      • 미생물의 첨가가 수중의 가용태 인산에 미치는 영향

        토양개량제에서 분리한 5종의 미생물을 토양이 1% 첨가된 YPD배지에 배양하면서 경시적으로 배양기의 pH, 생균 수, 가용성 인산등을 조사하였던 바 그 결과를 요약하면 다음과 같다. 배양기내의 pH는 aerobe의 JUFA03와 anaerobes의 JUFA05에서는 초기의 pH와 시간이 경과할수록 그다지 큰 차이를 보이지 않은 반면 aerobe의 JUFA01-02와 anaerobe의 JUFA04균에서는 배양시간이 경과함에 따라 점진적으로 저하되는 경향을 보였다. 호기성균인 JUFA01과 JUFA02는 시간이 경과함에 따라 용액중의 인산함량을 증가시킨 반면 JUFA03은 48시간까지 증가하다가 그 이후부터는 오히려 감소시켰다. JUFA01과 JHFA02는 배양기내의 대부분의 인산을 흡수하여 세포내 축적한 반면에 JUGA03, JUFA04 그리고 JUFA05 가용성 인산을 배양후 72시간이 경과한 후 각각 36.90%, 78.23% 및 96.08%만 이용하였다. 따라서 부영양화의 원인물질 중 수중의 인산을 이와 같은 미생물을 이용하면 감소시킬 수 있음이 확인되었다. Phosphate solubilizing microbes were isolated form the soil inhancer and selected 5 microorganisms among them, cultivated in YPD medium adding 1% of soil. The pH, number of cells, phosphate solubilizing activities with time of those microbes were analysed. As result, pH of the aerobes, JUFA01 and 02, and anaerobe JUFA04 decreased gradually with time, whereas the aerobe JUFA03 and anaerobe JUFA05 remained as initial pH. The amount of soluble phosphate released by the aerobes, JUFA01 and 02, increased with time where as that of JUFF03 increased up to 48 hours and after that decreased. The aerobes, JUFA01 and 02, utilized the most of phosphates in the medium and accumulated those in the cells, where as JUFA03, JUFA04 and JUFA05 discharged the free phosphate form in the medium as 24.07%, 45.29 %and 55.17 %after 72 hours repectively. Therefore, the certain microbes was proved to be suitable for the reducing the soluble phosphate in water which caused to eutrophication.

      • 통조림의 初溫 및 冷却이 殺菌에 미치는 影響

        살균전 통조림 내용물의 온도와 냉각처리 유무가 통조림의 살균에 미치는 영향을 조사하였다. 살균시간을 최소화하고 레토르트안과 통조림냉점의 두 곳에서 온도를 감지하여 치사율의 적산치를 계속적으로 이행할 수 있는 컴퓨터시스템을 이용하였다. 8% 벤토나이트 분산액을 301-5호관에 채우고 반복 실험을 통해서 내용물의 초온의 차이와 냉각수의 처리 유무 공히 살균시간에 뚜렷한 영향을 미쳤다. The effect of initial can temperatures and water cooling treatment on the cans for thermal sterilization was described. The accumulated lethal rate method based on microcomputer-based electronic system has introduced in this experiments to minimize the process time and provide precise temperature measurements at 2 points with continuous calculation of the lethal rate. According to a series of experiments which were performed by using 301-5 cans of 8% bentonite suspension, there was significant effect in accumulated lethal rate initiated in different temperatures and treated water cooling treatment.

      • 백금저항제를 이용한 report내의 온도계측 시스템

        The tempearture measuring system which consisted of 8 bit A/D converter, singnal conditioning circuits and two of Pt 100 thermal resister was developed and tested. The temperature measuring errors by using the digital temperature calibration curves for measuring the temperature of the inside of retort and the cold point of the can in the retort were ± 1.524 ℉ and ± 0.312 ℉ respectively. The relation between the bulb temperature and the converted digital values tested from 50℃ to 150℃ gave strait lines with slop of 0.996(the sensor installed inside retort) and 0.999(sensor mounted in the cold point) respectively.

