<P><B>Abstract</B></P><P><B>Aim: </B> We investigated the accuracy of liver stiffness measurement (LSM) in chronic hepatitis C (CHC) in a multicenter, prospective study in South Korea.</P><P><B>Methods: </B> Between June 2005 and July 2009, 91 CHC patients without a previous history of antiviral treatment, clinical evidences of cirrhosis, coinfection with other viruses, and heavy alcohol consumption and with alanine aminotransferase (ALT) ≤ 5x upper limit of normal, total bilirubin ≤ 1.5 mg/dL, sufficient liver biopsy quality (≥ 15 mm and more than six portal tracts), interquartile range to median liver stiffness (LS) value ratio ≤ 0.21, and more than 10 valid measurements, were recruited. The Batts and Ludwig scoring system was used for histologic assessment. Age–platelet index (API), aspartate aminotransferase (AST)–to–platelet ratio index (APRI), and age–spleen–platelet ratio index (ASPRI) were calculated. Area under the receiver operating characteristic curve (AUROC) was used to evaluate the performance of LSM and other noninvasive models.</P><P><B>Results: </B> The mean age was 47.9 years, and the mean LS value was 7.7 kPa (44 men and 47 women). LS value was highly correlated to the fibrosis stages (<I>r</I> = 0.835, <I>P</I> < 0.001). The AUROCs of LSM were 0.909 for ≥ F2, 0.993 for ≥ F3, and 0.970 for F = 4 and were superior to those of API (0.72, 0.858, and 0.948, respectively), APRI (0.780, 0.887, and 0.904, respectively), and ASPRI (0.713, 0.862, and 0.957, respectively). The optimal cutoff LS values were 6.2 kPa for ≥ F2, 7.7 kPa for ≥ F3, and 11.0 kPa for F = 4.</P><P><B>Conclusions: </B> Our data suggest that LSM can accurately assess liver fibrosis in patients with CHC and be applied in South Korea.</P>

We compared the nutritional requirements of whole larvae of the black seabream Acanthopagrus schlegeli, sweet fish Plecoglossus altivelis, olive flounder Paralichthys olivaeus and rock fish Sebastes schlegeli. The larvae were 20, 30, 14 and 5 DAH (or spawning) of black seabream, sweet fish, olive flounder and rock fish, respectively. Specific α-amylase activity (mU/mg protein) was highest (8,324.9 mU/mg protein) in rock fish larvae (P<0.05). Specific trypsin-like protease activity was highest (11,330.1 mU/mg protein) in black seabream larvae (P<0.05), which also exhibited the highest activity, 685.5 mU/mg dry weight (P<0.05). The specific activities per mg protein and mg dry weight of black seabream were the highest (187.4 mU/mg protein and 11.3 mU/mg dry weight, respectively) (P<0.05). A/P, P/L and A/L ratios of rock fish were 1.47, 90.3 and 133.1, respectively (P<0.05). We present here basic larval digestive enzymatic nutritional requirement data.

In this study, we developed a novel tool for purifying two mycotoxins, aflatoxin B1 (AFB1) and zearalenone (ZEN), in feed. This system utilized monoclonal antibodies (mAbs) against AFB1 and ZEN, and magnetic nanoparticles (MNPs). Among ten MNPs with different diameters and functional groups, a 100-nm diameter MNP (fMA) conjugated to an amine group (-NH2) was found to be optimum for coupling with mAbs. The optimal mAb concentrations for coupling to the fMA along with mycotoxin purification capacities of the fMA-mAb conjugates (fMA-AFB1 and fMA-ZEN) were determined. A comparison of mean recovery rates (from corn and product X feed) between the fMA-mAb conjugates and immunoaffinity columns (IAC-AFB1 and IAC-ZEN) showed that the rate for fMA-AFB1 (90∼92% and 81∼88%) was higher (p > 0.05) than that of IAC-AFB1 (81∼84% and 72∼78%) for AFB1 (5, 10, 15 ng/mL), and the rate for fMA-ZEN (99∼100% and 92∼94%) was significantly higher (p < 0.01) than that of IAC-ZEN (86∼88% and 81∼88%) for ZEN (10,25, 50 ng/mL) except at a concentration of 10 ng/mL, demonstrating the remarkable purification efficiency of the novel fMA-mAb method. Additionally, mycotoxin purification was much faster using our novel method (approx. 5 min) than the IAC-based technique (> 30 min). This study suggests that the novel purification system we developed would be a useful tool for monitoring and regulating mycotoxin contaminatio in feed, and replace IAC methods.
Association of IL-17RB Gene Polymorphism With Asthma

