        Improvement of Fast-Growing Wood Species Characteristics by MEG and Nano SiO<sub>2</sub> Impregnation

        Jabon (Anthocephalus cadamba) is a fast-growing wood species that is widely utilized for light construction and other purposes in Indonesia. The objectives of the current study were to determine the effects of monoethylene glycol (MEG) and SiO<sub>2</sub> nanoparticles (nano SiO<sub>2</sub>) impregnation treatment on the dimensional stability and density of jabon wood and to identify the characteristics of impregnated jabon wood. Wood samples were immersed in water (as untreated), MEG, 0.5% MEGSiO<sub>2</sub>, then impregnated by applying 0.5 bar of vacuum for 60 min, and then applying 2.5 bar of pressure for 120 min. The results showed that impregnation with MEG and Nano SiO<sub>2</sub> had a significant effect on the dimensional stability of jabon wood. Polymers can fill cell walls in wood indicated by increasing weight percentgain, antiswelling efficiency, bulking effect, and density, then decreasing in water uptake value. Jabon wood morphology by using SEM showed that MEG SiO<sub>2</sub> polymers can cover part of the pitsin the wood vessel wall of jabon. This finding was reinforced by EDX results showing that the silicon content was increased due to the addition of SiO<sub>2</sub> nano. The XRD diffraction pattern indicated that MEG SiO<sub>2</sub> treatment increased the degree of crystallinity in wood samples. Overall, treatment with 0.5% MEG SiO<sub>2</sub> led to the most improvement in the dimensional stability of 5-year-old jabon wood in this study.

        Impact of transgenic sugarcane overexpressing SoSPS1 gene on bacterial diversity, enzyme activity and minerals content in soil rhizosphere

        The transgenic sugarcane overexpressing SoSPS1 gene increased sugar content which may alter the root exudation that interact with soil rhizosphere microorganism. The microbial plays an important role in biochemical cycling and nutrient transformation in the soil. Therefore, it is important to investigate the potential risk of the transgenic sugarcane on soil ecosystem. This study is designed to evaluate the impact of overexpressing gene for sucrose-phosphate synthesis ( So SPS1) on the bacterial diversity, enzyme activity and nutrient contents. The transgenic sugarcane showed no signifi cant difference in the population of culturable bacteria in the soil rhizosphere compared to non-transgenic counterpart. The populations of total, nitrogen-fixing and phosphate-solubilizing bacteria were similar in transgenic and non-transgenic sugarcane, although the bacterial population tends to increase at later sugarcane growth stage may due to the rise in nutrition from the root exudation. The PCR analysis showed no detection of horizontal gene fl ow in the bacteria. However, urease activity was significantly decreased in the transgenic sugarcane concomitant with lower nitrogen content in the soil rhizosphere at a later growth stage might due to the transgenic sugarcane lines increased nitrogen absorption for higher growth rate. Furthermore, the nutrient contents, such as organic carbon, phosphorus, potassium, magnesium and calcium, slightly varied in the soil rhizosphere but did not affected by the transgenic sugarcane. The results suggested that the transgenic sugarcane overexpressing SoSPS1 gene may not aff ect the bacterial diversity, nutrient contents and soil ecosystem.

        Food packaging is an important part in supporting food safety rules. The fact that not all of food packaging materials meet the requirements of food packaging materials, even primary, secondary, and tertiary packaging, cause the food in the packaging is potentially contaminated. One of the primary packaging materials is a duplex carton which is a product of recycled paper. Duplex carton is the most widely used primary material because of its cheaper and more eco-friendly. However, recycled paper is likely to contain printing inks, adhesives, waxes, fluorescent dyes and bleaches, fillers, organochlorine compounds, aromatic hydrocarbons, volatile organic compounds, barrier and oil-resistant materials, amines and surfactants. One of the hazardous substances contained in primary food packaging especially duplex carton is mineral oil or mineral oil hydrocarbon (MOH). Contamination of mineral oil into food packaging can be derived from printing ink contained on recycled paper or from printing ink on the packaging itself. Animal studies show that digestion of small amounts of mineral oil compounds can accumulate in the body and damage the liver and blood vessels in the heart, lymph nodes, and can be carcinogenic. This study aims to determine the potency of mineral oil migration from recycled paper-based food packaging. Migration test method that was used was the Federal Reference Laboratory for Food Contacts at the Federal Institute for Risk Assessment (BfR). Based on the analysis result both from internal standard adition at the beginning and at the end of the process showed that there is a potency of mineral oil to migrate from recycled paper based food packaging in this case is duplex carton.

