      • 난관배양액이 처녀발생유기된 돼지난포란의 체외발달에 미치는 영향

        본 연구는 난관배양액이 돼지수정란의 체외발달에 미치는 효과를 규명키 위하여 수행하였다. 돼지 미성숙 난포란은 TCM-199, Ham's F-10 그리고 Whitten's 배양액에 10% 난포액과 호르몬(PMSG : 10IU/㎖, HCG : 10IU/㎖)을 첨가 20시간 배양하고 호르몬을 첨가하지 않는 배양액에서 20시간 추가 배양하여 총 40시간동안 배양하여 체외성숙을 유도하였다. 체외성숙후 0.1% hyaluronidase로 난구세포를 제거하고 15% FCS가 함유된 TCM-199으로 3회 세척하고 TCM-199에 15% FCS와 10% ethanol 혼합액에 세척한 난자를 옮겨 10분간 배양 처녀발생을 유기하였다. 처녀발생 6시간후 전핵형성율은 체외성숙배양액으로 TCM-199을 사용했을 때 56.4%, Ham's F-10의 경우 58.3%, Whitten's 배양액의 경우 74.0%를 보였다. 처녀발생 유기 48시간째 난할율은 TCM-199을 사용했을 때 45.7%, Ham's F-10에서 45.4%, Whitten's배양액에서 39.2%를 보였으며 세종류의 배양액에 POCM을 첨가 배양했을 때 TCM-199에 44.8%, Ham's F-10에서 45.4%, Whitten's배양액에서 43.7%로 나타났다. 처녀발생육 난자를 96시간 체외배양시킨 결과 상실배 발달율이 POCM을 첨가 했을 때 첨가하지 않은 시험구에 비하여 유의적으로 높았다(P<0.05) The effect of porcine oviductal conditioned medium(POCM) on in vitro development of chemically activated porcine oocytes was studied. Porcine oocytes were cultured in TCM-199, Ham's F-10 and Whitten's medium with hormonal supplements for 20h and 40h additional culture without hormonal supplements. After in vitro maturation, the denuded oocytes were washed 3 times with TCM-199 contaning 15%(v/v) ethanol to induce pathenogenetic activation. At 6h after activation, pronuclea formation rates were 56.4% in TCM-199, 59.3% in Ham's F-10 and 74.0% in Whitten's maturation medium. At 48h after activation, 45.7%, 45.4% and 39.2% of oocytes claved in TCM-199, Ham's F-10 and Whitten's culture medium, respectively. And 44.8%, 45.5% and 43.7% of oocytes were claved in TCM-199, Ham;s F-10 and Whitten's culture medium supplemented with POCM, respectively. The rates of moular were higher in culture medium with POCM than without POCM at 96h after activation.(P<0.05)

      • 난관액, 난관배양액 및 난포액이 돼지정자의 운동성과 수정능 획득에 미치는 영향

        본 연구는 돼지 정자의 운동성과 첨체반응에 대한 난관액, 난관배양액 및 난포액의 효과를 조사하였다. 수정능획득배양액에 난관액(10%)을 첨가 공배양했을 때 정자의 운동성은 처리구에서 유의적으로 높았다(P<0.05). CTC 염색방법으로 정자의 첨체반응을 조사한 결과 난관액과 난포액이 첨가된 수정능획득 배양액에서 1.5, 3, 4.5시간동안 전배양을 실시하였을 때 정자의 첨체반응이 대조구에 비하여 유의적으로 높았으며(P<0.05), 난관배양액에서 1.5, 3, 4.5시간동안 전배양을 실시하였을때도 대조구에 비하여 정자첨체반응이 유의적으로 높았다(P<0.05). 이러한 연구결과는 난관과 난포액등에 함유되어있는 어떤인자가 정자의 운동성과 첨체반응율을 높힌다고 사료된다. The effects of oviductal fluid, oviductal conditioned medium and follicular fluid on porcine sperm mortility and capacitation were studied. The addition of oviductal fluid, oviductal conditioned medium and follicular fluid to the prefertilization medium increased sperm mortility(P<0.05). Chloroterracycline(CTC) flourescence patterns were used to determine incidence of acrosome reaction. The proportion of acrosome reaction increased when spermatozoa were exposed for 1.5, 3 and 4.5h to oviductal fluid and follicular fluid(P<0.05). In sperm-POCM(porcine oviductal conditioned medium) co-culture period of 4.5 h, acrosome reaction rates was higher than other groups(P<0.05). These results suggest that the factor(s) in secreation from oviduct and follicular fluid increased sperm mortility and acrosome reaction.

      • Induction of nucleolin translocation by acharan sulfate in A549 human lung adenocarcinoma

        <P>Acharan sulfate (AS), isolated from the giant African snail Achatina fulica, is a novel glycosaminoglycan, consisting primarily of the repeating disaccharide structure α-D-N-acetylglucosaminyl (1 → 4) 2-sulfoiduronic acid. AS shows anti-tumor activity in vitro and in vivo. Despite this activity, AS is only weakly cytotoxic towards cancer cells. We examine the interactions between AS and cell-surface proteins in an effort to explain this anti-tumor activity. Using flow cytometry and affinity column chromatography, we confirm that AS has strong affinity to specific cell-surface proteins including nucleolin (NL) in A549 human lung adenocarcinomas. Surprisingly, we found the translocation of NL from nucleus to cytoplasm under the stimulation of AS (100 µg/ml) in vitro. Also, as NL exits the nucleus, the levels of growth factors such as bFGF and signaling cascade proteins, such as p38, p53, and pERK, are altered.       • Endocervical-like (Müllerian) mucinous borderline tumours of the ovary are frequently associated with the <i>KRAS</i> mutation

