        Expression of Thymosin α1-thymopentin Fusion Peptide in Pichia pastoris and its Characterization

        Thymopentin plays an important role in improving imbalanced immune systems of patients, however, it has a limited half-life in plasma. To get more stable and active thymopentin analogs, a fusion thymosin α1-thymopentin (Tα1-TP5) gene was synthesized and cloned into vector pGAPZαA. Tα1-TP5 fusion peptide was expressed in pichia pastoris and purified by metal chelating chromatography and gel filtration chromatography. The circular dichroism spectra (CD) indicated that the secondary structure of Tα1-TP5 fusion peptide is dominated by a-helix and random coil. In vitro analysis showed that the plasma half-life of Tα1-TP5 fusion peptide is 140 ±14 min, which is longer than that of TP5 (5.6±0.7 min) and Tα1 (127±11 min). The in vitro activity assay presented that Tα1-TP5 fusion peptide has greater activity in promoting proliferation of Kunming mouse splenocytes, and in vivo experiment it showed better activity in promoting the phagocytosis of macrophages and secretion of IL-2 than both Tα1 and TP5. Our findings suggest that Tα1-TP5 fusion peptide might be a potential therapeutic agent.

        PSD Integrated Calibration Method Based on Annunciator in Vacuum Environment

        In this paper, an integrated calibration method for position sensitive detector (PSD) of moving support mechanism in vacuum plasma environment is proposed. That is, the PSD is used to track the laser spot to restore the coordinate information of the signal. Firstly, a PSD integrated calibration principle is proposed to reduce the position error of the signaler by tracking laser points with PSD. Then on this basis, due to the spatial declination angle in the PSD imaging model, a spatial declination correction model was established to compensate the spatial declination error. Finally, there are errors occurred in the measurement because of the nonlinearity of PSD, and the improved backpropagation (BP) neural network calibration algorithm is adopted to further correct the errors through the Fletcher–Reeves linear search method to obtain the accurate coordinate information of the annunciator. MATLAB is used to verify the convergence of the calibration algorithm error. Simulation results show that 951 epochs need to be trained by using the traditional BP algorithm to make the position error converge to 0.001 mm. But when using the improved BP algorithm, only 44 epochs are needed.

        Efficient treatment of anthraquinone dye wastewater by adsorption using sunflower torus-like magnesium hydroxide microspheres

        Novel sunflower torus-like magnesium hydroxide (MH) microsphere particles were prepared by a facile one-step, self-assembly method. The synthesized products and the mechanism of adsorption of samples of the anthraquinone dyes reactive blue 19 (RB19) and alizarin red S (ARS) were analyzed by different modern characterization techniques, such as X-ray diffractometry (XRD), scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), energy dispersive X-ray spectrometry (EDS), the Brunauer-Emmett-Teller (BET) method and Fourier transform infrared (FT-IR) spectroscopy. The adsorptive potential of the as-prepared microspheres for the removal of RB19 and ARS in aqueous solution was evaluated. The effects of multiple condition parameters, including, adsorbent dosage, adsorption time, adsorption temperature, wastewater pH, rotating speed and sodium chloride concentration on the removal of the dyes from the wastewater were studied in detail. The effect of the structural and shape properties of the MH adsorbent on the dye adsorption performances was also studied. The results showed that sunflower torus-like MH was an effective adsorbent for dye removal. The removal rates of ARS and RB19 were 91.65% and 83.03%, respectively, under the optimized conditions. The maximum adsorption capacity of the microspheres was 349.85mg/g for ARS and 231.78mg/g for RB19 at 25 oC. The equilibrium adsorption experimental data of the microsphere adsorption conformed to the Freundlich isotherm for ARS and the Langmuir isotherm for RB19. The adsorption kinetics experimental studies showed that the pseudo-second-order and pseudo-first-order model perfectly fit for both ARS and RB19 microsphere adsorption. RB19 and ARS were absorbed on the sunflower torus-like MH surface via the formation of H-bonds. Thus, the sunflower torus-like MH microsphere particles are an effective purifying agent for the removal of the anthraquinone dyes ARS and RB19 from wastewater.

