Chymotrypsin was purified from jumbo squid hepatopancreas (HP) with 2.4-fold and yield 1.9%, and characterized with a molecular weight of 31 kDa, as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Chymotrypsin effect over collagen extracted from the mantle, fins and arms of the jumbo squid was evaluated. The enzyme exhibited the maximum activity at pH 7 and $65^{\circ}C$ using Suc-Ala-Ala-Pro-Phe-p-nitroanilide (SAAPNA) as a substrate and it was identified using the specific inhibitors N-tosyl-L-phenylalaninechloromethyl ketone (TPCK) and phenyl methyl sulfonyl fluoride (PMSF), showing residual activities of 6% and 0%, respectively. Furthermore, high activity was observed in the pH range of 4.0 to 8.0. Purified enzyme showed a moderate in vitro activity using muscle collagen as a substrate. Although further research is needed, the results suggest that the enzyme has a potential application where acidic or slightly alkaline conditions are needed.
A taxonomic revision of <i>Allium</i> (Alliaceae) in the Canadian prairie provinces

<P> The taxonomy, rarity, and conservation status of Allium L. is revised for the Canadian prairie provinces, based on analyses of herbarium specimens and fieldwork. Five species are recognized: Allium schoenoprasum L., A. geyeri S. Watson var. tenerum M.E. Jones, A. textile A. Nelson & J.F. Macbride, A. cernuum Roth, and A. stellatum Ker Gawler. Distribution maps and a key to species are provided, as well as complete descriptions of the species examined, including new illustrations, information on nomenclatural types, synonymies, and chromosomal and ecological data. A lectotype is designated for A. geyeri var. tenerum. In this study, A. geyeri var. geyeri reported from Alberta and Saskatchewan and ranked in these provinces as having rarity levels S2 and S1, respectively, by the Nature Conservancy, is excluded from the Canadian flora and the rare list of these provinces because it was misidentified from a herbarium specimen of A. textile. Allium tricoccum Solander in W. Aiton is regarded as a non-native species to Manitoba. The rarity and conservation status of Allium in the Canadian prairie provinces is as follows: (i) A. schoenoprasum, listed as S2 in Saskatchewan, is rare in Manitoba, although its rarity status has not been formally assessed in the province; (ii) A. geyeri var. tenerum is the rarest Allium taxon, with distribution restricted to the Waterton Lakes National Park areas of Alberta, and is currently listed as S2; and (iii) A. cernuum was re-evaluated and a rarity level of S1S2 was recommended for the species in Saskatchewan, particularly in its southwestern distributional habitat. </P>

The major components of the wastewater from the petroleum refineries are benzene, toluene and phenoland one of the techniques applied to the treatment of effluents is sorption using organo-functionalizedclay. The materials exploited in the present study are a family of surface-functionalized synthetic micasand their sorption capacities for non-ionic organic pollutants are analyzed. The organo-functionalizationof their surface provides them the capacity to sorb effectively non-ionic pollutants in the interface. Theiradsorption performance is a function of the alkylamonium properties such as the chain length, the massfraction and the organization of the organic cation in the interlayer space of the micas.

Present record is the first reported occurrence of the species in the Tropical Eastern Pacific biogeographic region (Gulf of California to southern Ecuadorian waters). Its large fan-like anal and dorsal fins and its counts of fin rays and vertebrae can distinguish the Pacific fanfish from the other species in the genus. This fish may have not yet been recorded in the region because its presence has been overlooked in the past because of rarity and lack of commercial value. This paper presents a mathematical model that allows representing the optical behavior of a solar parabolic dish concentrator and the thermal performance of a cavity receiver. A procedure and a graphical method for the design of dish/cavity systems are proposed. A parametric study of the main geometric variables is performed and the influence of climate variables on the thermal behavior of the system coupled to a Stirling engine is analyzed. The model considers errors of solar collector, intercept factor, reflected and emitted radiation,conduction, and convection heat losses. For the validation of the model, the results obtained were compared with theoretical and experimental results reported in the literature. The calculation of the radiation losses, emitted and reflected from the receiver presented errors of up to 14%, and the average error for the rest of the thermal losses, interception factor and the absorber’s temperature, was less than 3%. These results show that the proposed model can be used with sufficient certainty to design and optimize solar dish collectors with cavity receivers.
Surface analysis of GeC prepared by reactive pulsed laser deposition technique

Amorphous germanium carbide (a-Ge_(1-x)C_x) thin films were prepared by reactive pulsed laser deposition technique using several methane pressures. Surface analysis was performed by X-ray photoelectron spectroscopy (XPS) to examine the composition and elemental bonding at the surface of the material. Optical analysis was carried out by spectroscopic ellipsometry to study the optical constants (n and k) and other parameters of the film. Results indicate that the carbon atoms to be incorporated in the germanium lattice, forming a-Ge_(1-x)C_x alloy, for concentrations below about 10 atomic % where the Ge atoms are uniformly distributed. There is formation of graphitic agglomerates for higher carbon concentrations.

Fresh and preserved (type material) specimens of the black ghost chimaera Hydrolagus melanophasma were compared for morphometric characteristics. A molecular comparison was also performed on two mitochondrial gene sequences (12S rRNA and 16S rRNA gene sequences). While significant differences in measurements were found, the differences were not attributable to sexual dimorphism or the quality of the specimens, but to the sample size and the type of statistical tests. The result of the genetic characterization showed that 12S rRNA and 16S rRNA genes represented robust molecular markers that characterized the species.

Mycobacterium tuberculosis produces mycolic acids which are relevant for persistence, recalcitrance to antibiotics and defiance to host immunity. c-di-GMP is a second messenger involved in transition from planktonic cells to biofilms, whose levels are controlled by diguanylate cyclases (DGC) and phosphodiesterases (PDE). The transcriptional regulator dosR, is involved in response to low oxygen, a condition likely happening to a subset of cells within biofilms. Here, we found that in M. bovis BCG, expression of both BCG1416c and BCG1419c genes, which code for a DGC and a PDE, respectively, decreased in both stationary phase and during biofilm production. The kasA, kasB, and fas genes, which are involved in mycolic acid biosynthesis, were induced in biofilm cultures, as was dosR, therefore suggesting an inverse correlation in their expression compared with that of genes involved in c-di-GMP metabolism. The relative abundance within trehalose dimycolate (TDM) of α- mycolates decreased during biofilm maturation, with methoxy mycolates increasing over time, and keto species remaining practically stable. Moreover, addition of synthetic c-di-GMP to mid-log phase BCG cultures reduced methoxy mycolates, increased keto species and practically did not affect α-mycolates, showing a differential effect of c-di-GMP on keto- and methoxy-mycolic acid metabolism.
