      • S-159 Portal hypertensive gastropathy as a prognostic index in patients with liver cirrhosis

        Objective: Portal hypertensive gastropathy (PHG) is a frequently overlooked complication of liver cirrhosis (LC). The clinical implication as a prognostic factor of LC or predictive factor for the development of hepatocellular carcinoma (HCC) has not been established. The aim of this study was to assess the clinical significance of PHG in patients with LC. Materials and Methods: Patients with LC was prospectively enrolled and followed in a single tertiary hospital of Korea. Baseline hepatic vein pressure gradient (HVPG) was measured and esophagogastroduodenoscopy (EGD) was performed. The association of PHG with HVPG, survival and the development of HCC was evaluated. Results: A total of 587 patients were enrolled. The mortality rate was 20.3% (n=119) and HCC was developed in 9.2% (n=54) during the follow-up period (32.6±27.8 months). The grade of PHG was well correlated with HVPG [no PGH: median 9.2 (IQR: 7.2-16.7), mild PHG: 14.6 (10.1-19.3), and severe PHG: 17.3 (12.3-21.5), p<0.001], as well as with Child-Pugh class, MELD score or survival. However, it was not associated with the development of HCC. The grade of PHG (HR 3.29, 95% CI: 1.12-9.63, severe vs. No PHG) and Child-Pugh class (HR 3.53, 95% CI: 1.79-6.97, Child C vs. A) showed significant association with mortality. Conclusions: PHG is well correlated with portal hypertension. This can be used as a prognostic factor for LC, but not for the prediction of HCC.

      • Structural and Dynamics Study of DNA Dodecamer Duplexes That Contain Un-, Hemi-, or Fully Methylated GATC Sites

        <P>Methylation of DNA plays a regulatory role in DNA metabolism. The Escherichia coli DNA adenine methyltransferase methylates the N6 positions of adenines in the sequence 5'-GATC-3', which exists in the fully methylated state during most of the cell cycle. Just after DNA replication, however, the GATC sites transiently become hemimethylated, a condition that is indispensable for various cellular processes, such as negative modulation of replication initiation at oriC by SeqA. The lack of structural and dynamic information on DNA duplexes that contain fully methylated GATC sites makes it difficult to explain how hemimethylated GATC sites are recognized in vivo by proteins in a sea of fully methylated ones. Here, we used NMR spectroscopy to characterize the solution structure of a dodecamer DNA duplex that contained a fully methylated GATC site and the dynamics of the unmethylated, hemimethylated, and fully methylated GATC duplexes. Only the hemimethylated GATC duplex displays a unique major groove conformation, which is optimized for entrance into the cleft structure of SeqA. The apparent equilibrium constants for base-pair opening of the three differentially methylated GATC duplexes revealed that N6-methylation of the adenine residue affects the thermodynamics and kinetics of its own and neighboring base pairs. The equilibrium constants for base-pair opening of three GATC duplexes were determined using proton exchange catalyzed by TRIS. The two G.C base pairs of the hemimethylated GATC duplex displayed a faster base-pair opening rate and required less energy for the base-pair opening reaction than did those of the fully methylated one.</P>

        Foucauldian Discourse Analysis as an Epistemological position in Sport Policy Studies

        This article explores the usefulness of Foucauldian discourse analysis and the applications as an epistemological position for sport policy studies which have recently secured attentions in not only areas of sport studies but also public policy academia. For the purpose of the exploration, we discuss some Foucault’s theories applied in sport policy studies, recapitulating ontological and epistemological points. We argue that Foucauldian discourse analysis as an epistemological standpoint could enrich the terrain of sport policy studies in relation to: 1)sensitization of complex and structural dynamics of knowledge and power in sport policy processes, the related decision-making, and the implementation, which informed of Fouauldian research traditions; 2)acknowledgement of sport policy studies positions’ inherent beneficiaries, especially in qualitative and discursive researches, by which researchers could position themselves as a part of the social phenomena in question; 3)reconceptualization of sport policies as social phnomena and the related critical researches as a production of discourse and, simultaneously, the process of constructing discursive reality seeking the constant changes and the foundation for pursuit of truth, and the plurality in social science and sport policy study.

        Survey of Awareness and Concept of Insects in Korea

        To investigate the degree of individuals’ concept and awareness of insects, a survey study was conducted with students and adults living in Korea. The misconception rate for insects was about 50% for both students and adults, but it was lower for students and people who had experienced insect-related events than for adults and those who had not. The highest misconception rate was obtained in answer to a question about the basic structure of an insect. Most people had a high awareness of insects. Significant differences and correlations for the awareness of insects were found between students and adults, men and women, people who had experienced insect-related events and those who had not. The experience of an insect-related event most influenced awareness of insects. These results suggest that increasing people``s interest in insects and utilizing insects in treatment situations may be beneficial for the field of mental healthcare.

