        Travessia e Resiliência na Poética de Raduan Nassar : Travessia and Resilience in Raduan Nassar Poetics

        The author from São Paulo, Raduan Nassar, was an excellent opportunity for us to reflect on a poetics of reading, mainly targeting his book Menina a caminho. This article intends to sketch, within this poetics, an analysis of the psychosocial and political tensions in which the young heroine was directly and indirectly involved. From the assumption that every literary text involves a provocation to discover the ethical-political dynamics that move it, we think that the notion of resilience could support our response to such provocation. The protagonist is in the pre-adolescent phase, the text, a narrative of less than 100 pages, in which the girl undertakes an apprenticeship. Therefore, we cast the hypothesis of a novel of formation or education. However, since the originality of any character or literary fact is inferred from comparison with other characters and similar facts, they will come to witness works, authors and foreign critics.         Conventional and digital impressions for complete-arch implant-supported fixed prostheses: time, implant quantity effect and patient satisfaction

        PURPOSE. To evaluate and compare the effect of impression type (conventional vs digital) and the number of implants on the time from the impressions to the generation of working casts of mandibular implant-supported fixed completearch frameworks, as well as on patient satisfaction. MATERIALS AND METHODS. 17 participants, 3 or 4 implants, received 2 types of digital impression methods (DI) and conventional (CI). In DI, two techniques were performed: scanning with the scan bodies (SC) and scanning with a device attached to the scan bodies (SD) (BR 10 2019 026265 6). In CI, the making of a solid index (SI) and open-tray impression (OT) were used. The outcomes were used to evaluate the time and the participant satisfaction with conventional and digital impressions. The time was evaluated through the timing of the time obtained in the workflow in the conventional and digital impression. The effect of the number of implants on time was also assessed. Satisfaction was assessed through a questionnaire based on seven. The Wilcoxon test used to identify the statistical difference between the groups in terms of time. The Mann-Whitney test was used to analyze the relationship between the time and the number of implants. Fisher's test was used to assess the patient satisfaction (P <.05). RESULTS. The time with DI was shorter than with CI (DI, x~=02:58; CI, x~=31:48) (P <.0001). The arches rehabilitated with 3 implants required shorter digital impression time (3: x~=05:36; 4: x~=09:16) (P <.0001). Regarding satisfaction, the DI was more comfortable and pain-free than the CI (P <.005). CONCLUSION. Digital impressions required shorter chair time and had higher patient acceptance than conventional impressions.

        Depression and Mania Induce Pro-inflammatory Activation of Macrophages Following Application of Serum from Individuals with Bipolar Disorder

        Objective: Evidence has suggested that immune imbalance is involved with bipolar disorder (BD); however, its precise mechanism is poorly understood. This study investigated whether biochemical changes in the serum from BD patients could modulate the phenotype of cultured macrophages. Methods: Eighteen subjects with BD and five healthy individuals were included in this study. The human monocyte cell line U-937 was activated with phorbol 12-myristate 13-acetate (PMA) and polarization was induced with RPMI-1640 media supplemented with 10% serum from each patient for 24 hours. Gene expression of selected M1 and M2 markers was assessed by quantitative PCR. Results: Macrophages exposed to serum of manic and depressive BD patients displayed an increase of interleukin-1 (6.40±3.47 and 9.04±5.84 vs. 0.23±0.11; p<0.05) and tumor necrosis factor- (2.23±0.91 and 2.03±0.45 vs. 0.62±0.24; p=0.002 and p=0.004, respectively) compared to euthymic group (there was no difference between euthymic and controls). In parallel, U-937 macrophages treated with serum of patients in acute episode displayed a down-regulation of CXCL9 (0.29±0.20 vs. 1.86±1.61; p=0.006) and CXCL10 expression (0.36±0.15 and 0.86±0.24 vs. 1.83±0.88; p<0.000 and p=0.04) compared to the euthymia group. Conclusion: Our results are consistent with previous studies showing that changes in peripheral blood markers could modulate M1/M2 polarization in BD. The evidence of macrophages as source of inflammatory cytokines might be helpful to unravel how the mononuclear phagocyte system is involved in the etiology of BD.

