      • Impact of Home Education on Levels of Perceived Social Support for Caregivers of Cancer Patients

        Background: The healthcare needs of cancer patients are complex and persons involved in their caregiving process are faced with many issues that need to be addressed. The entire family and particularly the person taking on responsibility for patient care develop expectations from healthcare professionals, especially nurses. Objective: The study was conducted to evaluate the impact of a home education program provided to caregivers of cancer patients on the level of their perceived social support and problems in caregiving. Interventions/Methods: The caregivers of thirty seven cancer patients of 2,400 registered people in a family center were given an educational program in this descriptive and cross-sectional study twice a week for a month during the period of March 2011 - April 2011. Results: Of all caregivers, 56.8% were between the ages 36-40, 94.5% were female, 91.9% had received no education on caregiving, 81.0% stated that they mostly felt physically and mentally inadequate in their caregiving. Perceived Social Support from the family indicated a significant difference at $8.05{\pm}4.38$ before and $11.7{\pm}4.97$ after the education. A comparison of the mean scores of caregivers on emotional issues before and after the education revealed the following: spiritual distress scores were $2.54{\pm}0.69$ before and $2.44{\pm}0.43$ after the education; hopelessness scores, $2.24{\pm}0.59$ before and $2.23{\pm}0.38$ after the education; ineffective individual coping was $3.89{\pm}1.42$ before and $2.45{\pm}0.59$ after the education; competing needs in decision-making were $3.54{\pm}0.69$ before and $2.10{\pm}1.24$ after the education; depressive feeling were $3.01{\pm}1.53$ before and $2.02{\pm}0.99$ after the education (p<0.05). Conclusions: Positive effects of home education on levels of perceived social support and caregiving problems of caregivers of cancer patients were observed. Home educational programs for caregivers of cancer patients are important for both better understanding of the requirements of their patients and themselves.

      • Knowledge of Turkish Mothers with Children in the 0-13 Age Group about Cancer Symptoms

        Background: There have been few studies in Turkey on the incidence of childhood cancers. A mother's knowledge about signs and symptoms of cancer is important for early diagnosis, effective treatment, and improvement of life expectancy. This study was conducted with a group of mothers of children, aged 0-13, at a Family Health Center (FHC) in Turkey's Eastern Black Sea Region, to analyze their knowledge about cancer symptoms in childhood. Materials and Methods: The study group of this descriptive/cross-sectional research comprised 2,061 mothers, ages 19-49, at an FHC in the Eastern Black Sea Region in February 1, 2011 - June 1, 2011. Before the study, permission was obtained from the local ethics board and the institutions concerned. A value of p<0.05 was accepted as statistically significant. Results: Of the mothers, 34.9% were between the ages of 40-47, 40.5% had three children, 73.8% had no experience with children with cancer, 45.9% said they learned about cancer on television, 39.7% stated that the primary reason for childhood cancer was the mother's smoking during pregnancy, 68.8% said that early diagnosis would save a child, and 98% wanted to learn about childhood cancer. Conclusion: It was determined that the mothers' knowledge of cancer was deficient.

        The First Study on Bacterial Flora and Biological Control Agent of Anoplus roboris (Sufr., Coleoptera)

        The hazelnut leaf holer (Anoplus roboris Sufr,, Coleoptera: Curculionidae) is a devastating pest of hazelnut and oak trees. It causes approximately 20-30% economic damage to hazelnut production per year in Turkey. In the present study, in order to fond a more effective and safe biological control agent against A. roboris, we investigated the bacterial flora of the hazelnut leave holer, and tested them for insecticidal effects on it. According to morphological, physiological and biochemical tests bacterial flora were identified as Bacillus circulans (Ar1), Bacillus polymyxa (Ar2), Enterobacter sp. (Ar3) and Bacillus sphaelicus (Ar4). Insecticidal effects of bacterial isolates were performed on adult A. roboris. The highest insecticidal effect determined was 67% by B. sphaericus within eight days. The insecticidal effects of the other isolates (Ar1, Ar2 and Ar3) were determined as 33%, 47% and 47% within the same period, respectively.

