A Simultaneous Analytical Method for Duplex Identification of Porcine and Horse in the Meat Products by EvaGreen based Real-time PCR

Sakalar, Ergun,Ergun, Seyma Ozcirak,Akar, Emine Korean Society for Food Science of Animal Resource 2015 한국축산식품학회지 Vol.35 No.3

A duplex real-time polymerase chain reaction (PCR) based assay for the detection of porcine and horse meat in sausages was designed by using EvaGreen fluorescent dye. Primers were selected from mitochondrial 12S rRNA and 16S rRNA genes which are powerful regions for identification of horse and porcine meat. DNA from reference samples and industrial products was successfully extracted using the GIDAGEN® Multi-Fast DNA Isolation Kit. Genomes were identified based on their specific melting peaks (Mp) which are 82.5℃ and 78℃ for horse and porcine, respectively. The assay used in this study allowed the detection of as little as 0.0001% level of horse meat and 0.001% level of porcine meat in the experimental admixtures. These findings indicate that EvaGreen based duplex realtime PCR is a potentially sensitive, reliable, rapid and accurate assay for the detection of meat species adulterated with porcine and horse meats.

Comparison of retention forces with various fabrication methods and materials in double crowns

        Melahat Çelik Güven,Meral Tuna,Ergun Bozdağ,Gizem Nur Öztürk,Gulsen Bayraktar 대한치과보철학회 2017 The Journal of Advanced Prosthodontics Vol.9 No.4

PURPOSE. The purpose of this study was to analyze the retention force changes and wear behaviours of doublecrown systems over long-term use. MATERIALS AND METHODS. Ten groups, each consisting of six samples, were evaluated. Specifically, casting gold alloy primary crown - casting gold alloy secondary crown (AA), laser sintering primary crown - laser sintering secondary crown (LL), casting Cr alloy primary crown - casting Cr alloy secondary crown, (CC) zirconia primary crown - electroformed secondary crown (ZA), and CAD/CAM titanium alloy primary crown - CAD/CAM titanium alloy secondary crown (TT) groups were evaluated at cone angles of 4° and 6°. The samples were subjected to 5,000 insertion-separation cycles in artificial saliva, and the retention forces were measured every 500 cycles. The wear levels were analyzed via SEM at the beginning and end of the 5,000 cycles. RESULTS. In all samples, the retention forces increased when the conus angle decreased. The highest initial and final retention force values were found in the LL-4° group (32.89 N-32.65 N), and the lowest retention force values were found in the ZA6° group (5.41 N-6.27 N). The ZA groups’ samples showed the least change in the retention force, and no wear was observed. In the other groups, wear was observed mostly in the primary crowns. CONCLUSION. More predictable, clinically relevant, and less excursive retention forces can be observed in the ZA groups. The retention force values of the LL groups were statically similar to those of the other groups, except the ZA groups.

The stress analysis of a shear wall with matrix displacement method

        Ergun, Mustafa,Ates, Sevket Techno-Press 2015 Structural Engineering and Mechanics, An Int'l Jou Vol.53 No.2

Finite element method (FEM) is an effective quantitative method to solve complex engineering problems. The basic idea of FEM for a complex problem is to be able to find a solution by reducing the problem made simple. If mathematical tools are inadequate to obtain precise result, even approximate result, FEM is the only method that can be used for structural analyses. In FEM, the domain is divided into a large number of simple, small and interconnected sub-regions called finite elements. FEM has been used commonly for linear and nonlinear analyses of different types of structures to give us accurate results of plane stress and plane strain problems in civil engineering area. In this paper, FEM is used to investigate stress analysis of a shear wall which is subjected to concentrated loads and fundamental principles of stress analysis of the shear wall are presented by using matrix displacement method in this paper. This study is consisting of two parts. In the first part, the shear wall is discretized with constant strain triangular finite elements and stiffness matrix and load vector which is attained from external effects are calculated for each of finite elements using matrix displacement method. As to second part of the study, finite element analysis of the shear wall is made by ANSYS software program. Results obtained in the second part are presented with tables and graphics, also results of each part is compared with each other, so the performance of the matrix displacement method is demonstrated. The solutions obtained by using the proposed method show excellent agreements with the results of ANSYS. The results show that this method is effective and preferable for the stress analysis of shell structures. Further studies should be carried out to be able to prove the efficiency of the matrix displacement method on the solution of plane stress problems using different types of structures.

