Also, eddy current loss analysis and copper loss calculation are performed.

        혈소판제제는 출혈질환이나 혈소판의 기능장애 치료를 위해 사용되고 있다. 혈소판성분채집술은 혈액성분채집기를 사용하여 혈소판 성분을 채집하는 방법으로, 성분채집혈소판의 질 평가에는 다양한 지표들이 이용되고 있다. 채집된 혈소판의 생산성은 채집효율(collection efficiency)과 채집률(collection rates)로 평가되고 보관된 혈소판의 질은 in vitro 상에서 시각적 외양, 대사활성도, 양, 혈소판 수, 백혈구 수, 미세입자(microparticles), 그리고 다양한 혈소판 활성 표지자들로 평가된다. 혈소판 활성 표지자들은 여러 연구분야에서도 이용되고 있다. 수혈된 후의 혈소판의 질은 교정증가치(corrected count increment)와 혈소판회복퍼센트(percentage platelet recovery)로 평가된다. 본 논문은 혈소판성분채집술의 채집과정, 사용되는 장비, 성분채집혈소판의 질, 전반적인 효율성, 그리고 질평가 지표들에 대한 리뷰(review)와 함께 다른 혈액성분채집기 간의 기능의 차이점을 비교하였고, 또한 혈소판성분채집술은 구연산염 항응고제에 의한 부작용을 일으킬 수는 있지만 전혈 헌혈시에 실신의 전구증상이나 실신을 일으킬 위험이 있는 헌혈자들에게는 더 안전한 방법임을 보여주고 있다. Platelet products are used to treat hemorrhagic or platelet dysfunction diseases. Plateletpheresis involves collecting the platelet components of blood using an apheresis blood-collection system. Various indicators are available for evaluating the qualities of the apheresis platelets. The productivity of platelet collection is evaluated through both the collection efficiency and collection rates. Platelet storage quality can be evaluated in vitro using several indicators, including visual appearance, metabolic activities, volume, platelet count, white blood cell count, microparticles, and various platelet activation markers. Platelet activation markers have been used as indicators of storage quality in various studies. Post-transfusion platelet quality can be evaluated based on the corrected count increment and the percentage of platelet recovery. Although various studies have investigated the aspects of plateletpheresis, no article has systemically presented assessments of the platelet products obtained from different plateletpheresis devices. The present study provides a review of plateletpheresis, including the specifics of the process, the types of devices employed, the platelet quality, the overall efficacy, and the evaluation indicator qualities. Furthermore, the differences in functionality among the different apheresis devices are discussed. Although adverse reactions to the citrate anti-coagulant have been reported, apheresis processing may provide a safer option for donors who are at a high risk for presyncopal or syncopal reactions related to whole blood collection.

        겨우살이 Lectin이 흰쥐의 혈청지질 및 간조직에 미치는 영향

        본 연구에서 국내산 겨우살이 lectin이 흰쥐 간의 지방형성에 미치는 영향을 알아보기 위해 혈청 내 지질함량을 측정하였고, 이를 확인하고자 간조직의 지방형성 변화를 조사하였다. 렉틴의 분리는 sephadex G-150로 여과 및 sepharose 4B affinity chromatography 로 정제하였다. CCl₄를 복강 내에 투여시 72시간 경과 후 혈청 총cholesterol 및 triglyceride는 정상군 및 olive oil 투여군에 비해 유의하게 증가하였으며, lectin과 CCl₄를 동시 투여 하였을 때는 유의한 감소를 나타내였다. 흰쥐 간의 조직학적 관찰은 CCl₄투여군은 정상군 및 olive oil 투여군에 비해 지방변화 및 간세포 내에 많은 대소의 cytoplasmic vacuole가 보였으나, CCl₄와 lectin을 동시 투여시는 CCl₄ 투여군에 비해 cytoplasmic vacuole은 현저히 감소하였고, 이에 비례하여 혈청 cholesterol 및 triglyceride 측정치가 현저히 감소하는 결과를 나타내었다. 이상의 결과를 볼때 흰쥐에서 lectin이 혈청 지질을 감소시키거나 또는 조직학적 관찰에서 간조직의 지질형성을 억제시키는 효과가 있는 것으로 확인되었다. This study was carried out to investigate effect of serum lipid and liver tissue in lectin from Viscum album on rats. The lectin was purified by sepharose 4B affinity chromatography and gel filtration using sephadex G-150 with plant material from Viscum album. After 72 h of CCl₄ injection, there was a significant increase in serum total cholesterol and triglycerige levels relative to the control group. However, treatment of both Viscum album and purified lectin were significantly decreased lipid parameters against the CCl₄-induced. Histological observation of the liver section of rats challenged with CCl₄ produced a marked increase of cytoplasmic vacuoles in number, while rats administered olive oil alone did not alter the normal hepatic architecture. Histological observation of the liver section in rats treated 72 h with either Viscum album purified lectin or CCl₄-induced liver lipogenesis showed decreased numbers of cytoplasmic vaculoes and necrotic cells. These results suggest that Viscum album lectin has a significantly decreased lipid in serum or histological observation of the liver section decreased in lipogenesis in rats.