<P>BACKGROUND: Interleukin (IL)-17E is a member of the IL-17 family, which induces IL-4, IL-5, IL-13, and eotaxin in experimental animals via IL-17 receptor B (IL-17RB). The activation of IL-17RB amplifies allergic-type inflammatory responses by inducing Jun kinase (or JNK), p38 mitogen-activated protein kinase (or MAPK), and nuclear factor-kappaB. OBJECTIVES: We examined the association of polymorphisms in the IL-17RB gene with asthma susceptibility and investigated the effects of those polymorphisms on the transcription of various IL-17RB isoforms. METHODS: In total, 954 asthmatic patients or 265 healthy control subjects were screened for polymorphisms in IL-17RB by single-base extension. The messenger RNA expression IL-17RB in B-cell lines derived from patients was also measured by reverse transcription-polymerase chain reaction. RESULTS: Direct sequencing of 24 unrelated Korean DNA samples revealed 18 genetic variants, including four insertion/deletions and 14 single-nucleotide polymorphisms (SNPs). Six of the SNPs (-1465G>A, +5661G>A, +6297T>C [Y123Y], +13797C>T, +18661C>T, and +18965G>A) were used to screen a larger group of subjects. Intronic polymorphism +5661G>A was significantly associated with the development of asthma (p = 0.001); moreover, a minor allele of IL-17RB +5661G>A appeared at a lower frequency in the asthmatic patients than in the healthy control subjects (0.13 vs 0.19, respectively). The IL-17RB messenger RNA expression in B cells homozygous for IL-17RB+ 5661GG was significantly higher than that in B cells homozygous for IL-17RB+5661AA (p = 0.002). CONCLUSIONS: A rare allele of IL-17RB +5661G>A may have a protective role against the development of asthma via regulation at the level of transcription. The SNPs identified in this study may be used to develop markers to assess the risk of asthma.</P>

N/A Background: Diabetes mellitus is one of the major health problems in Korea. It has been suggested that there are quite many atypical IDDM patients who initially manifest as NIDDM, but require insulin eventually. However, little is known about the clinical characteristics of this type of diabetics in Korea. This study was undertaken to find out how many current IDDM patients bad a history of noninsulin requiring phase. Methods: We obtained medical histories retrospectively from 2300 patients who visited Asan Medical Center from June 1989 to July 1992. Fifty nine patients were classified as IDDM because their fasting plasma C-peptide levels were below 0. 13fmol/L (0.4ng/ml). The patients were classified further into the two groups: group A who required isulin within 1year after diagnosis or presented as debut DKA, and group B who had nonisulin requiring phase at least for 1year after the diagnosis. Results: Twenty six patients (44%) were classified as group A, and 27patients (46%) as group B. Median age of onset was 26years in group A, (range; 10-50) and 45 (range; 23-73) in group B. As a whole, 24patients (41percent) were diagnosed as diabetes over the age of 40. Two groups had similiar values in body mass index. History of DKA was found in 58percent in group A and 7percent in group B. All patients of group A were using insulin at the time of the study, while only 56percent of group B were using insulin. Conclusion: Our study showed: first, forty percent of IDDM patients developed their diabetes at the age over 40: second, about half of the patients had a non-insulin- requiring phase at least for one year, The lack of DKA, old age of onset, and the initial non-insulin-requiring phase suggest that these patients are different from the typical IDDM. This subclass of IDDM in Korea may be classified as slowly progressive IDDM.
