      • Exploration, Characterization and Profiling of Stress Resistance and Reproductive Organ Development Related Gene Families in Brassica rapa

        The genus Brassica includes a number of economically important crops that harbor a vast diversity in morphotype and adapted to grow in most part of the world. However, the Brassica crops are being affected by range of abiotic and biotic stresses that are considered as the main detriment of yield loss. To deal with this, it is necessary to give focuses in the genetic level for enhancing resistance against stresses. Exploitation of stress resistance gene family having important growth regulatory functions could be the most systematic approach to develop Brassica crops with higher stress tolerance. In this study, we identified different gene families‟ viz. MADS-box, Brassinazole Resistant (BZR), MYB and TIFY having stress resistant role with different growth regulatory functions. Primarily the above mentioned family genes were retrieved with their sequence information from the Brassica database. All the members of these gene families were thoroughly characterized by sequence analysis, comparison study and expression profiling after applying different abiotic and biotic stresses. Expressions of these family genes also studied in the different growth organs of B. rapa. And, this study identified set of genes with their constitutive expression patterns in different reproductive organs of B. rapa specially in flowers. In our study 167 MADS-box genes were identified, thereof 89 MIKCc and 11 MIKC* genes exhibited widespread expression in different growth organs. More importantly, from a low temperature-treated whole-genome microarray data set, 19 MADS-box, 15 BZR, 27 MYB and 21 TIFY genes were found to show variable transcript abundance in two contrasting inbred lines of B. rapa Chiifu and Kenshin. In further validation test through quantitative reverse transcriptase-polymerase chain reaction (qPCR), most of the gene families showed differential expression patterns, where inbred line Chiifu showed higher resistance against cold than Kenshin. Subsequently, those gene families were also tested against other environmental stresses like salt, drought and different exogenous hormone treatment. And, 90 genes (13 MADS-box, 15 BZR, 42 MYB and 20 TIFY genes) were identified with their corresponsive expressions against multiple stresses. In the biotic stress study, we have found that 10 MYB and 20 TIFY „JAZ‟ showed resistance against F. oxysporum f.sp. conglutinans thereof one MYB showed resistance against P. carotovoram subsp. caratovorum. In addition, our extensive expression profiling on different flower organ development revealed 18 MADS-box, 13 MYB and 20 TIFY genes with their constitutive expression patterns in male reproductive tissues, which might be an important basis for studying male sterility in B. rapa. Therefore, the extensive annotation and transcriptome profiling results of this study will be useful for understanding the role of MADS-box, BZR, MYB and TIFY genes for stress resistance and regulatory functions of growth and development of reproductive organs, which obviously provides the basis for functional characterization and exploitation of the candidate genes for target specific genetic engineering of B. rapa.
