The native Asian oyster, Crassostrea ariakensisis one of the most common and important Crassostreaspecies that occur naturally along the coast of East Asia. Molecular species diagnosis is a prerequisite for populationgenetic analysis of wild oyster populations because oysterspecies cannot be discriminated reliably using externalmorphological characters alone due to character ambiguity. To date there have been few phylogeographic studies ofnatural edible oyster populations in East Asia, in particularthis is true of the common species in Korea C. ariakensis. We therefore assessed the levels and patterns of moleculargenetic variation in East Asian wild populations ofC. ariakensis from Korea, Japan, and China using DNAsequence analysis of five concatenated mtDNA regionsnamely; 16S rRNA, cytochrome oxidase I, cytochromeoxidase II, cytochrome oxidase III, and cytochrome b. Twodivergent C. ariakensis clades were identified betweensouthern China and remaining sites from the northernregion. In addition, hierarchical AMOVA and pairwise USTanalyses showed that genetic diversity was discontinuousamong wild populations of C. ariakensis in East Asia. Biogeographical and historical sea level changes are discussedas potential factors that may have influenced thegenetic heterogeneity of wild C. ariakensis stocks acrossthis region.
