        Two New Bibenzyl Glucosides from Dendrobium chrysotoxum

        Two new bibenzyl glucosides, 3,3',4',5-tetramethoxybibenzyl-4-O-β-D-glucopyranoside (1) and 3,4,4',5- tetramethoxybibenzyl-3'-O-β-D-glucopyranoside (2), together with five known ones, chrysotobibenzyl (3), erianin (4), chrysotoxine (5), gigantol (6) and tristin (7) were isolated from the stems of Dendrobium chrysotoxum. The structures of those compounds were elucidated by extensive spectroscopic analysis. Moreover, compounds 1-7 were assessed for inhibitory activity of two enzymes-AChE (acetylcholine esterase) and BChE (butyrylcholine esterase).

        A Combination of a Microemulsion and a Phospholipid Complex for Topical Delivery of Oxymatrine

        Oxymatrine (OMT), a water-soluble drug, has a very low oral bioavailability because of its low membrane permeability and its biotransformation in the gastrointestinal tract. Formulated as an oxymatrine-phospholipid complex (OMT-PLC) can improve the lipid solubility and effectiveness of OMT. The purpose of this study was to explore the utility of the combination of a microemulsion and an OMT-PLC as a topical delivery vehicle for enhancing the absorption and efficacy of OMT. The solubility of OMT-PLC was determined and phase diagrams of microemulsions were constructed. Various microemulsion formulations were developed and characterized by their physicochemical properties, and their in vitro and in vivo permeability through skin. An optimal microemulsion (ME4), which presented as spherical droplets and consisted of 10.0% OMT-PLC, 8.0% isopropyl myristate, 30.0% Cremophor RH40/polyethylene glycol 400 (1:1) and 52.0% water, was selected. It possessed an average droplet size of 32.4 nm, a low viscosity of 113.7 mPa · s, and a high cloud point of 88°C. Compared to the control solution, ME4 provided better skin permeability in vitro and a higher retention ratio of OMT in skin in vivo. Moreover, ME4 significantly enhanced the antiproliferative activity of OMT on scar fibroblasts. These results indicate that the combination of a microemulsion and a phospholipid complex represents an effective vehicle for topical delivery of OMT.

        Voltammetric Investigation of Vitamin B_6 at a Glassy Carbon Electrode and Its Application in Determination

        The voltammetic behavior of Vitamin B6 (VB6) was studied at a glassy carbon electrode in phosphate buffers using cyclic, linear sweep and differential pulse voltammetry. The oxidation process was shown to be irreversible over the entire pH range studied (4.0-10.0) and was adsorption controlled. The adsorption amount of VB6 on the glassy carbon electrode was examined by chronocoulometry and the value of n, product of transfer coefficient and number of electrons transferred in the rate determining step, was determined from Tafel plot. VB6 was determined by differential pulse voltammetry and the peak current was found linearly with its concentration in the range of 3 10-7-2 10-4 mol L-1. The detection limit was 1 10-7 mol L-1. The procedure was successfully applied for the assay of VB6 in tablets.

        Overexpression of a Nitrile Hydratase from Klebsiella oxytoca KCTC 1686 in Escherichia coli and Its Biochemical Characterization

        The putative Co-type nitrile hydratase (NHaseK, consisting of α- and β-subunits) genes and the putative activator (17K) gene adjacent to the β subunit region were cloned from Klebsiella oxytoca KCTC 1686. 17K is essential for the functional expression of recombinant NHaseK in Escherichia coli; however, the expression level of 17K was very low when the 17K gene and NHaseK structural genes were expressed as a gene cluster in E. coli BL21(DE3). To improve the 17K expression level and NHaseK activity, the expression cassette was redesigned by placing the 17K gene and NHaseK structural genes under the control of different promoters in the pETDuet-1 expression vector, co-expressing the 17K gene with the gene cluster in a double plasmid or a single plasmid with a double promoter, and introducing an efficient Shine- Dalgarno sequence 5' to the17K gene. The specific activity of NHaseK was improved when 17K was co-expressed with the gene cluster, whereas the production of NHaseK protein decreased. The maximum activity was achieved when an efficient Shine-Dalgarno sequence was introduced 5' to the 17K gene: the expression level of 17K was significantly improved and the expression level of NHaseK did not decrease significantly. The maximum activity was about 63,480 ± 1915.6 U/L broth towards 3-Cyanopyridine. Recombinant NHaseK could hydrolyze a wide range of aliphatic, aromatic, and heterocyclic nitriles, and convert racemic nitriles to the corresponding S-amides, with E values ranging from 9 to 17. The enzyme had a temperature optimum of 35°C and exhibited remarkably stability below 35°C.

