Purpose: Retrospective audit of recurrent glioma patients treated by different fractionation schedules and to validate the modified Combs prognostic score in Indian patient cohort. Materials and Methods: Between Jan 2009 and June 2022, 66 recurrent gliomas patients treated with standard adjuvant treatment—radiation (RT) ± temozolomide (chemotherapy)—and re-treated with RT (± chemotherapy) were categorized as per modified Combs prognostic criteria and outcomes were compared. Results: Sixty-six patients with recurrent gliomas who received reirradiation (re-RT) were audited—53% males; 61% Karnofsky performance status (KPS) ≥80 at time of re-RT; median age 41.5 years (range, 6 to 70 years); 67% <50 years; primary histology low-grade glioma in 33% ; grade III 27%, grade IV 40%; initial median dose of 60 Gy equivalent dose in 2 Gy fractions (EQD2); maximum safe resection at recurrence 41%; mean and median follow-up 78 ± 51 months and 66 months. Mean time interval between RT was 46.4 ± 39 months. Mean planning target volume (PTV) volume in conventional RT (Conv-RT), hypofractionated RT (Hypo-RT), and ultra-hypofractionated RT (UF-RT) was 226.1 ± 140.7 mL, 162.8 ± 123.3 mL, and 143.3 ± 145.8 mL. Mean dose for Conv-RT, Hypo-RT, and UF-RT was 50 Gy (range, 40 to 60), 31 Gy (range, 20 to 40), and 20 Gy (range, 10 to 30). Mean overall survival (OS) in Conv-RT, Hypo-RT, and UF-RT cohort was 18.8 months (range, 2.4 to 76.8); 6.6 months (range, 2 to 17.4), and 13.9 months (range, 3 to 131.9). Median OS as per Combs criteria were 16.6 months (Group a), 24.6 months (Group b), 4.6 months (Group c), and 3 months (Group d). Significant survival benefit was with good KPS score (KPS >80 vs. <80; 20.46 vs. 5.25 months; p < 0.001), patients receiving salvage chemotherapy (20.46 vs. 6.96 months; p = 0.001), and patients received re-RT biological equivalent dose (BED3) >80 Gy (16.62 vs. 5.48 months; p = 0.03). Median overall survival (OS) in our patient cohort and Combs cohort in Group a was 16.6 and 19.5 months; Group b was 24.6 and 11.3 months; Group c was 4.7 and 8.1 months, and Group d was 2 and 5.5 months, respectively. Six months survival in our patient cohort and Combs cohort in Groups a, b, c, d were 100%, 92%, 34%, 17% and 94%, 79%, 70%, 41%, respectively. Twelve months survival in our patient cohort and Combs cohort in Groups a, b, c, d were 88%, 74%, 22%, 0% and 88%, 47%, 22%, 7%, respectively. Conclusion: Modified Combs prognostic factors predicts OS and is applicable in Indian subcontinent patient population.

Background: Heat-shock protein70 (HSP70) are intracellular protein chaperones, with emerging evidence of their association with various diseases. We have previously reported significantly elevated plasma-HSP70 (pHSP70) in pancreatic cancer. Current methods of pHSP70 isolation are ELISA-based which lack specificity due to cross-reactivity by similarities in the amino-acid sequence in regions of the protein backbone resulting in overestimated HSP70 value. Materials and Methods: This study was undertaken to develop a methodology to capture all isoforms of pHSP70, while further defining their tyrosine and serine phosphorylation status. Results: The methodology included gel electrophoresis on centrifuged supernatant obtained from plasma incubated with HSP70 antibody-coupled beads. After blocking non-specific binding sites, blots were immunostained with monoclonal-antibody specific for human-HSP70, phosphoserine and phosphotyrosine. Conclusions: Our novel immunocapture approach has distinct advantages over the commercially available methods of pHSP70 quantification by allowing isolation of molecular aggregates of HSP70 with additional ability to precisely distinguish phosphorylation state of HSP70 molecules at serine and tyrosine residues.

Rhetorical relations between two text fragments are essential information and support natural language processing applications such as Question - Answering (QA) system and automatic text summarization to produce an effective outcome. Question - Answering (QA) system facilitates users to retrieve a meaningful response. There is a demand for rhetorical relation based datasets to develop such a system to interpret and respond to user requests. There are a limited number of datasets for developing an Arabic QA system. Thus, there is a lack of an effective QA system in the Arabic language. Recent research works reveal that unsupervised learning can support the QA system to reply to users queries. In this study, researchers intend to develop a rhetorical relation based dataset for implementing unsupervised learning applications. A web crawler is developed to crawl Arabic content from the web. A discourse-annotated corpus is generated using the rhetorical structural theory. A Naïve Bayes based QA system is developed to evaluate the performance of datasets. The outcome shows that the performance of the QA system is improved with proposed dataset and able to answer user queries with an appropriate response. In addition, the results on fine-grained and coarse-grained relations reveal that the dataset is highly reliable.