      • 마이크로 컴퓨터를 이용한 pH측정 시스템에 관한 연구

        Analog pH meter에 microcomputer 계측 시스템을 구성하여 pH 계측의 신속, 정확 및 신뢰성을 부여하고 계측 데이터의 분석을 용이하게 하여서 식품공학의 다른 물리, 화학적 특성의 계측 및 multi-system 구성의 기초 연구를 수행하기 위하여 본 연구를 실시하였으며 그 결科를 요약하면 다음科 같다. 본 시스템은 16 chanel18 bit A/D converter, apple Ⅱ microcomputer, analog pH meter와 OP AMP으로 구성하였다. pH 완충욕액과 시스템에 나타난 Digital value의 관계는 y=0.06x -0.487로 나타났다. Analog-computer system과 digital pH meter에서 나타난 pH 값의 관계는 y=1.033x-0.303으로 나타났다. 이 시스템은 만족할만한 성과를 보였으며 이 analog-computer pH meter system에 의한 pH 측정은 식품공학적 multisystem 구성시 pH 연속 및 제어등에 유용하게 사용될 것으로 사료된다. In order to measure the pH accurate, rapid and reliable, the microcomputer based pH measuring system was studied. This system were consisted of 16 chanels 8 Bit A/D converter Apple Ⅱ Microcomputer, conventional analog pH meter and signal conditioner usign OP AMP. The relationship between pH buffer(y) and digital valuesx from microcomputer resulted as y=0.058x-0.2436 and the relationship between the pH values read in digital pH metery and analog-computer pH meterx resulted as y=1.033x-0.303. The performance of this system was succesful in measuring the pH by analog-computer system and it is considered that this system can be applied for measuring the pH continuously and its control for further multisystem of the food industries.

      • Microcomputer를 이용한 유량계측장치에 관한 연구(I)

        흐름유체에 열을 가하여 유량변화에 따른 온도변화를 Microcomputer로 계측하는 것을 이용하여 유체의 유량을 측정할수 있는 계측장치를 개발하여 그 성능을 실험적으로 조사한 결科는 다음科 같다. 1. A/D변환기 입력신호 파형의 통科 주파수를 3.88Hz 미만으로 하므로서 관내의 맥동, 외부로부터의 전자파 잡음등의 영향을 소거할 수 있었고, 이를 기준으로 신호처리 회로를 구성하였다. 2. 개발된 시스템의 유량측정은 2cc/sec-133cc/sec까지 가능하였으며, 실험에서 유량 Q와 디지털 값 X와의 관계식은 다음科 같이 나타났다. Q=11.542-0.1805X 3. 본 측정시스템은 저렴한 가격으로 간단하게 제작할 수 있지만 사용목적에 부합되는 유량의 한계에 따라서 측정장치가 보다 개선되어야 할 것으로 생각된다. In order to measure the flow rate in pipe or tube, the microcomputer-based flow measuring system was designed and tested. The system was consisted of 16 channel A/D converter, Apple Ⅱ microcomputer, signal conditioner using OP AMP and flow sensor consisting of a heater and two thermocouples setted in the pipe. The system could be measured the flow rate from 2 cc/sec to 133 cc/sec, but it had some problemes that measured flow rate involved high resolution error and digital value became function of square root of flow rate. Accordingly, the system should be suitable for low flow rate measurement if it exchange the sensor and signal conditioning unit as sensible and linear devices.

        저염 우렁쉥이 젓갈의 가공 및 갈변방지

        These studies were carried out to develop the low salt-fermented sea foods from ascidian. In present papers, we examined optimum processing conditions and quality improvement of the low salt-fermented ascidian. The optimum processing conditions of low salt-fermented ascidian was as followed. After deshelling and slicing a ascidian muscle in 5 mm width, and dipping in a 10% salt and 1% sodium erythorbate solution for 20 min. We dewatered it, and dipped it in 0.1% sodium bisulfite solution for 1 min, then added an anchovy sauce and rice gruel(1:1) mixture and finally fermented it at 5℃ for 15 days. The moisture and salinity contents of the low salt-fermented ascidian were 75.0~75.4% and 8.0 ~8.5%, respectively. During fermention at 5℃ for 20 days, amino-N contents of the low salt-fermented ascidian increased, and texture were softened gradually. The viable cell counts were 4.2~4.5×10^(4) CFU/g, and tended to decrease in the first stage during fermentation. The ratio of saturated fatty acids tended to increase in the early stage of fermentation, while that of polyunsaturated fatty acids decreased slightly during fermentation. In the results of chemical experiments and sensory evaluation, dipping treatment in 1% sodium erythorbate solution and 0.1% sodium bisulfite solution showed a good color and prevention of browning in the low salt-fermented ascidian, and also adding anchovy sauce and rice gruel mixture improved the taste of it.

