in normal cellular proliferation and differentiation, accumulate in renal failare. Alsa it has been suggested tht the synthesis and metabolism of polyamine are influence by acute changes of the general condition, such as acute renal failure, and are mediated by a calcium influx into the cells. The study was designed to study the protective effects of systemic verapamil pretreatment on renal function and the influence on polyamine metabolism in experimental ischemic ARF in cats. For these purpose, the experimental animals were divided into 2 groups. While Group I (n=5) was an ischemic ARF model by renal artery clamping for 60 minutes, Group II (n=5) was ischemic ARF with systemic verapamil (5 ml/min/kg) pretreatment. The creatinine clearance and plasma and urinary polyamine were measured in each group before and after the renal artery clamp. The results were as follows: 1) Creatinine clearance before and after the renal artery clamp were 10.64±7.18 ml/min/kg and 2.09±1. 70 ml/min/kg in Group I, 4.47±3.38 ml min/kg and 0.60±0.79ml/min/kg in Group II, respectively, So creatinine clearance decreased more significantly in Group II campared with Group I. 2) Plasma polyamine increased after ischemia in Group I. In group I, plasma levels of putrescine, spermidine, and spermine before ischemia were 4.75±0.40 nmol/ml, 0.69±0.09 nmol/ml, and 0.83±0.63 nmol/ml, were elevated to 7.17±2.91 nmol/ml, 9.83±1.46 nmol/ ml, and 2.64±1.14nmol/ml after ischemia. But in Group II, the plasma level of polyamine was not changed, and especially, spermine decreased significantly from 0.83±0.27 before ischemia to 0.49±0.23 nmol/ml after ischenmine (p=0.033). 3) Urine polyamine excretion decreased after ischemia in Group I and Group II. In Group II, urinary excretion of spermidine and spermine before ischemia, 0.13±0.10 nmol/min and 0.17±0.13nmol/min, decreased after ischemia to 0.01±0.01nmol/min (p=0.019) and 0.032±0.26 nmol/min (p=0.0257). 4) In renal tissue, spermine content vas highest. In Group II, preischemic spermine were 397.20 nmol/g and increased to 646.66nmol/g after ischema, But there were no significant changes in the polyamine contents in Group II. From these data, it was suggested that systemic verapamil pretreatment exerts no protective effect on ischemic ARF. Plasma polyamines are elevated in ischemic ARF, and verapamil may protect these elevations.

      • Effect of Chromatin Changes in the Germinal Vesicle on the Development of Porcine Embryos In Vitro

        In all the studies of mammalian species, chromatin in the germinal vesicle (GV) is initially decondensed with the nucleolus not surrounded by heterochromatin (the NSN configurations). During oocyte growth, the GV chromatin condenses into perinucleolar rings (the SN configurations) or other corresponding configurations with or without the perinucleolar rings, depending on species. During oocyte maturation, the GV chromatin is synchronized in a less condensed state before germinal vesicle breakdown (GVBD) in species that has been minutely studied. As not all the species show the SN configuration and gene transcription always stops at the late stage of oocyte growth, it is suggested that a thorough condensation of GV chromatin is essential for transcriptional repression. Because the GV chromatin status is highly correlated with oocyte competence, oocytes must end the NSN configuration before they gain the full meiotic competence and they must take on the SN/corresponding configurations and stop gene transcription before they acquire the competence for early embryonic development. In this study, we firstly investigated whether the layer of cumulus cells and size of oocytes could determine chromatin configurations in porcine oocytes. Using Hoechst3342 staining, the GV nucleolus and chromatin of porcine oocytes was classified into SN and NSN configurations. Next, we examined the changes in GV chromatin configurations during growth and maturation of porcine oocytes. In addition, the maturation and parthenogenetic development abilities in vitro were significant different between the SN and NSN configurations oocytes. These results indicated that chromatin changes in GV oocytes affect the development potential of parthenogenetic embryos.

        Effect of sous-vide cooking conditions on the physicochemical, microbiological and microstructural properties of duck breast meat