<P>A new donor–acceptor-conjugated organic small molecule, <B>BDT(TBT)</B><SUB><B>2</B></SUB>, comprised of benzo[1,2-<I>b</I>:4,5-<I>b</I>′]dithiophene and 2,1,3-benzothiadiazole units was designed and synthesized. The small molecule <B>BDT(TBT)</B><SUB><B>2</B></SUB> in its thin film showed an absorption band in the range of 300–700 nm with an absorption edge at 650 nm and an optical band gap of 1.90 eV. As estimated from the cyclic voltammetry measurements, the HOMO and LUMO energy levels of <B>BDT(TBT)</B><SUB><B>2</B></SUB> were −5.44 and −3.37 eV, respectively. The spin-coated thin film of <B>BDT(TBT)</B><SUB><B>2</B></SUB> exhibited p-channel output characteristics with a hole mobility of 2.7 × 10<SUP>–6</SUP>. <B>BDT(TBT)</B><SUB><B>2</B></SUB>, when explored as an electron-donor material in solution-processed bulk-heterojunction organic solar cells in conjunction with a PC<SUB>71</SUB>BM acceptor with an active layer thickness of 50–55 nm, generated a power conversion efficiency (PCE) of 1.18%. A more impressive PCE of ∼2.9% with a short-circuit current density (<I>J</I><SUB>sc</SUB>) of 7.94 mA cm<SUP>–2</SUP> and an open-circuit voltage (<I>V</I><SUB>oc</SUB>) of 0.89 V was achieved when the active layer of the cell was annealed at higher temperature (∼180 °C).</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/aamick/2012/aamick.2012.4.issue-12/am301841p/production/images/medium/am-2012-01841p_0008.gif'></P>

<P><I>meso</I>-Ethynyl extended aryl-picket calix[4]pyrroles 2 and 3 are designed and synthesized by directly anchoring arylethynyl groups at diametrically opposed <I>meso</I>-positions. The critical roles of direct ethynyl linkers are manifested through the isolation of unexpected host-anion conformers of <I>meso</I>-arylethynyl calix[4]pyrroles and a significant enhancement in halide binding affinities.</P>

<P>Washing processes, essential in most heterogeneous labeled assays, have been a big hurdle in simplifying the detection procedure and reducing assay time. Nevertheless, less attention has been paid to washing-free heterogeneous labeled assays. We report a purely washing-free immunosensor that allows fast, sensitive, and single-step detection of prostate-specific antigen in serum with low interference. Proximity-dependent electron mediation of ferrocenemethanol (Fc) between an indium–tin oxide (ITO) electrode and a glucose-oxidase (GOx) label allows us to discriminate between a bound and an unbound label: a bound label offers faster electron mediation than an unbound one. The electrooxidation of Fc at a low applied potential (0.13 V vs Ag/AgCl) and a low electrocatalytic ITO electrode and the oxidation of <SMALL>l</SMALL>-ascorbic acid by <SMALL>l</SMALL>-ascorbate oxidase minimize the effect of the interfering species. With a high concentration of glucose (200 mM), the signal and background levels are hardly dependent on the glucose-concentration variation in the sample. The washing-free immunosensor can detect a concentration of ca. 1 pg/mL for mouse IgG in phosphate-buffered saline and a concentration of ca. 10 pg/mL for prostate-specific antigen spiked in female serum after an incubation period of 10 min. <P>In washing-free electrochemical detection, various redox and reactive species cause significant interference. To minimize this interference, we report a washing-free electrochemical immunosensor using flavin adenine dinucleotide (FAD)-dependent glycerol-3-phosphate dehydrogenase (GPDH) and glycerol-3-phosphate (GP) as an enzyme label and its substrate, respectively, because the reaction of FAD-dependent dehydrogenases with dissolved O<SUB>2</SUB> is slow and the level of GP preexisting in blood is low (<0.1 mM). A combination of a low electrocatalytic indium–tin oxide (ITO) electrode and fast electron-mediating Ru(NH<SUB>3</SUB>)<SUB>6</SUB><SUP>3+</SUP> is employed to obtain a high signal-to-background ratio via proximity-dependent electron mediation of Ru(NH<SUB>3</SUB>)<SUB>6</SUB><SUP>3+</SUP> between the ITO electrode and the GPDH label. Electrochemical oxidation of GPDH-generated Ru(NH<SUB>3</SUB>)<SUB>6</SUB><SUP>2+</SUP> is performed at 0.05 V vs Ag/AgCl, at which point the electrochemical interference is very low. When a washing-free immunosensor is applied to cardiac troponin I detection in human serum, the calculated detection limit is approximately 10 pg/mL, indicating that the immunosensor is very sensitive in spite of the use of washing-free detection with a short detection period (10 min for incubation and 100 s for electrochemical measurement). In an attempt to further lower the HOMO of a cost-effective polymer poly(2,3-bis-(3-octyloxyphenyl) quinoxaline-5,8-dyl-alt-thiophene-2,5-diyl) (TQ) by adding F atoms onto the existing quinoxaline acceptor within the polymer backbone, we have synthesized two structurally identical fluorinated analogues of TQ (TQ-F (single F) and TQ-FF (double F)), except for the number of F atoms. The effects of inclusion of F atoms on the optical properties, nature of charge transport, and molecular organization are thoroughly investigated. The resulting two fluorinated polymers show a decrease in both the HOMO and the LUMO energy levels relative to non-fluorinated TQ. Moreover, the fluorination of the polymer backbone has lowered the HOMOs more than the LUMOs, slightly widening the energy bandgaps as the number of F atoms increases. Thus, use of these polymers in bulk-heterojunction (BHJ) solar cells, in all cases, leads to large V-OC values. The power conversion efficiency (PCE) of the optimized PSCs based on TQ-F reaches 4.41%. In addition, it is interesting to note that, despite TQ-FF having the PCE that is lower than that of TQ-F, an unprecedentedly high V-OC of 1.00 V is achieved, which is nearly equal to the highest V-OC values ever reported for polymers.
