전자빔을 이용한 폴록사머 하이드로젤의 제조 및 구강 점막부착성 약물전달을 위한 특성 분석

본 연구에서는 전자빔을 이용하여 폴록사머 하이드로젤을 제조하여 구강 점막부착성 약물전달체로서의 특성을 평가하고자 하였다. 온도감응성을 나타내는 폴록사머 고분자의 말단에 가교반응을 위한 비닐기를 도입 한 후, 방사선 가교반응을 이용하여 하이드로젤을 제조하였다. 점착성 향상을 위한 첨가제로 점착성 고분자인 카 보폴을 첨가하여 하이드로젤을 제조하였고, 카보폴이 점막부착성과 서방성 방출에 미치는 영향을 알아보았다. 결과적으로, 폴록사머 고분자의 말단 개질과 카보폴의 첨가에 의해 하이드로젤의 기계적 물성과 점막부착성이 향상되었고, 약물방출실험 결과 약물이 방출되는 속도가 지연되는 결과를 확인하였다. In this study, poloxamer hydrogels were prepared by electron beam irradiation and evaluated for potential application as a buccal mucoadhesive drug delivery system. Poloxamer, one of typical thermoresponsive polymers, was modified to have vinyl end groups for crosslinking reaction, and its hydrogels were fabricated by irradiation crosslinking reaction. Carbopol as a mucoadhesive polymeric additive was introduced to improve the mucoadhesive property of the hydrogels and its effect on the mucoadhesion and drug release properties was investigated. The results showed that the end group modification of poloxamer and the addition of carbopol improved mucoadhesive force and mechanical properties and led to a sustained drug release behavior.
"촛불집회"에 대한 보도경향과 이데올로기 -백선기의 기호네트워크구조 분석(SNA)과 담론구조 분석(DSA) 적용을 중심으로

This study was conducted on the assumption that news coverage of media affects construction of reality of audiences, and examines the coverage patterns of news about ``2008 Candlelight Rally`` and its ideological meanings. The objects of research are news report articles from April 1st to August 31st of 2008 in Chosun ilbo and Hankyoreh Shinmun. Focusing on Baek`s Semiotic Network Analysis(SNA) and discourse analysis, the research was conducted with the news coverage patterns and SNA of news texts, and discourse structure. The results are as follows. Difference of phase 1 through SNA is the cause of movement which ``Chosun ilbo`` indicates ``MBC PD Diary`` unlike ``Hankyoreh Shinmun``. In phase 2, Chosun ilbo shows some elements of ``Changes of features of Candlelight Rally``, ``suspicion of secret syndicates of Rally``, and ``Seriousness on diffusion of false information in Internet media``. However, unlike Chosun ilbo, they criticized ``conservative press`` and ``government`` insisting about ``usefulness of internet media``, ``voluntary participation of attendees`` and ``suspicion of syndicates``. In phase 3, Chosun ilbo and Hankyoreh Shinmun criticized each other`s distorted news coverage in relation with controversial issue about intentional distortion of ``MBC PD Diary`` and ``oppressing movement of conservative newspaper advertiser``. In phase 4, there appeared some elements meaning ``termination of Candlelight Rally`` in Chosun ilbo. Meanwhile, elements related to ``continuation of Candlelight Movement`` appeared in Hankyoreh Shinmun, so both two papers showed semiotic signs about the inclination of their ideologies. For the result of discourse analysis, there was a big difference between two papers. From first phase to fourth, Chosun ilbo dealt about ``negative discourse of Candlelight Rally`` and ``pro-government discourse``, and ``pro-pro US policies discourse``, while Hankyoreh Shinmun showed opposite discourses such as ``positive discourse of Candlelight Rally``, ``anti-government discourse``, and ``Anti-pro US policies discourse``. Eventually, two newspapers showed that whenever newspaper companies produce news articles, they made those coincide with their ideological features, or build new meanings through the process connecting with other events. Finally, these two newspapers mentioned that when public opinions made news about the question pending, they proceed their news to unite their Ideological Meanings or they made new ideological meaning.
Eudragit으로 코팅된 초다공성 하이드로젤의 제조 및 pH 의존형 팽윤거동

초다공성 하이드로젤은 다공성 공극구조를 이용하여 기존 수화젤의 팽윤성을 획기적으로 향상시킨 것으로, 빠른 팽윤거동과 높은 흡수율로 다양한 의약용 응용분야에 유용한 재료이다. 본 연구에서는 장용 코팅제인 Eudragit 계열 고분자들을 사용하여 poly(acrylic acid-co-acrylamide)계 초다공성 하이드로젤을 코팅함으로써 pH 의존성 팽윤거동을 보이는 초다공성 하이드로젤을 제조하고자 하였다. 서로 다른 pH 영역에서 작용하는 Eudragit L100과 S100을 이용하여 딥 코팅에 의해 표면을 코팅한 후 SEM을 이용해 공극구조를 관찰하고 pH에 따른 초다공성 하이 드로젤의 팽윤거동을 관찰하였다. Eudragit 계열 고분자들로 코팅된 하이드로젤은 낮은 pH 환경하에서는 팽윤이 억 제되다가, 특정 pH 이상에서 팽윤성이 향상되는 pH 의존성 팽윤거동을 보였고, 이러한 pH 의존성은 사용한 장용 코팅제용 고분자들의 pH 특성에 의존하였다. Superporous hydrogels (SPHs) with fast swelling and superabsorbent properties are useful materials in various biomedical fields, by improving the swelling properties of conventional hydrogels based on their unique porous structure. In this study, Eudragit polymers were used as coating materials to control the swelling properties of poly(acrylic acid-co-acrylamide) based SPHs by environmental pH. The SPHs were coated with Eudragit L100 and S100 that have different pH characteristics as enteric coating materials by a dip coating method, and their pH dependent swelling behaviors were observed in various pH environments. The swelling of SPHs was inhibited at a low pH range, but significantly enhanced above a characteristic pH of Eudragit polymers. This pH dependent swelling behavior of hydrogels could be modulated by the characteristics of the enteric coating polymers.