      • Impact resistant properties of Kagome truss reinforced composite panels

        This paper presents an experimental study exploring impact resistant properties of Kagome truss reinforced composite panels. Three types of panels with different materials and reinforcements, i.e., ultra-high-performance mortar, steel fiber, and Kagome truss, were designed and manufactured. High-velocity projectile impact tests were performed to investigate the impact response of panels with dimensions of 200 mm×200 mm×40 mm. The projectile used in the testing was a steel slug with a hemispherical front; the impact energy was 1 557 J. Test results showed that the Kagome truss reinforcement was effective at improving the impact resistance of panels in terms of failure patterns, damaged area, and mass loss. Synergy effects of a combination of Kagome truss and fiber reinforcements for the improvement of impact resistance capacity of ultra-high-performance mortar were also observed.

        Understanding Epistatic Interactions between Genes Targeted by Non-coding Regulatory Elements in Complex Diseases

        Genome-wide association studies have proven the highly polygenic architecture of complex diseases or traits; therefore, single-locus-based methods are usually unable to detect all involved loci, especially when individual loci exert small effects. Moreover, the majority of associated single-nucleotide polymorphisms resides in non-coding regions, making it difficult to understand their phenotypic contribution. In this work, we studied epistatic interactions associated with three common diseases using Korea Association Resource (KARE) data: type 2 diabetes mellitus (DM), hypertension (HT), and coronary artery disease (CAD). We showed that epistatic single-nucleotide polymorphisms (SNPs) were enriched in enhancers, as well as in DNase I footprints (the Encyclopedia of DNA Elements [ENCODE] Project Consortium 2012), which suggested that the disruption of the regulatory regions where transcription factors bind may be involved in the disease mechanism. Accordingly, to identify the genes affected by the SNPs, we employed whole-genome multiple-cell-type enhancer data which discovered using DNase I profiles and Cap Analysis Gene Expression (CAGE). Assigned genes were significantly enriched in known disease associated gene sets, which were explored based on the literature, suggesting that this approach is useful for detecting relevant affected genes. In our knowledge-based epistatic network, the three diseases share many associated genes and are also closely related with each other through many epistatic interactions. These findings elucidate the genetic basis of the close relationship between DM, HT, and CAD.

        Genetic Diversity and Population Structure of Kalopanax pictus (Araliaceae)

        Kalopanax pictus (Araliaceae) is a long-lived woody species primarily distributed throughout East Asia. This species is regarded as medically and ecologically important in Korea. We used starch gel electrophoresis to investigate its allozyme variation and genetic structure in samples from Korean populations (both natural and cultivated). Our analysis of 10 enzymes revealed 18 loci, of which 10 were polymorphic (55.6%). Genetic diversity at the species and population levels were 0.200 and 0.149, respectively, with the mean for cultivated populations (0.124) being lower than for natural (wild) populations (0.181). Asexual and sexual reproduction modes, perennial habitat, and longevity all were possible factors contributing to such high diversity. An indirect estimate of the number of migrants per generation (N_(m)=1.08) indicated that gene flow was not extensive among these Korean populations. Therefore, we suggest that geographical distance as well as reproductive isolation between wild and cultivated plants may play roles in shaping the population structure of this species.

        Characteristics of the Abdominal and Neck Flexor Muscles of Children with Cerebral Palsy

        Purpose: The purpose of this study was to compare the activities of the abdominal and neck flexor muscles of children with and without cerebral palsy (CP) while lifting the head in a supine position. Methods: The subjects were eight children with CP and eight children without the disease. The activities of the external abdominal oblique (EO), internal abdominal oblique (IO), rectus abdominis (RA), sternocleidomastoid (SCM), and RA/SCM muscles were collected by surface electromyography (EMG) when the children lifted their heads. A Mann-Whitney U test was used to compare the activity of each muscle during the head-lifting exercise. Statistical significance was accepted at p<0.05. Results: The activities of the EO, IO, and RA, and RA/SCM muscles differed significantly between the children with and without CP, but there was no significant between-group difference in the activity of the SCM muscle. Conclusion: These findings suggest that the abdominal muscles are not employed as much in the activities of children with CP compared to those without the disease. Additionally, those with CP were more dependent on the neck flexor muscle during the head-lifting exercise in a supine position.