        채택 빈도수에 의한 교육용 소프트웨어 품질평가 준거의 설정 및 분석

        교육용 소프트웨어가 갖는 여러 가지의 우수한 기능과 장점에도 불구하고 실제적으로 학습 현장인 학교와 가정에서 교육용 소프트웨어의 활용도가 저조한 현상은 교육용 소프트웨어가 교육 전문가나 컴퓨터 소프트웨어 개발 전문가의 입장에서 연구되고 개발되어 학습자의 요구사항이 교육용 소프트웨어에 제대로 반영되지 못한 결과로 볼 수 있다. 이에 따라 이 연구에서는 교육용 소프트웨어의 평가 도구들이 소비자의 요구를 얼마나 수용하고 있는지 알아보는 사전 연구의 일환으로서, 국내외에서 발표된 기존의 교육용 소프트웨어의 평가 도구들이 중요하게 다루고 있는 평가 준거는 무엇인지 교육용 소프트웨어의 평가 도구에서 채택된 빈도수를 통하여 조사 분석하였다. 그리고 이 분석 결과와 비교하여 교육용 소프트웨어의 실수요자인 학습자들은 교육용 소프트웨어에 대한 기존의 품질 평가 도구가 가지고 있는 품질 평가 준거를 어느 정도의 차이점을 가지고 중요하게 생각하는지에 대하여 조사하고 분석하였다. In spite of various good functionalities and merits of educational software products, they are generally not used widely in school and home, One of these reasons is which many of the educational software products are developed by educational professionals without adequate consideration to the requirement of learners. So, in this paper, in order to see the degree of how much learner’s requirement have been met in educational software products, we survey and analyze the evaluation criteria based on the adoption frequencies which have been searched out from domestic and foreign educational software evaluation tools. And, by the learner-centered survey, we compare and analyze the difference of quality estimation criteria for educational software between the viewpoints of students and the selected evaluation criteria treated as important in educational software evaluation tools.

      • Application of Stopping Rule of Pegylated Interferon-Alpha 2a Therapy to Korean Patients with Chronic Hepatitis B

        Aims: In pegylated interferon-alpha 2a (PEG-IFN) of chronic hepatitis B (CHB), the decline of hepatitis B surface antigen (HBsAg) titers after 12-week therapy has been considered as a potent predictor that can determine discontinuation of therapy. In this study, we evaluated treatment response in Korean patients with CHB after 12-week PEG-IFN therapy. Methods: 100 patients with CHB were prospectively analyzed from April 2015 to September 2016 in multi-centers. After 12-week use of PEG-IFN, we evaluated the changes of HBsAg, heapatitis B virus (HBV) DNA, and other liver-related markers in patients with CHB. Results: Of 100 patients with CHB, heapatitis B e antigen (HBeAg)- positive patients were 75, and HBeAg-negative patients were 25. After 12-week PEG-IFN therapy, the number of HBeAg-positive patients with HBsAg less than 20,000 IU/mL was 61 (82.43%), and the number of HBeAg-negative patents with declined HBsAg titers or decreased HBV DNA levels more than 2 log IU/mL was 19 (76.00%). In HBeAg-positive patients, the decline of HBV DNA and HBsAg levels after 12 weeks was 1.83 ± 2.05 log10 IU/mL (p < 0.0001) and 6452.33 ± 11266.74 IU/mL (p <0.0001), respectively. In HBeAg-negative patients, the decrease of HBV DNA and HBsAg levels after 12 weeks was 2.55 ± 2.11 log10 IU/mL (p < 0.0001) and 1940.71 ± 5071.62 IU/mL (p < 0.0001), respectively. The number of patients with HBsAg titers less than 1,500 IU/mL after 12 weeks was 14 (18.92%) in HBeAg-positive patients and 17 (68.00%) in HBeAg-negative patients, respectively. Conclusions: As applying 12-week stopping rule, early anti-viral response rate of PEG-IFN was confirmed in Korean patients with CHB, and we could apply this results to determine patients who will continue long-term PEG-IFN therapy in the early period of treatment.

        Upregulation of the serine palmitoyltransferase subunit SPTLC2 by endoplasmic reticulum stress inhibits the hepatic insulin response

        Endoplasmic reticulum (ER) stress is induced by various conditions, such as inflammation and the presence of excess nutrients. Abnormal accumulation of unfolded proteins leads to the activation of a collective signaling cascade, termed the unfolded protein response (UPR). ER stress is reported to perturb hepatic insulin response metabolism while promoting insulin resistance. Here, we report that ER stress regulates the de novo biosynthesis of sphingolipids via the activation of serine palmitoyltransferase (SPT), a rate-limiting enzyme involved in the de novo biosynthesis of ceramides. We found that the expression levels of Sptlc1 and Sptlc2, the major SPT subunits, were upregulated and that the cellular concentrations of ceramide and dihydroceramide were elevated by acute ER stress inducers in primary hepatocytes and HepG2 cells. Sptlc2 was upregulated and ceramide levels were elevated by tunicamycin in the livers of C57BL/6J wild-type mice. Analysis of the Sptlc2 promoter demonstrated that the transcriptional activation of Sptlc2 was mediated by the spliced form of X-box binding protein 1 (sXBP1). Liver-specific Sptlc2 transgenic mice exhibited increased ceramide levels in the liver and elevated fasting glucose levels. The insulin response was reduced by the inhibition of the phosphorylation of insulin receptor β (IRβ). Collectively, these results demonstrate that ER stress induces activation of the de novo biosynthesis of ceramide and contributes to the progression of hepatic insulin resistance via the reduced phosphorylation of IRβ in hepatocytes.