        Synthesis of Core−Shell Au@TiO<sub>2</sub> Nanoparticles with Truncated Wedge-Shaped Morphology and Their Photocatalytic Properties

        <P>Core−shell Au@TiO<SUB>2</SUB> nanoparticles with truncated wedge-shaped TiO<SUB>2</SUB> morphology have been synthesized successfully by a simple and flexible hydrothermal route. Morphological evolution of TiO<SUB>2</SUB> shells was investigated by transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HRTEM), and X-ray diffraction technique. It has been revealed that the truncated wedge-shaped TiO<SUB>2</SUB> shells experience an epitaxially segmented orientation growth. Also, the (101) crystal planes of TiO<SUB>2</SUB> crystals grow preferentially on the surface of gold nanocrystals stabilizing the heterointerfaces, then faster [001] growth results in the “budding” process occurs, producing growth sites on the initial deposition TiO<SUB>2</SUB> layers, where the TiO<SUB>2</SUB> crystals grow up into truncated wedge-shaped morphologies. It is also found that morphological evolution of TiO<SUB>2</SUB> shells is dependent on the produced F<SUP>−</SUP> ion concentration from hydrolyzed TiF<SUB>4</SUB> precursors. The produced F<SUP>−</SUP> ions not only facilitate the formation of well-defined wedge-like TiO<SUB>2</SUB> shells, but also contribute to the externally exposed truncated crystal {004} facets. As the representative photocatalyst, the catalytic activities of the resultant core−shell Au@TiO<SUB>2</SUB> nanoparticles were investigated by photoinitiated oxidation degradation of gaseous acetaldehyde. It has been indicated that the nanostructured core−shell Au@TiO<SUB>2</SUB> photocatalyst represents high photocatalytic activity when exposed to UV or visible light irradiation.         Syntheses, Crystal Structures, and Spectral Properties of Two Coordination Compounds Based on 1,2-Bis(benzimidazolyl)benzene

        Two new benzimidazolyl-containing complexes have been synthesized by reactions of Cu^II salts and 1,2-bis(benzimidazolyl)benzene (H_2bbbz) with two different dicarboxylate ligands.When phthalate acid (H2pt) was employed as secondary ligand, a 0Dmolecular complex Cu(H_2bbbz)(pt)·(H_2pt) (1)was furnished and when the secondary ligand was instead by a linear bridging ligand of terephthalic acid (H_2tp) a 1D zipper-like coordination polymer [Cu(H_2bbbz)(tp)·2(C_2H_5OH)·H_2O]n (2) was obtained, suggesting the structure-direction effect of the secondary dicarbxylate ligand. The preliminary investigation on the spectral properties of the complexes was also presented.

        Mechanical behaviors of concrete-filled rectangular steel tubular under pure torsion

        Pure torsion loading conditions were not frequently occurred in practical engineering, but the torsional researches were important since it’s the basis of mechanical property researches under complex loading. Then a 3D finite element model with precise material constitutive models was established, and the effectiveness was verified with test data. Parametric studies with varying factors as steel yield strength, concrete strength and sectional height-width ratio, were performed. Internal stress state and the interaction effect between encased steel tube and the core concrete were analyzed. Results indicated that due to the confinement effect between steel tube and core concrete, the torsional strength of CFT columns was greatly improved comparing to plain concrete columns. The steel ratio would greatly influence the torque share between the steel tube and the core concrete. Then the torsional strength calculation formulas for core concrete and the whole CFT column were proposed. The proposed formula could be simpler and easier to use with guaranteed accuracy. Related design codes were more conservative than the proposed formula, but the proposed formula presented more satisfactory agreement with experimental results.

      • Cytotoxic T Lymphocytes Elicited by Dendritic Cell-Targeted Delivery of Human Papillomavirus Type-16 E6/E7 Fusion Gene Exert Lethal Effects on CaSki Cells

        Human papillomavirus (HPV) is the primary etiologic agent of cervical cancer. Consideration of safety and non human leukocyte antigen restriction, protein vaccine has become the most likely form of HPV therapeutic vaccine, although none have so far been reported as effective. Since tumor cells consistently express the two proteins E6 and E7, most therapeutic vaccines target one or both of them. In this study, we fabricated DC vaccines by transducing replication-defective recombinant adenoviruses expressing E6/E7 fusion gene of HPV-16, to investigate the lethal effects of specific cytotoxic T lymphocytes (CTL) against CaSki cells in vitro. Mouse immature dendritic cells (DC) were generated from bone marrow, and transfected with pAd-E6/E7 to prepare a DC vaccine and to induce specific CTL. The surface expression of CD40, CD68, MHC II and CD11c was assessed by flow cytometry (FCM), and the lethal effects of CTL against CaSki cells were determined by DAPI, FCM and CCK-8 methods. Immature mouse DC was successfully transfected by pAd-E6/E7 in vitro, and the transfecting efficiency was 40%-50%. A DC vaccine was successfully prepared and was used to induce specific CTL. Experimental results showed that the percentage of apoptosis and killing rate of CaSki cells were significantly increased by coculturing with the specific CTL (p <0.05). These results illustrated that a DC vaccine modified by HPV-16 E6/E7 gene can induce apoptosis of CaSki cells by inducing CTL, which may be used as a new strategy for biological treatment of cervical cancer.