        Optimization of steel-concrete composite beams considering cost and environmental impact

        In the optimized structure sizing, the optimization methods are inserted in this context in order to obtain satisfactory solutions, which can provide more economical structures, besides allowing the consideration of the factors related to the environmental impacts in the structural design. This work proposes a mathematical model for the optimization of steel-concrete composite beams aiming to minimize the monetary cost and the environmental impact, using the Harmonic Search optimization method. Discrete variables were the dimensions of the steel profiles and the thickness of the collaborating slab of the composite steel-concrete beam. The proposed model was implemented in Fortran programming language and based on improvements in the structure of the optimization method proposed by Medeiros and Kripka (2017). To prove the effectiveness and applicability of the model, as well as the Harmonic Search method, analyzes were performed with different configurations of steel-concrete composite beams, in order to provide guidelines that make the use of these systems more streamlined. In general, the Harmonic Search optimization method has proved to be efficient in the search for the optimized solutions, as well as important considerations on the optimization of the monetary and environmental costs of steel-concrete composite beams were obtained from the developed examples.

        <P>Potyvirus HCPro is a multifunctional protein that, among other functions, interferes with antiviral defenses in plants and mediates viral transmission by aphid vectors. We have visualized in vivo the subcellular distribution and dynamics of HCPro from Potato virus Y and its homodimers, using green, yellow, and red fluorescent protein tags or their split parts, while assessing their biological activities. Confocal microscopy revealed a pattern of even distribution of fluorescence throughout the cytoplasm, common to all these modified HCPros, when transiently expressed in Nicotiana benthamiana epidermal cells in virus-free systems. However, in some cells, distinct additional patterns, specific to some constructs and influenced by environmental conditions, were observed: i) a small number of large, amorphous cytoplasm inclusions that contained α-tubulin; ii) a pattern of numerous small, similarly sized, dot-like inclusions distributing regularly throughout the cytoplasm and associated or anchored to the cortical endoplasmic reticulum and the microtubule (MT) cytoskeleton; and iii) a pattern that smoothly coated the MT. Furthermore, mixed and intermediate forms from the last two patterns were observed, suggesting dynamic transports between them. HCPro did not colocalize with actin filaments or the Golgi apparatus. Despite its association with MT, this network integrity was required neither for HCPro suppression of silencing in agropatch assays nor for its mediation of virus transmission by aphids.</P>

        Stroke Severity Score based on Six Signs and Symptoms The 6S Score: A Simple Tool for Assessing Stroke Severity and In-hospital Mortality

        Background and Purpose Ascertaining stroke severity and predicting risk of in-hospital mortality is crucial to advise patients and families about medical decisions. We developed and tested the validity of a new stroke score, the 6S Score (Stroke Severity Score based on Six Signs and Symptoms), for quantifying ischemic stroke severity and predicting in-hospital mortality. Methods We prospectively assessed 210 consecutive acute ischemic stroke patients. The cohort was further divided into a derivation (n=120) and a validation (n=90) sample. From a total of 10 stroke signs and symptoms, we selected those with likelihood ratio’s P<0.005. We tested the validity of the score for predicting in-hospital mortality by using receiver operating characteristic curves. We used a scatterplot and the Spearman’s test to evaluate the correlation between the 6S Score and the National Institutes of Health Stroke Scale as a marker of stroke severity. We used principal component and exploratory factor analyses for assessing qualitative aspects of the 6S Score. Results The C statistic for in-hospital mortality was 0.82 for the 6S Score and 0.86 for the National Institutes of health Stroke Scale, respectively, with no significant differences between each other (P=0.79). The correlation between both scores was strong (Spearman’s rho 0.68, P<0.001). The factor analyses showed a good balance between left/right hemispheres and anterior/posterior circulations. Conclusions The 6S Score may constitute a tool for easily assessing stroke severity and predicting stroke mortality. Further research is needed for further assessing its external validity.

        PUBLIC INVESTMENT, ECONOMIC PERFORMANCE AND BUDGETARY CONSOLIDATION: VAR EVIDENCE FOR THE FIRST 12 EURO COUNTRIES

        In a period of heightened concern about fiscal consolidation in the euro area, a politically expedient way of controlling the public budget is to cut public investment. A critical question, however, is whether or not political expediency comes at a cost, in terms of both long-term economic performance and future budgetary contention efforts. First, common wisdom suggests that public investments have positive effects on economic performance although the empirical evidence is less clear. Second, it is conceivable that public investment has such strong effects on output that over time it generates enough additional tax revenues to pay for itself. Obviously, it is equally plausible that the effects on output although positive are not strong enough for the public investment to pay for itself. In this paper, we investigate these issues empirically for the first twelve countries in the euro area using a vector auto-regressive approach. We conclude that the euro countries can be gathered in four groups according to the nature of the economic and budgetary impact of public investment. The first group includes Austria, Belgium, Luxembourg, and Netherlands, where the economic effects are either negative or positive but very small and, therefore, cuts will be harmless for the economy and effective from a budgetary perspective. The second group includes Finland, Portugal, and Spain, where public investment does not pay for itself and, therefore, cuts are an effective tool of budgetary consolidation although they are harmful for the economy. The third group includes France, Greece, and Ireland where public investment just pays for itself and therefore cuts are not an effective way of achieving long-term budgetary consolidation and are harmful for the economy. Finally, the fourth group includes Germany and Italy, where public investment more than pays for itself and, therefore, cuts are not only harmful for the economy but also counterproductive from a budgetary perspective.

