        Kinetic Approaches to Measuring Peroxiredoxin Reactivity

        Peroxiredoxins are ubiquitous thiol proteins that catalyse the breakdown of peroxides and regulate redox activity in the cell. Kinetic analysis of their reactions is required in order to identify substrate preferences, to understand how molecular structure affects activity and to establish their physiological functions. Various approaches can be taken, including the measurement of rates of individual steps in the reaction pathway by stopped flow or competitive kinetics, classical enzymatic analysis and measurement of peroxidase activity. Each methodology has its strengths and they can often give complementary information. However, it is important to understand the experimental conditions of the assay so as to interpret correctly what parameter is being measured. This brief review discusses different kinetic approaches and the information that can be obtained from them.

        MULTIFUNCTIONALIZED SPIONs FOR NUCLEAR TARGETING: CELL UPTAKE AND GENE EXPRESSION

        Superparamagnetic iron oxide nanoparticles (SPIONs) are used in many biological applications,which necessitate intracellular targeting. Here, we investigate intracellular localization andgene expression in HeLa cells after treatment with functionalized SPIONs. Functional groups investigated included positive amino propyl silane (APS), polyethylene glycol and targetingpeptides: nuclear targeting peptide (NTP) and/or cancer cell uptake promoting peptide (cRGD). Results revealed that the intracellular localization of SPIONs was strongly dependent on thesurface chemistry. Nuclear targeted SPIONs functionalized with only NTP or both NTP andcRGD were mostly localized in perinuclear endosomes with a small fraction entering the nucleus. The biocompatibility of cells after treatment was also dependent on surface chemistry, whereSPIONs functionalized with both NTP and cRGD exhibited a more signi¯cant reduction of cellproliferation compared to NTP or cRGD individually. Interestingly, gene expression aftertreatment with SPIONs was similar, regardless of the surface functionalization or intracellularlocalization. The results of this study showed that cellular uptake and intracellular localizationpredominantly depended on the surface chemistry, while gene expression exhibited a more genericresponse to SPION treatment.

      • College Students’ General Creativity as a Predictor of Cognitive Risk Tolerance

        Cognitive risk tolerance is an individual’s ability to formulate and express one’s ideas despite potential opposition or negative assessment in regard to reputation, integrity, and honor, which is needed for creativity, innovation and global competi-tiveness. The goal of this study was to further examine the degree of the relationship between cognitive risk tolerance and general creativity in various college majors. We hypothesized that higher levels of general creativity would predict higher levels of cognitive risk tolerance. Male (n = 676) and female (n = 487) college students from various college majors from engineering, psychology, music and English classes completed creative personality, creative temperament and cognitive risk tolerance measures. Both creative personality (r = .42) and creative temperament (r = .35) were predictors of higher cognitive risk tolerance. Engineering and social science majors had higher cognitive risk tolerance than other college majors. Older students and ethnic minorities (non-Caucasians) also demonstrated higher cognitive risk tolerance. Through cognitive risk tolerance, college students can utilize cognitive thinking skills that may contribute to innovation in the current global economic climate.

        Bitmap-Assisted Focused Ion Beam Fabrication of Combined Atomic Force Scanning Electrochemical Microscopy Probes

        Bitmap assisted focused-ion-beam (FIB) milling for the fabrication of combined atomic force scanning electrochemical microscopy (AFM-SECM) probes is presented. This technique facilitates the integration of frame-shaped nanoelectrodes recessed from the AFM tip. Thereby, the fabrication process for bifunctional AFM-SECM tips is significantly simplified while maintaining high reproducibility with reduced FIB milling time/cost. Additionally, different electrode geometries (frame- and disk-shaped) and a wide variety of ratios between electrode size and re-shaped AFM tip length can be easily maintained. Disk nanoelectrodes can be integrated with radii down to 50 nm after appropriate pre-modification of the AFM tip. Characterization and performance of bifunctional probes are demonstrated by using cyclic voltammetry at the tip-integrated electrodes and simultaneous imaging in contact mode AFM and feedback-mode SECM operation.

        Acinetobacter baumannii Biofilms: Variations Among Strains and Correlations with Other Cell Properties

        Acinetobacter baumannii is an opportunistic pathogen that causes serious infections in humans by colonizing and persisting on surfaces normally found in hospital settings. The capacity of this pathogen to persist in these settings could be due to its ability to form biofilms on inanimate surfaces. This report shows that although the ATCC 19606^T type strain and 8 different clinical isolates form biofilms, there are significant variations in the cell density and microscopic structures of these cell aggregates, with 3 of the isolates forming pellicles floating on the surface of stagnant broth cultures. PCR indicated that, like ATCC 19606^T, all 8clinical isolates harbor all the genetic components of the CsuA/BABCDE chaperone-usher pili assembly system, which is needed for biofilm formation on plastic. Pili detection in cells of all strains examined supports the presence and function of a pilus assembly system. However, only one of them produced the putative ATCC 19606^T CsuA/B pilin subunit protein. Hydrophobicity tests and motility assays also showed significant variations among all tested strains and did not result in direct correlations between the biofilm phenotype and cell properties that could affect biofilm formation on abiotic surfaces. This lack of correlation among these 3 phenotypes may reflect some of the variations already reported with this pathogen, which may pose a challenge in the treatment of the infections this pathogen causes in humans using biofilm formation on abiotic surfaces as a target.

        Structural analysis of amyloid aggregates by multifunctional fluorescence nanoscopy

        An oligomeric structure of amyloidb protein (Abtional nanoscopy, which incorporates uorescence correlation spectroscopy (FCS), atomic force microscopy (AFM) and uorescencelifetime imaging microscopy (FLIM), was used to characterize these oligomeric Ab structures. FCS data showed that various sizes ofAb aggregates exist simultaneously in the solution phase, spanning from nm tol m scale. Stable oligomeric Ab nanoparticles were pre-pared and their topological details of Ab b oligomers stainedwith FRET (uorescence resonance energy transfer) amyloids demonstrated that micelle and donut structures both exist in dierent pHsand temperatures.

        Duhng the last two centuries of the Ancien Regime, rnedicine and the monarchistic state progressively increased their hold over the law and institutions dedicated to insane people. Whereas at the beginning of the XVII^(th) century, families and charity institutions were still the main supports of mad people, on the eve of Revolution, most mentally sick persons were locked up and taken care of on "public powers" initiative. Allied to the "medical power", the absolute monarchy developed a kind of social assistance specifically intended for insane people, while their legal status tended to be standardized. The principles and practices established by the "Esquirol law" of June 30th 1838 already existed at the end of the Ancien Regime.

      • WBC Analysis Through Image Processing

        This study aims to develop a simple, cost effective and efficient neutrophil, lymphocyte and monocyte counter using image processing techniques. To achieve this, a microscope, with a CCD camera mounted on it, is interfaced with the system. The camera provides the live feed from the microscope which is the source of still images that are subjected to the software for assessment. OpenCV library is used in the image processing techniques. The process has two main modules ? cell detection and cell matching. In cell detection, the region of interest (ROI) is extracted and undergoes image matching by means of histogram method. Matching module is subdivided into three ? neutrophil matching, lymphocyte matching and monocyte matching. The result of the matching process is the classification of the detected cell which can be neutrophil, lymphocyte, monocyte or unclassified. Additionally, the total number of neutrophils, lymphocytes and monocytes including unclassified cells found per image and on all images are given.
