<P>Cyclodextrins (CDs) are produced from starch by cyclodextrin glucanotransferase (CGTase), which has cyclization activity. Specifically, alpha-CD is an important biomolecule, as it is a molecular carrier and soluble dietary fiber used in the food industry. Upon inspection of the conserved regions of the glycoside hydrolase (GH) 13 family amylases, the amino acids K232 and H233 of CGTase were identified as playing an important role in enzyme reaction specificity. A novel CD hydrolyzing enzyme, cyclodextrin glycosyl transferase.(CGTase)-alpha, was developed using site-directed mutagenesis at these positions. Action pattern analysis using various substrates revealed that CGTase-alpha was able to hydrolyze beta- and gamma-CD, but not alpha-CD. This selective CD hydrolyzing property was employed to purify a alpha-CD from alpha-CD mixture solution. The alpha-CD that remained after treatment with CGTase-alpha and exotype glucoamylase was purified using hydrophobic interaction chromatography with 99% purity.</P>

Objective. To estimate the cardiovascular (CV) and gastrointestinal (GI) risks of etoricoxib in the treatment of osteoarthritis (OA) compared to a placebo and other non-steroidal anti-inflammatory drugs (NSAIDs). Methods. A systematic review of randomized, controlled trials (RCTs) of etoricoxib were performed. Bayesian network meta-analysis was used over a duration of 12 weeks. The incidence of CV and GI events for a duration ≥26 weeks were also tabulated and presented using descriptive statistics. Results. From this search, 10 studies were identified. Of these, 6 and 5 RCTs that measured the CV and GI events at 12 weeks were included in meta-analysis. They showed that etoricoxib did not increase the CV events compared to the placebo or NSAIDs during the 12 week period (odds ratio [OR]=0.59 compared to celecoxib, OR=0.89 with ibuprofen, OR=0.70 with placebo, and OR=2.16 with naproxen). The risk of GI events was comparable to that of most comparators, with the exception of naproxen, which had a significantly lower risk of GI events (OR=0.18) during the 12 week period. For a duration ≥26 weeks, the incidence of CV and GI events with etoricoxib increased with increasing duration. Conclusion. Etoricoxib is an alternative short-term treatment option for OA, showing comparable CV and GI complications to other NSAIDs. Nevertheless, further studies will be needed to elucidate the long-term safety of etoricoxib in the treatment of OA. (J Rheum Dis 2017;24:293-302)
Differential Expression Patterns of Gangliosides in Micro-pig solid organs

Ganglioside are ubiquitous membrane component in mammalian cells and suggested to play important roles in various cell functions such as cell-cell recognition, differentiation and transmembrane signaling. These compounds are localized in a glycosphingolipid-enriched microdomain on the cell surface and regulated by the glycosphingolipid composition. However, the role that gangliosides play in immune rejection response by xenotransplantation is not yet clearly understood. In this study, differential expression patterens of gangliosides of Micro-Pig solid organs, which are heart, kidney, liver, pancreas and spleen. We examined expression level of high-performance thin-layer chromatography (HPTLC) showed that all of solid organs contained GM3, GM2, GM1 and GD1a as major gangliosides. Especially, ganglioside GD1b expressed in liver and pancreas, not in heart, kidney and spleen. Also, GT1b expressed in liver, kidney and spleen, not in heart and pancreas. As a results suggest that differential expression patterens of gangliosides of Micro-Pig solid organs are related immune rejection response in xenotransplantation by biomarker.

<P>In this study, <I>α</I>-glucanotransferase from <I>Bacteroides thetaiotaomicron</I> was expressed in <I>Escherichia coli</I> and characterized. Conserved amino-acid sequence alignment showed that <I>Bacteroides thetaiotaomicron α</I>-glucanotransferase (Bt<I>α</I>GTase) belongs to the glycoside hydrolase family 77. The enzyme exhibited optimal catalytic activity at 60°C and pH 3.0. Bt<I>α</I>GTase catalyzed transglycosylation reactions that produced only glycosyl or maltosyl transfer products, which are preferable for the generation of transglycosylated products with high yield. The 1-deoxynojirimycin (DNJ) glycosylation product G1-DNJ was generated using Bt<I>α</I>GTase, and the inhibitory effect of G1-DNJ was analyzed. A kinetic study of inhibition revealed that G1-DNJ inhibited <I>α</I>-glucosidase to a greater extent than did DNJ but did not show any inhibitory effects towards <I>α</I>-amylase, suggesting that G1-DNJ is a potential candidate for the prevention of diabetes.</P>
월별 드론 영상을 이용한 밴드 조합에 따른 수목 개체 및 수관폭 추출 실험

공간해상도가 높은 드론 영상은 수목 밀도가 높은 지역에서 추출 한계를 갖는 기존 연구의 대안으로 떠오르고 있다. 본 연구에서는 드론 영상으로부터 수목이 우거진 산림 지역 내 수목 개체를 추출하였다. 영상 분할 과정을 거쳐서 추출되는 수목 개체 인식을 위해, DSM(digital surface model), 그리고 R, G, B 밴드 모두를 조합한 경우와 각각을 분리 조합한 경우의 영상 분할 결과를 비교하였다. 또한, 낙엽수림의 수목 우거짐의 변화를 시기별⋅영상별로 실험하였다. 3, 4, 5월 영상 중 숲이 울창한 5월의 경우 현지 측량한 나무를 기준으로 한 수목 개체 추출율은 50%로 나타났고, 수관폭 정확도 분석 결과 RMSE(root mean square error)가 1.5미터 이하로 가장 좋은 결과를 보였다. 실험지역의 추출은 중간 나무, 작은 나무 2가지 크기로 추출하였으며 작은 크기의 나무가 추출 정확도가 더 높았다. 이를 바탕으로 수고 추출을 하고, 수관폭과 흉고직경간의 관계식을 이용하여 흉고직경을 추정한다면, 임목재적 추정 및 산림바이오매스 추정까지 가능할 것으로 보인다. Drone images with high spatial resolution are emerging as an alternative to previous studies with extraction limits in high density forests. Individual tree in the dense forests were extracted from drone images. To detect the individual tree extracted through the image segmentation process, the image segmentation results were compared between the combination of DSM and all R,G,B band and the combination of DSM and R,G,B band separately. The changes in the tree density of a deciduous forest was experimented by time and image. Especially the image of May when the forests are dense, among the images of March, April, May, the individual tree extraction rate based on the trees surveyed on the site was 50%. The analysis results of the width of crown showed that the RMSE was less than 1.5m, which was the best result. For extraction of the experimental area, the two sizes of medium and small trees were extracted, and the extraction accuracy of the small trees was higher. The forest tree volume and forest biomass could be estimated if the tree height is extracted based on the above data and the DBH(diameter at breast height) is estimated using the relational expression between crown width and DBH.