        An Investigation of Glyceollin I’s Inhibitory Effect on The Mammalian Adenylyl Cyclase

        글리세올린 I 은 다양한 피부 질환의 예방과 치료에 유용한 물질로 주목을 받아왔다. 그러나 피부 멜라닌 형성에 결정적 역할을 담당하는 포유류의 아데니닐 고리화 효소(이하 mAC)에 대한 직접적인 결합 형태와 상호 작용에 대한 연구는 현재까지 연구 보고된 사례가 없었다. 글리세올린 I 의 mAC 활성 부위에 대한 결합 작용 기작을 규명하기 위하여 우선 글리세올린 I과 SQ22,536 (mAC에 결합하여 mAC의 활성을 억제하는 것으로 이미 잘 알려진 단일 화합 물질)의 mAC에 대한 결합 친화도와 결합 형태를 비교 분석 하였다. 글리세올린 I은 mAC의 활성부위에 존재하는 Asp 1018, Trp 1020, Asn 1025와 각각 3개의 수소결합을 형성하는 것으로 분석되었고 SQ22,536은 mAC 활성부위의 Asp 1018, Asn 1025와 2개의 수소결합을 형성하여 글리세올린 I이 상대적으로 우월하게 결합하는 것으로 분석되었다. 글리세올린 I은 또한 포스콜린(forskolin)에 의해서 유도되는 세포내 멜라닌 형성 2차 신호전달 물질인 cyclic AMP의 생성과 이로인해 유발되는 단백질 인산화 효소 A의 인산화를 효과적으로 억제하는 것을 멜라노마 세포 실험을 통하여 확인하였다. 또한 글리세올린 I 을 장시간 세포에 투여하여도 세포의 생존률에는 영향을 주지 않음을 확인하였다. 본 연구 통하여 규명된 글리세올린 I의 mAC 활성 억제 효능 및 멜라닌 생성 신호전달 기작을 조절하는 성질을 이용하여 향후 흑색종과 같은 다양한 피부 질환 치료제 개발 및 미백화장품 핵심 물질 개발에 활용될 수 있을 것으로 사료된다. Glyceollin I has gained attention as a useful therapy for various dermatological diseases. However, the binding property of glyceollin I to the mammalian adenylyl cyclase (hereafter mAC), a critical target enzyme for the down-regulation of skin melanogenesis, has not been fully explored. To clarify the action mechanism between glyceollin I and mAC, we first investigated the molecular docking property of glyceollin I to mAC and compared with that of SQ22,536, a well-known mAC inhibitor, to mAC. Glyceollin I showed superiority by forming three hydrogen bonds with Asp 1018, Trp 1020, and Asn 1025, which exist in the catalytic site of mAC. However, SQ22,536 formed only two hydrogen bonds with Asp 1018 and Asn 1025. Secondly, we confirmed that glyceollin I effectively inhibits the formation of forskolin-induced cAMP and the phosphorylation of PKA from a cell-based assay. Long term treatment with glyceollin I had little effect on the cell viability. The findings of the present study also suggest that glyceollin I may be extended to be used as an effective inhibitor of hyperpigmentation.