        Purification and Characterization of the Bacteriocin Thuricin Bn1 Produced by Bacillus thuringiensis subsp. kurstaki Bn1 Isolated from a Hazelnut Pest

        A novel bioactive molecule produced by Bacillus thuringiensis subsp. kurstaki Bn1 (Bt-Bn1), isolated from a common pest of hazelnut, Balaninus nucum L. (Coleoptera: Curculionidae), was determined, purified, and characterized in this study. The Bt-Bn1 strain was investigated for antibacterial activity with an agar spot assay and well diffusion assay against B. cereus, B. weinhenstephenensis, L. monocytogenes, P. savastanoi, P. syringae, P. lemoignei, and many other B. thuringiensis strains. The production of bioactive molecule was determined at the early logarithmic phase in the growth cycle of strain Bt-Bn1 and its production continued until the beginning of the stationary phase. The mode of action of this molecule displayed bacteriocidal or bacteriolytic effect depending on the concentration. The bioactive molecule was purified 78-fold from the bacteria supernatant with ammonium sulfate precipitation, dialysis, ultrafiltration, gel filtration chromatography, and HPLC, respectively. The molecular mass of this molecule was estimated via SDS-PAGE and confirmed by the ESI-TOFMS as 3,139 Da. The bioactive molecule was also determined to be a heat-stable, pH-stable (range 6-8), and proteinase K sensitive antibacterial peptide, similar to bacteriocins. Based on all characteristics determined in this study, the purified bacteriocin was named as thuricin Bn1 because of the similarities to the previously identified thuricin-like bacteriocin produced by the various B. thuringiensis strains. Plasmid elution studies showed that gene responsible for the production of thuricin Bn1 is located on the chromosome of Bt-Bn1. Therefore, it is a novel bacteriocin and the first recorded one produced by an insect originated bacterium. It has potential usage for the control of many different pathogenic and spoilage bacteria in the food industry, agriculture, and various other areas.

        A Highly Pathogenic Strain of Bacillus thuringiensis serovar kurstaki in Lepidopteran Pests

        In order to detect and identify the most toxic Bacillus thuringiensis strains against pests, we isolated a B. thuringiensis strain (Bn1) from Balaninus nucum (Coleoptera: Curculionidae), the most damaging hazelnut pest. Bn1 was characterized via morphological, biochemical, and molecular techniques. The isolate was serotyped, and the results showed that Bn1 was the B. thuringiensis serovar, kurstaki (H3abc). The scanning electron microscopy indicated that Bn1 has crystals with cubic and bipyramidal shapes. The Polymerase Chain Reactions (PCRs) revealed the presence of the cry1 and cry2 genes. The presence of Cry1 and Cry2 proteins in the Bn1 isolate was confirmed via SDS-PAGE, at approximately 130 kDa and 65 kDa, respectively. The bioassays conducted to determine the insecticidal activity of the Bn1 isolate were conducted with four distinct insects, using spore-crystal mixtures. We noted that Bn1 has higher toxicity as compared with the standard B. thuringiensis subsp. kurstaki (HD-1). The highest observed mortality was 90% against Malacosoma neustria and Lymantria dispar larvae. Our results show that the B. thuringiensis isolate (Bn1) may prove valuable as a significant microbial control agent against lepidopteran pests.

        The statistical optimization of bacterial cellulose production via semi-continuous operation mode

        Bacterial cellulose (BC) is highly pure and has a higher crystallinity and molecular weight than plantcellulose. Therefore, BC can be used in many different areas such as biotechnology, pharmaceutical,cosmetics. Because of the price of BC, the productivity of BC is an important parameter for industrial scaleapplications. In this study, BC was produced in static culture using a semi-continuous operation mode;the conditions were optimized using response surface methodology (RSM). The collected BC wascharacterized by Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM),and tensile strength. Optimization parameters were selected as glucose concentration, surface area/volume ratio, surface area and incubation day intervals. The optimum values for incubation dayintervals, volume changing ratios, glucose concentrations and surface area/volume ratios were 7 days,66%, 50 g/L and 1.22 cm 1, respectively. BC productivity reached 0.284 g/L/day under optimalconditions, while the model equation proposed 0.289 g/L/day. RSM is essential for determining theoptimum values of parameters for BC production compared with the one-variable-at-a-time method. The semi-continuous operation mode is alternative and a good candidate for the industrial scaleproduction of BC.
