Although accumulating evidence points to theessential roles of nucleus-encoded chloroplast S1 domaincontainingproteins (SDPs) in chloroplast RNA metabolismand plant development, functions of chloroplast SDPs inabiotic stress responses are largely unknown. In this study,we investigated the role of a SDP (At1g12800) in Arabidopsisresponse to diverse abiotic stresses. Analysis of the sdpknockout mutant and complementation lines demonstratedthat loss of SDP function results in decreased survival rate ofArabidopsis under salt, heat, UV, or freezing stress, but notunder dehydration stress or ABA. In consistent with aprevious report demonstrating that SDP is involved inchloroplast rRNA processing, translation in chloroplasts wasimpaired in the sdp mutant. Expression of several nucleargenes involved in stress response and adaptation was alteredin the sdp mutant subjected to different abiotic stresses,suggesting that modulation of chloroplast translation affectsthe expression of nuclear genes under abiotic stresses. Thesedata reveal that chloroplast-localized SDP plays an importantrole in abiotic stress response by modulating chloroplasttranslation and the expression of nuclear genes possibly viaunidentified plastid-to-nucleus signaling.

Study on tissue culture of sweet potato (Ipomea batatas L.) invitro and effects of chitooligosaccharide on growth of the plant were investigated. Effects of condition of sterilization and concentration of plant regulators such as: N(6) - benzyl adenine (BA), napthyl acetic acid (NAA) and kinetin (KN) on the rate of bud formation and the growth of root and shoot of the sweet potato were studied. The results showed that 20% sodium hypochlorite and 30 min for treatment was suitable for sterilization of the sweet potato tissue; the rate of contamination was only 3.3% and 76.7% bud formation; results also showed optimal concentration of BA and KN for the growth of the plantlets invitro was 2 mg/l and 1 mg/l. Effect of chitooligosaccharide supplement on the growth of sweet potato was also studied. Mugashige and Skoog (MS) medium containing BA and KN supplemented 0, 10, 20, 30 and 50 ppm chitooligosaccharide. Results showed that 20~30 ppm chitooligosaccharide was the best for growth of the root and shoot of sweet potato, number of the plantlet roots increased by 30%, the length of root also increased 40% and fresh and dry biomass of roots increased 100% compared to the control (no supplement of chitooligosaccharide). Chitooligosaccharide also increased in the number of leaf, height and fresh and dry biomass of the plantlet shoots.
Modeling and Fault Tolerant Control of an Electro- Hydraulic Actuator

In the modern industry, electro-hydraulic actuators (EHAs) have been applied to various applications for precise position pressure/ force control tasks. However, operating EHAs under sensor faults is one of the critical challenges for the control engineers. For its enormous nonlinear characteristics, sensor fault could lead the catastrophic failure to the overall system or even put human life in danger. Thus in this paper, a study on mathematical modeling and fault tolerant control (FTC) of a typical EHA for tracking control under sensor-fault conditions has been carried out. In the proposed FTC system, the extended Kalman-Bucy unknown input observer (EKBUIO) -based robust sensor fault detection and identification (FDI) module estimates the system states and the time domain fault information. Once a fault is detected, the controller feedback is switched from the faulty sensor to the estimated output from the EKBUIO owing to mask the sensor fault swiftly and retains the system stability. Additionally, considering the tracking accuracy of the EHA system, an efficient brain emotional learning based intelligent controller (BELBIC) is suggested as the main control unit. Effectiveness of the proposed FTC architecture has been investigated by experimenting on a test bed using an EHA in sensor failure conditions.

This paper presents a simple and effective force reflecting joystick controller for applications to bilateral teleoperation in construction machinery. First, this controller is a combination of an advanced force reflecting gain tuner and two local adaptive controllers, master and slave. Second, the force reflecting gain tuner is effectively designed using recursive least square method and fuzzy logics to estimate directly and accurately the environmental characteristics and, consequently, to produce properly a force reflection. Third, the local adaptive controllers are simply designed using fuzzy technique and optimized using a smart leaning mechanism to ensure that the slave follows well any given trajectory while the operator is able to achieve truly physical perception of interactions at the remote site. An experimental master-slave manipulator is setup and real-time control tests are carried out under various environmental conditions to evaluate the effectiveness of the proposed controller.

A novel gene coding for an endo-β-1,4-mannanase (manA) from Bacillus subtilis strain G1 was cloned and overexpressed in P. pastoris GS115, and the enzyme was purified and characterized. The manA gene consisted of an open reading frame of 1,092 nucleotides, encoding a 364-aa protein, with a predicted molecular mass of 41 kDa. The β-mannanase showed an identity of 90.2-92.9% (≤95%) with the corresponding amino acid sequences from B. subtilis strains deposited in GenBank. The purified β- mannanase was a monomeric protein on SDS-PAGE with a specific activity of 2,718 U/mg and identified by MALDITOF mass spectrometry. The recombinant β-mannanase had an optimum temperature of 45oC and optimum pH of 6.5. The enzyme was stable at temperatures up to 50oC (for 8 h) and in the pH range of 5-9. EDTA and most tested metal ions showed a slightly to an obviously inhibitory effect on enzyme activity, whereas metal ions (Hg2+, Pb2+, and Co2+) substantially inhibited the recombinant β-mannanase. The chemical additives including detergents (Triton X- 100, Tween 20, and SDS) and organic solvents (methanol, ethanol, n-butanol, and acetone) decreased the enzyme activity, and especially no enzyme activity was observed by addition of SDS at the concentrations of 0.25-1.0% (w/v) or n-butanol at the concentrations of 20-30% (v/v). These results suggested that the β-mannanase expressed in P. pastoris could potentially be used as an additive in the feed for monogastric animals.