      • 한국인에서 혈청 중성지방, 복부비만과 허리둘레의 상관성연구

        Purpose: The purpose of this study was to determine the association between serum triglyceride, abdominal obesity, and waist circumference in Korean. Method: A total of 168 healthy examinees that enrolled in this study were examined at Gimcheon Medical Center from July 2012 to December 2013. Waist circumference (WC) and body mass index (BMI) of subjects were measured as well as blood pressure, fasting blood sugar level, High Density Lipoprotein (HDL-cholesterol), Low Density Lipoprotein (LDL-cholesterol), cholesterol, and C-reactive protein (CRP). Results: In men, WC(p<0.011) had a significant correlation with triglyceride and BMI(p<0.001) had a significant correlation with triglyceride. In women(p<0.001), WC and BMI had a significant correlation with triglyceride. In men, triglycerides (r=0.501, p<0.011) had a positive correlation with WC. However, CRP (r=0.407, p<0.001) had a positive correlation with BMI. In women, triglycerides (r=0.470, p<0.001) and CRP (r=0.367, p<0.001) had a positive correlation with WC. Conclusion: Therefore, WC and triglyceride may be the most effective anthropometric indices for screening Korean people in regards to obesity.

        he authors evaluated a recent introduced chemical analyzer, Biotechnica Instruments 1000 (BT-1000) by analyzing the levels of GOT, GPT, creatinine, cholesterol and triglyceride in both pooled serum and SER-T-FY control serum. The accuracy was good maintained within a range of significant levels. The linearity was well maintained within a range of medical significant levels. The within-run coeffi- cients of variations (CVs) at low and high concentrations were less than 6.5%. The between-run coefficients of variations at low and high concentrations were less than 7.9%. The coefficient correlation of GOT, GPT, creatinine, cholesterol and triglyceride levels from BT-1000 or Olympus AU 400, which is a usual instrument for determing the levels of clinical makers, were from 0.971 to 0.988. The accuracy, linearity, precision and correlation of BT-1000 were good. It is con-cluded that BT-1000 analyzer can be used for clinical laboratories.

        We have studied the assay method of plasma -Thromboglobulin This method the results were summarized as followings. 1) Plasma seperating must do within in 4hours with collection sample. 2) Plasma seperating is higher than 3000G in centrifugation velocity. 3) The maintenance must 4C for plasma seperating temperature. 4) Plasma seperating centrifugation time is higher than 20mins. 5) Ag-Ab incubation time is higher than 60mins. 6) The coefficient of variation in intraassay and interassay were all less than 11%. The assay recovery mean 100.1%.

      • 면역검사실에서 이월오염에 대한 영향의 분석

        The purpose of the carry-over recovery experiment in clinical immunology test is performed to estimate effect variation from high concentration to low concentration. In practice, carry over experiment is a problem only for analyte with a wide clinical range of analyte concentration, such that a minute degree of carry over could have significant clinical implications. One suggested method to study carry-over contamination is to run known high level patient samples, followed by known low level patient samples to see if the results of the low level material are affected significantly. If carry-over is detected, th team leader and laboratory physician in managing it the analyte concentration above which subsequent patient samples may be affected, and define this output value in the procedure. Carry-over experiment should be expressed as a percentage because the experimental objective is to estimate to reduce effect variation from high level patient sample to low level patient sample, which is a percentage type of error, Good carry-over is 0%. The results of carry-over experiment were Imunoglobulin G (-0.41%), Imunoglobulin A (-0.86%), third complement component (-0.47%) , fourth complement (0.19%) with turbidimetric immunoassay of DADE Behring . hepatitis B surface antibody (0%), hepatitis B surface antigen (-0.004%), hepatitis B core antibody imunoglobulin M (0.12%), hepatitis B core antibody (0%), hepatitis B envelope antigen (0.0043%), hepatitis B envelope antibody (0.012%), with chemiluminescent microparticle immunoassay (CMIA) of ABBOTT Diagnostic Division in Germany. We concluded that the results of carry-over experiment compared to the range of error allowable for the test. There were no item betond the 1% criterion for acceptable performance.