        영화 역사상 가장 높은 역사 지위를 지닌 영화 〈시민 케인〉독유의 영시 언어 창조성을 보여준 후 영화 창작 성공의 모범이 되였다. 그동안 수많은 영화인들로부터 부단히 학습하고 연구하였다. 〈미국영화 약사〉에서 말하기를 “1941년에 상영한 영화 〈시민 케인〉은 한 편의 결정적 영화이며 이는 인간의 소리가 있는 영화 역사에서 10년 동안 배운 기교를 종결하는 동시에 미래 발전 방향의 앞길을 인도해 주는 등대불이 되였다.”고 한다. 본 영화는 영화제작 천재 웰스의 첫 번째 스크린 작품이다. 서사 모식과 시청 언어상에서 선명한 실험 풍격, 창신 특정으로 인해 사람들의 영화에 대한 새로운 사고 인식을 가져다주었다. 본문은 〈시민 케인〉 서사 모식과 시청 언어 두 가지 방면에서 개시, 분석, 사고 줄거리, 카메라 운영, 구도 선택 및, 소리 운영 등 방면에서 영화 〈시민 케인〉의 창작 사고의 방향을 찾았다. Occupying a significant position in the history of films, Citizen Kane creatively set up a successful paradigm for subsequent films with its unique film languages, which has been learned by numerous film-makers. A Brief History of American Films said: “Citizen Kane, which premiered in 1941, is a crucial film, which concludes all skills in the past decade since sound film was created. In addition, it is also a beacon light that sheds light on the future development direction of films.” As the maiden film work of genius Orson Welles, Citizen Kane reflects a distinctive experimental style and innovation characteristics regarding its narrative model and visual-audio language, which enlightens people’s new thought and understanding of the film language. This thesis attempts to study the story plot, shot techniques, composition selection and sound application of Citizen Kane from two perspectives, including narration model and audio-visual speech, to explore its creation thinking.

        Removal of ammonia nitrogen from wastewater by three-dimensional electrode system based on solid waste containing iron

        In order to solve the problem of ammonia nitrogen pollution, three-dimensional electrode electrochemical oxidation technology is utilized to transform ammonia nitrogen. The influence of influent pH, power supply and electrolyte concentration on ammonia nitrogen removal by three-dimensional electrode method and its mechanism were discussed by single factor variable analysis. The particle electrode by emission scanning electron microscope (SEM) and energy dispersive spectroscopy (EDS). The results show that the removal efficiency of ammonia nitrogen by three-dimensional electrode is higher than that by two-dimensional electrode. When the aqueous solution is neutral, the direct and indirect oxidation reactions of ammonia nitrogen can reach the best state, and the removal efficiency is the highest. Within a certain range, the electrolytic efficiency will increase with the increase of electrolytic voltage and electrolyte concentration. The reduction of electrode spacing in a certain range can enhance the direct oxidation of ammonia nitrogen and improve the electrolysis efficiency of ammonia nitrogen. Under the conditions of pH 7, power supply voltage 14 V, electrolyte concentration 0.1 mol/L, electrode spacing 1 cm, electrolytic time 50 min, ammonia nitrogen can be completely removed from the three-dimensional electrode

        Unconditionally stable Gauge-Uzawa finite element methods for the Darcy-Brinkman equations driven by temperature and salt concentration

        In this paper, the Gauge-Uzawa methods for the Darcy-Brinkman equations driven by temperature and salt concentration \linebreak (DBTC) are proposed. The first order backward difference formula is adopted to approximate the time derivative term, and the linear term is treated implicitly, the nonlinear terms are treated semi-implicit. In each time step, the coupling elliptic problems of velocity, temperature and salt concentration are solved, and then the pressure is solved. The unconditional stability and error estimations of the first order semi-discrete scheme are derived, at the same time, the unconditional stability of the first order fully discrete scheme is obtained. Some numerical experiments verify the theoretical prediction and show the effectiveness of the proposed methods.

        Synthesis and Characterization of Low Viscosity Aromatic Hyperbranched Polyester Epoxy Resin

        Low viscosity aromatic hyperbranched polyester epoxy resin (HTBE) was synthesized by the reaction between epichlorohydrin (ECH) and carboxyl-end hyperbranched polyester (HTB) which was prepared from inexpensive materials $A_2$ (1,4-butanediol glycol, BEG) and $B_3$ (trimellitic anhydride, TMA) by pseudo one-step method. The molar mass of the HTB was calculated from its acid value by "Recursive Probability Approach". The degree of branching (DB) of the HTB was characterized by model compounds and $^1H$ NMR-minus spectrum technology, and the DB of the HTB was about $0.47{\sim}0.63$. The viscosity and epoxy equivalent weight of the HTBE were $3,600{\sim}5,000\;cp$ and lower than 540 g/mol respectively. The reaction mechanism and structure of the $AB_2$ monomer, HTB and HTBE were investigated by MS, $^1H$ NMR and FTIR spectra technology. The molecular size of HTBE is under 8.65 nm and its shape is ellipsoid-like as determined by molecular simulation.