        Objective: Sous-vide cooking offers several advantages for poultry meat, including enhanced tenderness, reduced cooking loss, and improved product yield. However, in duck meat, there are challenges associated with using the sous-vide method. The prolonged cooking time at low temperatures can lead to unstable microbial and oxidative stabilities. Thus, we aimed to assess how varying sous-vide cooking temperatures and durations affect the physicochemical and microbial characteristics of duck breast meat, with the goal of identifying an optimal cooking condition. Methods: Duck breast meat (Anas platyrhynchos) aged 42 days and with an average weight of 1,400±50 g, underwent cooking under various conditions (ranging from 50℃ to 80℃) for either 60 or 180 min. Then, physicochemical, microbial, and microstructural properties of the cooked duck breast meat were assessed. Results: Different cooking conditions affected the quality attributes of the meat. The cooking loss, lightness, yellowness, Hue angle, whiteness, and thiobarbituric acid reactive substance (TBARS) values of the duck breast meat increased with the increase in cooking temperature and time. In contrast, the redness and chroma values decreased with the increase in cooking temperature and time. Cooking of samples higher than 60℃ increased the volatile basic nitrogen contents and TBARS. Microbial analysis revealed the presence of Escherichia coli and Coliform only in the samples cooked at 50℃ and raw meat. Cooking at lower temperature and shorter time increased the tenderness of the meat. Microstructure analysis showed that the contraction of myofibrils and meat density increased upon increasing the cooking temperature and time. Conclusion: Our data indicate that the optimal sous-vide method for duck breast meat was cooking at 60℃ for 60 min. This temperature and time conditions showed good texture properties and microbial stability, and low level of TBARS of the duck breast meat.

        Forty eight strains of yeast were cultured in potato dextorse(PD) broth at 30˚C for 24 hr and centrifuged with 12,000 rpm for 20 min. After concentrated the cultures, antihypertensive angiotensin I-converting enzyme(ACE) inhibitory activities of its concentrates were investigated. Among them, the concentrates from Saccharomyces cerevisiae Y183-3 showed the highest ACE inhibitory activity of 71.8%. The ACE inhibitor from Saccharomyces cerevisiae Y183-3 was maximally produced when Saccharomyces cerevisiae Y183-3 cultured in PD broth at 30˚C for 36 hr.

        파 에너지 (자외선, 초음파)/과황산나트륨을 이용한 이부프로펜 분해특성 연구

        In this study, ibuprofen(IBP) degradation by the photochemical (UV/S2O82-) and sonochemical (US/S2O82-) processes was examined under various parameters, such as UV (10~40±5 W/L) and US (50~90±5 W/L) power density, optimum dosage of persulfate ion (S2O82-), temperature (20~60℃) and anions effect (Cl-, HCO3-, CO32-). The pseudo?first?order degradation rate constants were in the order of 10-1 to 10-5 min-1 depending on each processes. The synergistic effect of IBP degradation in UV/S2O82- and US/S2O82- processes could investigated, due to the generation of SO4- radical. This result can confirm from the produced H2O2 and SO42- concentration in each processes. IBP degradation rate affected by the S2O82- dosage, temperature, power and anion existence parameters. In particular, IBP degradation rate increased with the increase of the temperature (60℃) and applied power density (UV:40±5 W/L, US:90±5 W/L). On the other hand, anions effect on the IBP degradation was negative, due to the anion play as a the scavenger of radical.

        시뮬레이션을 활용한 공급사슬 내 재고관리 방법 비교 연구

        본 연구는 다양한 공급망 내 재고관리 방법 중 최적의 방법을 찾아내는 것을 목적으로 하고 있다. 비교대상 재고관리 방법은 Min-Max, 정기, 정량, 목표재고관리 방법으로 각 제품별 리드타임과 수요를 달리하여 구성하였다. 또한 공급사슬 망의 단계가 2단계인 경우와 3단계인 경우를 구분하여 공금망 단계를 줄였을 경우의 결과를 분석한다. 시뮬레이션 모형은 8개의 시나리오로 나뉘며, 각 시나리오는 주문방식과 단계로 구분할 수 있다. 시뮬레이션을 위한 입력 자료는 기존의 선행연구 결과와 실제데이터를 토대로 하였다. 시뮬레이션의 평가는 매출액, 총판매건수, 총배송비, 총결품량, 결품횟수, 결품손실금, 평균 재고량, 재고관리비, 순이익으로 실시한다. 시뮬레이션 결과 목표재고관리 방법이 가장 우수하게 나타났으며, 목표재고관리 시스템은 기존의 재고관리 방법을 사용한 시나리오보다 순이익 및 재고 등의 측면에서 우수한 결과를 나타냈다. 또한 기존의 공급사슬에서 한 단계 공급망을 줄였을 경우 수익이 증가하는 것을 확인할 수 있었다. 본 연구 결과는 목표재고관리 시스템을 추가적으로 고려하고 있다는 것과 공급사슬의 단계를 줄일 수 있는 개선 방안을 논의했다는 것에 의의를 둘 수 있다. This research focuses on the method of inventory management to find out the best way in various SCM. There are several comparable methods of inventory management like Min-Max, regular interval, fixed target inventory that have different lead-time and demand for each product. Also, the inventory performances between the case of three-stages and two-stages in SCM are analyzed. 8 scenarios for the simulation are suggested considering order ways and stages. Input data for the simulation is from the result of the previous studies and actual data in the market. The performance measurements are selected as sales revenue, total number of sales, total delivery cost, total inventory shortages, the number of inventory shortage occurrence, the loss for inventory shortages, average inventory, handling cost, net profit. According to the results of the simulation, fixed target inventory management is better than existing inventory management method by net profit and inventory and so on. Also, it was confirmed that the increase in profit is obtained when SCM stages is reduced from three-stages to two-stages in existing SCM. The result of the research provides significant meaning that newly introduced target inventory management system shows good performance and SCM performance can be improved by reducing the stages.