        When Tourist Audiences Encounter Each Other: Diverging Learning Behaviors of K-pop Fans from Japan and Indonesia

        Japanese hallyu fans who often travel to Korea after falling in love with Korean dramas or K-pop music are usually referred to as “tourist audiences.” More recently, K-pop tourist audiences come not only from Japan but also from Southeast Asia, China, Europe, and the Americas, expanding the nationality boundary of the concept. Although such tourist audiences are still predominantly female, the number of male K-pop tourist audience members is also growing slowly. In this study, we address the question of learning behavior among tourist audiences from different countries in the K-pop mecca of Seoul. Based on the notion of forward and retrospective learning, in-depth interviews with Japanese and Indonesian female K-pop fans who have encountered fans from other countries were conducted in order to delineate differing patterns of learning behavior. We find that forward learners from Indonesia actively engage in meeting Korean, Chinese, and Japanese fans, whereas retrospective learners from Japan are very reluctant to meet Chinese or Indonesian fans, although they were somewhat interested in meeting their North American or Western counterparts.

        From campus to legislatures: Women’s experience of electoral politics in Indonesia

        The path from student politics to electoral politics is a well-worn one across Asia. Literature examining this pathway overwhelmingly does so from the perspective of men, failing to adequately acknowledge women's relative inability to make the transition from campuses to legislatures in the same way. This article focuses on Indonesian women's experiences to explain why they remain underrepresented in politics and sheds light on the ways its student movements have shaped democracy. Through in-depth interviews with women who have and have not entered electoral politics after early political apprenticeships in student movements and focus group discussions with current students on potential political careers, we identify the barriers and opportunities faced by women in enhancing their political participation. We argue that the literature on student politics across Asia will remain incomplete without seriously attending to women's experiences. We thereafter argue for the need to identify ways to support them in their journey from campus to legislatures.

        Purchase Intention of Green Products Following an Environmentally Friendly Marketing Campaign: Results of a Survey of Instagram Followers of InnisfreeIndonesia

        In Indonesia, concern about environmental protection is increasing. Thus, companies not only adjust their products but also create promotion strategies with environmentally friendly messages, called green marketing. This study identifies the effect of green marketing on purchase intentions. A quantitative method with an explanatory design was conducted. Data was gathered online. In this study, the population was @InnisfreeIndonesia Instagram followers. Thus, a direct message with the survey invitation was sent to randomly selected Instagram followers. Additional followers were invited daily for about a month until 100 responses were collected. The majority of respondents were Indonesian females, age 21-25. The results show that green products partially influence purchase intentions. Once consumers consider environmental friendly issues before buying a skin care product, they may identify whether the product is toxic-free, environmentally friendly, and holds a certification that identifies it as such. Thus, potential consumers tend to seek information to set expectations about the product they want to buy, prioritize them over other products, and to refer the product to others.

        The Effect of Growth Condition on a Soluble Expression of Anti-EGFRvIII Single-chain Antibody in Escherichia coli NiCo21(DE3)

        Single-chain antibodies against epidermal growth factor receptor variant III (EGFRvIII) are potentially promising agents for developing antibody-based cancer treatment strategies. We described in our previous study the successful expression of an anti-EGFRvIII scFv antibody in Escherichia coli. However, we could also observe the formation of insoluble aggregates in the periplasmic space, limiting the production yield of the active product. In the present study, we investigated the mechanisms by which growth conditions could affect the expression of the soluble anti-EGFRvIII scFv antibody in small-scale E. coli NiCo21(DE3) cultures, attempting to maximize production. The secreted scFv molecules were purified using Ni-NTA magnetic beads and protein characterization was performed using SDS-PAGE and western blot analyses. We used the ImageJ software for protein quantification and determined the antigen-binding activity of the scFv antibody against the EGFRvIII protein. Our results showed that the highest percentage of soluble scFv expression could be achieved under culture conditions that combined low IPTG concentration (0.1 mM), low growth temperature (18℃), and large culture dish surface area. We found moderate-yield soluble scFv production in the culture medium after lactose-mediated induction, which was also beneficial for downstream protein processing. These findings were confirmed by conducting western blot analysis, indicating that the soluble, approximately 30-kDa scFv molecule was localized in the periplasm and the extracellular space. Moreover, the antigen-binding assay confirmed the scFv affinity against the EGFRvIII antigen. In conclusion, our study reveals that low-speed protein expression is preferable to obtain more soluble anti-EGFRvIII scFv protein in an E. coli expression system.