        <P>Kim K-R, Choi J, Hwang J-E, Baik Y-A, Shim J Y, Kim Y M & Robboy S J(2010) <I>Histopathology</I><B>57</B>, 587–596<B>Endocervical-like (Müllerian) mucinous borderline tumours of the ovary are frequently associated with the <I>KRAS</I> mutation</B></P><P>Aims: </P><P>Clinicopathological aspects of the endocervical-like mucinous borderline tumour of the ovary (EMBT), including higher frequencies of bilaterality, endometriosis and hormone receptor reactivity, and often admixtures of various Müllerian-type epithelia, closely resembles endometrioid tumour more than mucinous borderline tumour of the intestinal type (IMBT). Thus, the aims of this study were to determine whether EMBT is really a subtype of mucinous borderline tumours, as shown in the current classification system, and to determine the best classification for EMBT.</P><P>Methods and results: </P><P>The clinicopathological and immunohistochemical features of 17 EMBTs were analysed, including oestrogen receptor (ER), progesterone receptor (PR), <I>PTEN</I>, cytokeratins (CK) 7 and 20, and <I>&bgr;-catenin</I>. Additionally, mutational analyses of the <I>KRAS</I> (exon 1) and <I>PTEN</I> genes (all nine exons) were performed in all cases, and the results were compared with literature findings for IMBT and endometrioid tumours. Twelve patients (71%) were confirmed histologically to have endometriosis in one or both ovaries. In seven cases, gradual transitions from endometriotic foci to the EMBT were identified. Immunohistochemically, all cases were reactive for ER and PR, with no nuclear expression of &bgr;-catenin. CK7 positivity was strong in all patients, whereas there was no reactivity for CK20. <I>PTEN</I> reactivity was diffuse in the nuclei of epithelial and underlying stromal cells. Sixty-nine per cent showed <I>KRAS</I> mutations in exon 1 and codon 12, but no <I>PTEN</I> mutation was identified in any of the nine exons.</P><P>Conclusion: </P><P>Our study suggests that EMBT has features of both mucinous and endometrioid tumours and is an additional tumour type arising in endometriosis. While clinicopathological features of EMBTs are closer to endometrioid tumours, they still have molecular characteristics closer to IMBTs.</P>

      • LEF1-mediated MMP13 gene expression is repressed by SIRT1 in human chondrocytes

        <P>Reduced SIRT1 activity and levels during osteoarthritis (OA) promote gradual loss of cartilage. Loss of cartilage matrix is accompanied by an increase in matrix metalloproteinase (MMP) 13, partially because of enhanced LEF1 transcriptional activity. In this study, we assessed the role of SIRT1 in LEF1-mediated MMP13 gene expression in human OA chondrocytes. Results showed that MMP13 protein levels and enzymatic activity decreased significantly during SIRT1 overexpression or activation by resveratrol. Conversely, MMP13 gene expression was reduced in chondrocytes transfected with SIRT1 siRNA or treated with nicotinamide (NAM), a sirtuin inhibitor. Chondrocytes challenged with IL-113, a cytokine involved in OA pathogenesis, enhanced LEF1 protein levels and gene expression, resulting in increased MMP13 gene expression; however, overexpression of SIRT1 during IL-113 challenge impeded LEF1 levels and MMP13 gene expression. Previous reports showed that LEF1 binds to the MMP13 promoter and transactivates its expression, but we observed that SIRT1 repressed LEF1 protein and mRNA expression, ultimately reducing LEF1 transcriptional activity, as judged by luciferase assay. Finally, mouse articular cartilage from Sirt1-/presented increased LEF1 and MMP13 protein levels, similar to human OA cartilage. Thus, demonstrating for the first time that SIRT1 represses MMP13 in human OA chondrocytes, which appears to be mediated, at least in part, through repression of the transcription factor LEF1, a known modulator of MMP13 gene expression.       • THE CDF ARCHIVE:<i>HERSCHEL</i>PACS AND SPIRE SPECTROSCOPIC DATA PIPELINE AND PRODUCTS FOR PROTOSTARS AND YOUNG STELLAR OBJECTS

        <P>We present the COPS-DIGIT-FOOSH (CDF) Herschel spectroscopy data product archive, and related ancillary data products, along with data fidelity assessments, and a user-created archive in collaboration with the Herschel-PACS and SPIRE ICC groups. Our products include datacubes, contour maps, automated line fitting results, and best 1D spectra products for all protostellar and disk sources observed with PACS in RangeScan mode for two observing programs: the DIGIT Open Time Key Program (KPOT_nevans_1 and SDP_nevans_1; PI: N.. Evans), and the FOOSH Open Time Program (OT1_jgreen02_2; PI: J. Green). In addition, we provide our best SPIRE-FTS spectroscopic products for the COPS Open Time Program (OT2_jgreen02_6; PI: J. Green) and FOOSH sources. We include details of data processing, descriptions of output products, and tests of their reliability for user applications. We identify the parts of the data set to be used with caution. The resulting absolute flux calibration has improved in almost all cases. Compared to previous reductions, the resulting rotational temperatures and numbers of CO molecules have changed substantially in some sources. On average, however, the rotational temperatures have not changed substantially (<2%), but the number of warm (T-rot similar to 300 K) CO molecules has increased by about 18%.</P>