        Interaction between Neuronal Depolarization and MK-801 in SH-SY5Y Cells and the Rat Cortex

        Objective: The interaction between MK-801, a model of psychosis and KCl-induced depolarization or electroconvulsive shock (ECS), a therapeutic model of electroconvulsive therapy (ECT), was investigated in SH-SY5Y cells and the rat frontal cortex. Methods: SH-SY5Y cells were pretreated with 1 μM MK-801 for 15 min, followed by cotreatment with 100 mM KCl for 5 min. MK-801 was reintroduced after the KCl was washed out, and the samples were incubated before harvesting. For the experiments in rats, male Sprague-Dawley rats were treated with MK-801 followed by ECS. Immunoblot analyses of glycogen synthase kinase 3β (GSK3β) (Ser9), AKT (Ser473) and extracellular legulated kinase (ERK)1/2 in SH-SY5Y cells and the rat frontal cortex were performed. Results: KCl-induced neuronal depolarization resulted in the transient dephosphorylation of AKT (Ser473) and GSK3β (Ser9), followed by increased phosphorylation of the enzymes in SH-SY5Y cells. Cotreatment with MK-801 and KCl inhibited the initial dephosphorylation of AKT and GSK3β produced by KCl-induced neuronal depolarization. Similarly, ECS resulted in the transient dephosphorylation of AKT (Ser473) and GSK3β (Ser9), whereas cotreatment with MK-801 inhibited the initial dephosphorylation of AKT (Ser473) and GSK3β (Ser9) produced by ECS in the rat frontal cortex. No significant interaction was observed between MK-801 and KCl in the dephosphorylation of ERK1/2. Conclusion: These results suggest that an antagonistic interplay between MK-801 and neuronal depolarization by KCl or ECS is involved the regulation of AKT (Ser473) and GSK3β (Ser9) phosphorylation.

      • Sol-Gel방법에 의해서 성장시킨PbTiO₃박막의 결정화와 물리적 성질

        Ferroelectric PbTiO3 thin films coated on SiO2/Si(100) and ITO glass substrates were prepared by sol-gel processing of metal alkoxide solutions and solvents. The use of sol-gel method for preparing various functional films makes it possible for the homogeneous distribution of elements at the molecular level, precise composition control, lower temperature synthesis, large surface coating and high optical quality. First of all, the XRD investigation shows that perovskite phase of polycrystalline PbTiO3 was formed by four times coating of the concentrated solution and by the heat treatment over 500, 600℃(O2 gas and air) and over 10, 20, 30 min. Surfaces of thin films were crack-free and uniform and the average grain size investigated by SEM was 0.25∼1.00 ㎛. Dielectric constant of PbTiO3/ITO glass thin film were 380 ∼ 5.25 in the region of 1 kHz∼ 10 MHz at room temperature(30℃), respectively. From the hysteresis loop of PbTiO3/ITO glass thin film, a remnant polarization of 0.382 μC/cm2 and a coercive filed of 23.85 kV/cm were obtained. At the frequency(5 kHz), C-V measurements of films in MFIS and MFM configurations showed well-behaved C-V characteristics and an injection type hysteresis. The capacitance of flat band(CFB) was 60 pF. The band gap energies of the thin films depend on the assumption of direct or indirect transition. Using one layer structure model, we calculated refractive index, extinction coefficient, absorption coefficient and band gap energy of thin film in the wavelength range of 300 to 800 nm. The band gap energy, refractive index and extinction coefficient at λ = 6325 Å of the PbTiO3 thin film were 3.7 eV, 1.96 and 0.1 respectively.

      • 횡문근융해증과 경한 신손상을 동반한 급성 A형 간염 1예

        A 48-year-old male visited the emergency room of the authors’ hospital due to nausea, vomiting, and myalgia for four days. Acute hepatitis A was identified from the serologic marker of the hepatitis A virus. Mild elevation of the serum creatinine and creatinine phosphokinase(CPK) suggested rhabomyolysis, which was confirmed with the serum aldolase, myoglobin, and urine myoglobin. With supportive care, both the liver and renal func- tions were recovered gradually and fully. This case shows that rhabdomyolysis can be one of the mechanisms of renal complication in cases of acute symptomatic hepatitis A.