      • Expression of Root-Related Transcription Factors Associated with Flooding Tolerance of Soybean ( <i>Glycine max</i> )

        <P>Much research has been conducted on the changes in gene expression of the model plant <I>Arabidopsis</I> to low-oxygen stress. Flooding results in a low oxygen environment in the root zone. However, there is ample evidence that tolerance to soil flooding is more than tolerance to low oxygen alone. In this study, we investigated the physiological response and differential expression of root-related transcription factors (TFs) associated with the tolerance of soybean plants to soil flooding. Differential responses of PI408105A and S99-2281 plants to ten days of soil flooding were evaluated at physiological, morphological and anatomical levels. Gene expression underlying the tolerance response was investigated using qRT-PCR of root-related TFs, known anaerobic genes, and housekeeping genes. Biomass of flood-sensitive S99-2281 roots remained unchanged during the entire 10 days of flooding. Flood-tolerant PI408105A plants exhibited recovery of root growth after 3 days of flooding. Flooding induced the development of aerenchyma and adventitious roots more rapidly in the flood-tolerant than the flood-sensitive genotype. Roots of tolerant plants also contained more ATP than roots of sensitive plants at the 7th and 10th days of flooding. Quantitative transcript analysis identified 132 genes differentially expressed between the two genotypes at one or more time points of flooding. Expression of genes related to the ethylene biosynthesis pathway and formation of adventitious roots was induced earlier and to higher levels in roots of the flood-tolerant genotype. Three potential flood-tolerance TFs which were differentially expressed between the two genotypes during the entire 10-day flooding duration were identified. This study confirmed the expression of anaerobic genes in response to soil flooding. Additionally, the differential expression of TFs associated with soil flooding tolerance was not qualitative but quantitative and temporal. Functional analyses of these genes will be necessary to reveal their potential to enhance flooding tolerance of soybean cultivars.</P>

        Nanoemulsions with Chloroaluminium Phthalocyanine and Paromomycin for Combined Photodynamic and Antibiotic Therapy for Cutaneous Leishmaniasis

        Background: Photodynamic therapy (PDT) using chloroaluminium phthalocyanine (ClAlPc) and paromomycin sulfate (PM) can be effective against New World Leishmania species involved in cutaneous leishmaniasis (CL). The aim of this study is to assay the skin permeation and the antileishmanial effects of a nanoemulsion (NE) containing both ClAlPc and PM in experimental CL by Leishmania (Viannia) braziliensis. Material and Methods: Cremophor ELP/castor oil-based NEs were prepared by a low-energy method and characterized for their physicochemical parameters. The NEs were used to deliver both ClAlPc and PM to leishmania cells. The in vitro toxicity of NEs were tested in vitro against L. (V.) braziliensis and THP-1 cells. The in vivo toxicity was assessed in non-infected BALB/c mice. Ex-vivo permeation and retention studies using healthy mice skin were also conducted. Finally, the in vivo activity of NE-PM+ClAlPc after PDT was tested in BALB/c mice infected with parasites. Results: NEs are colloidally stable with average droplet diameter of 30 nm, polydispersity index (PDI) below 0.2, and zeta potential near zero. Both promastigotes and intracellular amastigotes treated with NE-PM, NE-ClAlPc and NE-PM+ClAlPc were inhibited at >50%, >95%, >88%, respectively, after PDT with a phototoxic index (PI) >1.2. No skin ClAlPc permeation was observed. In contrast, PM skin permeation was 80-fold higher using PMloaded NE formulation in comparison to aqueous PM solution. Topical treatment with NE formulations showed no signs of local toxicity or genotoxicity. In addition, concentrations of PM between 27.3 - 292.5 μM/25 mg of tissue were detected in different organs. In vivo, the NE-PM+ClAlPc treatment did not reduce skin lesions. Conclusion: The Cremophor ELP/castor oil NE formulation increases the permeation of PM through the skin and can be used to co-deliver PM plus ClAlPc for combined PDT protocols. However, the lack of efficacy in the in vivo model evidences that the therapeutical scheme has to be improved.