Free vibration and buckling analysis of the impacted hybrid composite beams

Emin Ergun,Yasin Yilmaz,Hasan Çallioğlu 국제구조공학회 2016 Structural Engineering and Mechanics, An Int'l Jou Vol.59 No.6

The aim of this experimental study is to investigate the free vibration and buckling behaviors of hybrid composite beams having different span lengths and orientation angles subjected to different impact energy levels. The impact energies are applied in range from 10 J to 30 J. Free vibration and buckling behaviors of intact and impacted hybrid composite beams are compared with each other for different span lengths, orientation angles and impact levels. In free vibration analysis, the first three modes of hybrid beams are considered and natural frequencies are normalized. It is seen that first and second modes are mostly affected with increasing impact energy level. Also, the fundamental natural frequency is mostly affected with the usage of mold that have 40 mm span length (SP40). Moreover, as the impact energy increases, the normalized critical buckling loads decrease gradually for 0

Serum Chitotriosidase Activity in Pulmonary Tuberculosis: Response to Treatment and Correlations with Clinical Parameters

        Gulhan Cakır,Seyfettin Gumus,Ergun Ucar,Hatice Kaya,Ergun Tozkoparan,Emin Ozgur Akgul,Bulent Karaman,Omer Deniz,Ismail Kurt,Metin Ozkan,Hayati Bilgic 대한진단검사의학회 2012 Annals of Laboratory Medicine Vol.32 No.3

Background: Chitotriosidase is an accepted marker of macrophage activation. In this study, we investigated serum chitotriosidase levels in pulmonary tuberculosis (PTB). Methods: Forth-two patients with PTB and 30 healthy subjects were enrolled in the study. The radiological extent of PTB, radiological sequela after treatment, and the degree of smear positivity were assessed. Chitotriosidase levels were measured by a fluorometric method. Results: The serum chitotriosidase levels of the PTB patients were significantly higher than those of the control subjects (39.73±24.97 vs. 9.63±4.55 nmol/mL/h, P<0.001). After completion of the standard 6-month antituberculous treatment, chitotriosidase levels in PTB patients significantly decreased (10.47±4.54 nmol/mL/h, P<0.001). Chitotriosidase levels correlated significantly with the radiological extent of PTB, degree of smear positivity, and post-treatment radiological sequela score (r=0.439, r=0.449, and r=0.337, respectively). Conclusions: This study demonstrated that serum chitotriosidase levels increase in PTB; therefore, chitotriosidase can be used as a marker of disease activity, severity, and response to treatment. Image Restoration by Lifting-Based Wavelet Domain E-Median Filter

        Sema Koç,Ergun Er?lebi 한국전자통신연구원 2006 ETRI Journal Vol.28 No.1

In this paper, we propose a method of applying a liftingbased wavelet domain e-median filter (LBWDEMF) for image restoration. LBWDEMF helps in reducing the number of computations. An e-median filter is a type of modified median filter that processes each pixel of the output of a standard median filter in a binary manner, keeping the output of the median filter unchanged or replacing it with the original pixel value. Binary decisionmaking is controlled by comparing the absolute difference of the median filter output and the original image to a preset threshold. In addition, the advantage of LBWDEMF is that probabilities of encountering root images are spread over sub-band images, and therefore the e-median filter is unlikely to encounter root images at an early stage of iterations and generates a better result as iteration increases. The proposed method transforms an image into the wavelet domain using lifting-based wavelet filters, then applies an e-median filter in the wavelet domain, transforms the result into the spatial domain, and finally goes through one spatial domain e-median filter to produce the final restored image. Moreover, in order to validate the effectiveness of the proposed method we compare the result obtained using the proposed method to those using a spatial domain median filter (SDMF), spatial domain e-median filter (SDEMF), and wavelet thresholding method. Experimental results show that the proposed method is superior to SDMF, SDEMF, and wavelet thresholding in terms of image restoration.
